Abstract

Salah satu peranan bakteri tanah adalah sebagai pemacu pertumbuhan tanaman. Penelitian ini bertujuan untuk mempelajari respon pertumbuhan tanaman jagung dan kedelai terhadap pemberian konsorsium bakteri indigenous asal lahan kering Pulau Lombok pada perlakuan tanah steril dan nonsteril dan perbedaan kapasitas lapang air media tanam. Tanah diambil dari lahan kering Lombok Utara. Tanah steril dipersiapkan dengan cara mensterilisasi tanah lahan kering menggunakan autoklaf. Konsorsium bakteri dipersiapakan dengan mencampur 15 inokulum isolat bakteri rhizosfer asal lahan kering Pulau Lombok. Inokulasi konsorsium dilakukan dengan teknik inokulasi tanah. Kapasitas lapang air media tanam diatur 25%, 50%, dan 75%. Penelitian dilakukan pada skala rumah kaca. Parameter pertumbuhan yang diamati berupa jumlah daun, lebar daun, panjang daun, panjang tanaman, berat basah dan berat kering biomassa bagian atas tanaman. Hasil penelitian menunjukkan bahwa konsorsium bakteri mampu memacu pertumbuhan tanaman. Kemampuan tersebut lebih tampak pada tanaman yang ditumbuhkan pada media tanah steril dibandingkan pada tanah nonsteril.

Abstract

Composition and abundance of culturable bacteria of four soil samples (Ktr50II, D50II, G50II, and A50II) were analyzed. The soil samples were collected from maize rhizosphere that planted in dryland Lombok Island. Each soil sample give different growth performance of maize in greenhouse experiment. This study was to investigate the relation of maize growth performance with culturable bacterial community of their rhizosphere and the effect of culture media on number of bacterial isolates recovery. The rhizosphere bacteria were cultured and isolated on commercial media (SEA) and non commercial modification media (NAln, NAln-SE and NAln- RE). Thirty four strains rhizosphere bacteria were obtained from four maize rhizophere soil samples. D50II is the soil sample that caused the better growth performance to the maize, contrary to Ktr50II. D50II has significantly highest number of culturable bacterial types, while significantly lowest on Ktr50II. In D50II, at least 17 bacterial isolates contributed to better growth performance in maize and have relative abundance of dominant isolate not more than 35.34%. In comparing the rhizosphere bacteria recovered using different culture media, bacteria cultivated on SEA have different growth characteristic compared with bacteria cultivated on NAln, NAln SE and NAln-RE. Six bacterial isolates showed antagonistic ability when grew on SEA but not in all of three media. Compared with commercial media, non commercial modification media can increase total isolates recovery about 70.6%.

Abstract

Materi mikrobiologi sebagai bagian dari mata pelajaran Biologi SMA seringkali tidak bisa dilaksanakan praktikumnya, disebabkan alat dan bahan yang tidak tersedia di sekolah maupun prosedur yang cukup sulit &/ lama untuk dilakukan di laboratorium. Hal ini membuat materi mikrobiologi menjadi sulit dipahami oleh siswa SMA. Kegiatan Pengabdian Kepada Masyarakat ini bertujuan memberikan pelatihan/ penyegaran bagi guru-guru Biologi SMA pada praktikum Mikrobiologi Dasar dan Mikrobiologi Terapan yang bisa dilaksanakan di laboratorium Biologi pada tingkat SMA. Kegiatan ini dilaksanakan di SMA Negeri I Jonggat, Lombok Tengah, selama 3 hari (13-15 September 2024). Materi yang diberikan adalah materi mikrobiologi dasar berupa tahapan dalam isolasi, kultur dan identifikasi bakteri tanah dan materi mikrobiologi terapannya berupa Pembuatan Yogurt dan Roti Dengan Peralatan yang Terdapat di Dapur Rmah Tangga. Kegiatan ini diikuti oleh para guru biologi SMA dan para siswa SMA yang tertarik dalam praktikum mikrobiologi. Hasil yang didapatkan pada kegiatan ini adalah beberapa para siswa mampu membuat kultur dan pewarnaan mikroba tanah dan mengidentifikasi mikroba yang dapat diisolasi dari tanah dengan cara yang sederhana. Para siswa juga mampu membuat minuman dan makanan fermentasi dengan cara yang sederhana menggunakan bahan dan alat yang tersedia di dapur rumah tangga.

Abstract

Kawasan Hutan Pusuk Pass terkenal sebagai kawasan ekowisata di Lombok Barat. Selain dihuni oleh monyet ekor panjang, Hutan Pusuk juga dihuni oleh berbagai jenis flora endemik. Salah satu diantaranya adalah pohon gaharu dari spesies Gyrinops versteegii. Tidak dipungkiri bahwa pohon gaharu merupakan salah satu daya tarik kawasan tersebut. Populasi pohon gaharu yang sudah sangat langka memerlukan upaya pelestarian. Pengabdian masyarakat ini bertujuan untuk melakukan restorasi hutan dengan penanaman dan pemupukan bibit pohon gaharu di Hutan Pusuk Pass Lombok Barat. Kegiatan dilaksanakan dengan metode aksi kolaboratif yang melibatkan mahasiswa dan dosen Program Studi Biologi FMIPA Universitas Mataram serta masyarakat lingkar hutan. Sebanyak 100 bibit pohon gaharu berhasil ditanam diikuti dengan pemupukan untuk meningkatkan kelulushidupan bibit. Kegiatan berjalan lancar dengan adanya kerjasama yang baik dari semua unsur yang terlibat. Melalui aksi ini diharapkan semakin menambah rasa tanggung jawab terhadap kelestarian pohon gaharu di daerah ekowisata Hutan Pusuk Pass.

Abstract

Busy activities make people prefer to consume ready-to-eat food which sometimes does not guarantee the level of cleanliness, so that cases of food poisoning often occur. Bacillus cereus is a bacteria that causes food poisoning which is usually found in rice. To overcome this problem, research was carried out to find new sources of medicine from endophytic fungi, namely fungi that live in plant tissue. The addition of clinical drugs derived from fungi has attracted interest in the discovery of new secondary metabolites derived from endophytic fungi. This research aims to (1) obtain endophytic fungal isolates isolated from red ginger rhizomes, (2) obtain endophytic fungal isolates that are antibacterial against Bacillus cereus. The method used is a laboratory experiment by randomizing and repeating the samples. The results showed that four isolates of endophytic fungi were successfully isolated from red ginger rhizomes with varying inhibitory abilities between isolates. The highest percentage of inhibitory power based on the in vitro dual culture assay was a cream colored isolate of 100 ± 0.0%.

Abstract

This research is important to conduct in order to study and explore the macrofungi of the Basidiomycota group in the environment of Mataram University, which can serve as a supporting reference in macrofungi learning and also serve as an initial step in macrofungi conservation. The research is descriptive-exploratory in nature, and the sampling was carried out using the roam method in habitats representing macrofungi of the Basidiomycota division. The results of the study revealed 16 species of macrofungi from the Basidiomycota division, including Schizophyllum commune, Pleurotus ostreatus, Termitomyces sp., Coprinus sp1., Coprinus sp2., Auricularia auricula, Geastrum sp., Pycnoporellus sp., Trametes sp., Trametes sp.2, Ganoderma applanatum, Ganoderma lucidum, Ganoderma sp1., Lentinus tigrinus, Lentinus sp., Picnoporellus sp., and Coprinellus sp. These macrofungi inhabit decaying wood with environmental temperature ranging from 31-32°C, air humidity of 77-79%, and soil pH ranging from 5-6, indicating an acidic environment.

Abstract

Organic waste management is an important step in supporting environmental sustainability and food security. This study aims to utilize household waste such as stale rice, banana peels, and bread as fermentation media to produce Local Microorganisms (MOL) that have the potential to become decomposers, biofertilizers, and organic pesticides. The study was conducted for 9 days at the Faculty of Mathematics and Natural Sciences, University of Mataram, with various media cover treatments (plastic, rice paper, and banana leaves) and natural media. Observations were made every two days by observing physical changes such as texture, color, aroma, and growth of microorganisms. The results showed that the type of media and cover treatment affected the development of microorganism colonies. Media with loose covers tended to have more bacterial growth, while tightly closed media showed fungal dominance. Banana peels produced black to pink mycelium, while bread produced fungal colonies such as Aspergillus sp. and Penicillium sp. Media with stale rice tended to produce a distinctive sour tape odor. In conclusion, household waste can be utilized to produce MOL containing decomposer microorganisms with variations in microbial content according to the media and treatment. This study provides an environmentally friendly solution in managing household organic waste into organic liquid fertilizers that are rich in nutrients.

Abstract

Bacterial leaf blight (BLB) is a significant disease of rice plants. It is caused by the Gram-negative bacterium Xanthomonas oryzae and often leads to yield losses. Excessive use of synthetic chemicals to control BLB has raised concerns over resistance and chemical residues. Therefore, natural alternatives are needed, such as Euphorbia hirta, which is traditionally used as a medicinal plant. This study aimed to determine the inhibitory power of E. hirta extracts against X. oryzae and to identify the presence of flavonoids, tannins, and saponins. Plant extracts were prepared by ethanol maceration, hot-water infusion (~90 °C), and fresh squeezing. Antibacterial activity was evaluated by the well diffusion method on Mueller-Hinton Agar with concentrations of 25–100% (v/v), using chloramphenicol as positive control. Inhibition zones were measured after 24–120 h. The results showed that all preparations inhibited X. oryzae, with ethanol extract producing the strongest activity (10.65–15.94 mm), followed by infusion (3.09–6.31 mm) and fresh-juice (1.50–4.85 mm), compared to chloramphenicol (21.40 mm). Phytochemical tests confirmed flavonoids, tannins, and saponins in all extracts. In conclusion, E. hirta demonstrates antibacterial potential against  , with ethanol extract being the most effective.

Abstract

Treating leaf blight in rice, which is caused by the bacterium Xanthomonas oryzae, using anti-bacterial agents from synthetic chemical compounds often causes environmental problems, so it is necessary to study the use of natural anti-bacterial agents. The pecut kuda plant (Stachytarpheta jamaicensis) has the potential to contain anti-bacterial properties because the leaves are often used by the public as a medicine to heal fresh wounds. This research aims to determine the anti-bacterial power of the S. jamaicensis plant against X. oryzae bacteria. S. jamaicensis leaves were extracted in stages using a series of successive solvents, namely hexane, DCM, and ethanol. Each fraction of S. jamaicensis leaf extract was tested for its inhibitory power against the growth of X. oryzae on MHA medium using the well method. The inhibitory variable observed was the diameter of the clear zone formed during 5 x 24 hours of incubation. The clear zone diameter data was analyzed to determine the inhibitory power. The results showed that each fraction of S. jamaicensis leaf extract had inhibitory power against X. oryzae. The S. jamaicensis plant can be developed as a source of natural anti-bacterial which are bacteriostatic, especially against X. oryzae.

Abstract

Kembang Kuning resort is one of the Mount Rinjani National Park Resorts that have high biodiversity, so it has the potential for plant growth-promoting bacteria. Bacillus sp. is one species of bacteria that has great potential as a plant growth-promoting bacteria. The purpose of this study was to isolate and determine the ability of phosphate solubilization, nitrogen-fixing and anti-fungal activity of Bacillus spp. isolated from the Primary Forest of Kembang Kuning Resort, Gunung Rinjani National Park. This research is an exploratory study consisting of the isolation of Bacillus sp., phosphate solubilization test with spot inoculation method on Pikovskaya Agar media, qualitative nitrogen fixing test using Jensen's Nitrogen - Free media, and anti-fungal activity test against Fusarium sp. with dual culture method. There were 21 isolates of the genus Bacillus successfully isolated in this study. The phosphate solubilization test showed that 16 bacterial isolates could solubilize phosphate, where the highest phosphate solubilization index was produced by isolate P3.1 namely 0,52. A total of one isolate, namely isolate P3.1, can fix nitrogen on Jensen's Nitrogen-Free medium. In the anti-fungal test against Fusarium sp., three bacterial isolates were obtained that were able to inhibit the growth of Fusarium sp., where the highest antagonism was produced by isolate P2.8 which was 46,66%. Bacillus spp. isolated from Kembang Kuning Resort has potential as PGPB in terms of crop protection.
