Abstract

The addition of growth regulator is one of the critical success factors in in vitro cultures. 2,4-D as a plant regulator in media can stimulate the cell division, enlargement of the explants and promotes the formation and growth of callus. The purpose of this study was to determine the time of callus formation and to determine the best concentration of 2.4-D in inducing the growth of callus from leaf explants of Vanda sp. This research was conducted by experiment with completely randomized design, which consists of six levels of treatment concentration of 2,4-D i.e. 0 ppm; 1 ppm; 1.5 ppm; 2 ppm; 2.5 ppm; and 3 ppm. The parameters observed were the percentage of callus formation and the form of callus from Vanda sp leaf explants. The results were statistically analyzed by using MINITAB program version 17. Analysis of variance (ANOVA) was performed and the difference between means score/value was separated by F test at p < 0.05. The results showed that 2,4-D treatment give significant effect (F 5,12 = 3,20; p = 0,046 < 0,05) on the callus growth time and its percentage. Application of 2 ppm 2.4-D was the best concentration for accelerating the callus growth time (14.3 days after planting) and increasing the percentage of callus formation (83.3%). Most of callus type were proliferative callus (36.11%) and senescence callus (11.11%). The results of this research are very important to grow the callus from Vanda leaves orchid explant because it is very diffucult to grow.

Abstract

Conventionally, cultivations of Nepenthes are conducted by using seeds, cutting, and filial separation. However, there are many obstacles come both from time and technical aspect. In vitro culture is an alternative way for cultivating  N. ampullaria (Jack,). One of a technique of in vitro culture is micro cutting. BAP (6-benzyl amino purine) growth regulator could be added to optimize the growth of  N. ampullaria microcutting. The purpose of this research was to determine the effect of  BAP on the growth of Nepenthes microcutting. This research was done experimentally using Completely Randomized Design (CRD). BAP treatment consisted of 5 concentrations: 0; 0.5; 1; 1.5; 2 (ppm), each treatment were multiplied 4 times. The parameters observed were: a time of bud initiation, time of root initiation, total of leaves, total of new buds, total of roots, length of leave, length of root, and height of bud. The data obtained were analyzed with ANOVA (Analisis of Variance) and continued with 5% and 1% LSD (Least Significant Different)  test. The result showed that addition of BAP affected the growth of  N. ampullaria microcutting in total leaves, length of leave, and total of buds. LSD test proved that 0.57 ppm of BAP was optimal concentration to increase total buds, whit the value reached of 3.86. Here, we found that BAP can be utilized to enhanche N. ampullaria growth on in vitro culture. The benefit of this study is to conserve N. ampullaria in vitro using BAP at  0.57 ppm.

Abstract

ABSTRACT Cadmium is one of the heavy metals that has nephroxic effect and &nbsp;dangerous to &nbsp;human health, especially welding workers. If cadmium enters the body will bind to metallotionein protein.&nbsp; This process can increase the formation of free radical compounds, thereby induced lipid peroxidation which damage kidney organ and may cause impaired the renal function. Decreased renal function is characterized by an increase of ?2-Microglobulin and creatinine level. The aim of this study are to know effect of Cd exposure to renal function indicated by increasing of Cd, creatinine and ?2-M level. Research design used case control. Research subjects consist of thirty welding workers in Purwokerto with a period of working more than 3 years as case subjects and control subjects come from Pamijen village Baturaden. &nbsp;Data of Cd, creatinine and ?2-M level were analyzed by t independent. Result of this research showed that in case subjects occur an increase of Cd, creatinine and ?2-M level and have higher level compare with control subjects.It can be concluded that Cd exposure has an effect on renal disfunction which indicated by increasing&nbsp; blood Cd level, creatinine and ?2-M level. &nbsp; &nbsp; &nbsp; &nbsp; &nbsp; &nbsp;&nbsp;

Abstract

Patchouli (Pogostemon cablin Benth.) is an important plant used by industrial facilities to absorb cadmium (Cd) in polluted land. We performed an experiment using plant medium polluted with both Cd and lead (Pb) with added humic acid. The aims of this study were to 1) determine the effects of humic acid in growth medium contaminated with Cd and Pb on the absorption of Cd and phenol content in patchouli, and 2) determine the Cd tolerance level of the growth media. A completely randomized factorial design was used for the experiment with two factors. The heavy metals were a combination of pure PbNO3 and Cd (PC) with a ratio 1 : 1, and included five concentrations: PC0 (without PbNO3 and without Cd); PC1 (250 ppm PbNO3 + 250 ppm Cd); PC2 (500 ppm PbNO3 + 500 ppm Cd); PC3 (750 ppm PbNO3 + 750 ppm Cd); PC4 (1,000 ppm PbNO3 + 1,000 ppm Cd) and humic acid concentration (0; 6,000; 12,000; and 18,000 ppm). Each treatment was replicated three times. The parameters observed were plant biomass, Cd absorption, and phenol content. The application rate of humic acid to the plant medium containing heavy metals influenced the growth of patchouli, Cd absorption, and phenol content. An application rate of 12,000 ppm of humic acid reduced the toxicity of the heavy metals and increased the dry biomass and phenol content of patchouli.

Abstract

Based on a preliminary study conducted in PAUD Terpadu Joykids Makassar, it was found that the material provided for students in introducing English vocabulary based on theme is still limited. In this case, the researcher intended to develop thematic English material for students. This study was Research and Development adapting ADDIE model that consists of Analysis, Design, Development, Implementation and Evaluation. In developing the material the researcher concerned on developing “plant” theme material. There were 1 teacher, 2 experts and 20 students involved in this research. This study found out the need of students at PAUD Terpadu Joykids over English material. The data of students need then become the inventory need in designing and developing the material. Moreover, the material consisted of English vocabulary integrated with the theme, number, picture, simple sentence, and artwork.

Abstract

This Study aims at designing English material at PAUD Terpadu Joy Kids Makassar. A preliminary study conducted in PAUD Terpadu Joykids Makassar shows that the material for students in Intorducing English vocabulary based on theme is still limited. The material consists only one page of vocabulary without any other kind of activity that might encourage and facilitite them in recognizing the English vocabulary. In this case the researcher intended to develop thematic English material for students.This study was Reserach and Development adapting ADDIE Model that consists of Analysis, Design,Development,Implementation and Evaluation. In developing the material the researcher concerned on developing "Plant" theme material. Abstract

Character building is the most important thing to do as it is a striving system which underly behavior (Freud).   Even more, good character is more to be praised than outstanding talent. Most talents are to some extent a gift. Good character, however, is not given to us. we have to build it peace by peace by thought, choice, courage, and determination. So important it is that it is said that if there is no more character every thing is lost (Mahatma Gandhi). The best way to build it is  to develop the function of all individual potential, including cognitive, affective, and psychomotoric aspects simultanously in the context of socio-cultural interaction (in family, in  school, and in society). Character is gained by nature and nurture. It can be done begin from the golden age to the old one through the three character building components: moral knowing, moral feeling, and moral action as suggested by Lickona. They make it possible since human beings, as the best-formed creature of all, are the ones and the only creature posessing culture, and that, they can educate and be educated in terms of the model of person of character. Everybody must have character. Therefore, to apply Lickona's, one must  empower her/his language, in which she/he/ perform her/his competence in using language creativity (Chomsky) in both ordinary  and literary language. In this case, Buginese language is used as the sample.

Abstract

AbstrakThe aim of this research is: (1) to identify the forms and categories of Indonesian words that are absorbed into Buginese sentences and (2) to reveal the reasons for the use of Indonesian elements into Buginese sentences by Facebookers in the social media. Data on this qualitative research obtained from social media ‘Facebook’. The data source of this research is the Facebookers who are members of the MABBASA UUGIE KU PESBU’ group, November 2013 to April 2014 edition. Data analyzed are Buginese sentences consisting of three to five examples of Buginese sentences containing Indonesian elements in the form of words, phrases or clauses taken purposively. Furthermore, the analysis was carried out with grounded research strategies. The results of this research indicate that (1) Buginese language can survive as a means of communication within Buginese ethnic groups when writing on the social media ‘Facebook’, due to they have obtained vocabulary contributions from Indonesian in the form of the basic word, affixation word, and phrase. In word categorization, the loan words consist of nouns, verbs, adjectives, and conjunctions. Then, (2) the use of the Indonesian language elements has four main reasons, namely (a) filling in the blanks, (b) adding equivalence variations, (c) clarifying the meaning, and (d) interference. Reasons (a) to (c) can take the form of code-mixing and code-switching. AbstrakPenelitian ini bertujuan: (1) mengidentifikasi bentuk dan kategori kata bahasa Indonesia (bI) yang  terserap ke dalam kalimat-kalimat bahasa Bugis (bB) dan (2) mengungkap alasan-alasan penggunaan unsur-unsur bI tersebut ke dalam kalimat bB oleh para Facebooker di media sosial. Data penelitian kualitatif ini diperoleh dari media sosial Facebook. Sumber data penelitian ini ialah para Facebooker yang menjadi anggota grup MABBASA UUGIE KU PESBU’ edisi bulan November 2013 s.d. bulan April 2014. Data yang dianalisis ialah kalimat-kalimat ber-bB yang terdiri atas tiga sampai lima contoh kalimat ber-bB yang berisi unsur-unsur bI, yang berupa kata, frasa, atau klausa, yang diambil secara purposif. Selanjutnya, analisis dilakukan dengan upaya grounded research. Hasil penelitian ini menunjukkan bahwa (1) bB dapat bertahan hidup sebagai sarana perhubungan intern suku Bugis dalam komunikasi tulisan sosial Facebook karena memperoleh sumbangan kosakata bI yang berbentuk kata dasar, kata berimbuhan, dan frasa atau ungkapan. Dari segi kategorisasi kata, unsur-unsur serapan tersebut terdiri atas kata benda, kata kerja, kata sifat, dan kata sambung. Kemudian, (2) penggunaan unsur-unsur bI tersebut memiliki empat alasan utama, yaitu (a) mengisi kekosongan, (b) menambah variasi kesepadanan, (c) memperjelas pemaknaan, dan (d) interferensi. Alasan (a) sampai dengan (c) dapat mengambil bentuk campur kode dan alih kode.

Abstract

There have been several studies discussing the novel Sali: The Story of a Dani Woman. These studies generally use sociological literature, feminist literary criticism, and gender perspectives. This paper comes with a different perspective, namely the ethnolinguistic approach. This paper describes the linguistic forms in the novel that represent the Dani culture and reveals the cultural meaning of the linguistic forms in the novel. The linguistic data is taken entirely from the novel and the meanings obtain apart from the novel are also from other sources as listed in the references. The results of data analysis show that linguistic forms indicating the Dani culture include the level of words, phrases, clauses, or sentences. Furthermore, the cultural meaning of language forms in terms of words indicating Dani culture, namely: (1) Abambuk which means tribal chief, Dani tribal leader. (2) Pig which means a dowry given to a girl or a widow to marry. (3) Hunting which means the dominant male activity to show his self-esteem. (4) Ebeai which means a house that a wife/woman, children and husband/men may enter to gather. (5) Fugima which means a place to end life/everything for a woman/wife who has given up on her fate. 6) Honai which means a traditional house located in silimo. (7) Koteka which means penis genital cover has many meanings. The variation indicates that the more varied the koteka, the higher the class of men who wear it. (8) Noken which means a replacement for the ballot box which means a form of appreciation for the cultural values and local wisdom of the Dani community. (9) Pilamo which means a place to limit women's movement. This was a house that only men could enter to ensure that they and the war equipment were free from the dirty blood of women. (10) Sali which means wife indexer. (11) Silimo which means a family residence consisting of honai, ebeai, and pilamo, the three of which are far from each other in a certain area. Furthermore, the linguistic forms in the form of phrases are (1) Chewing greedily which means eating food in an unethique way. (2) Tribal war which means the implementation of a war caused by a member of the Dani community was being killed. (3) Polygamous marriage which means the family of the prospective wife. (4) Cutting a child's finger which means a meaningful representation of emotional pain which is important for the process of grieving the death of the child's parents. It is a symbol of grief at the loss of a family member. (5) The cry of mourning which means that family members have to cry for a long time as hard as possible to mark the mourning of being left dead by a member of a communal system 6) Cremation, or the burning of a corpse which means an attempt to take the white ashes of the corpse. (7) Relative-exchange marriage which means marry to the wife's sister in order to get a replacement from the pig keeper, the caretaker of his children and himself. Finally, the forms of clauses/sentences indicating Dani culture are: (1) Keeping the younger sister away from her twin means avoiding bad things. The younger sister in the twins is believed to be the daughter of the devil, so she must be exiled, otherwise a disaster will come that will harm the family. (2) Leaving sali on the rocks in Fugima which means the owner has died as a form of surrendering to the problems of life that surrounds her so that in a very tragic way her life is ended.

Abstract

The aim of this study was to investigate the pragmatic failure in English and Arabic students at the UIN Alauddin Makassar's Faculty of Adab and Cultural Studies. The data for the study were gathered through observation, distribution of questionnaires in the form of a discourse completeness test (DCT), and interviews. Quantitative and qualitative research data were collected in this research. This study employed three statistical models or techniques: descriptive statistics, association rules (a priori algorithm), Chi Square, and qualitative analysis. The results shos that there are three kinds of pragmatic failure that produced by the English and Arabic students such as sociopragmatic, pragmalinguistic, and psychopragmatic aspects. In addition, The researcher suggests to the next researcher to study more deeply about the types of pragmatic failure in other fields.
