Abstract

Snake fruit is a tropical fruit that is currently in great interest by the public. The This fruit is known to have high antioxidants so it can prevent an increase in cholesterol and other plasma lipids. Moreover, it can also prevent diarrhea. This study investigated the effective which one of the effective doses (25%, 50%, and 100%) from snake fruit extract against the ESBL-Escherichia coli (E. coli). This study was an experimental study with a post-test-only control group design in Microbiology Laboratory, Universitas Prima Indonesia. To investigate the antibacterial activity of snake fruit extract, this study was used disc diffusion methods which used five different groups including positive control (meropenem), negative control (DMSO), 100%, 50%, and 25% of snake fruit extract. The antibacterial assay showed that it required 100% snake fruit extract to inhibit the growth of ESBL-E. coli with average inhibition zone diameter of 7.33 ± 0.58 mm. The lower concentration extract did not show any antibacterial activity like the negative control group. The positive control group showed the most potent antibacterial activity with an average inhibition zone diameter of 36.33 ± 0.58 mm. Thus, it can be concluded that the snake fruit has antibacterial activity against ESBL-E. coli, but it was not as good as the meropenem. Abstrak: Buah salak merupakan buah tropis yang saat ini banyak diminati oleh masyarakat. Buah ini dikenal memiliki antioksidan yang tinggi sehingga dapat mencegah peningkatan kolesterol dan berbagai lipid plasma. Selain itu, juga dapat mencegah diare. Penelitian ini menyelidiki dosis mana yang efektif (25%, 50%, dan 100%) dari ekstrak salak terhadap ESBL-Escherichia coli (E. coli). Penelitian ini merupakan penelitian eksperimen dengan desain post test only control group di Laboratorium Mikrobiologi Universitas Prima Indonesia. Untuk mengetahui aktivitas antibakteri ekstrak buah salak, penelitian ini menggunakan metode difusi cakram yang menggunakan lima kelompok berbeda yaitu kontrol positif (meropenem), kontrol negatif (DMSO), ekstrak buah salak 100%, 50%, dan 25%. Uji antibakteri menunjukkan bahwa diperlukan 100% ekstrak salak untuk menghambat pertumbuhan ESBL-E. coli dengan diameter zona hambat rata-rata 7.33 ± 0.58 mm. Ekstrak konsentrasi rendah tidak menunjukkan aktivitas antibakteri seperti pada kelompok kontrol negatif. Aktivitas antibakteri paling kuat ditunjukkan oleh kelompok kontrol positif dengan rata-rata diameter zona hambat 36.33 ± 0.58 mm. Dengan demikian dapat disimpulkan bahwa salak memiliki aktivitas antibakteri terhadap ESBL-E. coli, tetapi tidak sebaik meropenem.

Abstract

Penyakit Diabetes Melitus merupakan masalah kesehatan global yang ditandai dengan hiperglikemia, seringkali disebabkan oleh kerusakan sel beta pankreas. Pengobatan diabetes seringkali memiliki efek samping dan biaya yang tinggi, sehingga penelitian mengenai terapi alternatif, seperti ekstrak alami diperlukan. Penelitian ini bertujuan untuk mengevaluasi efektivitas ekstrak bawang putih tunggal (Allium sativum) terhadap kadar gula darah (KGD), HbA1c, dan gambaran histopatologi pankreas pada tikus (Rattus norvegicus) yang mengalami dislipidemia. Penelitian ini menggunakan desain eksperimen dengan desain kelompok pre-test dan post-test menggunakan 24 ekor tikus yang dibagi menjadi empat kelompok perlakuan. Data dianalisis dengan uji normalitas, uji One-way ANOVA, uji Post-Hoc Tukey HSD, uji Kruskal-Wallis, dan Mann-Whitney. Hasil statistic menunjukkan perbedaan KGD yang signifikan antar kelompok pada hari ke-0 (P. 0.001) dan hari ke-28 (P. 0.001). Namun, pada uji Post-Hoc Tukey HSD, tidak ditemukan perbedaan signifikan antara kelompok kontrol positif dengan kelompok dosis 200 mg/kgBB (P. 0.499) dan 400 mg/kgBB (P. 0.866) pada hari ke-28. Pada parameter HbA1c, uji ANOVA menunjukkan tidak ada perbedaan signifikan antar kelompok pada seluruh haripengamatan (P. > 0.05). Secara visual, gambaran histopatologi menunjukkan adanya perbaikan pada struktur pankreas. Secara statistik, pemberian ekstrak bawang putih tunggal tidak menunjukkan perbedaan yang signifikan terhadap kadar KGD, HbA1c, maupun histopatologi pankreas pada tikus yang diuji.

Abstract

This study investigated the hepatoprotective efficacy of ethanol-extracted single garlic (EEBPT) on fatty liver and hepatic enzymes in rats. A true experimental design with pretest-posttest control group approach was employed. Twenty-four male Wistar rats (Rattus norvegicus) were randomly allocated into four groups: negative control (distilled water), positive control (simvastatin 2.1 mg/kg body weight), low-dose EEBPT (200 mg/kg body weight), and high-dose EEBPT (400 mg/kg body weight). All animals received high-fat diet supplementation for four weeks. Serum aspartate aminotransferase (SGOT/AST) and alanine aminotransferase (SGPT/ALT) were measured on days 0, 14, 21, and 28. Hepatic histopathology was assessed using hematoxylin and eosin staining. One-way ANOVA with Tukey post-hoc test and Kruskal-Wallis test were applied for statistical analysis (p<0.05). Results demonstrated that high-dose EEBPT significantly reduced SGOT and SGPT levels equivalent to simvastatin treatment and ameliorated hepatic steatosis, hepatocyte ballooning, and inflammatory infiltration. Ethanol-extracted single garlic at 400 mg/kg body weight exhibited hepatoprotective effects comparable to conventional statin therapy through antioxidative and anti-inflammatory mechanisms. Garlic extract represents a promising natural hepatoprotective agent for NAFLD management

Abstract

Diabetes mellitus is a global health problem with a growing prevalence. Blood glucose levels that remain high can cause damage to various body tissues. including the pancreas. Durian seeds contain bioactive compounds such as alkaloids. triterpenoids. phenolics. and flavonoids that can reduce blood sugar levels. This study is an experimental study with a posttest only controlled group design conducted at the Pharmacology Laboratory of the Faculty of Pharmacy. University of North Sumatra and the Anatomical Pathology Laboratory of the Royal Prima Medan Hospital for more than 3 months. From the results of this study. durian fruit seed extract (Durio zibethinus Murr) in the phytochemical compound test contained alkaloids. saponins. flavonoids. and glycosides. With doses of 150 mg. 300 mg. and 450 mg proven to reduce blood sugar levels (KGD) in streptozotocin-induced Wistar rats. The highest dose in reducing blood sugar levels (KGD) with a dose of 450 mg. The group with a dose of 300 mg showed the best results in increasing the diameter of the islets of Langerhans.

Abstract

Parkinson's disease is a neurodegenerative disorder characterized by loss of dopamine and formation of α-synuclein aggregates in the substantia nigra pars compacta (SNpc). This study aims to evaluate α-synuclein levels in the plasma of rotenone-induced Wistar rats as a model of Parkinson's disease and the therapeutic effect of Andaliman fruit extract. The design of this study was experimental with 25 male Wistar rats divided into five groups, negative control group (K-, only given distilled water), positive control (K+, induced rotenone 3 mg/kgBB/day for two weeks), and three treatment groups (P1, P2, P3) induced rotenone and given Andaliman extract orally at a dose of 150 mg/kgBB, 300 mg/kgBB, and 450 mg/kgBB respectively for 14 days. Measurement of levels was performed using ELISA on day 14 after induction and day 28 after therapy. The results of one-way ANOVA analysis showed that Andaliman extract significantly reduced α-synuclein levels at all doses compared to the positive control (p=0.000). The decrease in α-synuclein levels indicates the potential of Andaliman fruit extract in reducing Parkinson's disease-related pathology in mouse models.
