Abstract

Objective: To evaluate the relationship between the number of LH receptor and the success of oocyte maturity in the process of in vitro maturation (IVM). Method: This experimental study was conducted in the Permata Hati Infertility Clinical Laboratory, Dr. Sardjito General Hospital, Yogyakarta, with the samples of 300 oocytes obtained through collecting immature cow’s oocytes from the abattoir and grouped the oocytes into 3 (three) groups based on the pattern of oocyte cumulus cells on the vesicle germinal stage 2 - 8 mm with three layers of cumulus cell. The sample of the cumulus cells from these three groups were taken and the LH receptor examination was done with immunohistochemistry. After that, the IVM process was performed to the three groups and its development for 24 hours was evaluated. Its maturation quality was evaluated with the emergence of the first polar body (1PB) and compared to the other groups and related to the number of LH receptor in the three groups. Result: The result of this study indicated that the oocyte cumulus cells showed a difference of function during IVM process. The maturity rate in this study showed that the number of LH receptor was related to the morphological pattern of oocyte cumulus cells with oocyte maturity. The maturity of the cumulus cells which 100% covered the oocyte was higher than that of the cumulus cells which > 50% and < 30% covered the oocytes, namely, 74% compared to 60% and 12%. The result of this study also showed that the average number of LH receptors in the three groups (A, B, and C) was 183.4, 78.8, and 24.0 respectively. A significant difference was found in the three groups (p < 0.0001). When related to IVM maturity, this difference showed that the bigger number of oocyte cumulus cells influenced the oocyte maturity. Conclusion: The number of LH receptor can be used as a prediction to determine the success of oocyte maturation in the process of in vitro maturation. Abstract

AbstrakBiji ketumbar (Coriandrum Sativum L) adalah tanaman yang diperkenalkan dari daerah Mediterranean, dan Timur Tengah, di Negara Indonesia biji ketumbar pada umumnya di gunakan sebagai bumbu masakan. Biji ketumbar juga banyak dimanfaatkan untuk penyakit diabetes, anti-inflamasi, antioxidant, anti fungal, antibiotik baik bakteri maupun jamur. Pada penelitian ini ekstraksi biji ketumbar dilakukan dengan cara maserasi menggunakan pelarut etanol 70%. Ada beberapa tingkatan konsentrasi yang digunakan yaitu 50%, 75%, 100%, dengan control positif gentamicin 10 mcg dan control negatif Aquadest steril. Teknik pengujian dilakukan sebanyak 5 kali pengulangan dengan metode difusi cakram. Pada hasil skrining fitokimia ekstrak biji ketumbar didapatkan adanya senyawa flavonoid, saponin, tannin, alkaloid, fenolik, glikosida, dan triterpenoid. Dari hasil pengujian ekstrak biji ketumbar dengan konsentrasi 100% didapatkan hasil bahwa ekstrak biji ketumbar sangat berpotensial menghambat pertumbuhan bakteri Staphylococcus aureus dengan hasil rata-rata diameter 8,20 mm dengan daya hambat tergolong besar

Abstract

Amount of 207 million (83.6%) women who give birth simultaneously are diagnosed with GDM in 2019. Prevalence of GDM reaches 1.9-3.6% in Indonesia. Amount of 40-60% of GDM will suffer from postpartum Non-Dependent Insulin Diabetes Mellitus. Hence, this study was aimed to look for the distribution of blood sugar levels among pregnant women who are overweight. This research was performed among pregnant women in the Obstetrics and Gynecology polyclinic of Royal Prima Medan Hospital against 41 people as the sample with a cross-sectional model (Longitudinal study). The aspects evaluated in this study were age, gestational age, BMI, occupation, income, recent education, history of gestational DM, Macrosomia, and obstetrics. All research data was analyzed with SPSS 25 by spearman correlation. The trend of random et blood sugar levels of pregnant women as respondents was 100 mg/dL, while the BMI tendency was 31.72 kg/m2. Based on these parameters, they had a correlation with each other among overweight pregnant women (Value P = 0.007) that was moderate (R = 0.417). Blood sugar levels were moderate correlated with BMI among pregnant women with excess body weight.

Abstract

Biji ketumbar (Coriandrum Sativum L) memupakan tanaman yang berasal dari Mediterranean, dan Timur Tengah, masyarakat Indonesia umumnya di manfaat kan sebagai bumbu masak. Biji ketumbar sering juga digunakan untuk penyakit diabetes, diuretic, hypolipidemia, anti- fungal, antibiotic baik jamur maupun bakteri, antioxidan, dan anti-inflamasi. Ekstraksi biji ketumbar dilakukan dengan cara maserasi menggunakan pelarut etanol 70%. Konsentrasi yag digunakan 50%, 75%, dan 100%, dengan control positif gentamisin 10mcg (OXOID) serta control negative (aquadest steril). Pengujian dilakukan dua kali pengulangan dengan metode difusi cakram. Skrining fitokimia ekstrak biji ketumbar menunjukan adanya senyawa alkaloid, saponin, tannin, flavonoid, fenolik, Triterpenoid, dan glikosida. Hasil pengujian menunjukan bahwa ekstrak biji ketumbar mampu menghambat petumbuhan bakteri pseudomonas aeruginosa. Ekstrak biji ketumbar yang mampu menghambat pertumbuhan bakteri pseudomonas aeruginosa adalah konsentrasi 100% dan rata-rata diameter 8,20 mm dengan daya hambat tergolong besar.

Abstract

Skin aging caused by oxidative stress that leads to the formation of wrinkles, pigmentation, and asteatosis. On the other hand, the elastase enzyme also can influence the elasticity of the skin that leads to sagging. Moringa leaves (Moringa oleifera Lamk) has been reported contain some active compounds like flavonoid that has antioxidant and anti-aging potential. This study was presented to determine the potential of moringa leaves extract (MLE) in scavenging DPPH radicals and inhibiting the elastase enzyme. MLE was obtained by using maceration methods. The potential of MLE in scavenging DPPH radicals and inhibiting elastase enzyme was determined using spectrophotometric methods. MLE has IC50 value of DPPH scavenging activity also elastase inhibitory activity at 109.67 + 0.83 µg/mL and 159.67 + 7.95 µg/mL. MLE has antioxidant and anti-aging potential as indicated by DPPH scavenging activity and elastase inhibitory. However, MLE has moderate DPPH scavenging activity and low elastase inhibitory activity.

Abstract

Asparagus is a vegetable that contains phenolic compounds with antioxidant properties that scavenges aging-triggering free radicals. This study aimed to investigate the components and anti-aging potentials of Ethanol Extract form Asparagus (EEA). The study was performed in February 2020 at the Pharmacy Laboratory, University of North Sumatera. The EEA was obtained through maceration using 96% ethanol. An antioxidant assay was performed and the total phenol and flavodoid content were determined using the spectroscopic method. Three gel formulas with different concentrations of EEA was prepared (F1: 1.5%, F2: 2.5%, and F3: 3.5%), and F0 was used as control. The parameters evaluated were moisture, oil content, texture, collagen, wrinkle, pigment, sensitivity, and pore. The result showed that asparagus had a moderate antioxidant activity (IC50: 118,992) with the total phenol and flavonoid contents of 15,9407 mg GAE/g and 3,2286 mg QE/g extract, respectively. The highest aging activities was seen in F3 (3.5%), followed by F2 (2.5%) and F1 (1.5%). The percentage of moisture, oil, texture, collagen, wrinkle, spot, sensitivity, and pore recovery were found to be 40.15%, 49.73%, 71.76%, 17.70%, 70.93%, 49.34%, 42.56% and 25.31%, respectively. Hence, it can be concluded that the EEA Gel at the highest concentration (3.5%) has a high content of phenol and flavonoid which can improve the skin moisture, oil content, texture, collagen, wrinkles, spots, sensitivity, and pores, which promotes anti-aging activities.

Abstract

The change in the pattern and timing of menopause is an exciting and exploratory phenomenon that requires more detailed research to be carried out using hormonal contraception. This study was aimed to explore the relationship between hormonal contraceptive use and menopausal age. This study used a cross-sectional design. The study was conducted at Royal Prima General Hospital Medan from May to August 2021. A total of 68 research subjects participated in this study with the following inclusion criteria: menopausal women, aged 50-60 years, using hormonal contraceptives (pills, injections, implants), able to remember history previous contraceptive use. This study uses primary data obtained directly from guided interviews with respondents. Participants were dominated by ≥ 55 years old women with 57 people (83.81%), 62 Muslims (91.2%), menarche age >13 years as many as 28 people (41.2%). The number of children 2-4 were 32 people (47.1%) with an average menopause age of 51.93 and the smallest for 1 was 8 people (11.8%) with an average menopause age of 50.37. There were 49 users of hormonal contraception (72.1%) and 48 people (70.6%). In conclusion, we found that the use of hormonal contraceptives is significantly related to menopausal age.
