Abstract

Prepared slide of plant tissue needs to be staining to facilitate observations under microscope. Laboratorium activities in schools usually use synthetic dyes which expensive and can be damaged the student. Therefore the exploration of alternative dyes need to be established, such as utilizing of red dragon fruit (Hylocereus castaricensis Britt. Et R.). This study aims to (1) find out the best concentration of dragon fruit peel extract for staining plant tissue prepared slide and (2) to develop the practical guide related to plant tissue observation. The qualitative research used different concentration  of red dragon fruit peel extract, namely: 30%, 40%, 50%, 60%, 70%, 80%, 90%, and 100% with 3 repetitions. Data were obtained from observation photos of prepared slide. The result showed that the most contrast prepared slide was used red dragon fruit extract in 60% concentration. The result use to arrange practical guide in observation of plant tissues which is validated by material expert. The validation result showed “very good” criteria (86.01%).

Abstract

α-Terpineol is a natural compound of terpenoid alcohols class. However, it can be synthesizedfrom α-pinene of turpentin content. α-Terpineol has been reported as potential anticancer agentdue to its activity on inhibition of cells growth and induction of tumor cell death. However, itsanticancer activity in HeLa cervical cancer cells line has never been studied, yet. The aim of thisstudy was to evaluate the cytotoxicity of α-terpineol and its effects to apoptosis and cell cycle. This was a quasi-experimental study with post-test only with non-equivalent control groupdesign. Cytotoxicity of á-terpineol was evaluated using MTT cell viability assay. The effect of α-terpineol on cell apoptotis was tested using acridine orange-ethidium bromide staining method, whereas its effect on cell cycle was evaluated by flowcytometry method. The results showedthat α-terpineol had cytotoxicity against HeLa cell with an IC50 value about 12.46 μg/mL.Furthermore, α-terpineol induced the HeLa with an IC50 value about 13.12 μg/mL. Cell accumulationat G1 phase during cell cycle after incubation with α-terpineol (52.78)was observed. In conclusion,α-terpineol is potential as an anticancer due to its ability to induce cell apoptosis and to inhibitthe cell cycle at G1 phase.

Abstract

Observations in this study were about variations in the combination of solutions of  fruit Basella alba Linn, tubers  of Curcuma longa Linn, leaves Dracaena angustifolia as natural alternative dyes, with variations in the order of administration of solutions that produced contrasting preparations of plant tissue preparations of spinach stems (Amaranthus spinosus L). The study aimed to determine the effect of interaction variations from the combination of  Basella alba, Linn fruit solution, tubers of Curcuma longa Linn, Dracaena angustifolia leaves on the level of clarity of plant tissue preparations, variations in the order of giving the best solution can be used as natural alternative dyes and the results of the study can be used as a medium for high school biology learning. This study was presented using qualitative descriptive data processing using 6 treatments with 3 replications with 2 negative control without using dye and positive control using safranin dye. This research was conducted at the Integrated Science Laboratory, Muhammadiyah Metro University. Based on the data analysis, the results showed that the variation in the order of administration of the solution affected the clarity of the preparations. Variations in the order of tubers of Curcuma longa Linn solution,  Basella alba, Linn fruit, Dracaena angustifolia leaves gave the best effect on the clarity of plant tissue in stems of spinach (Amaranthus spinosus L). The results of this study can be used as a medium for learning biology in the form of preserved preparations.Kata kunci: Jaringan tumbuhan, buah binahong, kunyit, daun suji, Pewarna alami

Abstract

In the environment of campus Muhammadiyah University of Metro, students care in maintaining the cleanliness of the campus can be said to be lacking. Many objects are thrown away at random, such as: candy wrappers, plastic snack foods, and various papers in the campus environment. This objectives of the research to obtain an empirical of the influence of scientific experience and environmental knowledge on environmental awareness. The research method used is ex post facto. The research was conducted in the Biology Education Department, Faculty of Teacher Training and Education, Muhammadiyah University of Metro. Implementation of research from March to July 2016. The population of this study is the students of Biology Education Departmen who take the course of Environmental Knowledge in the even semester of Academic Year 2015/2016 amounted to 124 people. Research sample is taken with proportional random sampling as much 68 people. Data analysis techniques using Two-Way Variance Analysis. Conclusion of the research are follow: 1. There is influence of scientific experience to environmental awareness, 2. There is influence of environmental knowledge to environmental awareness, and 3. There is no influence of scientific experience and environmental knowledge to environmental awareness.

Abstract

Macroalgae is one component of the ecosystem that has an important role inwaters, especially on the coast of Pisang Island. The role of macroalgae formarine biota such as being a breeding ground and providing food. The PisangIsland Government plans that Pisang Island become one of the natural touristdestinations. Surely the region will experience a transition into a touristattraction and will have an influence on the ecosystem at sea. So before thetransition occurs, macroalgae research needs to be held on Pisang Island. Aqualitative descriptive research method with a purposive sampling techniqueusing a combination of transect lines. The data obtained were analyzed withShannon Winner. The objectives of the study of macroalgae on Pisang Island are: (1)Determine the species of macroalgae in Pisang Islands. (2) Knowing the resultsof research can be used as a poster. The results of macroalgae research onPisang Island found 10 species grouped into 3 divisions named Chaetomorphacrassa, Boergesenia forbesii, Avrainvillea erecta, Valonia aegagrophillia, Ulvalactuca, Sargassum polycytum, Turbinaria ornate, Fucus sp, Padina australis,Valonia aegagrophillia, Ulva lactuca, Sargassum polycytum, Turbinariaornate, Fucus sp. The results of the study were made into posters that can beused in learning and for the public. Posters contain information such asclassification of macroalgae, morphology, habitat benefits and there areoriginal photographs of macroalgae found. The results of poster validation, from material experts and design experts get a percentage of 96% with thecriteria "Very Good" and valid for use. Validation was carried out by 2Lecturers at the University of Muhammadiyah Metro.

Abstract

Cell or tissue dyes that have been used are synthetic dyes where the chemicals are relatively expensive and carcinogenic in nature. Alternativedyes can be used are natural dyes that have a high affinity for the componentsof cells or tissues. The aim of this study was to determine the contrast level ofmuscle tissue preparations stained using solution (Beta vulgaris L) anddetermine the concentration of solution (Beta vulgaris L) which had the besteffect on the level of muscle tissue contrast. This type of research is an experimental study with qualitativedescriptive data processing, the design uses 6 treatment variations of beetrootconcentration concentration (20%, 50%, 60%, 75%, 90%, 100%) and 2treatments as a control (without coloring and eosin coloring) with 3replications. Data obtained by direct observation using a microscope anddocumented with a camera and then conducted a review by biologists. Theparameter observed was the contrast of the color of the muscle tissuepreparation. The results showed that the contrast level of (Columna livia)skeletal muscle tissue preparations stained using solution (Beta vulgaris L) wasbest at a concentration of 60% was very clear criteria and very contrasted withthe average value obtained 2, 74. The results of this study were compiled as asource of learning biology in the form of practical instructions for class XI highschool students in the structure and function of tissue in animals with anaverage percentage worth of 86%.
