Abstract

The inhibition of Sclerotium rolfsii as causal agent of soybean damping off by using chitinolytic bacteria were studied in Laboratory of Observation Pest and Disease UPT-BPTPH 1 and Laboratory of Microbiology, Department of Biology, Faculty of Mathematics and Natural Sciences, University of Sumatera Utara, Medan. The aim of this research was to investigate  the ability of chitinolytic bacteria to inhibit the growth of S. rolfsii. Six isolates of chitinolytic bacteria were used in this study. The inhibition zone between colony of the bacteria and the fungus was measured. The result showed that the bacterial isolates has different ability in inhibiting the growth of S. rolfsii. Enterobacter sp. BK15 and Bacillus sp. BK13 were the most effective in inhibiting the growth of S. rolfsii with inhibition zone of 3.70 cm and 3.75 cm, respectively. The less effective was shown by Enterobacter sp. PB17 with inhibition zone of 0.10 cm, while Enterobacter cloacae LK08 have no ability to inhibit S. rolfsii. The seeds soaked in bacterial suspension by  Bacillus sp. BK13 dan Enterobacter sp. BK15 were able to reduce damping off by 44.4% and 50%, respectively.   Keywords: Bacillus sp., damping off, Enterobacter sp., Sclerotium rolfsii, soybean

Abstract

Ability of watermelon endophytic bacteria to suppress development of leaf spot disease caused by Colletotrichum sp. A studi on assay of endophytic bacteria to control Colletotrichum sp., causal agent of leaf spot disease on watermelon, was conducted. Colletotrichum sp. was isolated from infected leaf of leaf spot disease, while endophytic bacteria were isolated from stem, leaf, and root of watermelon healthy plant. Antagonistic assay was conducted by dual culture method. Hyphal abnormalities as a result of antagonistic assay was observed using light microscope. To determine the ability of endophytic bacterial isolates to control leaf spot disease, watermelon seeds were treated by dipping the seed in endophytic bacterial suspension. Seven endophytic bacterial isolates showed to inhibit Colletotrichum sp. to some extent. Two isolates DS 01 and BS 01 showed relatively high inhibition zone compared to others, therefore were choosen for further study. Abnormal hyphae such as broken, lysis, twisted, curled, and swollen hyphae were recorded as the result of antagonistic assay. Watermelon seed treatment revealed that dipping the seed in endophytic bacterial suspension of DS 01 and BS 01 reduced leaf spot disease to 12 and 24%, but inhibited seed growth by 12% and 44%, respectively. It seemed that all treatments showed to reduce seedling performance i.e seedling height, leaf number, and dry weight, compared to that of (-) control. DS 01 however seemed to increase dry weight of watermelon plants.

Abstract

Assay on ability of endophytic bacteria isolated from peanut to inhibit Sclerotium sp. growth in peanut seedlings. A study on assay of ability of endophytic bacteria to inhibit Sclerotium sp. in peanut seedling has been done. The bacteria were isolated from peanut healthy plants, while Sclerotium sp. was isolated from infected peanaut plant. Antagonistic assay was conducted by dual culture method. In vivo assay of inhibiting Sclerotium sp. was conducted by dipping peanut seed in bacterial solution, and planting the seed in soil:compost (3:1) growing media. Six endophytic bacterial isolates showed to inhibit the growth of Sclerotium sp. in vitro. LN1 seemed to inhibit more of Sclerotium sp., while LN5 showed to inhibit less. Two potential isolates LN1 of gram-negative and LN2 of gram-positive using for further study showed to decrease more of dumping off. It also seemed that the isolates increased the seedling height, number of leaves, and dry weight.

Abstract

Air kelapa adalah senyawa alami kompleks yang sering digunakan sebagai pengatur tumbuh alami.   Penggunaan air kelapa sebagai bahan organik merupakan alternatif dari penggunaan bahan sintetis pada tanaman. Tujuan penelitian ini adalah untuk mengetahui pengaruh konsentrasi limbah dalam air kelapa dan air cucian beras serta lama perendaman terhadap perkecambahan biji kacang hijau. Parameter yang diamati adalah tinggi globul, jumlah radikula, dan panjang radikula. Berdasarkan hasil penelitian, air kelapa dan air cucian beras berpengaruh terhadap tinggi bulu dimana bulu tertinggi terdapat pada kombinasi (20 ml air kelapa + 80 ml air cucian beras dan perendaman 2 jam). Panjang radikula terpanjang ditemukan pada kombinasi (10 ml air kelapa + 90 ml air cucian beras dan perendaman 6 jam). Jumlah akar paling banyak terdapat pada kombinasi (50 ml air kelapa + 50 ml air cucian beras dan perendaman 6 jam).

Abstract

Dermatophytes are a group of fungi that cause nail abnormalities. This study aims to determine dermatophyte fungal infections in patients with nail mycoses. The method used in this study is descriptive and the sample was taken by purposive sampling with the criteria of damaged toenails, with characteristics: nails are not shiny, brownish to black from the base to the tip then the nails are examined directly with 10% KOH and cultured using media Sabouraud Dextrose Agar (SDA). The fungi isolated on slanted agar containing SDA and each fungal species were then identified using preparations for microscopic examination. The results showed that from 30 samples, 8 (eight) nail samples were positive for dermatophytes, namely: 5 (five) people infected with the fungus Tricophyton mentagrophytes species, 2 (two) people infected with the Trichophyton rubrum fungus, and 1 (one) person infected with Epidermophyton floccosum..

Abstract

Air pollution, primarily originating from transportation and industrial activities, has become a major concern in the context of environmental health. This study aims to investigate the relationship between air pollution and the morphological characteristics of leaves as well as the stomatal density of the pulutan plant (Urena lobata L.) in polluted environments within the industrial area of Medan and unpolluted environments in Percut Sei Tuan, Medan, North Sumatra. The research method employed is ex-post facto with a laboratory approach to the polluted industrial area. Experimental results indicate that the stomatal density on pulutan leaves in the polluted area, specifically in the Industrial Zone of Medan, is 17.27/mm2, whereas in the unpolluted area, namely Percut Sei Tuan, it is 896.81/mm2. This difference provides insights into the negative impacts of air pollution on pulutan plants, highlighting the importance of maintaining air quality for ecosystem health and human well-being.

Abstract

Endophytic bacteria are bacteria that live within plant tissues and are capable of producing secondary metabolites in the form of bioactive compounds similar to those produced by their host plants. This study aims to isolate and characterize endophytic bacteria from the roots of kecombrang (Etlingera elatior (Jack) R. M. Smith) and to test their ability to inhibit the growth of Staphylococcus aureus. The research was conducted using a quantitative descriptive method through several stages, including sample preparation, sterilization of equipment and media, isolation, characterization, and testing. The results identified three isolates of endophytic bacteria from the roots of kecombrang, namely AK1, AK2, and AK3, which were shown to be gram-positive cocci. Inhibition tests indicated that these endophytic bacteria exhibit antagonistic properties against Staphylococcus aureus. The isolate coded AK1 had an inhibition zone of 9.6 mm, AK2 had the lowest inhibition zone of 6.6 mm, and AK3 had the largest inhibition zone of 13.5 mm.

Abstract

Basil is a plant that can be used as a traditional medicine. One part of the basil plant used as an alternative medicine is the leaves. Basil leaves contain active compounds, including flavonoids, saponins, tanins, and essensial oils. These active compounds is able to prevent the presence of pathogenic bacteria that can accelerate food spoilage, so that basil leaves can be used as an alternative natural preservative. The objective of the study was to determine the ability of basil leaves as a natural preservative for  mackerel. The research was carried out in vitro, by observing the physical, chemical, and microbiological characteristicts of mackerel which were treated 8 hours of soaking basil leaf extract. Variations in the concentration of basil leaves used were 0, 25, 50, 75 and 100%. The results showed that the physical quality of the 8-hour storage samples showed changes in the physical quality of the fish, as seen from the color, eyes, gills, mucus, aroma and texture. The microbioloical quality of fish was analyzed by the total plate number method. The total number of bacteria decreased as the concentration of basil leaf extract increased

Abstract

The people of Bandar Setia Village experienced the impact of the implementation of PPKM due to regulations delimiting community activities. The limited agricultural land in the village also affects the fulfillment of food needs, especially vegetables. The purpose of this community service is to train the Forum Peduli Sesama Seniman Indonesia Cabang Tembung group representing the community of Bandar Setia in terms of hydroponic farming as an alternative to providing food stocks for the community in the village during the pandemic. The specific target of the training is the hydroponic model that can be made by the people of Bandar Setia village. The method used mixing counseling, training, hands-on practice, and work evaluation. Hydroponics manufacturing uses a mixing of NFT (nutrient film technique) and wick system methods. Vegetables harvested using hydroponics can be used for personal needs or sold according to the needs of the people of Bandar Setia village.

Abstract

Yayasan Ekosistem Lestari (YEL) in Bahorok Research Station in Bahorok Sub-district, Langkat District, is a very important location for this work. The hilly terrain of the area provides a beautiful landscape and varied flora and fauna with the main attraction being the corpse flower (Amorphophallus sp). With a preserved natural environment and unique ecological features, this research station allows other parties to participate in protecting the area in various conservation campaign, one of which is the installation/establishment of conservation information boards. Conservation information boards can raise public awareness about the importance of protecting forests and their natural environment. Information on ecosystems, flora, fauna, management, and the importance of conservation can be conveyed through these information boards. Information boards are widely used as an educational tool for local communities and visitors to the forest at the research station. They can learn about forest ecology, the negative impacts of deforestation, and sustainable ways of utilizing forest resources. Information boards can also be used to invite community participation in conservation efforts, which is expected to improve the quality of knowledge and understanding of conservation areas for communities, researchers, and tourists. Therefore, the installation/establishment of conservation information boards needs to be carried out at Bahorok Research Station, Batu Katak as one of the campaign and solutions to maintain the natural conditions in the forest and river.
