Abstract

Synthetic bone graft material was developed as a scaffold that serves to template the formation of bone. The ideal scaffold should be biodegradable, osteoindukctive, and osteoconductive. Silica proven to support and promote bone growth. Rice husk contains silica is quite high. The purpose of this study is to analyze the amorphous silica from rice husk as synthetic bone graft material (scaffold), particularly on osteoblast activity in vitro. The research was conducted by isolating silica from rice husk. The results of isolation would be conditioned on the culture medium of primary osteoblasts. Primary osteoblast cultures derived from rat calvaria wistar age of 2 days, growing in Alpha-Modified Eagle Medium (α-MEM) (as a negative control), α-MEM in conditioning with silica 58s (as a positive control) and α-MEM that conditioned with silica from rice husk (as treatment group). Osteoblast activity was observed by analyzing the expression of alkaline phosphatase, using Alkaline phosphatase Kits at days 7 and 14. The results obtained that the rice husk silica were significantly (p <0.05) has the expression of alkaline phosphatase higher than the control group and potitif group. This may imply that the amorphous silica of rice husk affects the increase in osteoblast activity in vitro. It was concluded that rice husk silica increases osteoblast cell activity that is potentially as scaffold for bone graft synthetic material.

Abstract

Gingivitis is an inflammatory disease in gingiva without any loss of attachment caused by plaque. Gingivitis is reversible, if not treated it can develop into periodontitis. Gingivitis-causing bacteria include Phorpyromonas gingivalis, acting as secondary bacteria in plaque formation at the secondary colonization stage by attaching themselves to monolayers that have been formed by primary bacteria. Herbal use in Indonesia is still a trend until now with the slogan back to nature, one of the easy to find and useful is essential oil from curcuma rhizomes containing xanthorrizol and curcumin. They are antibacterial that can inhibit the formation of plaques that cause gingivitis. The purpose was to analyze the potential of curcuma essential oil toothpaste in inhibiting the formation of plaque and gingivitis. 27 wistar mice were divided into 9 groups, namely negative control group, positive control, treatment applied curcuma essential oil toothpaste concentration of 0.5%, 1%, 2%, 4%, 8%, 16% and 32%. Each mouse induced P. gingivalis 0.02 ml on molar sulkus one and two, 24 hours later performed tooth brushing with toothpaste according to the group, the next 24 hours were observed in day 3, 5 and 7 using plaque index according to Park and Katz and gingiva index according to Lee. Treatment and observation are carried out 3 repetitions. The results of the plaque index score and gingiva index in the treatment group were fewer than the negative controls. Curcuma essential oil toothpaste can inhibit the formation of plaque and gingivitis by decreasing the plaque index and gingiva index in P. gingivalis- induced wistar mice.

Abstract

Streptococcus sanguinis has strong adhesion with salivary pellicles which causes other oral bacteria to attach more and has contribution in causing caries and periodontal disease. S. sanguinis mediated the adhesion of S. mutans to dental pellicle receptors so inhibiting the interaction between S. sanguinis and S. mutans would prevent the risk of dental caries. Purple leaf (Graptophyllum pictum) is one of the medicinal plants or natural ingredients that are useful as antibacterials. The purpose of this study was to examine antibacterial activity of purple leaf extract against S. sanguinis. This experimental laboratory study with a pre-posttest control group design was conducted by dilution method on treatment groups, purple leaf extract with concentrations of 25%, 12.5%, 6.25% and 3.12%. Visual and spectrophotometer examination was observed before and after incubation. The result showed that purple leaf extract with concentrations of 6.25% and 3.12% and control + groups had not turbidity and decreased absorbance value. Furthermore, the One Way Anova parametric statistical test and also Post-Hoc Least Significant Difference showed that there were significant differences between the study groups. The purple leaf extract (Graptophyllum pictum L. Griff) had antibacterial activity against S. sanguinis. The antibacterial test for extracts that are dark in color can be continued using the TVC (Total Viable Count) method to provide a quantitative estimate of the concentration of bacterial cells in the sample in units of CFU/ml.

Abstract

Streptococcus mutans is a major species that causes dental caries due to its acidogenic and aciduric properties. Coffee beans contain several compounds, such as caffeine, phenols, tannins, trigonelline, chlorogenic acid, and caffeic acid, which are known for their antibacterial properties. Robusta instant coffee powder is a stable product that is not easily contaminated with bacteria, fungi, and yeast. It is a zero-waste and soluble material. This study aimed to analyze the antibacterial activity of robusta instant coffee powder (RICP) against S. mutans. The antibacterial activity of RICP against S. mutans was tested using three antimicrobial susceptibility tests: 1) disk diffusion method, 2) minimum inhibitory concentrations (MIC), and 3) minimum bactericidal concentration (MBC). The results of the disk diffusion method test showed that the RICP concentration of 0.2 g/mL to 0.8 g/mL against S. mutans had an inhibition zone of 31.14 mm to 42.01 mm. MIC and MBC values of RICP against S. mutans were both 0.0313 g/mL. In conclusion, robusta instant coffee powder (RICP) demonstrates antibacterial activity against the acidogenic and aciduric S. mutans bacteria.
