Abstract

Penelitian ini bertujuan untuk mengkaji pengaruh penggunaan formalin dan periode simpan terhadap viabilitas dan pertumbuhan awal benih kelengkeng. Rancangan percobaan berdasarkan rancangan acak lengkap faktorial dengan 4 ulangan. Faktor pertama adalah formalin (0%, 0,5%, 1% dan 2%). Faktor kedua adalah periode simpan dengan 3 taraf (tanpa penyimpanan, 15 dan 30 hari). Parameter yang diamati adalah daya kecambah, kecepatan berkecambah, Jumlah daun dan tinggi bibit kelengkeng. Data yang diperoleh dianalisis ragam taraf 5% dan apabila berpengaruh nyata (p<0,05), dilanjutkan dengan uji jarak berganda Duncan (DMRT) pada taraf 5%. Hasil penelitian menunjukkan bahwa meningkatnya kadar formalin, nyata menurunkan daya kecambah dan kecepatan berkecambah, sedangkan kadar formalin 0,5% dengan 30 hari periode simpan optimal meningkatkan jumlah daun dan tinggi tanaman. Kata kunci : kelengkeng, formalin, periode simpan, viabilitas, pertumbuhan awal

Abstract

The method of thyme stem cutting needs to be developed to produce better roots to support growth. The study aims to examine the response of thyme stem cuttings to various concentration of IBA and time soaking. The research was carried out on January – March 2020 in screen house PT. INDMIRA, Sleman, Yogyakarta and the Physiology and Plant Breeding Laboratory of the Faculty of Animal and Agricultural Science, Diponegoro University. The method used was a completely randomized design with a factorial 4×3 with 5 replications. The first factor was a various concentration of IBA: A0 (0 ppm), A1 (100 ppm), A2 (250 ppm), and A3 (500 ppm). The second factor was the time soaking: B1 (5 minutes), B2 (10 minutes), and B3 (15 minutes.) The results showed that IBA concentration has no significant effect for root dry weight, while the time soaking has significant effect for all parameters. Soaking thyme stem cuttings at an IBA 250 ppm for 10 minutes gave the best results on plant length, the number of leaves, root length, and root dry weight, but soaking in water (without IBA) for 5 minutes was able to give the best number of percentage of life cuttings (85%) so that giving IBA had no needed.

Abstract

Silica fertilizer is applied to the soil for the purpose of reducing the effects of salinity stress. This research study was conducted to examine the effect of fertilization ratio with different sources of silica to soybean growth and production on saline soil. The methods used was Completely Randomized Factorial Designs 2 x 6 with 3 replications. The first factor was soil salinity levels (A), consists of A1 (±3 dS/m) and A2 (±6 dS/m). The second factor was different silica fertilizer aplications (B) which consists of  B0 (without silica), B1 (100% zeolite), B2 (75% zeolite, 25% liquid silica), B3 (50% zeolite, 50 % liquid silica), B4 (25% zeolite, 75% liquid silica), and B5 (100% liquid silica). Parameters observed in this research were plant height, number of leaves, number of pods, pod weight, number of seed and seed weight. Data was analyzed by analysis of variance followed by Duncan's Multiple Range Test. The result showed that the application of silica fertilizer through soil and leaves (50% zeolite, 50 % liquid silica) improved the growth and yield of soybean on soil with moderately saline (3 dS/m). application of silica fertilizer (50% zeolite, 50 % liquid silica) improved the growth and yield of soybean. The higher soil salinity reduced soybean growth and yield. Moreover, the utilization of silica minimized the decrease in soybean growth and yield.

Abstract

Maize has a wide diversity. The diversity of maize exceeds that of any other species and is used and conserved by traditional maize farmers. The way to find information about maize diversity is to evaluate the maize genotype through plant characterization. Plant characteristics include observing the qualitative and quantitative traits to identify and select appropriate parental species for use in forming or developing varieties. This study aimed to examine the diversity of agronomic traits in 12 genotypes of colored maize; Meanwhile, Bonanza (sweet maize) was used as a control. An augmented design with the basic design of a completely randomized design was used as a research experiment. From the agronomic data, cluster analysis between characters was carried out based on the Euclidean distance coefficient using the MVSP application to determine the relationship between genotypes. The observations of quantitative and qualitative traits showed that the characteristics of plant height, seed color, stem color, tassel color, and cob hair had heterogeneous diversity. However, sweetness, cob color, seed type, and seed row arrangement had homogeneous diversity in the male and female flowering age characters. Based on the dendrogram results, the twelve genotypes tested and the control showed that these genotypes were different varieties in one genus, i.e., Zea L. because they had a similarity value above 80%.

Abstract

Kentang merupakan tanaman yang mengandung tingkat karbohidrat tinggi sehingga menjadi pangan alternatif pengganti beras. Kultur jaringan menjadi teknik perbanyakan tanaman kentang yang sering digunakan karena dapat menghasilkan bibit kentang dalam jumlah banyak sekaligus. Namun, permasalahan yang sering terjadi pada kultur jaringan adalah kontaminasi oleh mikroorganisme. Penanganan kontaminasi bisa dilakukan dengan menambahkan bahan bersifat menekan pertumbuhan mikroorganisme seperti pegagan. Pegagan mengandung senyawa triterpenoid yang bersifat antimikrobial dan antifungal. Tujuan penelitian adalah untuk mengkaji efektivitas konsentrasi ekstrak daun pegagan (Centella asiatica L.) sebagai anti kontaminan pada pertumbuhan kultur jaringan kentang var. Tedjo MZ. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) faktor tunggal dengan empat kali ulangan. Faktor yang digunakan adalah konsentrasi ekstrak daun pegagan; C0: 0%, C1: 2,5%, C2:5%, C3: 7,5% dan C4: 10%. Parameter yang diamati adalah panjang akar, panjang cabang, jumlah daun, tinggi planlet, waktu pertama kontaminasi, persentase kontaminasi jamur dan bakteri serta persentase keberhasilan kultur jaringan. Hasil penelitian menunjukkan pemberian ekstrak pegagan pada berbagai konsentrasi belum memberikan dampak signifikan pada seluruh parameter. Pemberian ekstrak pegagan pada media planlet kentang terlihat menghambat pertumbuhan planlet kentang daripada perlakuan kontrol, hal ini dapat disebabkan zat senyawa yang terdapat pada ekstrak pegagan bersifat toksik bagi planlet. Perlakuan kontrol memberikan hasil terbaik pada parameter waktu pertama kontaminasi, persentase kontaminasi jamur dan bakteri, persentase keberhasilan kultur jaringan, panjang akar, panjang cabang, jumlah daun dan tinggi planlet.

Abstract

Corn is the number two stable crop in the world with increasing demand in line with population growth. The use of superior hybrid corn varieties is one of the solution in meeting the needs of corn throught plant breeding activities. The experimental design used was a monofactor randomized block design (RAK), with the object is 12 prosprective line of hybrid corn varieties from PT Tunas Widji Inti Nayottama (RD 422, RD 471, RD 491, RD 496, RD 551, RD 519, RD 523, RD 536, RD 586, RD 591, RD 602, RD 60), and control varieties is Bisi-18 and P31. The parameters observed included number of plants grown, high of plant, high of ear, lenght of ear, diameter of ear, weight of ear, weight of cob, number of seed rows pear earm number of seed per row, weight of 100 seeds, yield, seed moisture content, physiological maturity, and potential harvest. Variance analysis data and continued with the independent T test. The result showed that 7 corn lines which have the desired properties are RD 422, RD 471, RD 551, RD 519, and RD 603. The level of stability according to the coefficient of diversity, all of the test lines had low stability (uniform), except for RD 491 on the character of anxious weight (29.23%) and RD 602 on the character of the number of seeds per row (39.36%).

Abstract

Dendrobium sp. is well known as the largest genus in orchid, every region has its own characteristics. The aim of the research was to examine the interaction effect of sucrose and IBA (indole 3-butyric acid) on root initiation of Dendrobium sp. in vitro culture. This study used a completely randomized design (CRD) factorial design 3x4 with 3 replications. The first factor was sucrose concentration (S) : S1 (20 g/l), S2 (30 g/l) and S3 (40 g/l). The second factor was IBA concentration (I) : I0 (0 mg/l), I1 (1 mg/l), I2 (2 mg/l) and I3 (3 mg/l). The parameters observed were number of roots, length of root, initiation root period, height of plantlets, number of shoots and culture success percentages. Data was analysed using analysis of variance and further test using LSD (least significance different) with a 5% level. The result showed that the concentration of sucrose 40 g/l had a significant effect on number of roots, length of root, height of plantlets and number of shoots. Concentrations of IBA 2 mg/l had a significant effect on height of plantlets. Iinteraction of both sucrose and IBA had no significant effect on all of the parameters. Two objects of culture were contaminated by fungi. Suggest for this study was to increase sucrose and IBA concentrations for the best result combination

Abstract

Kegiatan ini bertujuan untuk meningkatkan keterampilan, pemahaman, serta pengetahuan Ibu-Ibu PKK dusun Semen dalam mengolah eceng gondok dan keong mas menjadi pupuk organik cair. Kegiatan dilakukan dengan dua metode yakni daring dan luring. Kegiatan yang dilaksakan secara luring meliputi observasi dan persiapan dilakukan dengan wawancara secara langsung dengan petinggi desa dan pembuatan modul serta video tutorial. Pelatihan dilakukan secara luring dengan metode demo pembuatan pupuk organik cair. Metode pendampingan dilakukan secara daring melalui grup whatsapp mengenai perkembangan fermentasi pupuk yang telah dibuat. Evaluasi kegiatan dilakukan secara daring meliputi pengisian angket menggunakan google form untuk mengetahui pemahaman Ibu-Ibu PKK pasca kegiatan. Berdasarkan pengisian kuisioner menunjukkan bahwa Ibu PKK sudah memahami cara mengolah eceng gondok dan keong mas menjadi pupuk organik cair. Setelah kegiatan pelatihan dilaksanakan, pupuk organik cair ini dapat digunakan oleh Ibu PKK harapan setelah program adalah terdapat perubahan pola perilaku Ibu PKK dalam menggunakan pupuk tanaman yang berlaih dengan pupuk buatannya sendiri.

Abstract

Red or purple yardlong bean is one type of yardlong bean that is starting to be developed but has a bitter taste due to its antioxidant content. Improvements in the quality of red or purple yardlong beans can be achieved through crossbreeding. The effective selection of crossbreeding can be seen from the diversity and selection progress. The research purpose was to assess genetic diversity and estimate the value of the selection progress of yardlong beans (Vigna unguiculata (L.)) F6 generation from a crossing between Fagiola IPB and Aura Hijau. The research was conducted at the Agrotechnopark, Diponegoro University, Semarang. The study was arranged in a completely randomized design (CRD) with an unequal number of replications. The results showed that all characters except the number of seeds per plant had low genetic diversity. The high heritability and value of genetic advances were found in antioxidant content. All characters had selection progress except for fresh harvest age and weight of 100 seeds. Partial correlation analysis showed a positive correlation between the character number of dry pods per plant with the number of leaves, the number of seeds per plant, seed weight per plant, and antioxidant content, also the character number of seeds per plant with several leaves and seed weight per plant. A negative correlation showed between the character number of leaves with seed weight per plant and antioxidant content and the character number of seeds per plant with antioxidant content.

Abstract

Teak is a wood-producing plant that is in great demand by the community. However, teak seeds are challenging to germinate due to dormancy, so special treatment is needed before planting. This research aimed to determine the effect of hydrochloric acid and soaking time on breaking the dormancy of teak seed to improve its viability. The first factor was the concentration of hydrochloric acid with four treatment levels, i.e., 0 M, 0.5 M, 1 M, and 1.5 M. The second factor was submerging time which was 12, 18, and 24 hours. Parameters observed included the percentage of germination, maximum growth potential, vigor index, germination rate, growth simultaneity, plant height, number of leaves, and wide leaf. The data obtained were analyzed statistically using ANOVA and followed the DMRT test with a level of 5%. The results showed that hydrochloric acid significantly affected the percentage of germination, maximum growth potential, vigor index, germination rate, growth simultaneity, plant height, and width of the leaf. In addition, soaking time treatment had a significant effect on the wide leaf. The research concluded that hydrochloric acid 0.5 M and 12 hours of soaking time increased the growth of teak seed compared to the control by 29.64% on a percentage of germination, 29% on germination rate, and 41.66% on growth simultaneity.
