Abstract

Background and objective.The response of rice plants to drought stress was preceded by physiological response in the form of reduction of transpiration rate to reduce water loss by closing stomata, reducing stomata, and decreasing leaf surface area by leaf rolling. Therefore, it is necessary to research the morphological character and physiological character of plants such as leaf area analysis and stomata index to find out their response to drought stress and without drought. Based on this, this research is aimed to know the area and index of stemata leaves of rice plants (Oryza sativa L.) varieties of Cisokan and Batang Piaman due to drought stress.Methodology. Drought treatment was performed by withholding water for 12 days in greenhouse.Data were taken destructively on days 0, 4, 6 and 12 after treatment. Result. Drought stress had an effect on decreasing leaf area and stomata index on both varieties. The lowest leaf area is found in Cisokan variety. Likewise with the stomata index, the lowest yield after 12 days of treatment was found in the Cisokan variety. Low leaf area and stomatal index are known to be more resistant to drought because they can reduce the rate of transpiration in plants. Keywords:    Drouhgt stress, Leaf Area, Stomata index

Abstract

Abstract. The purpose of this research to determined the effect of Sargassum sp. liquid organic fertilizer in the growth of spinach plant (Amaranthus hybridus L.) using hydroponic. This research used Completely Randomized Design with six treatments and four repetitions ie: A= comparison (AB mix 10 mL/L of water), B= liquid organic fertilizer 6 mL/L of water, C= liquid organic fertilizer 8 mL/L of water, D= liquid organic fertilizer 10 mL/L of water, E= liquid organic fertilizer 12 mL/L of water, and F= liquid organic fertilizer 14 mL/L of water. Parameter observed was height of plants, number of leaves, leaf surface area, fresh weight of plants, and dry weight of plants. Data was analyzed by using the Analysis of Variants (ANOVA)  and Duncan. The result showed that application of Sargassum sp. liquid organic fertilizer has been  an effect on height of plants, number of leaves, leaf area, fresh weight of plants, and dry weight of spinach plant (Amaranthus hybridus L.). However, the effect was not as expected to increase the growth of spinach plant.

Abstract

Pakcoy (Brassica rapa L.) is a type of vegetable plant that belongs to the Brassicaceae family. Content contained in Pakcoy are calories, protein, fat, carbohydrates, fiber, Ca, P, Fe, vitamins A, vitamins B and vitamins C. The high demand for vegetables, especially Pakcoy, is not matched by the extent of agricultural land for farming. One of the solutions that can be used for the cultivation of Pakcoy plants is hydroponically which does not require a large area of land, can also be done in the yard or terrace of the house. Coffe grounds can be used as organic fertilizer in a hydroponic system and can be an alternative that can be used to replace inorganic fertilizers. This study uses a CRD (Completely Randomized Design) with treatments consisting of 5 levels: A1B1 (1 dose AB Mix), A2B2 (½ dose AB Mix+5 mL liquid organic fertilizer coffe grounds), (½ dosis AB Mix+10 mL liquid organic fertilizer coffe grounds), (5 mL liquid organic fertilizer coffe grounds) and (10 mL liquid organic fertilizer coffe grounds). The result showed that the provision of hydroponic nutrients of coffe grounds\had an effect on the growth of Pakcoy, but the growth was lower than that of AB Mix.Pakcoy (Brassica rapa L.) merupakan salah satu jenis tanaman sayuran yang termasuk dalam famili Brassicaceae. Kandungan yang terkandung di dalam pakcoy adalah kalori, protein, lemak, karbohidrat, serat, Ca, P, Fe, vitamin A, vitamin B dan vitamin C. Tingginya permintaan sayuran khususnya pakcoy tidak diimbangi dengan luasnya lahan pertanian untuk bercocok tanam. Salah satu solusi yang bisa digunakan untuk budidaya tanaman pakcoy adalah secara hidroponik yang tidak membutuhkan lahan yang luas, bisa juga dilakukan di pekarangan atau teras rumah. Ampas kopi dapat digunakan sebagai pupuk organik dalam sistem hidroponik dan dapat menjadi alternatif yang dapat digunakan untuk menggantikan pupuk anorganik. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan perlakuan yang terdiri dari 5 taraf : A1B1 (1 dosis AB Mix), A2B2 (½ dosis AB Mix+5 mL pupuk organik cair ampas kopi), (½ dosis AB Mix+10 mL cair pupuk organik ampas kopi), (5 mL pupuk organik cair ampas kopi) dan (10 mL pupuk organik cair ampas kopi). Hasil penelitian menunjukkan bahwa pemberian nutrisi hidroponik ampas kopi berpengaruh terhadap pertumbuhan Pakcoy, namun pertumbuhannya lebih rendah dibandingkan dengan AB Mix.

Abstract

As mustard greens contain various benefits that are beneficial to health, because the mustard greens contain vitamins and minerals that play an important role in the process of growth and metabolism of the body. One way to grow vegetables in a narrow area is to use a hydroponic system. Hydroponics is a way of farming without using soil media but uses water as a medium and uses nutrients as nutrients for plant growth. Hydroponic cultivation is one of the efforts to obtain quality agricultural products, healthy, and free of pesticides. However, the use of hydroponic nutrients for now is still sourced from chemical fertilizers. Therefore, one solution to reduce the use of inorganic fertilizers can be used organic fertilizers, namely liquid organic fertilizer which is more easily absorbed by plants because the elements in it have been decomposed. One of the wastes that can be used as liquid organic fertilizer is coffee grounds waste. This study aims to determine the use of liquid organic fertilizer coffee grounds (Coffea arabica L.) as hydroponic nutrients in mustard plants (Brassica juncea L.). This research was conducted from February to April 2020, located at the Integrated Research Laboratory and the wire house of the Department of Biology FMIPA UNP. This study uses a completely randomized design (RAL) with 5 treatments and 3 replications. Data were analyzed using the Analysis of Variance (ANOVA) and DNMRT follow-up tests at the 5% level. The results showed that various concentrations of hydroponic nutrient solution with liquid organic fertilizer based on coffee grounds had a significant effect on increasing the growth of mustard plants (Brassica juncea L.) both from plant height, number of leaves and leaf area. Keywords: Coffea Grounds, Hydroponics, Liquid Organic Fertilizer, Plant SawiSebagai sayuran sawi mengandung berbagai khasiat yang bermanfaat bagi kesehatan, karena pada sayuran sawi mengandung vitamin dan mineral yang berperan penting dalam proses pertumbuhan dan metabolisme tubuh. Salah satu cara untuk penanaman sayuran pada lahan yang sempit adalah dengan menggunakan sistem hidroponik. Hidroponik merupakan cara bercocok tanam tanpa menggunakan media tanah tetapi menggunakan air sebagai media dan menggunakan unsur hara sebagai nutrisi untuk pertumbuhan tanaman. Budidaya secara hidroponik merupakan salah satu upaya untuk memperoleh produk pertanian yang berkualitas, sehat, dan bebas pestisida. Namun penggunaan nutrisi hidroponik untuk saat ini masih bersumber pada pupuk kimia. Oleh karena itu salah satu solusi untuk mengurangi penggunaan pupuk anorganik tersebut dapat digunakan pupuk organik yaitu pupuk organik cair dimana lebih mudah terserap oleh tanaman karena unsur-unsur di dalamnya sudah terurai. Salah satu limbah yang dapat dijadikan pupuk organik cair yaitu limbah ampas kopi. Penelitian ini bertujuan untuk mengetahui pemanfaatan pupuk organik cair ampas kopi (Coffea arabica L.) sebagai nutrisi hidroponik pada tanaman sawi (Brassica juncea L.). Penelitian ini telah dilakukan bulan Februari sampai April 2020, bertempat di Laboratorium Penelitian Terpadu dan rumah kawat Jurusan Biologi FMIPA UNP. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 5 perlakuan dan 3 ulangan. Data dianalisis menggunakan uji Analisis Varians (ANOVA) dan uji lanjut DNMRT pada tingkat 5%. Hasil penelitian menunjukkan bahwa berbagai konsentrasi larutan nutrisi hidroponik dengan pupuk organik cair berbahan dasar ampas kopi memberikan pengaruh yang nyata  untuk meningkatkan pertumbuhan tanaman sawi (Brassica juncea L.) baik dari tinggi tanaman, jumlah daun dan luas daun.

Abstract

Peroksidase (POD) merupakan enzim antioksidan yang memiliki beragam fungsi dalam siklus hidup tanaman, salah satunya adalah sebagai pertahanan dalam melawan ROS dengan mengkatalisis konversi H2O2 menjadi udara dan O2 . Kemampuan aktifitas enzim POD dalam mengatur kandungan H2O2 memungkinkan enzim tersebut dapat mempertahankan tanaman dari cekaman. Metode yang dapat digunakan untuk amplifikasi gen POD salah satunya yaitu kuantitatif reverse transcription- PCR (qRT-PCR). Metode ini memerlukan beberapa komponen penting salah satunya yaitu primer ( forward dan reverse ). Primer yang digunakan dalam amplifikasi gen harus spesifik terhadap gen target sehingga dapat mengenali dan menempel pada gen target yang diinginkan. Penelitian ini bertujuan untuk mendesain primer yang sesuai untuk amplifikasi gen POD menggunakan teknik qRT-PCR. Primer dirancang menggunakan perangkat PrimerQuest. Primer yang telah dirancang kemudian dianalisis untuk spesifikasinya dengan geneious prime . Kemudian spesifikasi primer di cek menggugakan primer BLAST. Hasil desain primer dengan kriteria terbaik untuk amplifikasi gen POD yaitu Forward POD 5'-AAATGCGTCGATCTACTGTACCT-3' dan Reverse POD 5'-GTGTTGAAAATGGCAATAAACCGG-3'
