Abstract

The purpose of this study was to analyze the inhibitory action of ursolic acid (UA) as an antitubercular agent by computational docking studies and molecular dynamics simulations. The effect of UA on the cell wall of Mycobacterium tuberculosis (MTB) was evaluated by using Scanning Electron Microscopy (SEM). UA was used as a ligand for molecular interaction and investigate its binding activities to a group of proteins involved in the growth of MTB and the biosynthesis of the cell wall. Computational docking analysis was performed by using autodock 4.2.6 based on scoring functions. UA binding was confirmed by 30 ns molecular dynamics simulation using gromacs 5.1.1. H37Rv sensitive strain and isoniazid-resistant strain were used in the SEM study. UA showed to have the optimum binding affinity to inhA (Two-trans-enoyl-ACP reductase enzyme involved in elongation of fatty acid) with the binding energy of -9.2 kcal/mol. The dynamic simulation showed that the UA-inhA complex relatively stable and found to establish hydrogen bond with Thr196 and Ile194. SEM analysis confirms that UA treatment in both sensitive strain and resistant strain affected the morphology cell wall of MTB. This result indicated that UA could be one of the potential ligands for the development of new antituberculosis drugs.

Abstract

Eleutherine americana (E. americana) is a medicinal plant commonly found on the island of Borneo, Indonesia. This plant is known to have several biological activities. However, anthraquinone residues are generally present as contaminants. This study was aimed to develop a method of determining the levels of 9,10-anthraquinone in plant extracts and fractions using High Performance Liquid Chromatography (HPLC). The research aims to optimize the mobile phase, the system suitability test, and the system validation. The optimal mobile phase was acetonitrile:distilled water 1:1 v/v with a flow rate of 1.25 mL/min. The validation result shows that the linearity was obtained with a correlation coefficient (r) of 0.9995 and an r2 coefficient of 0.9991. The estimated limits for detection and quantification values were 0.178 and 0.594 µg/mL, respectively. In the intraday and inter-day accuracy test, the coefficient of variance for reference was 0.627 and 0.774, while the results for the sample were 2.966 and 2.658. The percentage recovery rate for reference was between 98.976–101.452%, and for the sample, the result was 89.191–94.667%. The average 9,10-anthraquinone content in the acetate fraction of E. americana plant was 9.799 µg/g ± 5.243.

Abstract

A number of teenagers experience various levels of mental disorders, ranging from mild stress to Generalized Anxiety Disorder (GAD). According to the World Health Organization (WHO), approximately 264 million people suffer from anxiety disorders, including GAD, and this condition is a significant leading cause of suicide incidents, ranking sixth among serious mental health issues. From a societal perspective, Cognitive Behavioral Therapy (CBT) has the advantage of cost-effectiveness by minimizing the use of medications and enhancing productivity in education for adolescents receiving CBT. This study aims to conduct a systematic review of the existing literature on GAD in adolescents, using the PICO format method and formulating relevant research questions (RQ). Based on research that has been done, CBT has a relevant effect on some cases of GAD experienced by adolescents, although in some conditions CBT needs to be combined with antidepressants.

Abstract

Abstract

Tuberculosis (TB) continues to pose a global health challenge, mainly due to low case detection rates in high-burden countries like Indonesia. Community pharmacies (CP), the health facilities most commonly visited by individuals experiencing tuberculosis symptoms, play an essential role in early detection and referral to health facilities. This narrative review examines the role of community pharmacists in TB case detection through pharmacy-based interventions in developing countries with high TB burden. The following databases were used to identify articles that met the inclusion and exclusion criteria: PubMed and Scopus. Based on three eligible studies from India, Pakistan, and Cambodia, pharmacy-based TB case detection programs were associated with increased case detection and referral of presumptive TB cases. The included studies reported that structured interventions, including pharmacist training, referral systems, monitoring, and public-private collaboration, may support TB case detection in high-burden settings. However, several barriers were identified, including uncertainty about referral pathways, financial constraints, and concerns about service quality at referral facilities. Given the limited evidence from only three studies, these findings should be interpreted cautiously, and further research is needed to assess the effectiveness and sustainability of pharmacy-based TB case-detection programs. CP engagement may contribute to TB case detection when reinforced by well-defined, structured referral pathways and integration with public-private TB control initiatives.
