Abstract

Fish utilize energy from carbohydrate sources in small amounts because of the low secretion of the enzyme amylase for digestion. One alternative is to increase the availability of exogenous digestive enzymes by utilizing bacteria from the digestive tract that have amylolytic activity. This study aims to isolate amylolytic bacteria in the digestive tract of Red Snapper as a probiotic candidate. This study used several methods, namely amylolytic test on starch, test for resistance to acid conditions (pH 3), observation of bacterial growth for 30 hours, antagonistic test against pathogenic bacteria, test for attachment to the stomach, gram staining test, and pathogenicity test on shrimp. Screening of amylolytic bacteria in the digestive tract of red snapper produced five isolates, namely A1, A2, A3, A4, and A5. From the results of the amylolytic test, only three isolates were further tested, namely A1, A2, and A4. The three isolates were able to survive in acidic conditions of pH 3 and pH 7 for eight hours with density values ​​(OD) in isolates A1 (0.630), A2 (0.597), and A4 (0.601). Isolates A1, A2, and A4 respectively produced antagonistic values ​​with inhibition zones of 8.4 mm, 7.5 mm, and 10.3 mm. Next. the ability to attach bacteria to isolates A1 (6.6 x 102 cfu/ml), A2 (11.8 x 102 cfu/ml) and A4 (16.5 x 102 cfu/ml). After that, the pathogenicity test was carried out aimed at proving that the isolate was not pathogenic to the host. Based on the results of the study, there were three bacterial isolates (A1, A2, and A4) in the digestive tract of red snapper that had met the requirements for probiotic candidates in fish/shrimp and the best was A4 isolate.

However, it has a slow growth. The purpose of this research was to find the impact of giving feeding variations, namely D. magna that enriched ascorbic acid on improving the angelfish growth and its immune response. The research method used was Completely Randomized Design (CRD) using four treatments and three replications. The dosage of ascorbic acid consisted of 0 mg/L, 50 mg/L, 100 mg/L, and 150 mg/L. The fish was fed twice a day and applied with ad libitum. The result showed the addition of ascorbic acid had impacted its growth, and it produced SGR 2.393±0.003 %/day and challenge tests. The Survival Rate (SR) after challenge test was 79.99±1.000 %. In conclusion, the highest amount of growth and survival was treatment D (150 mg/L).

Abstract

Green seaweed (Ulva reticulata) is found in many parts of Indonesia. The potential of green seaweed is not widely known to the public, even though it is considered a parasite on other types of seaweed. The purpose of this study was to determine the antibacterial compounds contained in green seaweed and their effectiveness against Vibrio parahaemolyticus bacteria. Green seaweed was extracted by maceration method and then qualitative and quantitative tests were carried out. After that, the minimum inhibitory concentration test was carried out and continued with the minimum bactericidal concentration test and the inhibition test with 5 treatments and 3 replications. The data from the qualitative and quantitative test of seaweed, the minimum inhibitory concentration and the minimum bactericidal were analyzed descriptively, while the data from the inhibition zone test was analyzed statistically by ANOVA. The results of qualitative and quantitative tests showed that green seaweed contained flavonoids (6.4909 mg/g), tannins (70.7500 mg/g) and saponins (443.7286 mg/g). The results of the minimum inhibitory concentration test showed that the concentration of 3.125% was the minimum concentration to inhibit bacterial growth. In the inhibition test, it can be seen that P4 (6%) is the best treatment. The results of statistical analysis show that the treatments given are significantly different, except that P4 (6%) and P5 (7,5%) are not significantly different. From this study it can be concluded that green seaweed extract is effective in inhibiting the growth of Vibrio parahaemolyticus bacteria.

Abstract

However, there are still obstacles in the cultivation process, namely the threat of disease, one of which is the Motile Aeromonas Septicaemia (MAS) disease caused by Aeromonas hydrophila bacteria. The countermeasure can be done by using active compounds found in plants, including banana stems. This study aimed to determine the effectiveness of giving banana stem extract to carp culture media and treating A. hydrophila disease. This research used an experiment with a completely randomized design. The treatments tested were the addition of banana stem extract at a dose of 5% (treatment A), 10% (treatment B), 15% (treatment C), and antibiotics (treatment K-), and without extract treatment (treatment K+). Each treatment was repeated three times. The results showed that the administration of banana stem extract significantly affected the fish's erythrocytes, leukocytes, and blood glucose. Leukocyte differential observation showed that kepok banana stem extract had a significant effect on eosinophils, lymphocytes, and neutrophils but no significant effect on monocytes. In analyzing fish blood, the optimal values were 7.640.190a cells/mm3 for leukocytes, 3.600.116d cells/mm3 for erythrocytes, 656.028b mg/dl for glucose, 74.670.577d% for lymphocytes, 5.670.577a% for neutrophils, and 00a% for eosinophils. Abstract

The cultivation of vannamei shrimp (Litopenaeus vannamei) has become one of the most economically important sectors of aquaculture worldwide. However, challenges such as disease, declining water quality, and dependence on antibiotics have driven the search for more sustainable alternative solutions. The use of probiotics in feed has emerged as an innovative and environmentally friendly approach to improving shrimp health and production efficiency. Probiotics function by enhancing microbial balance in the digestive tract, strengthening the immune system, and improving nutrient absorption, which ultimately positively impacts the growth and survival of shrimp. This study highlights the importance of probiotic supplementation in shrimp feed cultivated under large-scale intensive systems. The method used in this study was an experimental method with two treatments: without probiotics (P0) and with probiotics (P1). The observed parameters included shrimp biomass growth (MBW), daily growth (SGR), and survival rate (SR). The results showed that the average MBW values were 8.83g (P0) and 8.92g (P1). The daily growth rate was 0.27%/day (P0) and 0.29%/day (P1). The FCR value was 1.2 for P0 and 0.9 for P1. Furthermore, the survival rate (SR) was 60% for P0 and 71% for P1. This study concludes that the use of probiotics in shrimp farming positively impacts the growth and survival of vannamei shrimp.Keywords: Effective; Lactobacillus Sp; Mortalities

Abstract

One of the biodiversity that Indonesia has and should be proud of is the diversity of freshwater ornamental fish species, one of which is koi kohaku fish (Cyprinus carpio). The purpose of this study was to determine the effect of color in telang flower extract to improve the color quality of koi kohaku fish and to determine the best dose of telang flower extract in improving the color of koi fish. This study used 4 different treatments, namely A (10%) bayang flower extract, B (15%) bayang flower extract, C (20%) bayang flower extract, and D (control). The parameters observed were the brightness of fish color, final weight, length growth, SR, and water quality, which will be analyzed using ANOVA. Measuring fish color using a modified toca color finder (M-TCF) tool where this tool has a color column column marked with a score value from 1 - 60. The results of this study indicate that the addition of telang flower extract to commercial feed affects the color quality of koi kohaku fish. The quality of color severity in Kohaku koi fish is highest in treatment C (20%) with a score value of 35 on fish color, for treatment D (control), B (15%), and A (10) each has a score value of 32, 34, 33 on fish color. Mixing bayang flower extract into commercial feed can improve the color of koi kohaku fish, the best color improvement was found in the treatment of adding 20% bayang flower extract.

Abstract

AbstrakIkan gurami merupakan ikan yang memiliki tingkat minat konsumsi tinggi, namun pertumbuhannya lambat. Upaya peningkatan produksi gurami (Osphronemus gouramy) dapat dilakukan dengan cara pemuasaan. Pemuasaan berarti pemberian pakan dengan selang waktu yang ditentukan dengan harapan ikan dapat mengalami peningkatan nafsu makan setelah dipuasakan. Peningkatan konsumsi pakan setelah dipuasakan disebut hiperfagia, yakni kondisi dimana nafsu makan ikan meningkat selama beberapa waktu 2-3 hari setelah ikan dipuasakan pada periode tertentu dan akan kembali ke keadaan normal. Selama dalam keadaan hiperfagia dapat memberikan pasokan nutrisi yang cukup untuk memenuhi kebutuhan metabolisme setelah ikan dipuasakan sehingga dapat meningkatkan pertumbuhan. Penelitian ini bertujuan untuk mempercepat pertumbuhan dengan menghemat pakan. Penelitian dilakukan skala laboratorium menggunakan metode eksperimen. Setiap akuarium berisi ikan gurami 10 ekor dengan panjang sekitar 8-10 cm. Perlakuan berjumlah 4 dengan ulangan 5 kali. Data SGRw (spesific growth rate weight), SGRl (spesific growth rate length) dan PER (protein efficiency ratio) diambil 10 hari sekali kemudian dilakukan uji analisis anova dan uji homogenitas. Hasil penelitian SGRw menunjukan hasil yang tidak berpengaruh namun P3 (makan 3 hari puasa 1 hari) mendekati kontrol sebesar 4.42±0.75% dan SGRl menunjukan hasil yang sama sedangkan PER menunjukan hasil yang tidak berbeda nyata dengan nilai P3 mendekati P4 sebesar 19.84±5.42%. Pemuasaan pada ikan gurami dengan interval waktu berbeda tidak berpengaruh terhadap pertumbuhan dan rasio efisiensi protein.Kata kunci: ikan gurami; pemuasaan; pertumbuhan; protein; hiperfagiaAbstractGourami is a fish that has a high level of consumption, but it growth is slow. Efforts to increase the production of gurami (Osphronemus gouramy) can be done by fasting. Fasting means feeding with a specified time interval in the hope that fish can experience an increase in appetite after fasting. Increased feed consumption after fasting is called hyperphagia, a condition in which fish appetite increases for a period of 2-3 days after the fish is fasted for a certain period and will return to its normal state. During the state of hyperphagia can provide a sufficient supply of nutrients to meet metabolic needs after the fish is fasted to increase growth. This study aims to accelerate growth by saving feed. The study was conducted on a laboratory scale using experimental methods. Each aquarium contains 10 fish of gourami with a length of about 8-10 cm. The treatment amounted to 4 with replications 5 times. SGRw data (specific growth rate weight), SGRl (specific growth rate length) and PER (protein efficiency ratio) were taken once every 10 days and then performed ANOVA analysis test. The results of the SGRw study showed no effect but P3 approached the control of 4.42 ± 0.75% and SGRl showed the same results while PER showed results that were not significantly different from the P3 value approaching P4 of 19.84 ± 5.42%. Fasting in gourami with different time intervals does not affect growth and protein efficiency ratio.Keywords: gourami; fasting; growth, protein; hyperphagia

Abstract

Permintaan pasar yang tinggi pada ikan manfish sangat sulit terpenuhi dikarenakan pertumbuhan ikan manfish yang lambat. Solusi untuk memperbaiki pertumbuhan ikan manfish yaitu mengetahui padat tebar yang baik untuk ikan manfish serta ditambah penerapan teknologi resirkulasi yang di harapkan pertumbuhan ikan menjadi lebih cepat. Berdasarkan permasalahan itu maka dilakukannya penelitian ini untuk melihat respon ikan terhadap pertumbuhan. Metode yang digunakan dalam penelitian ini adalah metode Rancangan Acak Lengkap. Penelitian ini menggunakan 4 perlakuan (P1 (3 ekor/L dengan 25 liter air yaitu 75 ekor/wadah), P2 (6 ekor/L dengan 25 liter air yaitu 150 ekor/wadah), P3 (9 ekor/L dengan 25 liter air yaitu 225 ekor/wadah), dan P4 (12 ekor/L dengan 25 liter air yaitu 300 ekor/wadah) yang masing-masing perlakuan diulang sebanyak 3 kali ulangan. Hasil penelitian menunjukkan bahwa nilai rerata SGR (specific growth rate) yang terbaik adalah pada pelakuan P1 dengan pelakuan 3 ekor/L dengan air 25 liter yaitu 75 ekor/wadah, dengan rerata SGR sebesar 5,12 %. Laju pertumbuhan panjang yang terbaik adalah pada pelakuan P1 dengan pelakuan 3 ekor/L dengan air 25 liter yaitu 75 ekor/wadah, dengan rerata laju pertumbuhan panjang sebesar 1,40 %. Kelangsungan hidup (survival rate) terbesar adalah pada perlakuan P1 dengan pelakuan 3 ekor/L dengan air 25 liter yaitu 75 ekor/wadah, dengan rerata kelangsungan hidup (survival rate) sebesar 86,22%. Secara statistik pada penelitian ini mendapatkan hasil berpengaruh nyata terdapat pertumbuhan dan kelulusan hidup.Kata kunci: ikan manfish; resirkulasi; petumbuhan; kelulusan hidup High market demand for manfish is challenging to fulfill due to the slow growth of manfish. The solution to improve the growth of manfish is knowing good stocking density for manfish and adding the application of recirculation technology, which is expected to accelerate fish growth. Based on these problems, this study was conducted to see the response of fish to growth. The method used in this study is a completely randomized design method. In this study, we were using four treatments (P0 (3 fish/L with 25 liters of water is 75 fish/aquarium), P1 (6 fish/L with 25 liters of water is 150 fish/aquarium), P2 (9 fish /L with 25 liters water which is 225 fish/aquarium), and P3 (12 fish/L) with 25 liters of water, i.e., 300 fish/aquarium), each of which is repeated three times. The results show that the best SGR (specific growth rate) was in the treatment P1 with the treatment of 3 fish/L with 25 liters of water, which is 75 fish/aquarium, with a mean SGR 5.12%. The best high growth rate was at P1 treatment with three fish/L treatment with 25 liters of water, which is 75 fish/aquarium, with a high growth rate 1.40%. The most significant survival rate was in treatment P1 with the treatment of 3 fish/L with 25 liters of water, which is 75 fish/aquarium, with a mean survival rate amounting 86.22%, statistically in this study getting influential results or there were growth and survival rate.Keywords: manfish; recirculation; growth; survival rate

Abstract

In nature, there are many types of bacteria that can act as probiotics. These bacteria can come from soil, water, plants or even the digestive tract of animals. This study aims to analyze the biochemical characteristics of probiotic candidate bacteria originating from the digestive tract of giant prawns. The method used is a descriptive explorative method to perform biochemical analysis of the types of probiotic bacteria derived from jerbung shrimp. The biochemical analysis included the Catalase test, Oxidase test, Urea test, Citrate test, TSIA, Methyl red, Voges-Proskaeurs, and sugar fermentation test (D-Glucose, Sucrose, Inositol, Manisitol. Laboratory analysis results showed that the isolated bacteria had morphological characteristics rod-shaped, gram-positive, does not produce catalase and oxidase enzymes, but can produce citric enzymes. These bacteria are acidic, capable of fermenting glucose, sucrose, inositol, and mannitol. The conclusion of this study, probiotic candidate bacteria from the test results showed compatibility with Lactobacillus sp bacteria.Keywords: isolation; probiotics; shrimp
