Abstract

Hisbiscus surattensis L the local name “tamoenju” is utilized by ethnic Sumari (Central Sulawesi) as an antidiabetic herbal, but de nitive studies on ef cacy and safety are lacking. Therefore, this study was aimed to investigate the antidiabetic activity of ethanolic extract of the leaves and fractions using in vitro and in vivo models. Ethanol extract (EE) of the leaves of H. surattensis L was prepared by maceration. The ethanol extract was separated into three fractions by liquid/liquid extractions to yield n-hexane (HF), ethyl acetate (EAF) and water fraction (WF). Firstly, the extract and fractions were tested for its inhibitory activity on α-glucosidase in vitro. The results showed IC50 EE, HF and WF more than 5000 ppm while the IC50 fraction of EAF 3888.34 ppm. Then, oral glucose tolerance test (OGTT) of the extract/fractions were carried out in normal mice. On OGTT, the ethanol extract, EAF, and WF at doses of 150 and 300 mg/kg BW were able to inhibit increasing the level of glucose in the blood. The results showed that the extract and fraction of H. surattensis L leaves in vitro had no effect as α-glucosidase inhibitor but in vivo had the hypoglycemic effect especially on the fraction of ethyl acetate and the water fraction. Abstract

Penelitian mengenai formulasi tablet effervescentsari jahe. Penelitian ini bertujuan untuk mengetahui pengaruh penggunaan variasi konsentrasi sumber asam dan basa terhadap mutu fisik tablet.  Metode yang digunakan yaitu metode granulasi basah. Empat variasi formula masing-masing dengan konsentrasi asam basa 50%, 60%, 65% dan 70%. Evaluasi mutu fisik granul meliputi ujikadar air, uji kandungan lembab, uji kompresibilitas, uji waktu alir dan sudut diam. Evaluasi tablet pada hari 1 dan 21 untuk mengetahui stabilitas mutu fisik tablet yang meliputi uji keseragaman bobot, uji kekerasan tablet, uji kerapuhan tablet dan uji waktu larut. Data yang diperoleh dibandingkan dengan Farmakope Indonesia dan literatur lain lalu dianalisis dengan uji oneway ANOVA dan uji t-student. Hasil yang diperoleh menunjukkan penggunaan variasi konsentrasi asam basa tidak berpengaruh terhadap stabilitas mutu fisik tablet yang meliputi keseragaman bobot, kekerasan, kerapuhan tetapi berpengaruh terhadap waktu larut  tablet effervescent jahe. Konsentrasi asam basa yang memenuhi mutu fisik tablet yang optimum yaitu formula 4 dengan konsentrasi asam basa 70% (asam sitrat 8,58%, asam tartrat 27,17% dan  natrium bikarbonat 34,45%).

Abstract

Gedi hijau (Abelmoschus manihot (L.)Medik ) mengandung flavonoid, alkaloid, tannin, dan saponin yang mampu melindungi mukosa lambung. Penelitian ini bertujuan untuk mengetahui aktivitas gastroprotektif serta untuk mengetahui dosis efektif ekstrak etanol daun gedi hijau dalam pemberian efek gastroprotektif terhadap tikus putih jantan yang diinduksi dengan aspirin. Ekstrak diperoleh dengan cara maserasi menggunakan pelarut etanol 96%. Sampel terdiri dari 24 ekor tikus putih jantan (Rattus norvegicus L.) dibagi dalam 6 kelompok. Perlakuan diberi per oral selama 3 hari meliputi: Kelompok kontrol normal diberi 0,5% Na-CMC; kelompok kontrol negatif diberi 0,5% Na-CMC + Aspirin 600 mg/kg BB; kelompok kontrol positif diberi suspensi Sukralfat 360 mg/kg BB + Aspirin 600 mg/kg BB; kelompok dosis uji 1 diberi ekstrak etanol daun gedi hijau 100 mg/kg BB + Aspirin 600 mg/kg BB; kelompok dosis uji 2 diberi ekstrak etanol daun gedi hijau 200 mg/kg BB + Aspirin 600 mg/kg BB; kelompok dosis uji 3 diberi ekstrak etanol daun gedi hijau 300 mg/kg BB + Aspirin 600 mg/kg BB. Pada hari ke 3, tikus dibedah 4 jam setelah pemberian aspirin, diambil organ lambung untuk dilakukan perhitungan indeks ulser dan persen inhibisi. Data dianalisis menggunakan uji statistik Kruskal Wallis dan uji statistik Mann Whitney dengan ? = 0,05. Hasil penelitian menunjukkan bahwa ekstrak daun gedi hijaudapat mencegah kerusakan mukosa lambung tikus yang dipapar aspirin dengan dosis efektif adalah 300 mg/kg BB yang sebanding dengan sukralfat

Abstract

Hantap (Sterculia coccinea Jack) merupakan salah satu tumbuhan di Sulawesi Tengah yang mempunyai potensi sebagai antioksidan sehingga dapat dimanfaatkan untuk mengobati berbagai macam penyakit. Penelitian ini bertujuan untuk menentukan nilai IC50 ekstrak etanol daun hantap dengan metode DPPH dan LC50 dengan metode BSLT serta mengetahui kandungan kimianya.Ekstrak dibuat dengan metode maserasi menggunakan pelarut etanol 96%.Penentuan kandungan kimia dilakukan dengan menggunakan pereaksi identifikasi dankromatografi lapis tipis (KLT).Uji aktivitas antioksidan ekstrak etanol daun hantap diperoleh nilai IC50 sebesar 6,48 ppm dan vitamin C (kontrol positif) 2,71 ppm, sedangkan aktivitas sitotoksiknya dihasilkan nilai LC50 sebesar 591,561 ppm. Hasil identifikasi dengan pereaksi identifikasi dan KLT menunjukkan ekstrak mengandung flavonoid, fenol, saponin, steroid, alkaloid, dan tanin. Berdasarkan hasil uji, ekstrak etanol daun hantap dapat dikatakan memiliki  aktivitas antioksi dan yang sangat kuat, sedangkan aktivitas sitotoksiknya masuk kategori sedang.

Abstract

Eleutherine palmifolia L. Merr having antioxidant activity, its because of contain flavonoid, fenolat and tanin. These compounds have effect in  preventing of free radical which cause premature aging. Research on ethanol extract of E. palmifolia L. Merr which formulated into antioxidant cream have been done by applying variation of TEA emulgator and stearic acid. That variation  were F1 (2% : 6%), F2 (3% : 12%) and F3 (4% : 18%). Determination of antioxidant activity  was done based on DPPH methode, and the Value is 98,210%. Furthermore, evaluation on  physical characteristics was done based on organoleptic test, homogenity, pH, viscosity and stability of cream preparation. Observation was done during 21 days. Obtained data were then analyze by student-t test in comparing of the value between fresh and 21 days after storage. the pH of cream F1, F2 and F3 were not showed significant decrease, while DPPH reduce percent of F1, F2 and F3 were showed significant decrease.  In contrast, viscosity of cream F3 was significant decrease. Antioxydant activity extract based on DPPH reduce percent value of F1, F2, F3 at the first day were 96,822%, 97,123% and 96,896%, respectively. Whereas that value  at 21nd days after storage  were 89,036%, 89,726% and 90,564%, respectively.  Our result showed that E. palmifolia L. Merr ethanol extract  can be formulated as a cream preparations. F2 physical properties was  fullfill standart quality, which it was used TEA emulgator and stearic acid proportion of 3% : 12%.

Abstract

This research is about anti inflammatory activity of cactus fruits (Opuntia elatior Mill.) extract gel in rats (Rattus norvegicus L.) at induced lamda carragenan in purpose to determine anti inflammatory activity, and concentration of cactus fruits extract in the gel that have an anti inflammatory activity which is equal to sodium diclofenac 1% activity, and to determine the effect of variations in the concentration of cactus fruit extract (Opuntia elatior Mill) on the physical stability of the gel. Extract was made by remaseration method using 70% ethanol. Physical stability test was conducted based on organoleptic, pH, viscosity and homogeneity of the gel. Test on Antiinflammatory activity were divided into 5 groups of treatment. Each rats induced by using lambda carrageenan. The first group of rats was given a gel without active ingredient as a negative control, groups of 2nd, 3rd and 4th rats were given gel cactus fruit extract in concentration of 5%, 10% and 15 % respectively, and 5th groups of rats were given diclofenac sodium gel as a positive control. Diameter and volume of inflammation were measured during 6 hours with 30 minute interval. Colected data were statistically analyzed using ANOVA. The results showed that concentration of cactus fruit extract in proportion of 5%, 10% and 15% in the gel preparation showed different conditions on organoleptic observations and homogeneity even though there was no different in the pH and viscosity during 14 days of storage. The results on anti-inflammatory activity testusing concentration of 5%, 10% and 15% have a recuperation effect on inflammation Gel contining 10% of extract has anti inflammatory activity which equal to 1% of sodium diclofenac activityi.e. the percentage of diameter and volume of  inflammation were 63,75% and 20,93%.

Abstract

Telah dilakukan penelitian mengenai formulasi tablet antioksidan ekstrak bawang hutan (Eleutherine bulbosa mill.urb) dengan tujuan untuk mengetahui pengaruh penggunaan variasi konsentrasi ekstrak bawang hutan pada mutu fisik dan aktivitas antioksidan tablet, mengetahui konsentrasi ekstrak yang memenuhi mutu fisik yang paling optimum dan mengetahui aktivitas antioksidan (IC50) dari tiap formula. Ekstrak dibuat dengan metode maserasi menggunakan pelarut etanol 96%. Proses pembuatan tablet dengan metode granulasi basah. Dilakukan uji mutu fisik granul dan uji mutu fisik tablet. Uji aktivitas antioksidan dilakukan secara in-vitro menggunakan metode DPPH pada formula 1, formula 2, formula 3 dan kontrol negatif dengan penggunaan Vitamin C sebagai kontrol positif. Data hasil pengukuran dianalisis secara statistik menggunakan metode ANOVA. Hasil penelitian menunjukkan bahwa penggunaan variasi konsentrasi ekstrak bawang hutan dengan konsentrasi 3,57%, 7,14% dan 10,71% pada sediaan tablet memperoleh mutu fisik tablet yang baik pada penyimpanan selama ± 1 bulan. Hasil uji aktivitas antioksidan dengan IC50 yang diperoleh pada hari ke-1 dan hari ke-28 kontrol negatif, formula 1, formula 2, dan formula 3 berturut-turut adalah 2110 ppm dan 2210 ppm; 126,18 ppm dan 135,73 ppm; 91,00 ppm dan 95,45 ppm; 46,87 ppm dan 50,93 ppm; dibandingkan dengan IC50 Vitamin C sebesar 4,93 ppm. Formula 1 mempunyai mutu fisik tablet yang paling optimum. Aktivitas antioksidan dari tiap formula masuk dalam kategori dari sangat kuat sampai sedang.

Abstract

Tomato (Solanum lycopersicum L.) represent an edible fruit which is easily available and known as a source of antioxidant. This research was aimed to test the antioxidant activity of tomato extract in olive oil and water. Antioxidant activity was determined using superoxide dismutase (SOD) and lipid peroxidation (LPO) activity test, ex vivo in Swiss Webster male mice. Result showed that olive oil tomato extract increased significantly (p<0.01) superoxide dismutase (SOD) activity and inhibited lipid peroxidation. The highest activity against superoxide dismutase (SOD) activity (0.85%) and inhibited lipid peroxidation in liver (74.67 %) was given by tomato extract in olive oil at 0.075 mL/kg bw (p<0.01). The highest inhibition of lipid peroxidation in kidney was given by tomato extract in water at dose of 150 mg/kg bw (20.27 %, p<0.01).

Abstract

Prolonged use of nonsteroidal anti-inflammatory drugs (NSAIDs) can trigger ulcers and bleed in the gastric mucosa. Pepolo stem bark (Bischofia javanica Blume) was a traditional plant that has been empirically used to treat gastric ulcers. This plant contains secondary metabolites such as alkaloids, flavonoids, tannins, saponins, and triterpenoids with gastroprotective potential. This study aims to determine the gastroprotective effect of pepolo stem bark extract (PSBE) against gastric ulcers of Wistar rat induced by Aspirin. The study used 24 rats grouped into six groups. Aspirin (150 mg/kg BW) was administered orally to all the groups except normal control. After 4 h of induction, normal and negative control received Na CMC 0.5%, while the positive group received Omeprazole (3.6 mg/kg BW). The remaining low, middle, and high dose groups received PSBE (100, 200, and 300 mg/kg BW). The treatment was given orally for eight days. On the 9th day, the rats were dissected and tested on the stomach organs. The parameters measured were scores based on the severity of rat peptic ulcers, ulceration index, and percentage protection ratio. The scoring data were analyzed using the Kruskal-Wallis non-parametric test and post hoc the Mann-Whitney test. The administration of PSBE showed the ability to reduce the ulcer index and increase the percentage of ulcer inhibition significantly compared to the negative control (p<0.05). This study showed that PSBE could be a promising gastroprotective herbal medicine.Key words: Aspirin, pepolo stem bark, gastric ulcer
