Abstract

A study was conducted to formulate semen diluent of pigswith a better quality, cheap and easy toprepare using egg yolk of various poultries such as chickens, ducks and quails in combinationwithastaxanthin, a potent antioxidant. The research design used was a completely randomized factorialdesign with three different types of sperm diluents and four levels of astaxanthin concentration. Spermdiluents used were phosphate duck egg yolk, phosphate quail egg yolk and yolks phosphate supplementedrespectivelywith 0,002%, 0,004% and 0,008%astaxanthine. The treated sperm were strored at 5oC for 48hours. The sperm qualities were examined for progressive motility, spermatozoa abnormalities, viabilityand plasma membrane integrity. The result showed that sperm diluents of using duck egg yolk phosphatein combination with 0.002% astaxanthinresulted in the highest progressive motility, viability and plasmamembrane intact while abnormalities spermatozoa is lowest. It can be concluded that phosphate duck eggyolk sperm diluents with the addition of 0,002% astaxanthinappeared to be able to maintain the qualityof pig sperm stored at 5oC for 48 hours.

Abstract

This research was conducted to produce a formula of diluent for the manufacture of frozen semen of Gembrong goat. Yolk phosphate was used as a basic diluent with the addition of anti-cold shock, antioxidants and combination of anti-cold shock-antioxidant. The research design used a completely randomized design with three kinds of treatments; T1: the addition of lactose 0.6% (anti-cold shock), T2: the addition of astaxanthin 0.004% (antioxidant), and T3: a combination of 0.6% lactose-Astaxanthin 0.004% (combination of anti-cold shock and antioxidants). The addition of DMSO 6% was used in each treatment as intracellular cryo protectants. The freezing process was done with the gradual freezing with conventional techniques. Examination of the quality of semen by thawing prior included progressive motility, viability, and abnormalities. The results showed that anticoldshock-antioxidant combination (0.6% lactose -Astaxanthin 0.004%) produced the best semen quality with progressive motility, viability, and less abnormalities as follow 44.00 + 3.46%, 59.00 + 1.85%, and 14.00 + 0.76% respectively. It was concluded that the addition of a combination of 0.6% lactose-Astaxanthin 0.004% on the phosphate yolk diluent with 6% DMSO as intracellular produced best quality of frozen goat Gembrong semen that suitable for use in artificial insemination and in vitro fertilization.

Abstract

A study was carried out to determine the effect of Pregnant Mares Serum Gonadotrophin (PMSG) on the weigth of ovary, the number of developing follicles and the onset egg production in ducks with delayed egg production. In this study, as many as 40 Bali ducks were used and they were divided into 4 groups in a completelty randomized design i.e. control group (TO) treated with NaCl 0,9%, group T1 treated with PMSG dose 5 IU per duck, group T2 treated with PMSG dose 10 IU per ducks and group T3 treated with PMSG dose 20 IU per duck intramuscularly. The weight of ovary increased significantly with the increasing dose of PMSG, i.e dose 0 IU (21,04 + 3,90 gram), dose 5 IU (25,40 + 5,31 gram), dose 10 IU (49,52 + 5,05 gram) and dose 20 IU (59,30 + 4,66 gram). The number of developing folilicles also increased significantly with the increasing dose of PMSG, i.e dose 0 IU (1,4 + 0,5 follicles), dose 5 IU (2,4 + 1,34 follicles), dose 10 IU (5,8+2,07 follicles) and dose 20 IU (8,6+1,67 follicles). In addition, the onset of egg production was shortened significantly with the increasing dose of PMSG, i.e. dose 0 IU (19,0+1,58 days), dose 5 IU (16,8+1,3 days), dose 10 IU (14,2+1,92 days) and dose 20 IU (9,6+14 days). PMSG treatment was proven to increase the egg production capability of ducks with delayed egg production.

Abstract

The aim of this study to examine the addition of cryoprotectant glycerol and dimethylsulfoxide (DMSO) in astaxanthin phosphate duck egg yolk on the quality of frozen boar semen. Each of the cryoprotectants distinguished its concentration ie : 4%, 6%, 8%, 10%, and 12% for freezing of semen. Semen was collected from one Landrace boar then performed macroscopically and microscopically. Semen was divided into 10 tubes each diluted with the duck egg yolk astaxanthin phosphate extender diluent then stored at 20oC for 2 hours, centrifugation for 15 minutes (2000 rpm) then taken the pellet with 1 ml of supernatant. The pellets were diluted with astaxanthin phosphate duck egg yolk extender containing various concentration (4, 6, 8, 10, and 12%). Semen was packed with 0.5 mL straw and performed equilibration for 2 hours (4oC). do aeration above liquid N2 vapor for 10 minutes, then dipped in liquid N2. Semen was stored in liquid nitrogen container for 24 hours , and then quality of spermatozoa was evaluated post thawing. The results of the study showed that addition of cryoprotectan glycerol and DMSO were no difference significant (p>0,05) on the quality of frozen semen, but cryoprotectant concentrations (4%, 6%, 8%, 10%, and 12%) significantly affected (p <0.05) the quality of frozen semen. Glycerol with a concentration of 4% produces the best quality frozen semen, whereas DMSO with a concentration of 6% produces the best quality frozen semen.

Abstract

This study aims to maintain the quality of pig semen for longer during storage at 15oC, in an effort to support artificial insemination programs with the addition of Bovine Serum albumin (BSA) to diluent Beltsville Thawing Solution (BTS). This study uses a completely randomized design with five treatment groups, each To = semen was diluted with BTS without the addition of BSA ; T1 = with the addition of 5 mg BSA/mL diluent; T2 = with the addition of 10 mg BSA/mL diluent; T3 = with the addition of 15 mg BSA/mL diluent; T4 = with the addition of 20 mg BSA/mL diluent. Each treatment was repeated five times so that the number of samples used was twenty-five. The diluted cement is stored at 15oC for 72 hours then observing the quality of cement includes: progressive motility, dead spermatozoa, abnormalities, and intact plasma membranes. The data obtained were analyzed by analysis of variance, if there were differences followed by Duncan’s test. The results showed, addition of BSA concentration of 10 mg/mL and 15 mg/mL of diluent gives the same effect on the quality of cement during storage and significantly better (p <0.05) when compared to the addition of 0 mg/mL, 5 mg/mL and 20 mg/mL diluents. It can be concluded, the addition of BSA 10 mg/mL BTS diluents can maintain the most optimal quality of pig semen against progressive motility, dead spermatozoa, abnormalities and intact plasma membranes.

Abstract

The objective of this study was to determine the addition of lactose on egg yolk phosphate diluentsstored at 5°C to the plasma membrane intact, acrosome membranes intact of green jungle fowlspermatozoa, fertility and hatchability of chicken eggs. This study used completely randomized designwith three treatment groups. Treatment I (T0): egg yolk phosphate diluents only (control), treatment II(T1): egg yolk phosphate diluents + lactose 0.3%, and treatment III (T2): egg yolk phosphate diluents +lactose 0.6 %. Each group was replicated nine times. Semen was collected from eight green jungle fowl andevaluated both macroscopically and microscopically. Semen was diluted with each treatment with a singleinsemination dose (0.5 mL) with a concentration of 150 million progressively motile cells. Once diluted,semen was stored at 3-5°C for 48 hours and then was observed for the plasma membrane of intact,acrosome membrane of intact, and some semen were inseminated on local female chicken village forfertility, and hatchability eggs observation. The results showed that the addition of lactose 0.3% and 0.6%providing significant increase (P <0.05) on plasma membrane intact, acrosome membrane intact, fertilityand egg hatchability when compared to controls. However the addition of 0.3% and 0.6% lactose did notshowed a significant effect (P> 0.05). This study concluded that the addition of lactose on egg yolk phosphatediluents for storing green jungle fowl spermatozoa at 3-5oC for 48 hours can improve the plasma membraneintact, acrosome membranes intact, fertility and hatchability of chicken eggs.
