Abstract

The widespread use of jamu, a traditional Indonesian herbal medicine, has been accompanied by a surge in adulteration with synthetic drugs to enhance efficacy and sales. This study aimed to detect the presence of glibenclamide, a common adulterant, in antidiabetic jamu product using TLC. This study employed three herbal medicine samples marketed for lowering blood glucose levels, with glibenclamide BPFI serving as a reference standard. Thin-layer chromatography, employing a mobile phase consisting of chloroform, cyclohexane, ethanol, and glacial acetic acid (9:9:1:1), was utilized for the analysis. Thin-layer chromatography (TLC) analysis revealed a standard Rf value of 0.4 for glibenclamide, producing a distinct blue-violet spot when visualized with 10% sulfuric acid. The results indicated that the tested herbal medicine samples did not contain the pharmaceutical substance glibenclamide. This conclusion was drawn from the absence of a corresponding Rf value for glibenclamide in the herbal samples when compared to the reference standard.

Abstract

The rhizosphere is the zone between the root surface and soil. Rhizosphere bacteria can produce metabolites that are antibacterial because they produce phytotherapeutic compounds that have the same properties as those produced by the plant. The purpose of this study was to isolate secondary metabolite compounds from the rhizosphere bacteria of cats’ whisker plants (Orthosiphon stamineus) as antibacterial agents against Escherichia coli and Staphylococcus aureus. This study began with the isolation of rhizosphere bacteria, purification, bacterial characterization, bacterial fermentation, and antibacterial tests. The results of the isolation of Orthosiphon stamineus rhizosphere bacteria obtained as many as six isolates, with each isolate coded as RKK-1, RKK-2, RKK-3, RKK-4, RKK-5, and RKK-6. The results showed that RKK-5 had an inhibition zone against Escherichia coli with a diameter of 8.5 mm. Based on these results, it can be concluded that RKK-5 has a moderate response to Escherichia coli.

Abstract

This study investigates the potential of white turmeric (Curcuma zedoaria Rosc.) rhizome extract as a cost-effective treatment for managing diabetes, focusing on its impact on pancreatic histopathology and blood glucose levels in diabetic Wistar rats. The research employed a randomized pre-test and post-test control group design, with rats induced with alloxan to simulate diabetes. The treatment groups received varying doses of white turmeric extract (250 mg/kg BW, 500 mg/kg BW, and 750 mg/kg BW) for 14 days. Histopathological analysis of the pancreas revealed that the white turmeric extract groups showed no signs of edema, inflammation, or necrosis, indicating a protective effect. Blood glucose level measurements demonstrated that the 500 mg/kg BW dose exhibited the most optimal results in lowering blood glucose levels. Statistical analysis using ANOVA and Kruskal-Wallis tests indicated significant differences between groups. These findings suggest that white turmeric extract, particularly at a dosage of 500 mg/kg BW, holds promise as a complementary treatment for managing diabetes by improving pancreatic health and reducing blood glucose levels. Further research is warranted to explore the underlying mechanisms and optimize its application.

Abstract

Ovarian cancer is a serious disease that affects the ovaries, and its early detection is challenging due to vague symptoms often dismissed as minor ailments. Currently, natural sources have gained attention for their potential role in anticancer treatment. This study aimed to utilize network pharmacology to explore the potential targets and mechanisms of Zanthoxylum acanthopodium in the treatment of ovarian cancer. This study utilized the KNApSAcK and Swiss Target Prediction to identify active compounds and target genes. Additionally, ovarian cancer-specific target genes were sourced from the GEO database. To identify possible key target genes, the network interaction between protein-protein using the STRING database and visualized them in Cytoscape. Subsequent analysis using Gene Ontology (GO) and the Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways enabled us to focus on primary therapeutic targets. Our investigation into Zanthoxylum acanthopodium revealed 10 active compounds that pass Lipinski rule of five and oral bioavailability with acceptable pharmacokinetic profiles, 88 therapeutic targets, and identified 5 hub genes: SRC, CCNB2, MMP9, PTGS2, and PTPRC, which are strongly associated with ovarian cancer progression. Pathway enrichment analysis highlighted several pathways significantly related to the pathogenesis of ovarian cancer. This study elucidates the therapeutic potential and mechanisms of action of Z. acanthopodium as a promising candidate for ovarian cancer treatment. However, further research, including both in vitro and in vivo studies, is necessary to understand its molecular mechanisms comprehensively.
