Abstract

Carcinogenesis can be involved in the malfunctioning of programmed cell death Most of the anticancer drug in current use induce apoptosis in susceptible cells. The fact that disparate agent interacting with different targets seem to induce cell death through some common mechanism suggest that anticancer activity is determined by the ability of inhibiting cell growth.Pentagamavunon-1 (PGV-1) is one of the curcumin analogue which showed to have potency in inhibiting proliferation of human breast carcinoma cell T47D. The effect on T47D growth is associated with cell cycle arrest in G2/M phase at the concentration of 2.5 mM, followed by hyperploidy. Our data on polymerization assay, indicate PGV-1 interact with tubulin in different manner from taxol. PGV-1 inhibit tubulin polymerization on cell culture while taxol stabilized tubulin polymerization. Immunostainning data on cell treated with PGV-1 showed slightly tubulin condensation, while cell treated with taxol showed tubulin condensation distinctly at 12 minutes after releasing from depolymerization agent.In conclusion, PGV-1 represent a new microtubule inhibitor and has the potential to be developed for anticancer drugKey words: Pentagamavunon-1, T47D, tubulin, antimitotik

Abstract

The initial research of Jati Belanda leaves extract (JBE) showed the inhibition of breast cancer cell growth (T47D). The phytochemistry screening showed that JBE contain flavonoid, alkaloid, polifenol, and volatile oil. The development of anticancer drugs need the molecular mechanism investigation in order to produce cancer-targeted drugs. The objective of this research is to determine the molecular mechanism of JBE cytotoxicity effect towards cancer cell lines. This research began with cytotoxicity asssay in vitro of JBE towards some cancer cell lines by MTT method. JBE was given in some variety of concentration. The result of this study showed that JBE do not contain tirosid, and contain flavonoid in the concentration of 0,976%. The result of cytotoxicity assay towards MCF-7, HeLa, T47D and Vero showed IC50 value 36,50; 58,02; 53,36; 1806,22 dan 2451,65 µg/mL respectively. It is concluded that JBE have a strong potency to inhibit the growth of WiDr cancer cell line.Keywords : jati belanda, T47D cells, cytotoxicity

Abstract

Several studies show that some plants and fruits could protect human body from free radical danger exposure. Zingiberaceae family has some chemical substances that have antiradical activity, such as phenolic compound and flavonoid. The aim of this study is to determine the correlation of total phenolic compound towards its antiradical activity. Antiradical activity assay was determined by DPPH method (1,1 difenil-2-pikrilhidrazil). The total phenolic compound is determined by spectrofotometry using Folin-Ciocalteu reagent and counted as GAE (Gallic Acid Equivalent). Based on this study, the total phenolic concentration of ethanolic extract of jahe (Zingiber officinale (L.) Rosc), kencur (Kaemferia galanga L.), lengkuas (Languas galanga), lempuyang gajah (Zingiber littorale) and lempuyang pahit (Zingiber americana) is 73.99 mg/g; 23.848 mg/g; 37.841 mg/g; 49.725 mg/g and 40.802 mg/g respectively. The radical scavenging activity (IC50) of jahe (27.38 µg/mL), lengkuas (93,27 µg/mL), lempuyang pahit (105,08 µg/mL), meanwhile lempuyang gajah and kencur show low activity of radical scavenging.Keywords: Zingiberaceae, total phenolic compound, antiradical, DPPH

Abstract

Due to severe side effect and non-specific chemotherapeutic agent, screening and discovery for cancer therapy are still working, especially from natural resources. Traditionally, people used herbal medicine either to prevent or cure diseases. One of herbal that commonly used in Indonesia is Alpinia galangal. Previous study stated that active compound is acetoxy chavicol acetate (ACA) and active as anticancer. This research aimed to determine ACA concentration and cytotoxic activity of Alpinia galanga extract (AGE) from three local markets on HeLa, MCF7 and T47D cell lines. The galangal used from three local markets namely Pasar Legi Surakarta, Beringharjo Yogyakarta, and Wonogiri. The extraction was performed by maceration using 96% ethanol as solvent. ACA quantitation using UV spectrophotometer at λ = 208.5 nm. Samples were prepared by liquid-liquid extraction using an ethyl acetate. Cytotoxic activities were performed by MTT assay. The result showed that the concentration of ACA of AGE from the three local markets were 3.798; 0.035; and 0.009 % w/w, respectively. Cytotoxic activity, describes as IC50 value, on HeLa cell line of AGE from three local markets, in order were 13.26; 36.32 and > 100 µg/ mL. Meanwhile, AGE from Pasar Legi on MCF7 and T47D cell lines have IC50 value of 15.80; 12.50 µg/ mL, respectively. In contrast, two other samples have IC50 values of greater than 100 µg/ mL. The highest activity was from the highest concentration of ACA on the samples.Key words: Alpinia galanga, HeLa, T47D, MCF7 and Acetoxy chavicol acetate

Abstract

AbstrakTeripang Holothuria atra merupakan biota laut yang banyak ditemukan di perairan Indonesia yang termasuk dalam filum Echinodermata dan berpotensi sebagai antikanker. Penelitian ini bertujuan untuk mengetahui sitotoksisitas dan induksi apoptosis ekstrak etanol teripang H. atra secara in vitro terhadap beberapa sel lestari. Pengujian sitotoksisitas dilakukan dengan metode MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) menggunakan sel HeLa, T47D, WiDr dan sel normal Vero, sedangkan uji induksi apoptosis dilakukan terhadap sel dengan hasil uji sitotoksisitas terbaik menggunakan metode flowcytometry dan double staining. Hasil penelitian menunjukkan bahwa ekstrak etanol  teripang H. atra mampu menghambat pertumbuhan sel kanker HeLa, T47D dan WiDr. Nilai IC50 ekstrak H. atra terhadap ketiga sel tersebut masing-masing sebesar 41,06±4,21; 20,89±1,55; 26,50±4,43 µg/ml tetapi esktrak tersebut memiliki sitotoksisitas  yang lebih rendah terhadap sel Vero (IC50 sebesar 128,00). Analisis flowcytometry dan double staining pada sel T47D memperlihatkan bahwa ekstrak etanol teripang H. atra mampu menginduksi apoptosis pada sel tersebut. Cytotoxicity and Apoptosis Induction of Ethanol Extract Holothuria atra Jaeger, 1833 on Cancer CellsAbstractHolothuria atra is the marine organism found in Indonesian waters which is included in the phylum of Echinodermata and has the potential as an anticancer. This research aims to determine the cytotoxicity and apoptosis induction of ethanol extract of sea cucumber  H.  atra in vitro on cancer cell lines. This cytotoxicity test was conducted by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay using HeLa, T47D, and WiDr cancer cell lines while apoptosis induction test was conducted on cell which had the best cytotoxicity result by flowcytometry and double staining. The results showed that ethanol extract of H. atra was able to inhibit the growth of HeLa, T47D and WiDr cell lines with IC50 of 41,06±4,21; 20,89±1,55; 26,50±4,43 µg/ml respectively, however, the extract had lower cytotoxicity to normal Vero cells. Flowcytometry and double staining analysis showed that ethanol extract of sea cucumber H.  atra was able to induce apoptosis in T47D cell.

Abstract

Kemangi merupakan tumbuhan yang banyak digunakan masyarakat serta memiliki aktivitas sitotoksik. Penelitian ini bertujuan untuk mengetahui adanya aktivitas antikanker kemangi dalam hal menghambat pertumbuhan serta menginduksi kematian secara terprogram sel MCF-7. Daun kemangi berupa serbuk dimaserasi dengan pelarut etanol kemudian dihilangkan pelarutnya dengan menggunakan rotary evaporator. Uji sitotoksisitas dilakukan dengan metode MTT, didapatkan nilai IC50 ekstrak etanol daun kemangi sebesar 174,46 µg/mL. Kadar yang digunakan dalam uji antiproliferatif adalah di bawah nilai IC50 yaitu 150 dan 75 µg/mL. Uji antiproliferatif dilakukan dengan metode MTT. Sel diinkubasi pada waktu yang berbeda-beda yaitu 24; 48; 72 jam. Hasil uji didapatkan adanya aktivitas penghambatan pertumbuhan sel oleh ekstrak etanol daun kemangi terhadap sel MCF-7 dengan nilai doubling time yaitu 530,61 jam pada sampel kadar 150 µg/mL; 147,88 jam pada sampel kadar 75 µg/mL; 125,58 jam pada kontrol sel. Selanjutnya dilakukan uji induksi apoptosis dengan kadar ekstrak etanol kemangi yang digunakan sebesar IC50. Metode yang digunakan adalah metode double staining. Setelah dipapari sampel, sel diberi reagen campuran Etidium Bromida-Akridin Oranye dan diamati di bawah mikroskop fluorescent. Hasil uji menunjukkan adanya aktivitas induksi kematian sel MCF-7 secara terprogram oleh ekstrak etanol daun kemangi yang ditandai dengan sel yang berfluoresensi berwarna oranye serta kondisi sel yang tidak utuh.

Abstract

Tanaman parijoto mempunyai banyak khasiat untuk tubuh manusia. Satu di antaranya sebagai agen antikanker. Tujuan penelitian ini guna mengetahui berapa kadar flavonoid dan fenolik serta aktivitas sitotoksik yang terjadi pada ekstrak etanol tanaman parijoto terhadap sel kanker serviks HeLa. Digunakan bagian daun dan buah dari tanaman parijoto. Daun dan buah parijoto dimaserasi selama 2 hari dalam pelarut etanol 95% lalu dipekatkan hingga terbentuk ekstrak kental. Senyawa yang terkandung di ekstrak etanol daun dan buah parijoto dianalisis menggunakan kromatografi lapis tipis. Hasil KLT membuktikan dimana ekstrak daun dan buah parijoto didapati senyawa flavonoid dan fenolik. Penetapan kadar flavonoid dan fenolik diuji dengan metode spektrofotometri UV-Vis. Kadar flavonoid dan fenolik pada ekstrak etanol daun parijoto adalah 56,62 mg QE/g sampel dan 159,61 mg GAE/g sampel. Pada uji sitotoksik dengan menggunakan metode MTT assay, diperoleh ekstrak etanol daun dan buah parijoto mempunyai aktivitas sitotoksik moderat dengan IC50 masing-masing 457,08 µg/mL dan 363,07 µg/mL.

Abstract

Perubahan bentuk fasilitas pelayanan kesehatan di Pondok Pesantren Modern Muhammadiyah Boarding School (PPM MBS) Yogyakarta dari Usaha Kesehatan Sekolah (UKS) menjadi Pos Kesehatan Pesantren (Poskestren) sesuai Permenkes RI Nomor 1 Tahun 2013 pada September 2022 lalu membawa berbagai konsekuensi, diantaranya adalah perubahan struktur organisasi dan tata kelolanya. Dalam Permenkes Nomor 1 Tahun 2013 mengenai Pedoman Penyelenggaraan dan Pembinaan Poskestren, juga diatur mengenai bentuk dari struktur organisasi dan tata kelola (SOTK) Poskestren. Kegiatan PkM ini bertujuan melakukan pendampingan kepada pengelola Poskestren PPM MBS Yogyakarta dalam menyesuaikan dan merevitalisasi struktur organisasi yang bermula dari UKS menjadi struktur organisasi sesuai aturan Kemenkes RI. Kegiatan dilakukan melalui workshop berupa pemaparan materi mengenai peran manajemen bagi optimalisasi mutu pelayanan kesehatan di pesantren; SOTK Poskestren sebagai dasar pengembangan pengelolaan fasilitas pelayanan kesehatan; SOTK UKS PPM MBS Yogyakarta (existing condition) sebelum menjadi Poskestren; mekanisme penyusunan SOTK dan deskripsi tugas Poskestren PPM MBS Yogyakarta. Selanjutnya dari kegiatan workshop ditindaklanjuti dengan diskusi dan pendampingan identifikasi jabatan yang disesuaikan kebutuhan poskestren, penyusunan struktur organisasi, penyusunan kualifikasi pejabat pada setiap bagian jabatan, dan penyusunan deskripsi tugas pejabat pada setiap bagian jabatan pada struktur organisasi yang telah terbentuk. Dengan adanya penyesuaian struktur organisasi poskestren yang dilengkapi dengan kualifikasi dan deskripsi tugas pejabat ini, diharapkan pengelolaan Poskestren PPM MBS Yogyakarta semakin optimal, sehingga pelayanan kesehatan pada para santri menjadi semakin lebih baik.Kata Kunci : struktur organisasi, deskripsi tugas, poskestren

Abstract

This study aims to determine the knowledge and application of pharmacist ethical practices in the city of Surakarta. This research is a type of descriptive research. This research was conducted in all pharmacies and hospitals in the Surakarta City area. Data collection will take place in November 2023-January 2024. The population in this study was 489 and the error rate was 5%, so the sample taken was 205 people. The selection of respondents in this study was carried out by non-probability sampling techniques with a sampling technique, namely purposive sampling. The instrument used in this study was a questionnaire. Data analysis of this study was carried out by descriptive analysis on SPSS software. Based on the results of the study, pharmacists were dominated by good knowledge as many as 132 respondents (64%) while for the application of pharmacist ethical practices in Surakarta City was dominated by the application of ethics as many as 122 respondents (60%). This study concludes that pharmacists in Surakarta City have good knowledge of ethical practices and sufficient application of ethical practices.

Abstract

This study aims to determine the knowledge and application of pharmacist ethical practices in Pekalongan Regency. This study is a type of observational descriptive study, the population in this study is pharmacists who practice in Pekalongan Regency as many as 129 using purposive sampling, the sample in this study was obtained as many as 100 respondents. The instrument used in this study was a questionnaire. This research will be conducted in Pekalongan district from October to December 2023. Data analysis of this study was carried out by descriptive analysis on SPSS software. The results showed that out of 100 respondents, 97 respondents (97%) had good knowledge about pharmacist ethical practices. However, out of 100 respondents, 52 respondents (52%) dominated the application of pharmacy ethical practices in the less category. It can be concluded that although pharmacists have good knowledge in pharmacist ethical practices, in the application of pharmacist ethical practices compared to the best, which is less.
