Abstract

Tomato is a horticultural commodity that has many benefits, it not only functions as a fruit vegetable but is also often used as a complement to spices, food coloring ingredients and fresh drinks because of its nutritional content. To increase tomato production, paklobutrazol is needed. Paklobutrazol has a function to inhibit the biosynthesis of gibberellins, so that the administration of these substances causes stunting elongation and stimulates flower induction. While GA3 serves to encourage the development of seeds, elongation of stems, encourage flowering and fruit development. This research was conducted with the aim of assessing the effect of Paklobutrazol and GA3 on growth and flowering on tomato plants and determine the concentration of Paklobutrazol and GA3 which are appropriate for the growth and flowering of tomato plants. This research was carried out experimentally using a completely randomized design (CRD) 2 factor factorial pattern. Each treatment combination was repeated 3 times so that there were 48 experimental units. The concentrations of paklobutrazol used were 0 ppm, 250 ppm, 500 ppm, and 750 ppm, while the concentration of GA3 used was 0 ppm, 50 ppm, 100 ppm and 150 ppm. The parameters observed in this study were plant height (cm), flowering time (days), and number of flowers (fruit). The data obtained were analyzed by Analysis of Variance with a confidence level of 95%, and continued with the Smallest Significant Difference test (LSD) at an error rate of 5%. The results showed that administration of paklobutrazol had an effect on inhibiting the height of tomato plants, and GA3 had an effect on accelerating flowering of tomato plants. Paklobutrazol with a concentration of 750 ppm is the best concentration to inhibit plant height, and GA3 with a concentration of 150 ppm is the best concentration to spur tomato flowering.

Abstract

A research of the influence of media and BAP concentrations on raja banana micro shoots growth has been carried out from November 2005 until July 2006. During the research, a split plot design has been used. The main plot was the kind of media; i.e. MS and Gamborg. The sub plot was BAP concentrations (K); i.e. K0: 0 .M, K1: 5 .M, K2: 10 .M, K3: 15 .M, K4: 20 .M and K5: 25 .M. On the plantlet formation, a factorial treatment pattern on CRD has been used. The result showed that both MS and Gamborg media could be used in planting raja banana micro shoots. The best growth of the micro shoots appeared with the addition of 15 .M BAP into the media. Abstract

A research with aims of obtaining which better between MS and Gamborg (B5) as the media in raja banana in vitro culture and knowing how much BAP needed in order to gain a most optimal growth of raja banana in in vitro culture. The research has been done in Plant Physiology Laboratory Biology Faculty General Soedirman University from April up to July 2010. Completely Randomized Design has been used with split plot design. As the main plot were media; i.e. MS media (M1) and Gamborg (B5) media (M2). As the sub plot were BAP concentration (K); consist of K0: 0 2M, K1: 5 2M, K2: 10 2M, K3: 15 2M, K4: 20 2M and K5: 25 2M. Each treatment repeated three times and each repeating treatments consist of three sub-samples. It could be concluded from the result that raja banana in vitro culture might be done in MS or Gamborg (B5) media. The best concentration of BAP in stimulating the forming and the growth of raja banana shoots in in vitro culture was 15 2M. Key words : BAP, Gamborg (B5) medium, MS medium, Raja banana shoot raja

Abstract

The research was carried out to determne the best concentration of kinetin and activated charcoal to stimulate the shoot growth of Ambon Nangka banana cultivar. This research used a Completely Randomised Design (CRD) arranged factorials. The first factor was the concentration of kinetin with 4 levels i.e. 15 µM, 20 µM, 25 µM and 30 µM. The second factor was the concentration of activated charcoal with 4 levels i.e. 0 gL-1; 1 gL-1; 2 gL-1; and 3 gL-1. Each treatment combination was repeated 3 times, which resulted in 48 experimental units. The parameters measured were the shoot emergence time, number of shoots, shoot length, number of roots and root length. The data were analyzed using an analysis of variance at 95% and 99% level of confidences. The research results showed that the interaction between kinetin and activated charcoal had no effect on shoot length of Ambon Nangka banana cultivar. It was also found that the addition of 20 µM kinetin without any added activated charcoal resulted in the highest shoot number of Ambon Nangka banana cultivar.Keywords: Ambon Nangka, kinetin, activated charcoal

Abstract

Cabai merupakan komoditas hortikultura yang memiliki peran penting dalam memenuhi kebutuhan pangan di Indonesia. Hal ini dikarenakan cabai dapat dikonsumsi dalam keadaan segar atau olahan. Paklobutrazol merupakan salah satu zat penghambat tumbuh atau retardan yang peranannya menyebabkan nutrisi dan energi tanaman akan diarahkan mencapai fase generatif lebih cepat, sehingga dapat meningkatkan produksi. Tujuan yang ingin dicapai pada penelitian ini adalah mengetahui pengaruh paklobutrazol dan GA3 terhadap pertumbuhan dan pembungaan cabai dan menentukan konsentrasi paklobutrazol dan GA3 yang tepat terhadap pertumbuhan dan pembungaan cabai . Metode penelitian dilakukan secara eksperimental dengan Rancangan Acak Lengkap (RAL) pola perlakuan faktorial. Faktor pertama yaitu konsentrasi Paklobutrazol (P) dengan 4 taraf yaitu P0 : 0 ppm, P1 : 250 ppm, P2 : 500 ppm, dan P3 750 ppm. Faktor kedua yaitu konsentrasi GA (G) dengan 3 taraf yaitu G0 : 0 ppm, G1 : 50 ppm, G2 : 100 ppm dan G3 : 150 ppm. Kombinasi kedua faktor menghasilkan 16 pelakuan. Setiap kombinasi perlakukan diulang sebanyak 3 kali, sehingga terdapat 48 unit percobaan. Parameter yang diamati dalam penelitian ini adalah tinggi tanaman (cm), jumlah daun, waktu pembungaan (hari) dan jumlah bunga. Data yang diperoleh dianalisis dengan Analisis Ragam dengan tingkat kepercayaan 95% dan dilanjutkan uji Beda Nyata Terkecil (BNT) pada tingkat kesalahan 5%. Hasil penelitian menunjukkan bahwa pemberian Paklobutrazol dan GA3 berpengaruh pada pertumbuhan dan pembungaan tanaman cabai apabila diaplikasikan secara mendiri. Paklobutrazol dengan konsentrasi 250 ppm dan GA3 dengan konsentrasi 50 ppm merupakan konsentrasi terbaik untuk pertumbuhan dan pembungaan pada tanaman cabai . Abstract

Anggrek Coelogyne pandurata Lindl atau anggrek hitam merupakan anggrek endemik dari Kalimantan. Anggrek tersebut mempunyai keunikan yakni sepal berwarna hijau dan labelum yang berwarna hitam. Perbanyakan secara alami tingkat keberhasilannya sangat kecil maka perlu di lakukan kultur in vitro. Kultur in vitro merupakan kultur yang menggunakan eksplan daun untuk memacu pertumbuhan kalus. Pertumbuhan kalus dapat dipacu dengan auksin (IAA) dan sitokinin (BAP). Perpaduan antara konsentrasi auksin dan sitokinin yang seimbang diharapkan bisa memacu pertumbuhan kalus. Tujuan dari penelitian ini adalah untuk mengetahui pengaruh penambahan IAA dan BAP terhadap pertumbuhan kalus anggrek C. pandurata Lindl dan mengetahui interaksi IAA dan BAP yang terbaik terhadap pertumbuhan kalus anggrek C. pandurata Lindl. Metode penelitian dilakukan yang digunakan adalah eksperimental dengan rancangan acak lengkap menggunakan pola faktorial, faktor pertama adalah kosentrasi IAA dengan 4 taraf: 0, 1, 2, 3 mg/L. Dan faktor kedua adalah konsentrasi BAP dengan 4 taraf perlakuan: 0, 1, 2, 3 mg/L, sehingga ada 16 perlakuan, masing-masing perlakuan diulang 3 kali sehingga ada 48 unit percobaan. Parameter yang diamati adalah: waktu munculnya kalus, tebal kalus, jenis kalus, berat kalus dan presentasi terbentuknya kalus. Data yang diperoleh dianalisis menggunakan ANOVA dan dilakukan uji lanjut DMRT dengan tingkat kepercayaan 95%. Hasil penelitian menunjukkan pemberian IAA dan BAP dapat mempengaruhi pertumbuhan kalus eksplan daun anggrek C. pandurata Lindl. Pengaruh secara interaksi terdapat pada wantu munculnya kalus, tebal kalus dan Prosentase hidup kalus. Serta mampu memacu perkembangan kalus ke arah proliferatif. Sedangkan pemberian IAA secara mandiri dapat memacu penebalan kalus. Interaksi yang terbaik dalam pertumbuhan kalus eksplan daun anggrek Coelogyne pandurata Lindl. adalah A2B2 (IAA 2 mg/L dan BAP 2 mg/L) dan A2B3 (IAA 2 mg/L dan BAP 3 mg/L).
