Abstract

Salinity stress on productive rice fields in coastal areas will have a negative impact on productivity through osmotic stress and nutrient deficiencies. Salinity stress causes high levels of Na and Cl in the soil, thus inhibiting phosphate absorption. Utilization of indigenous P-solubilizing bacteria from saline land is an alternative, environmentally friendly technology. The purpose of this study was to isolate and characterize P-solubilizing bacteria from saline rice fields. Soil samples were taken from the rice rhizosphere in Nyamplungsari Village, Peraturkan District, Pemalang Regency. Isolation and characterization were carried out at the Agronomy & Horticulture Laboratory, Faculty of Agriculture, UNSOED. The variables observed included P solubility index, P solubilizing ability, IAA production, and bacterial identification using the 16S RNA method. The results of the study obtained 7 isolates of P-solubilizing bacteria that had the ability to solubilize P and produce IAA. Isolate KF is a P-solubilizing bacterium that has the highest P-solubilizing ability and is identified as Priestia megaterium strain NRRL B-350. Priestia megaterium strain NRRL B-350 is a species of P-solubilizing bacteria that has the potential to be developed as a specific biological fertilizer for saline soil to increase the growth and yield of rice plants under saline stress.

Abstract

A plant breeding program for turmeric (Curcuma longa L.) development is required to fulfil the pharmaceutical industry's demand. Understanding genetic diversity is the initial step in selecting the parents in plant breeding. Selection assisted by molecular markers, specifically Simple Sequence Repeat (SSR), can be used to obtain genetic information. However, genetic information on turmeric based on SSR markers in Indonesia is still limited. This study aimed to determine the genetic diversity and relationship of six accessions of turmeric using 4 (four) SSR primers. This study was conducted from October 2023 to January 2024 at the Laboratory of Plant Breeding and Biotechnology, Faculty of Agriculture, Universitas Jenderal Soedirman. The result showed that the CuMiSat-19, CuMiSat-20, and CuMiSat-29 primers were informative based on the electrophoresis band polymorphism. The CuMiSat-19 primer generated the highest PIC value (0,88), followed by CuMiSat-20 (0,80) and CuMiSat-29 (0,74). While CuMiSat-23 did not produce polymorphism, the PIC value was 0. The dendrogram classified six turmeric accessions into the two clusters with a Jaccard correlation coefficient range of 0.11-0.33. Cluster I consisted of accession A (Ponorogo), accession C (Gresik), accession D (Indramayu), and accession B (Semarang). Cluster II grouped accession E (Banyumas) and accession F (Bogor). Based on the polymorphism and similarity coefficient, turmeric accessions have a wide range of genetic diversity. Turmeric in different clusters is a potential germplasm that can be utilized to broaden genetic diversity in plant breeding programs.

Abstract

Jahe merah merupakan jahe yang lebih pedas dibandingkan dengan jenis jahe lainnya, yang dapat dimanfaatkan dalam industri obat-obatan. Keterbatasan bibit menjadi permasalahan utama dalam budidaya jahe merah. Informasi tentang keragaman genetik ddan fenotipik jahe merah diperlukan untuk menentukan metode terbaik dalam menghasilkan varietas unggul baru melalui program pemuliaan tanaman. Penelitian ini bertujuan untuk mengetahui keragaman genotipe, fenotipe, dan heritabilitas 7 (tujuh) aksesi jahe merah yang dikoleksi dari 7 (tujuh) daerah di Pulau Jawa. Hasil penelitian menunjukkan bahwa 7 (tujuh) aksesi jahe merah yang diteliti, berdasarkan 4 (empat) karakter agronomi kuantitatif yang diamati, mempunyai ragam genetik yang sempit dan ragam fenotipe yang luas. Berdasarkan nilai duga heritabilitas, diketahui bahwa tinggi tanaman dipengaruhi oleh faktor genetik, sedangkan luas daun, jumlah daun, dan diameter batang dipengaruhi oleh faktor genetik dan lingkungan.

Abstract

Orphanages play an important role in replacing the role of parents in nurturing, looking after, and providing guidance to children so that they become adults who are valuable and responsible for themselves and society in the future. The goal of the community service was to increase the interest and knowledge of teenagers at the Gelora Indah Mosque Orphanage, North Purwokerto, regarding vertical-culture (verti-culture) cultivation techniques. Verticulture technique was chosen because of the potential for narrow yard land, which can be used as land for agricultural crop production, especially plants that are suitable for vertical cultivation. This activity can be used as a provision for achieving food self-sufficiency at the household level and as a basis for producing agricultural products that can be used as input for entrepreneurship from an early age. The community service was conducted from March to May 2023 with all the teenagers of Gelora Indah Mosque Orphanage. The methods were a pre-test, a transfer of knowledge about verti-culture cultivation techniques, cultivation practices, harvesting, and finally, post-tests for the last evaluation. This activity can increase participants' interest and knowledge about the concept of verti-culture by 83.34%, about the needs for verti-culture installation by 66.67%, and about the benefits of verti-culture by 100%. Activities in the form of transfer of knowledge about post-harvest handling and processing, as well as marketing techniques for processed and fresh agricultural products, are needed to support the long-term benefits of this activity.

Abstract

An orphanage is a place of residence and care, both for orphans and for abandoned children. The Gelora Indah Mosque Orphanage in Purwokerto is inhabited by thirteen teenagers. The potential of the second floor attic of this orphanage has not been maximized. With the condition of the yard land that is not spacious and to equip the teenagers with soft skills, one technology that can be applied is the vertiminaponic technique. This technique combines plant cultivation (hydroponics) and fish (aquaculture) in one place/land that is arranged vertically. Community service activities are carried out using the lecture method followed by cultivation assistance. The results of community service show an increase in knowledge and concepts about vertiminaponic cultivation of the orphanage teenagers by 83.34% and 49.58%. Meanwhile, when viewed from the aspect of knowledge about equipment and stages in vertiminaponic cultivation, there is an increase of 65.26% and 70.32%. This activity is an initiation in order to increase food independence for the orphanage teenagers through diversification of processed products from cultivation.

Abstract

Bandingan Village, Rakit District, Banjarnegara Regency, Central Java Province, has dry lands, resulting in relatively low agricultural productivity. Given these conditions, the drip irrigation system is an alternative solution for efficient water use. The community service activity on the socialization of the drip irrigation system was aimed to increase the knowledge of the women farmers group (KWT) regarding the application of the drip irrigation system. This activity was part of the Community Service Program of Universitas Jenderal Soedirman for the period July - August 2025, which have been carried out through initial observation, assesment (pre-test), the transfer of knowledge about the drip irrigation system, discussions, practices, then final assessment (post-test), and as well as analysis data of the evaluation. Eighteen members of the women farmers group actively participated in this program. Data analysis was carried out using a paired T-test in Microsoft Excel. Overall, the data analysis showed a significant increase in knowledge after the socialization, particularly in understanding (definition), advantages (benefits), tools used for installation, and how drip irrigation works. The increase in knowledge of drip irrigation was 95.83%, the benefits aspect reached 90.28%, the knowledge aspect regarding installation tools was 100%, and the aspect regarding how it works was 88.33%. Thus, this activity increases public knowledge about the concept, benefits, and application of the technology. This knowledge can be applied to improve water use efficiency in the agricultural land that tends to be dry in this village.

Abstract

Sistem Informasi Personel Polri (SIPP) 2.0 merupakan sistem berbasis web yang dirancang untuk mengelola data personel secara digital dan terintegrasi dalam lingkup Polri. Aplikasi ini membantu Biro SDM dalam proses pembinaan dan pengelolaan data sumber daya manusia secara efisien, akurat, dan tepat waktu. Tujuannya adalah untuk melakukan analisis terhadap sistem informasi SIPP 2.0 yang digunakan di Biro SDM Polda Jambi dengan menggunakan pendekatan SWOT. Hasil dari analisis menunjukkan bahwa sistem ini memiliki kekuatan dari sisi kelengkapan data dan kemudahan akses, namun juga ditemukan beberapa kelemahan seperti ketergantungan pada jaringan internet dan potensi kesalahan input. Dengan melihat peluang pengembangan dan tantangan keamanan, sistem ini dapat dioptimalkan lebih lanjut untuk mendukung transformasi digital Polri.

Acid-dry land is a suboptimal land area widely distributed in Indonesia, which has the potential to be developed as cropland to increase crop productivity. However, the low phosphorus availability in this land type often constrains plant growth, necessitating the use of microbial phosphate-solubilizing agents. To address this, the capability of the isolated rhizobacteria strain in phosphate solubilization was improved through a mutagenesis by Ethyl Methane Sulphonate (EMS). Three isolates of indigenous wild-type rhizobacteria of acid-dry land were isolated from cardamom, lemon grass, and bamboo rhizosphere. Mutagenesis of these wild-types yielded three mutant isolates, including KPG_m, SRH_m, and BMB_m, which exhibited higher Phosphate-Solubilization Indices (PSI) compared to their parental strains. The PSI values for the mutants were 2.2, 3.4, and 2.0, respectively, showing a marked improvement over the wild-type values of 2.0, 1.75, and 1.25. These results indicate an enhancement in phosphate-solubilizing ability driven by EMS mutagenesis. All the isolates were then characterized and showed similar traits to those of the Genus of Bacillus. According to the hypersensitive reaction assay, the six isolates showed a negative reaction, indicating they are non-pathogenic. Based on these findings, the mutants have the potential to be developed as biofertilizer agents due to they are non-pathogenic for plants. In addition, the mutants have a moderate PSI value (2<PSI<4) and effective solubilization rate that may improve phosphate solubilization process in acid-dry land.
