Abstract

Abstract. Diltiazem Hydrochloride (HCl) is commonly used in hypertension therapy for its beneficial effects, such as reducing blood vessel pressure resistance and relaxing the heart muscles. In an in-vivo study, Diltiazem HCl is known to be decrease by 60% of its bioavailability due to first-pass metabolism. Thus, it is necessary to develop a new administration route, some of which is buccal drug delivery route in mucoadhesive buccal film. Moringa gum is a polyuronide polymer with a hydroxyl group in its side chains, giving the potential use as a mucoadhesive polymer. However, it has poor folding endurance and its release kinetics study has not been investigated. Polyvinyl alcohol (PVA) is a water-soluble polymer with hydroxyl side chain. PVA is known for its high tensile strength and flexibility, though it can be affected by humidity. The aim of this research is to determine the right amount of moringa gum and PVA to obtain a film with good physical characteristics and mucoadhesive residence time. In this study, the optimization of moringa gum and PVA was carried out using factorial design. Mucoadhesive buccal film was prepared by solvent casting method, the films were later evaluated and the obtained responses were analyzed by Software Design Expert 11.0.0. The optimization result shows the film containing the amount of moringa gum 2% and PVA 5% can produce the optimum formula. The mucoadhesive buccal film formula has surface pH of 6.083, mucoadhesive residence time of 445.055 min, swelling index of 4.067, and 87.9913% drug release in in-vitro drug release study after 420 min following the Higuchi model release kinetics. Therefore, it can be concluded that the obtained optimum formula of mucoadhesive buccal film can be used in the preparation of mucoadhesive buccal film Diltiazem HCl.. Abstract

The liquisolid technique is one way to increase the drug dissolution rate, which has low solubility in water and can improve drug flow properties. This research aims to determine the optimum composition of a combination of polymer PVP K-30 and SSG on a liquisolid simvastatin tablet with a factorial design. The factorial design method is used to determine the effect of the concentration of two factors, namely PVP K-30 and SSG, on the response of hardness, friability, disintegration time, and dissolution. PVP K-30 acts as a binder, and SSG works as a disintegration, determines the optimum formula using expert design software version 1. The result showed that all formulas meet the requirements of hardness values (4-8 kg), friability (<1%), disintegration time of no more than 15 minutes, and dissolution in 30 minutes. The use of PVP K-30 concentration of 6 mg and SSG 24 mg indicated the optimum formula with a desirability value of 0,806.

Abstract

Antioxidants can prevent premature skin aging caused by free radicals. Cocoa pod husk extract can be used as an antioxidant in the form of a peel-off gel mask. The peel-off gel mask forms a thin, occlusive film that supports antioxidant absorption in a short time. The optimal concentration of gelling agent is necessary to make a good peel-off gel mask. The aim of this study is to determine the optimal concentrations of xanthan gum and HPMC in the preparation of a cocoa pod husk extract peel-off gel mask. The optimization was performed using the Simplex Lattice Design (SLD) method with two factors, which is the concentration of xanthan gum and HPMC, the optimum formula was tested for antioxidant activity using the DPPH method. The result indicated that the use of xanthan gum and HPMC had an impact on changes in formulation viscosity, pH, adhesion, and peeling time. The optimum formula consisted of 0.945% xanthan gum and 0.555% HPMC which was predicted to have a viscosity of 62.754 dPa.s, a pH of 5.091, an adhesion of 165.922 seconds, and a peeling time of 1765.391 seconds. Its antioxidant activity is classified as moderate with an IC50 value of 143.664 ppm ± 1.348. Keywords: peel-off gel mask; Theobroma cacao L.; gelling agent Abstrak Antioksidan dapat mencegah penuaan dini yang disebabkan radikal bebas. Ekstrak kulit buah kakao dapat dimanfaatkan sebagai antioksidan dengan pembawa berupa masker gel peel-off. Masker gel peel-off akan membentuk lapisan film tipis yang bersifat oklusif sehingga membantu penyerapan antioksidan dalam waktu singkat. Konsentrasi gelling agent yang optimum diperlukan untuk membuat masker gel peel-off yang baik. Penelitian ini dilakukan untuk mengetahui konsentrasi optimum gelling agent xanthan gum dan HPMC dalam masker gel peel-off ekstrak kulit buah kakao. Optimasi dilakukan dengan metode Simplex Lattice Design (SLD) menggunakan dua faktor, yaitu konsentrasi xanthan gum dan HPMC, formula optimum diuji aktivitas antioksidannya menggunakan metode DPPH. Hasil analisis SLD menunjukkan penggunaan xanthan gum dan HPMC berpengaruh terhadap perubahan viskositas, pH, daya lekat, dan peeling time sediaan. Formula optimum yang terpilih terdiri dari 0,945% xanthan gum dan 0,555% HPMC. Prediksi nilai respon viskositas sebesar 62,754 dPa.s, pH sebesar 5,091, daya lekat sebesar 165,922 detik, dan peeling time sebesar 1765,391 detik. Hasil uji antioksidan menunjukkan aktivitas yang tergolong sedang dengan nilai IC50 sebesar 143,664 ppm ± 1,348. Kata Kunci: masker gel peel-off, Theobroma cacao L., gelling agent

Abstract

Excessive exposure to ultraviolet radiation can cause various skin problems. This problem can be prevented by using cosmetic products that contain sunscreen ingredients. In this study, optimization of the combination of quercetin and avobenzone in sunscreen spray lotion was carried out using the factorial design method. The aim of this study was to determine the optimum concentrations of avobenzone and quercetin which could provide optimum physicochemical properties and effectiveness of in vitro sunscreen spray lotion. The formula is made with a low concentration of 0.5% quercetin and an upper limit of 3%, while avobenzone has a low level of 2% and an upper level of 4%. Based on the research that has been done, the optimum formula prediction is obtained with a concentration of 0.934% quercetin and 4% avobenzone. The optimum formula has a desirability value of 0.958 and shows the predicted value of software to have an SPF in vitro value of 33.311, a %TE 0.058%, a %TP 0.002%, a viscosity of 10 dPa.s, and a pH of 5.689. The results of the analysis using the t-test (one sample t-test) with a 95% level of confidence between the software prediction results and the observation results showed a value of > 0.05, which means that there was no significant difference between the software prediction results and the observation group.

Abstract

Abstract: Cetirizine dihydrochloride (Cetirizine diHCL) is an effective antihistamine to treat allergies. Cetirizin diHCL tablets are a dosage form that is commonly on the market. However, people who experience allergies will have symptoms of difficulty swallowing which make it difficult to take cetirizine diHCl tablets. Therefore, it is necessary to make cetirizine diHCl in the form of orally disintegrating tablets (ODT) which can dissolve in saliva in a few seconds without chewing tablets or drink water. The proportion of binder and super disintegran was used in ODT preparations must be taken into calculation because it will greatly affect the physical characteristics of the tablets. This study aims to optimize the super disintegran Croscarmelose Sodium and pregelatinasi corn starch binders using factorial design methods. The responses studied were flow rate, hardness, brittleness, wetting time, and disintegration time. The increased Croscarmelose Sodium factor reduces the flow rate and hardness, increases brittleness, and decreases wetting and disintegration times. The increased factor of pregelatinasi corn starch will increase the flow rate and hardness, reduce friability and increase wetting time and disintegration time. The optimum formula was obtained from the concentration of Croscarmelose Sodium 6.613 mg and pregelatinasi corn starch 20 mg which resulted in a flow rate response of 13.438 g/second, hardness 4.646 kg, friability 0.815%, wetting time 63.667 seconds, and disintegration time 18.641seconds.

Abstract

Moringa gum is a natural polymer obtained from Moringa oleifera plant and has been used as a stabilizer, binder, disintegrant, and controlled release matrix. Moringa gum has a polyuronide group, which can potentially be a mucoadhesive agent. This study used moringa gum as a mucoadhesive agent in diltiazem HCl mucoadhesive buccal film. This research aimed to determine the effect of the combination of moringa gum and HPMC on surface pH, swelling index, and in-vitro mucoadhesive residence time. Mucoadhesive buccal films were evaluated for weight and thickness uniformity, folding resistance, drug content, surface pH, swelling index, in-vitro mucoadhesive residence time, and characterization in the form of FTIR and release studies. All formulations met the tests of uniformity of weight and thickness, folding resistance, and drug content. The results showed FB as the optimum formula with a surface pH of 5.803±0.101, a swelling index of 7.031±0.134, and a residence time of 505.67±4.51 minutes. FTIR showed no interaction, and the release study showed 82.197±1.178% release at 480 minutes. In conclusion, a combination of moringa gum and HPMC mucoadhesive buccal film has been successfully prepared with moringa gum as a mucoadhesive agent that increases the residence time.
