Abstract

Musa acuminata cultivar pisang ambon lumut is a native climacteric fruit from Indonesia. Climacteric fruit ripening process is triggered by the gaseous plant hormone ethylene. The rate limiting enzyme involved in ethylene biosynthesis is ACC synthase (ACS) which is encoded by ACS gene family. The objective of this study is to identify MA-ACS gene family in M. acuminata cultivar pisang ambon lumut and to study the MA-ACS1 gene expression. The result showed that there were nine M. acuminata ACS gene family members called MA-ACS1–9. Two of them (MA-ACS1 and MA-ACS2) were assessed using reverse transcriptase PCR (RT-PCR) for gene expression study and it was only MA-ACS1 correlated with fruit ripening. The MA-ACS1 gene fragment has been successfully isolated and characterized and it has three introns, four exons, and one stop codon. It also shows highest homology with MACS1 gene from M. acuminata cultivar Hsian Jien Chiao (GenBank accession number AF056164). Expression analysis of MA-ACS1 using quantitative PCR (qPCR) showed that MA-ACS1 gene expression increased significantly in the third day, reached maximum at the fifth day, and then decreased in the seventh day after harvesting. The qPCR expression analysis result correlated with the result of physical analysis during fruit ripening. Abstract

Fusarium wilt which is caused by the soil-borne fungus, Fusarium oxysporum f.sp. cubense (Foc), is one of important diseases in banana plant. Foc tropical race 4 (Foc4) is the most pathogenic race which could infect various banana cultivars including Cavendish cultivar which was previously considered as resistant cultivar. Molecular detection of Foc using PCR analysis is indispensable to determine the race of Foc. We demonstrate a faster DNA isolation procedure described in previous method by substituting sodium acetate precipitation with ammonium acetate precipitation without affecting the result. Based on our experience, some fungal genomes were troublesome to be amplified. We suggested pre-treatment step prior to amplification procedure by incubating fungal DNA in 65°C for 10 minutes for any samples of fungal genome, including stubborn samples, before mixing into PCR mix reagent. PCR reagents should be tested for stubborn samples since some of the reagents were unable to amplify the desired DNA fragment. Pre-treatment and the choice of robust PCR reagent should be taken into consideration for a reliable and consistent Foc4 molecular detection result.

Abstract

Waste disposal and clean water management are some of the common social issues arising in densely populated settlements, including Muara Baru flats, Jakarta. Some programs have been carried out to overcome these two issues in Muara Baru, such as the construction of a Rain Water Harvesting (RWH) facility and the provision of Trash Banks. Nevertheless, adequate support for the residents is still needed to boost their awareness of proper clean water management and waste disposal and to encourage the formation of urban agriculture, which is expected to elevate their living standards. Therefore, a community empowerment program in Muara Baru was developed, consisting of three sections: education on clean water management, compost making training, and urban farming. The success of this program was then measured using questionnaires and focus group discussions. A majority of residents responded positively to the program and were willing to actively participate by preparing the land, making compost, planting seeds, or caring for the harvest. There were indeed some obstacles hampering the program's success, such as lack of commitment from the local community, climate change, water availability, and arable land availability. To overcome these barriers and to promote the program’s sustainability, some suggestions are to be proposed, including proper selections of plant types and provision of support and motivation for the local community.

Abstract

COVID-19, a source of distress in most countries, has encouraged world leaders to issue necessary policies to resolve this global pandemic. An alarming rate of infections in the community has pushed the Indonesian government to come up with the Large-Scale Social Restriction (Pembatasan Sosial Berskala Besar/PSBB). The faculty of Biotechnology of Atma Jaya Catholic University of Indonesia has since initiated a community service to halt the spread of COVID-19 by supplying simple hand washing facilities around Pasar Genteng Traditional market in Suradita, Cisauk. Prior to the installations, the traditional market lacked proper sanitizing facilities. Clean water sources such as taps are commonly situated far away from the marketplace. Traders and visitors crowd these areas each day, exponentially increasing the risk of spreading COVID-19. The crowded populations of the traditional market are a motivating factor for this community service, which aims to increase the public awareness in the prevention of COVID-19. Activities of this community service includes installations of hand washing facilities along with educational infographics of proper sanitizing procedures. A total of 21 participants attended the community service while maintaining physical distancing conditions and were encouraged to use the hand wash facilities at the end of the service. The community service was successful, providing impactful benefits towards the traders and visitors, with a raised community awareness of the importance of personal hygiene. The facilities are well maintained after 4 weeks and exemplary community awareness is displayed from their initiative that showed from the monitoring and evaluation results.  ABSTRAK:COVID-19, yang merupakan sumber penyebab keresahan berbagai negara di dunia telah menyebabkan pemerintah dunia mengeluarkan kebijakan untuk mengatasi pandemi yang terjadi secara global ini. Data di Indonesia menunjukkan angka terinfeksi yang terus meningkat sehingga pemerintah mengeluarkan kebijakan berupa Pembatasan Sosial Berskala Besar (PSBB). Oleh karena itu, Fakultas Teknobiologi Universitas Katolik Indonesia Atma Jaya berinisiatif melakukan kegiatan pengabdian masyarakat untuk menghambat tingkat penyebaran COVID-19 melalui penyediaan sarana cuci tangan sederhana di Pasar Genteng. Pasar tradisional ini terletak di kompleks Suradita, Cisauk. Sebelum kegiatan pemasangan, pasar ini kekurangan fasilitas sanitasi yang layak. Sumber air bersih seperti air sumur terletak jauh dari pasar dan belum ada akses air ledeng. Setiap hari pedagang dan pengunjung memadati pasar sehingga meningkatkan risiko penyebaran COVID-19. Oleh karena itu, kegiatan pengabdian masyarakat ini difokuskan pada pembuatan dan pemasangan fasilitas cuci tangan sederhana yang dilengkapi dengan infografis tentang cara mencuci tangan yang benar serta pembagian masker kain. Serah terima perangkat cuci tangan dan acara pembukaan dihadiri oleh 21 orang peserta dengan tetap memperhatikan protokol kesehatan. Kegiatan pengabdian masyarakat melalui penyediaan sarana cuci tangan sederhana di Pasar Genteng, Cisauk telah berlangsung dengan baik dan dirasakan manfaatnya oleh para pedagang dan pengunjung pasar. Sarana cuci tangan sederhana ini diharapkan dapat meningkatkan kesadaran masyarakat tentang pentingnya menjaga kebersihan diri dalam rangka pencegahan penyebaran COVID-19. Sarana cuci tangan tersebut masih dalam kondisi baik setelah empat minggu dan kesadaran masyarakat juga cukup baik yang terlihat dari hasil monitoring dan evaluasi.

Abstract

 AbstractAuricularia auricula is a wood mushroom that is commonly consumed and is in demand by the people as a food ingredient because it contains many nutrients and health benefits. Cultivation of A. auricula was carried out using growing media and controlled environmental conditions. This study objective to determine the optimal combination of composting time and the rice husks addition in the A. auricula production. The treatment in this study was the composting time of the growing media for 0, 2, 4, 6, and 8 days, as well as the addition of rice husks as much as 0, 25, and 50%. This research consisted of the preparation of growing media, spawning and incubation of A. auricula, and harvesting of fruit bodies. The success of mushroom cultivation is seen from the value of biological efficiency in each treatment. The combination treatment of 8 days composting time with the addition of 50% rice husk resulted in A. auricula production as much as 52.37%, while the combination of 4 days composting time treatment and the addition of 25% rice husks resulted in the best production of A. auricula, as it produced the mushroom weight of 340 ± 16.95 g with biological efficiency value as much as 68% for five harvest durations within four months after first watering. The treatment of composting media tie and the addition of rice husks to A. auricularia’s were successfully carried out.AbstrakAuricularia auricula merupakan jamur kayu yang umum dikonsumsi dan diminati oleh masyarakat sebagai bahan pangan karena mengandung nutrisi dan banyak manfaat. Budi daya A. auricula dilakukan dengan menggunakan media tanam dalam kondisi lingkungan yang terkontrol. Penelitian ini bertujuan menentukan kombinasi perlakuan waktu pengomposan dan penambahan sekam padi yang optimum terhadap produksi A. auricula. Perlakuan pada penelitian ini adalah waktu pengomposan media tanam selama 0, 2, 4, 6, dan 8 hari, serta penambahan sekam padi sebanyak 0, 25, dan 50%. Penelitian ini tersusun atas pembuatan media tanam, pembibitan dan inkubasi A. auricula, dan pemanenan tubuh buah. Keberhasilan budi daya jamur dilihat dari nilai efisiensi biologi pada setiap perlakuan. Kombinasi perlakuan pengomposan 8 hari dengan penambahan sekam padi 50% menghasilkan produksi A. auricula sebesar 52,37%, sedangkan kombinasi perlakuan waktu pengomposan 4 hari dan penambahan sekam padi 25% menghasilkan produksi A. auricula terbaik karena menghasilkan berat jamur sebanyak 340 ± 16,95 g dengan nilai efisiensi biologi sebesar 68% selama lima durasi panen dengan waktu empat bulan setelah dilakukan penyiraman pertama. Secara keseluruhan, perlakuan waktu pengomposan media dan penambahan sekam padi pada media pertumbuhan A. auricula berhasil dilakukan.

Abstract

Abstract— Andaliman (Zanthoxylum acanthopodium DC.) is one of the Indonesian spices that is not easy to be cultivated outside its natural habitat because it requires lower temperatures for its growth. This research aimed to determine the culture technique of andaliman plants using in vitro tissue culture techniques to solve the difficulties of plant cultivation outside its natural habitat. The explants used are seeds and nodal segments. Using seeds, methods were divided into seeds collection and sterilization and initiation. Methods using nodal segments explants were divided into sample collection, nodal sterilization, initiation, and shoot multiplication. Seed sterilization using 60 °C water which was replaced every six hours had lower contamination rate compared to other methods. Unfortunately, seeds germination and dormancy breaking were not exhibited in this research. Nodal segment sterilization using 0,1% (b/v) mercury chloride (HgCl2) exposed for seven minutes and addition of 100 IU mL-1 nystatin showed fewer contaminations. Multiplication using Murashige-Skoog (MS) media with addition of 1 ppm NAA and 5 ppm BAP (MSN1B5) produced nine shoots and produced callus. Overall, both explant sterilizations were successful, but it was recommended to use nodal segments as the explant due to success in producing andaliman plantlet in vitro. Keywords: andaliman, HgCl2, nystatin, plant tissue culture, woody plant medium Abstrak— Andaliman (Zanthoxylum acanthopodium DC.) adalah salah satu rempah Indonesia yang sulit untuk dibudidayakan di luar habitat aslinya karena membutuhkan suhu rendah untuk pertumbuhannya. Penelitian ini bertujuan untuk menentukan teknik perbanyakan tanaman andaliman menggunakan teknik kultur jaringan secara in vitro dengan memperhatikan beberapa faktor seperti larutan sterilisasi yang digunakan dan zat pengatur tumbuh (ZPT) yang diberikan. Eksplan yang digunakan adalah biji dan nodus. Metode penelitian dengan eksplan biji meliputi koleksi buah, germinasi, dan inisiasi eksplan. Metode penelitian dengan eksplan nodus dibagi menjadi koleksi bibit, inisiasi nodus, dan multiplikasi nodus. Sterilisasi permukaan biji menggunakan air hangat bersuhu 60 °C yang diganti setiap enam jam memiliki tingkat kontaminasi yang paling rendah dibandingkan dengan metode lainnya, tetapi germinasi dan pemecahan dormansi tidak terjadi pada penelitian ini. Sterilisasi permukaan nodus menggunakan larutan merkuri klorida (HgCl2) 0,1% (b/v) dengan durasi perendaman selama tujuh menit dan 100 IU mL-1 nistatin pada media menunjukkan tingkat kontaminasi yang lebih rendah. Multiplikasi menggunakan media Murashige-Skoog dengan penambahan 1 ppm NAA dan 5 ppm BAP (MSN1B5) menghasilkan organ tunas dalam jumlah yang paling banyak yaitu sebanyak sembilan tunas dan terbentuk kalus. Secara keseluruhan, sterilisasi kedua eksplan berhasil dilakukan, tetapi lebih disarankan menggunakan eksplan nodus karena dapat menghasilkan tunas andaliman secara in vitro. Abstract

Hidroponik dapat menjadi salah satu sarana pembelajaran tambahan bagi para siwa/i untuk mencapai pembangunan berkelanjutan/sustainable development goals (SDGs). Dengan adanya kegiatan pengabdian kepada siswa/i SMP Athalia yang dilakukan oleh tim dosen Fakultas Teknobiologi bersama mahasiswa, perangkat hidroponik yang tadinya tidak dapat dimanfaatkan karena tanaman terkena air hujan sehingga menyebabkan tanaman merunduk dan minimnya pengetahuan terkait hidroponik dapat teratasi. Pelatihan dilaksanakan secara luring yang meliputi teori dan praktik teknik hidroponik yang diharapkan dapat memaksimalkan pemanfaatan fasilitas hidroponik yang telah ada. Hasil monitor selama empat minggu dengan menggunakan tanaman sawi hijau, bayam merah, dan sawi pagoda dengan parameter tinggi daun, jumlah daun, dan bobot basah tanaman menghasilkan bayam merah memiliki pertumbuhan terbaik dengan bobot basah sebesar 17,26 gram. Evaluasi dan kuesioner kegiatan dilakukan dalam bentuk Microsoft Forms. Evaluasi pengetahuan serta pemahaman dari para siswa/i menunjukkan perubahan signifikan yang dapat dilihat dari peningkatan jumlah jawaban benar para siswa/i yang sebelumnya kurang tepat menjadi hampir keseluruhan siswa/i benar. Keseluruhan kegiatan workshop sudah berlangsung dengan cukup baik dan memadai, baik dalam hal perlengkapan, kesiapan, persiapan, maupun dari segi panitia.

Abstract

Soil microorganisms, especially diazotrophic bacteria, are vital for ecosystem sustainability, significantly contributing to nitrogen cycling and biodiversity conservation. Understanding the impact of different land-use practices on soil microorganisms, especially synthetic fertilizer addition, is essential for sustainable agriculture. This study compares soil properties, bacterial densities, and responses to fertilization in secondary forest soils and adjacent oil palm plantation soils located in PT Kerry Sawit Indonesia, Central Kalimantan. A greenhouse experiment was conducted using both secondary forest and oil palm plantation soils to evaluate the impacts of different synthetic fertilizers on nitrogen-fixing bacteria and soybean agronomic performance. Total soil bacteria and diazotrophic bacteria, essential for nitrogen fixation, were analyzed through microbiological assays and qPCR focusing on the nifH gene. Our findings show that oil palm plantation soils had higher levels of nitrogen, phosphate, and nitrate, while secondary forest soils had a higher abundance of diazotrophic bacteria. Furthermore, excessive nitrogen fertilization was found to reduce microbial density, negatively impacting plant sustainability, highlighting the importance of customized fertilizer management. The study highlights the intricate connections between land-use practices and microbial populations, providing valuable insights for balancing agricultural productivity with ecological sustainability.

Abstract

The Villa Ciomas RW 10 Farmer Group (PokTan Vici 10), actively engaged in urban farming in Ciomas Rahayu Village, Bogor, West Java, has been utilizing a small plot of land located next to the Cisindangbarang River. Their urban farming area already includes a small fishpond but lacks aquaponics equipment. The community service team aims to enhance the utility of the urban farming facilities by installing aquaponics equipment, providing workshops on basic hydroponics and aquaponics knowledge, exploring its business potential, and offering hands-on hydroponics and aquaponics cultivation practice. Throughout the program, the team also conducts monitoring and guidance to ensure successful implementation and address any challenges encountered during the training program. The hydroponic/aquaponics equipment installation was successful; however, nutrient supply from the fishponds only was initially suboptimal, prompting the addition of hydroponic nutrients. Hydroponic techniques were also introduced for vegetable cultivation. Based on a questionnaire analysis conducted before and after the training, the program positively impacted participants, significantly enhancing their knowledge and aquaponic skills (Wilcoxon signed-rank test, p<0.05). In conclusion, the aquaponics equipment was successfully installed, and participants gained foundational knowledge and skills in aquaponics, enabling them to independently manage aquaponics systems through to harvest. Looking ahead, the community members plan to explore business opportunities in selling hydroponics/aquaponics products.

Abstract

Variety development in ornamental plants increased rapidly due to the high demand during the COVID-19 outbreak. Therefore, analyzing the genetic diversity of ornamental plants is essential to assist breeders in improving and developing new cultivars with desirable traits. In this study, the genetic diversity of Philodendron species was analyzed. P. erubescens, P. 69686, P. burle-marx, P. lacerum, and P. micans were selected as the most popular species cultivated in Indonesian nurseries. These cultivars were assessed both genetically using Random Amplified Polymorphic DNA (RAPD) markers and morphologically. The genetic relationship among Philodendron spp. species was analyzed using DARwin 6.0 software, and a dendrogram was constructed based on the UPGMA algorithm. Based on the two selected RAPD primers, the five samples showed no clear genetic relatedness to each other, and the primers were insufficient to construct a single consensus dendrogram.
