Abstract

Abstrac Neutrophils are the first non-specific immune cells that successfully overcome the presence of antigens, starting with attachment and then terminate the antigen by phagocyte process. Mangosteen peel extract (Garcinia mangostana L.) possesses a compound that contains pharmacological activity. The purposes of this study were to determine the benefits of manufactured mangosteen peel extract toward increased neutrophil microbicidal activity and the most effective concentration in increasing neutrophil microbicidal activity exposed to S. mutans. Microbicidal activity of neutrophil cells was tested by firstly taking blood isolate neutrophils from peripheral veins of healthy people, secondly incubated with mangosteen peel extract, thirdly exposed to S. mutans, fourthly subcultured on a BHI-A medium, then incubated for 24 hours, and finally calculated the number of colonies by using colony counter. Results of the data were analyzed by using Kolmogorov-Smirnov, Levene, Kruskal-Wallis and Mann-Whitney. The results showed each treatment group had significant difference with value of 0,001 (p<0,05). The treatment group incubated with the mangosteen peel extract had less number of colonies compared to the group which was not given mangosteen peel extract. In conclusion, the mangosteen peel extract could increase neutrophils microbicidal activity that was exposed to S. mutans.. Abstract

Background: The prevalence of periodontal disease in Indonesia is 60%. Chronic periodontitis is one of periodontal disease which affects Indonesian. The primary cause of periodontal disease is bacterial plaque. Adhesions and accumulation of bacterial plaque can cause inflammation and destruction of periodontal tissue following by increasing of gingival crevicular fluid flows and releasing of its components, one of the components is phosphorus that excreted from sulcular epithelium. Soluble phosphorus along blood stream is from gingival crevicular fluid which the flow is increased and excreted by sulcular epithelium. Objective: To know phosphorus levels in the gingival sulcus of chronic periodontitis. Methods: The subjects of the study were patients who came to the Dental Hospital University of Jember and occupied as inclusion and exclusion criteria. The subjects who had inclusion criteria were divided into 2 groups based intra-oral examination; gingivitis and chronic periodontitis. Gingival crevicular fluid sampling was taken from tooth with gingivitis or chronic periodontitis using paperpoint. Phosphorus level measurement used UV-Vis Spectrophotometer with a wavelength of 883 nm. The results were analyzed by Mann-Whitney test. Results: There were phosphorus in the gingival crevicular fluid of chronic periodontitis and it higher than gingivitis. Conclusion: There is phosphorus level in the gingival crevicular fluid in patients with chronic periodontitis.   Keywords: Chronic periodontitis, gingival crevicular fluid, phosphorus

Abstract

Pendahuluan: Sutra cocoon laba-laba merupakan bahan alternatif yang diketahui sering digunakan oleh masyarakat tradisional untuk menghentikan pendarahan dan menutup luka. Cocoon laba-laba terdiri dari asam amino glisin dan alanin yang berkontribusi pada adhesi dan proliferasi sel. Tujuan penelitan ini adalah menganalisis potensi gel ekstrak sutra cocoon laba-laba Argiope modesta 5% terhadap jumlah sel fibroblas dan kepadatan kolagen pada penyembuhan luka gingiva. Metode: Penelitian experimental laboratories dengan the post- test only control group design. Sampel yang digunakan sebanyak 35 ekor tikus wistar jantan, dibagi secara acak menjadi 3 kelompok yaitu (a) kelompok kontrol negatif, diaplikasikan gel plasebo; (b) kelompok perlakuan, diberi gel ekstrak cocoon laba-laba 5%; (c) kelompok sehat, tidak diberi perlukaan maupun terapi. Lima belas ekor tikus kelompok kontrol negatif dan 15 ekor tikus kelompok perlakuan dibagi menjadi 3 sub kelompok hari pengamatan, yaitu hari ke-3, ke-5 dan hari ke-7 yang terdiri dari 5 ekor tikus setiap sub kelompok. Kelompok tikus sehat terdiri dari 5 ekor tikus. Perlukaan dibuat pada mukosa gingiva M1 kiri rahang bawah, kemudian dilakukan aplikasi gel. Hewan coba di euthanasia untuk selanjutnya dibuat sediaan histologi dan dilakukan pengamatan sel fibroblas dan kolagen. Hasil: Terdapat perbedaan yang signifikan (p=0,01) rerata jumlah fibroblas kelompok perlakuan (18,67±0,72) dibanding kelompok kontrol negatif (12,08±0,32) dan kelompok sehat (9,17±0,43) pada hari ke-7. Rerata kolagen berbeda signifikan (p=0,01) pada kelompok perlakuan (245,12±1,07) dibanding kelompok kontrol negatif (231,53±0,90) dan kelompok sehat (248,75±1,27) pada hari ke-7. Simpulan: Aplikasi gel ekstrak cocoon laba-laba (Argiope modesta) 5% terbukti meningkatakan jumlah sel fibroblas dan kepadatan kolagen.Kata kunci: cocoon; laba-laba; glisin; alanin; fibroblast; kolagen ABSTRACTIntroduction: Spider cocoon silk is an alternative material that traditional people often use to stop bleeding and close wounds. Spider cocoon is composed of glycine and alanine amino acids which con-tribute to cell adhesion and proliferation. This study aimed to analyze the potency of the 5% Argiope modesta spider silk extract gel on the number of fibroblasts and collagen density on gingival wound healing. Methods: Experimental laboratory research with the post-test only control group design. The samples used were 35 male Wistar rats, randomly divided into three groups, namely (a) negative control group, placebo gel was applied; (b) the treatment group was given 5% spider cocoon extract gel; (c) healthy group, not given any injury or therapy. Fifteen rats in the negative control group and 15 rats in the treatment group were divided into three subgroups on the day of observation, namely, on the third, fifth and seventh days, which consisted of five rats in each subgroup. The healthy rat group consisted of five rats. An injury was made on the mandibular left M1 gingival mucosa, and then the gel was applied. Experimental animals were euthanized for further histology preparation, and observations of fibroblasts and collagen cells were carried out. Results: There was a significant difference (p=0.01) in the mean number of fibroblasts in the treatment group (18.67±0.72) compared to the negative control group (12.08±0.32) and the healthy group (9.17±0, 43) on the seventh day. The mean collagen was sig-nificantly different (p=0.01) in the treatment group (245.12±1.07) compared to the negative control group (231.53±0.90) and the healthy group (248.75±1.27) on the day of seventh. Conclusions: The application of spider cocoon extract gel (Argiope modesta) 5% increases the number of fibroblasts and collagen density.Keywords: cocoon; spider; glycine; alanine; fibroblasts; collagen

Abstract

Background: celery leaves contain antibacterial substances that can be used as medicine. These contents include flavonoids, saponins and tannins. Purpose: the aim of this study was to determine the antibacterial effect of celery leaves (Apium graveolens L.) on the growth of Porphyromonas gingivalis (P. gingivalis). Methods: celery leaves were extracted by maceration method with 96% ethanol. The antibacterial test method against P. gingivalis using the well diffusion method on BHI-A media. The concentrations of celery leaf extract used in this study were 6.25%, 12.5%, 25%, 50% and 100%. Result: The results showed that celery leaves extract (Apium graveolens L.) didn’t have the ability to inhibit the growth of Porphyromonas gingivalis. Conclusion: celery leaves didn’t have antibacterial effect against Porphyromonas gingivalis in 6,25%, 12.5%, 25%, 50% and 100% concentrations.

Abstract

Obesity rate is increasing from year to year, both men and women in the group and increase the level of obesity is due to changes in lifestyle, especially diet. Obesity plays a major risk factor of chronic diseases, chronic diseases and various forms of oral cancer. Periodontal disease is a chronic inflammation in the tissues supporting the teeth. The purpose of this paper would like to know the relationship of obesity to periodontal disease. Obesity is likely to suffer from diabetes mellitus approximately three times more than that is not obese. The main complications of diabetes mellitus is periodontal disease. People who are obese have a possibility of periodontal disease in the oral cavity. The conclusion is an association between obesity and periodontal disease occur in the oral cavity.

Abstract

Almost people in the world got periodontal disease. It is about 50 % of adult population. Porphyromonas gingivalis (P. gingivalis) is anaerobic negative gram bacteria and played role in periodontitis, especially chronic periodontitis. Periodontal disease can be prevented by decreasing of plaque accumulation with oral rinse. Oral rinse can use traditional herbal, such as green and red betel leaf. Aim of this study was to know difference of antibacterial effectivity of green (piper betle l) and red (piper crocatum) betel leaf extract toward P. gingivalis. This research used well diffusion methods. There used 24 samples and divided 3 groups. First group used red betel leaf extract, second group used green betel leaf extract and third group used sterile aquadest. The samples were incubated for 24 hours in room temperature 37°C. Then, they were measured by inhibition zone by calipers. The result showed diameters of inhibition zone of green betel leaf was larger than red betel leaf and sterile aquadest. One way Anova test showed there was significant difference between the groups. The conclusion showed green betel leaf extract had antibacterial activity more than red betel leaf extract toward P. gingivalis.

Abstract

Level of oral hygiene have a close relationship with periodontal health status; where good oral hygiene will minimize the occurrence of periodontal disease. Aims of this study were to assess the oral hygiene status and periodontal health patients that come to Department of Periodontics of Jember University Dental Hospital at August 2009 until August 2010. This type of research is an observational study of clinical (cross sectional). A total of 155 sample (87 males and 66 females) were examined by using Oral Hygiene Index-Simplified (OHI-S) and Periodontal Index. Good oral hygiene was seen in 11% of total sample examined, 54,2% had fair oral hygiene and rest 34,8% showed poor oral hygiene. 0,6% of total sample examined had healthy periodontal, 18,1% had simple gingivitis, 41,3% had initial of periodontal disease destructive, 37,45 had periodontal disease destructive and 2,6% had terminal disease. Oral hygiene status and periodontal health examination revealed that majority of sample had fair oral hygiene and periodontal disease destructive.

Abstract

Dental caries and periodontal disease has been experienced by about 90% of the people of Indonesia. Porphyromonas gingivalis is a major cause of periodontal disease particularly periodontitis. Streptococcus viridans is a cause of the disease and root canal pulp. Both microorganisms are gram-negative and gram-positive anaerobic bacteria. Periodontal disease and dental caries can be prevented by reducing bacteria on teeth using antibacterial material. The red betel leaf extract as traditional plants can be used as an antibacterial material. The purpose of this study was to determine differences in the effectiveness of antibacterial extract of red betel leaf (piper crocatum) in Porphyromonas gingivalis and Streptococcus viridians. This study used 16 diffusion wells and divided into 2 groups: group I Porphyromonas gingivalis were given red betel leaf extract and Streptococcus viridans group II were given red betel leaf extract. The wells diffusion incubated for 24 hours at 37 ° C and it measured the inhibition zone diameter. The results showed that the average diameter of inhibition zone Porphyromonas gingivalis were given red betel leaf extract is greater than Streptococcus viridans. Independent T-test results showed significant differences in both the test group (p <0.05). Conclusion The study showed that the antibacterial power of red betel leaf extract in Porphyromonas gingivalis is more effective than the red betel leaf extract in the Streptococcus viridians.

Abstract

Background: Periodontal disease is caused by the interaction between pathogenic bacteria and plaque, without the accumulation of plaque, periodontal disease will not appear. Plaque will be visible one to two days if no oral cleaning steps are taken. The combination of antimicrobial therapy such as antibiotics and mouthwash as well as mechanical therapy such as scaling and root plain gives more effective results. However, if done in the long term it will cause resistance. To overcome this, alternative materials are needed that are used to inhibit bacterial growth, one of which is robusta coffee beans. The content of robusta coffee bean extract contains phenolic and flavonoid compounds that function as antibacterials that work by disrupting the function of the cytoplasmic and cytoplasmic membranes of bacteria.Method: In this study, 6 groups were used, namely KN, K-, K+, KP 25%, KP 50%, and KP 75%. The gingivitis model was made with wire using ligation technique for 3 days, then toothpaste was applied, brushing process, and disclosing agent was given, and the plaque index calculation was PHP (Personal Hygiene Performance). The data obtained were then analyzed using the Kruskall-Wallis test followed by the Mann-Whitney test.Result: The Robusta coffee bean extract toothpaste group at a concentration of 75% experienced a significant reduction in plaque compared to the negative control group and the 25% treatment group (p<0.05), but there were no significant differences or had the same ability when compared to the experimental group. 50% treatment group, normal control group and positive control group.Conclusion: The 75% concentration of Robusta coffee bean extract found in toothpaste is proven to be effective in inhibiting the formation of dental plaque.

Abstract

ABSTRAK Pendahuluan: Alat orthodontik cekat adalah salah satu alat yang digunakan untuk perawatan maloklusi. Bagian-bagian alat tersebut menempel pada gigi yang menyulitkan dalam pembersihan gigi, sehingga akan memudahkan akumulasi plak dan dapat menyebabkan gingivitis. Tujuan penelitian ini adalah menganalisis  hubungan antara kebersihan mulut dengan kesehatan gingiva pada pemakai alat orthodontik cekat. Metode: Jenis penelitian observasional dengan rancangan penelitian cross-sectional study. Subjek penelitian mahasiswa Fakultas Kedokteran Gigi Universitas Jember yang  memakai alat orthodontik cekat yang diambil dengan metode purposive sampling. Data penelitian diambil dengan kuesioner, pengukuran tingkat kebersihan mulut dengan OHI-S (Green dan Vermilion), dan kesehatan gingiva dengan Indeks Gingiva (Loe dan Silness). Hasil: Sebanyak 17% subjek memiliki tingkat kebersihan rongga mulut baik, 57% sedang, 26% buruk, dan rerata tingkat kebersihan mulutnya adalah sedang. Sebanyak 89% subjek mengalami gingivitis  ringan, 11% sedang, tidak ada yang mengalami gingivitis berat, dan rerata status kesehatan gingivanya adalah gingivitis ringan. Uji korelasi Pearson menunjukkan nilai korelasi (r=0,665) yang artinya terdapat hubungan yang kuat antara kebersihan mulut dengan kesehatan gingiva.  Uji regresi linier didapatkan nilai r2= 0,442 yang berarti kebersihan mulut berhubungan kuat dengan kesehatan gingiva sebesar 44,2%. Simpulan: Semakin buruk tingkat kebersihan mulut pada pemakai alat orthodontik cekat maka status kesehatan gingivanya juga semakin buruk. Kata kunci: kebersihan mulut; kesehatan gingiva; alat orthodontik cekat; OHI-S; indeks gingiva.ABSTRACTIntroductions: A fixed orthodontic device is one of the tools used for the treatment of malocclusion. The parts of the tool attach to the teeth which make it difficult to clean the teeth, so that it will facilitate the accumulation of plaque and can cause gingival inflammation.  To determine whether there is a relationship between oral hygiene and gingival health in students of the Faculty of Dentistry, University of Jember who use fixed orthodontics. Methods: Observational study with a cross–sectional study design. The research subjects were students of the Faculty of Dentistry, University of Jember who used fixed orthodontic appliances which were taken using purposive sampling method. The research data was collected by using a questionnaire, measuring the level of oral hygiene with OHI-S (Green and Vermilion), and gingival health using the Gingival Index (Loe and Silness). Results: 17 % of subjects had good oral hygiene levels, 57 % were moderate, 26 % were poor, and the average level of oral hygiene was moderate. As many as 89 % of subjects had mild gingival inflammation, 11 % were moderate, no one had severe gingival inflammation, and the average gingival health status was mild gingival inflammation. The Pearson correlation test showed a correlation value (r = 0.665) which means that there is a strong relationship between oral hygiene and gingival health. In the linear regression test the value of r2 = 0.442 means that the effect of oral hygiene on gingival health is 44.2 %. Conclusion: In users of  fixed orthodontics, the level of oral hygiene is better, the health status of the gingiva is getting better.Keywords: fixed orthodontics; gingival disease; gingival index; malocclusion; OHI–S.
