Abstract

Okra fruit has a flavonoid, phytosterol and pectin content, which is potential as an anticholesterol agent. The pharmacological activity of ethanol extract 70 % of okra fruit (EEOF) can be improved by forming nanoemulsion. This study aimed to prepare an ethanol extract 70 % and nanoemulsion of okra fruit and to prove the cholesterol inhibition in vitro EEOF is smaller than the nanoemulsion of EEOF. The okra fruit was extracted by kinetic maceration, the EEOF was analyzed by a phytochemical screening test and cholesterol inhibition assay in vitro using Liebermann-Burchard reagent. The anticholesterol activity was expressed as IC50 values. Nanoemulsion of EEOF prepared by spontaneous emulsification method using capmul, propylenglycol, glycerin (1 : 2.5 : 2 mL). The result of phytochemical screening of okra fruit simplicia and EEOF were contained flavonoid, steroid, triterpenoid, saponin, and coumarin. The resulting nanoemulsion has a 134.7 nm in diameter with a potential zeta value of -26.72 mV. EEOF has IC50 of 764.11 µg/mL and nanoemulsion of EEOF have IC50 of 712.50 µg/mL. Nanoemulsion can improve anticholesterol IC50 value because it was penetrated the gap between cells so that the delivery and bioavailability were higher.

Abstract

Introduction:Sargassum.sp is one of the marine biota published as antiaterosclerosis, but compound toxicity data need to be explored for safety. Method: Virtual screening has been done with MVD software from active compounds contained in sargassum where has activity as antiaterosclerosis with the mechanism of hypolipidemic effects. Test compounds in sargassum include: fucoidan, rhamnose, fucose, galactose, fucoxantin, alginate, phlorofucofuroeckol A, phloroglucinol, phlorotannin, with HMG-COA Reductase-Simvastatin adenosine receptors with a 1HW9 /PDB code. Its toxicity is predicted using pkCSM (online). Results: The value RS, RMSD value as a result of the docking simulation carried out on said compounds, the following results are obtained: fucoidan (-110,420; 1,478), rhamnose (-72,081; 1,629), fucose (-98,408; 1,546), galactose (-95,757; 5,187), fucoxantin (-106,297; 2,161), alginate (-84,674; 2,897), phlorofucofuroeckol A (-106.701; 2,809), phloroglucinol (-103,140; 2,142), phlorotannin (-48,826; 7,750). The prediction results of toxicity showed fucoidan, rhamnose, fucose, galactose, fucoxantin, alginate, phlorofucofuroeckol A, phloroglucinol and phlorotannin not toxic with LD50 0.95-2.482g / kg. Conclusion:Based on RS values, fucoidan, fucoxantin and phlorofucofuroeckol A compounds contained in brown seaweed were predicted to have activity as antiaterosclerosis. Compounds in brown seaweed can also be predicted to be relatively non-toxic with a value of LD50 0.95-2.482g / kg.

Abstract

Daun keji beling (Sericocalyx crispus (L.) Bremek) [Sinonim Strobilanthes crispa (L.)] merupakan salah satu tanaman yang secara empiris digunakan oleh masyarakat untuk membantu pengobatan. Menurut penelitian daun keji beling diketahui mempunyai khasiat sebagai antidiabetes. Penelitian ini bertujuan untuk mengidentifikasi golongan senyawa yang terkandung dalam ekstrak, mutu ekstrak dan aktivitas penghambatan enzim α-Glukosidase secara in vitro. Serbuk simplisia diekstraksi dengan pelarut etanol 70% secara maserasi kinetik, kemudian dilakukan penapisan fitokimia, penetapan mutu ekstrak dan uji aktivitas penghambatan enzim α-Glukosidase. Hasil penapisan menunjukkan ekstrak mengandung senyawa flavonoid, saponin, steroid dan triterpenoid. Hasil penetapan mutu ekstrak menunjukkan bentuk ekstrak kental berwarna hijau kehitaman; rasa pahit; kadar sari larut air 60,46%; kadar sari larut etanol 73,45%; kadar abu total 15,06%; kadar abu tidak larut asam 3,10%; kadar abu larut air 11,29%; kadar air 7,68%; susut pengeringan 9,25%; sisa pelarut 0,38%; kadar Pb 0,4941 bpj; kadar Cd 0,0222 bpj; Angka Lempeng Total (ALT) 4,52x102 koloni/g; Angka Kapang Kamir (AKK) terlalu sedikit untuk dihitung (TSUD); dan kadar flavonoid total 2,39%. Hasil uji aktivitas penghambatan enzim α-Glukosidase pada akarbose dan ekstrak kental keji beling menunjukkan IC50 masing-masing sebesar 50 bpj dan 86,2 bpj. Dapat disimpulkan ekstrak kental daun keji beling memenuhi persyaratan mutu ekstrak dan memiliki aktivitas sebagai penghambat enzim α-Glukosidase. Keji beling memiliki potensi untuk dikembangkan lebih lanjut sebagai obat herbal antidiabetes.

Abstract

Antioxidants have an important role in inhibiting free radicals, where free radicals can cause a hereditary disease. Natural medicines that are efficacious as an antioxidant are Gotu kola and kelor leaves. This study aimed to obtain the potential antioxidant compounds in Gotu kola herb extract, kelor leaves extract and the combination using the DPPH method. This research was started by extracting simplicia with 96% ethanol, followed by preparing combination of Gotu kola extract and moringa extract ratio (1:1), (1:2), (1:3), (2:1), (3:1) which was carried out at a concentration of 20, 40, 60, 80, and 100 µl/mL. Furthermore, the antioxidant activity was tested by UV Vis spectrophotometry. Results showed that the IC50 value for the Gotu kola herb extract was 76.66 ± 0.56. Kelor leaves extract 82.66 ± 2.32, a combination of Gotu kola: Moringa (1:1) 74.07 ± 1.16, a combination of Gotu kola: Moringa (1:2) 86.56 ± 1.11, a combination of Gotu kola: Moringa (1:3) 65.09 ± 3.03, a combination of Gotu kola: Moringa (2:1) 56.65 ± 1.61, a combination of Gotu kola: Moringa (3:1) 67.58 ± 2.77. The best potential antioxidant activity was showed in the combination of Gotu kola: Moringa (2:1).

Abstract

Kesadaran masyarakat untuk melaporkan efek samping kosmetik masih sangat rendah, padahal kosmetika digunakan setiap hari oleh berbagai lapisan masyarakat mulai dari bayi hingga lanjut usia, baik laki-laki dan perempuan. Ditambah inovasi kosmetik termasuk penggunaan bahan-bahan baru, relatif lebih cepat dibanding sediaan farmasi lainnya. BPOM sudah mengeluarkan Peraturan BPOM No. 26/2019 tentang Mekanisme Monitoring Efek samping Kosmetik (MESKOS), bahkan membentuk agent of change Pelaporan Efek Samping Kosmetik untuk meningkatkan efektivitas pelaporan. Tujuan: Pengabdian kepada Masyarakat yang terintegrasi ini bertujuan untuk memberikan informasi kepada konsumen mengenai : 1) 3) mengenalkan tools serta cara melaporkan efek samping ke Badan Pengawas Obat dan Makanan (BPOM). Metode: diawali dengan pemetaan calon peserta baik komunitas dan individu, penyesuaian materi agar mudah diikuti peserta yang beragam, dan memperhatikan susunan acara agar peserta mengikuti acara hingga selesai, dengan membuat Post Test menjadi bentuk permainan yang dilombakan. Kegiatan PKM dilaksanakan tanggal 18 Juni 2022, diikuti oleh lebih dari 400 peserta baik melalui zoom serta live streaming melalui youtube https://youtu.be/sVmhgSPvZ4I. Kesimpulan: hasil evaluasi dari 222 peserta individu atas manfaat, efektivitas nara sumber dalam menyampaikan materi serta nilai keseluruhan mendapat rata-rata diatas 4,6 skala 5, sedangkan hasil pengukuran kepuasan mitra kerjasama dari 3 komunitas diperoleh nilai 5 skala 5. Acara ini diunggah dibeberapa media social seperti IG, FB dan mendapat respons positif

Abstract

Mahogany (Swietenia macrophylla King.) is one of the plants used as an antidiabetic agent. 96% ethanol extract of S. macrophylla stem bark can inhibit the α-glucosidase enzyme. Methanol and aqueous extracts of S. macrophylla stem bark can reduce rat blood glucose levels induced by streptozotocin. Molecular docking is one of the most widely used structural-based drug design strategies due to its wide application in the analysis of binding energy and molecular interactions between drug compounds and receptors. This study aims to identify the compounds in the stem bark of S. macrophylla and analyze the molecular anchoring of the compounds produced by the aid. The research was carried out by means of the water fraction of the stem bark of S. macrophylla and was purified by column chromatography. Fraction 2 was identified by LCMS-MS. Molecular docking using AutoDock 4.2.6. The results of Fraction 2 showed the presence of catechins, evodionol, swietemacrophyllanin, swietenitin K and β-sitosterol. The results of an in-silico study showed that swietemacrophyllanin provided a better affinity than acarbose with a binding energy value of -8.57 kcal/mol and an inhibition constant of 0.52 µM. Forms hydrogen bonds with catalytic residues (Glu277 and Asp352) from the active site of the α-glucosidase enzyme. Swietemacrophyllanin has potential as an antidiabetic agent

Abstract

Dengue Hemorrhagic Fever (DHF) is an abnormal health condition caused by the dengue virus through the bite of the Aedes aegypti mosquito. The use of repellent to prevent mosquito bites can provide protection against DHF. DEET, a synthetic repellent with usage that has spread throughout the world, is reported to have toxic effects and cause irritation. Leaves of iler and patchouli oil are plants of the Lamiaceae family with repellent activity. This study aimed to obtain a repellant gel combination of iler leaf extract and patchouli oil. Gel preparations are made with a combination of Iler Leaf Extract and Patchouli Oil at concentrations of 12.5%, and 2%. Physical and chemical quality parameter test, stability test, acute dermal irritation test, and repel effectiveness test were tested on Aedes aegypti mosquitoes. Gel preparations showed a homogeneous physical appearance, greenish-brown color with an aromatic distinctive odor, a viscosity value of 16000 cPs, pH 6.32, and thixotropic flow properties. Gel preparations are effective as a mosquito repellent for 3 hours with a protection power of 56.82%. The gel preparation storage temperature, besides the resulting gel, does not irritate, and it is physically stable at 4 ± 2oC and 25 ± 2oC and is slightly less stable at 40 ± 2oC.
