Abstract

The aquaculture system in the maintenance of dumbo catfish is widely practiced by the community due to several advantages it offers. However, in terms of fish health, there is a risk of infection by Pseudomonas bacteria present in the environment. This study aims to isolate and identify Pseudomonas sp. bacteria in dumbo catfish (Clarias gariepinus) raised in a semi-intensive manner. A total of three samples of dumbo catfish were taken from ponds in Beurangong Village, Kuta Baro District, and nine samples from ponds in Dham Pulo Village, Ingin Jaya District, Aceh Besar Regency. The criteria for the samples were fish that clinically showed symptoms of disease caused by Pseudomonas sp. Each catfish had samples taken from the skin, liver, spleen, intestines, and gills for subsequent bacterial isolation and identification using the Carter method. Bacterial isolation was conducted using Nutrient Broth (NB) media. Identification utilized Plate Count Agar (PCA) media, Gram staining, and biochemical tests. The results showed bacterial growth in the NB media, and Pseudomonas bacteria were successfully isolated. The identification results on PCA media indicated Pseudomonas sp., and the biochemical tests identified it as Pseudomonas aeruginosa. This study concludes that in dumbo catfish raised semi-intensively, Pseudomonas sp. bacteria were isolated and identified in Beurangong Village, while in Dham Pulo Village, the identified species was Pseudomonas aeruginosa.

Abstract

Pathogenic Escherichia coli is a type of bacteria that has the ability to cause disease in hosts, including humans and animals, although in general E. coli acts as a normal flora in the digestive tract. This study aimed to detect the presence of pathogenic E. coli in the intestines of ducks sold at Lambaro Market, Aceh Besar. E. coli is a normal flora bacteria in the digestive tract of animals and humans, but some strains are pathogenic and can cause zoonotic diseases. The presence of pathogenic E. coli in poultry products has the potential to be a source of infection for humans through consumption of contaminated food. This study used 15 duck intestine samples obtained from cecum, to identify the presence of pathogenic E.coli. The results showed that 13 samples produced clear colonies on Sorbitol MacConky Agar (SMAC), indicating the inability of bacteria to ferment sorbitol, which correlated to the presence of pathogenic E. coli. In contrast, two samples showed pink colonies indicating the presence of nonpathogenic E. coli. The conclusion of this study was the cecum of ducks sold at Lambaro Market, Aceh Besar positive for pathogenic E. coli.

Abstract

Bandotan leaves (Ageratum conyzoides) are one of the traditional medicinal plants that are trusted empirically by the community to treat various diseases. Several studies report the potential of bandotan leaves as an antibacterial because they contain the chemical compounds trepanoids, flavonoids, alkaloids, essential oils, and tannins. This study aims to determine the effectiveness of bandotan leaves on the growth of Salmonella enterica serovar typhimurium bacteria in vitro. This research used the Kirby Bauer disk diffusion method with the antibiotic chloramphenicol as a positive control, CMC 1% as a negative control, and an ethanol extract of bandotan leaves with concentrations of 25%, 50%, and 75%. Re-identification of bacteria was carried out using Gram staining, and effectiveness testing was carried out on Mueller Hinton Agar (MHA) media. The results of the study on Gram staining showed uniform morphological characteristics of Salmonella enterica serovar Typhimurium bacteria and the diameter of the inhibition zone formed at concentrations of bandotan leaf ethanol extract of 25% (8.02 mm ± 0.41), 50% (8.37mm ± 0.69) and 75% (8.78mm ± 0.78). It can be concluded that the ethanol extract of bandotan leaves at concentrations of 25%, 50%, and 75% has a weak category of inhibitory activity against the growth of Salmonella enterica serovar Typhimurium bacteria in vitro.

Abstract

The purpose of this research was to identify avian influenza (AI) virus using serological and molecular methods on poultry which suspected as AI infected in Aceh province. This study used 37 samples of tracheal and cloacal swabs and organs from various species of poultry that were collected from several districts/cities in Aceh. Samples were collected and put into transport media and stored at 4 C before sending to the laboratory. Samples were inoculated in specific pathogen-free of embryonated chicken egg with the age of 9-11 days for further serological and molecular examination. From 37 samples which infected to embryonated chicken egg then followed by hemagglutinin agglutination test/hemagglutinin inhibition revealed that 7 samples were positively infected with AI virus. The amplification result of specific matrix gene primer was followed by electrophoresis on 2% agarose gel which were obtained in the form of a deoxyribonucleic acid (DNA) band at 276 bp for matrix gene and 1.725 bp for H5 gene for all isolates test. In conclusion, the virus which caused the death of various types of poultry in Aceh province is avian influenza A virus subtype H5.Key words: avian influenza virus, H5N1, serologic, matrix, heamaglutinin

Abstract

Tujuan dari penelitian ini adalah mengidentifikasi keberadaan gen M, H5, dan N1 virus avian influenza (AI) melalui metode single step multiplex reverse transcriptase-polymerase chain reaction (RT-PCR), sebagai acuan untuk peneguhan diagnosis secara molekuler virus AI di Provinsi Aceh. Penelitian ini menggunakan 11 isolat virus AI asal Provinsi Aceh yang diperoleh dari Laboratorium Balai Penyidikan dan Pengujian Veteriner (BPPV) Regional I Medan di Sumatera Utara dari tahun 2006-2008. Penelitian dilakukan di Laboratorium Virologi BPPV Regional I di Medan, Sumatera Utara. Amplifikasi terhadap gen matriks (M) virus AI menggunakan metode simplex RT-PCR. Hasil simplex RT-PCR terhadap gen M diperoleh 10 isolat yang menunjukkan pita deoxyribonucleic acid (DNA) pada 276 bp dan satu isolat yang tidak muncul, kemudian dilanjutkan dengan metode single step multiplex RT-PCR menggunakan pasangan primer gen penyandi protein N1, H5, dan M. Produk PCR 131 bp (N1), 189 bp (H5), dan 276 bp (M) muncul sebagai hasil elektroforesis dari semua isolat virus AI. Semua virus AI yang mewabah dari tahun 2006-2008 di Provinsi Aceh termasuk ke dalam virus influenza A subtipe H5N1.

Abstract

Blang Krueng Village has potential in the field of animal husbandry, but the livestock raised is still traditional, so that many livestock lack feed, this causes the supply of minerals to the livestock's body to decrease. Urea molasses multinutrient block (UMMB) is one of the additional feeds with a high content of nutritional supplements, especially minerals for ruminants in solid form. The purpose of the service activity is to increase community income based on the development of the ability of the farmer community by increasing the knowledge and skills of farmers regarding technology for making nutrient block and capa leaf cream to increase their cattle production. The method that used in this activity is to use the approach of community participation methods in development or Participatory Rural Appraisal (PRA). The results of this activity showed that 95% of participants are interested in practicing, developing, and transmitting their knowledge of how to make UMMB and capa leaf cream to members of farmer groups around them. The conclusion, farmers will share the knowledge that has been obtained during the service to other farmer group members as the target group of service activities.

Abstract

The activities of the Lamgapang Village community so far have done a lot of home industry businesses, but the results obtained are still relatively small, this is because the products produced are still unable to compete in the market. So that many people are still low-income and fall into the category of underprivileged families. The purpose of the activity is to increase community income based on developing Community Development activities for the Lamgapang Aceh Besar Community in Empowering Creative Economy Based on Local Wisdom. Provide motivational training and skills in plant cultivation using hydroponics and development of livestock health based on farmer empowerment. The method that will be used in this activity is using the Participatory Rural Appraisal approach. Partners participate in carrying out direct activities, especially in terms of production and analyzing problems in the field. The results of the service activities that have been carried out include: 1) The service team has succeeded in making hydroponic tools and planting hydroponic mustard plants; 2) For service activities based on livestock health, an examination of diseases in livestock and treatment for livestock has been carried out. The conclusion is that the community has been able to plant mustard greens using hydroponic tools and also the breeders already have the knowledge of how to raise livestock properly and correctly.
