Abstract

The aim of this study was to determine genetic diversity of Calpastatin gene (CAST) in Pasundan cattle. Forty-four fresh blood samples were collected from UPTD BPPT Beef Cattle Ciamis West Java. Genomic DNA samples were extracted using High Salt method. A 523 bp fragment of Calpastatin gene was successfully amplified using PCR method. Genotyping of CAST gene was conducted by PCR-RFLP method using RsaI restriction enzyme (GT^AC). Genotypes and alleles were analyzed using software Cervus 3.0.7. Parameters were observed i.e genotypes and alleles frequencies, heterozygosity observed (Ho) and expected (He), Hardy Weinberg Equilibrium (HWE), and Polymorphic Information Content (PIC). Result showed that three variant genotypes of GG, GC and CC were found and two variant alleles of G and C. Allele G was found higher (0.77) than allele C (0.23). Population of Pasundan cattle was found polymorphism and in the Hardy Weinberg Equilibrium. Polymorphic Information Content (PIC) value showed in a moderate (0.290) condition. Values of Heterozygosity observed and expected were 0.409 and 0.355 respectively. This research concludes that there is polymorphism of CAST gene in Pasundan cattle population and has genetic diversity. This result could be used as early genetic information in exploration of Pasundan cattle.

Abstract

Telah dilakukan penelitian mengenai pengujian aktivitas antibakteri dari ektrak air daun kecombrang (Etlingera elatior) yang bertujuan memberikan bukti ilmiah keunggulan tanaman kecombrang sebagai bahan pangan fungsional. Pengujian antibakteri dilakukan dengan metode difusi cakram dan identifikasi komponen kimia dengan alat GCMS. Ekstrak air daun kecombrang bersifat antibakteri E. Coli (zona hambat 10 mm/100%), S. Aureus (zona hambat 8,663 mm/20%.  Ekstrak air daun kecombrang memiliki komponen utama 2,3-butanadiol (tR=5,28, area=29,38, kemiripan 90 %) dan fenol (tR= 6,83,area=2,26, kemiripan 90 %). Abstract

Endophytic fungi live in healthy tissues of many plants, including in medicinal plant such as Asiatic pennywort (Centella asiatica). These fungi exist in different parts of the plant as symbionts. The study aimed to isolate endophytic fungi from various parts of Asiatic pennywort of Malaysia accession and characterize their nature.  Three individual plants of Asiatic pennywort (3 months-old) were obtained from the Sringanis Medicinal Garden in Bogor. The endophytes were isolated on Malt Extract Agar. The community structures of the endophytes were analyzed based on their diversity, colonization, dominance index, and relative frequency of occurrence of the isolated endophytic fungi. A total of 78 isolates have been obtained from three individual plants and clustered into 22 morphotypes consisted of 18 morphotypes of Ascomycota and 4 morphotypes of Basidiomycota divisions. The stolons harbored more endophytes (22.9 %) followed by leaf (16.7 %), root (11.8 %), and petiole (7.6 %). The diversity index was classified as medium category with the highest result (1.91) was found in the root, followed by leaf (1.79), stolon (1.75), and petiole (1.29). The most dominant endophytes were identified as Ceratobasidium sp., Colletotrichum sp, and Fusarium sp. Ceratobasidium sp. has the highest dominance index (0.02). UPGMA cluster analysis grouped the endophytic fungi into distinct clusters based on the plant parts origin. This study implied that stolon was the the most suitable part of Asiatic pennywort for isolating endophytic fungi. Further study is required to examine the role of the endophytic fungi to produce secondary metabolites in Asiatic pennywort.

Abstract

AbstrakBiji pepaya memiliki rasa yang pahit, pedas, dan beraroma menyengat sehingga kurang diminati untuk diolah. Bahan tersebut belum banyak dimanfaatkan secara optimal, padahal potensinya sangat besar untuk dijadikan sebagai bahan pengawet makanan. Oleh karena itu, perlu dilakukan penelitian untuk menguji keamanannya. Penelitian ini bertujuan untuk membandingkan efek toksisitas subakut serbuk biji pepaya ‘Bangkok’ dan ‘California’ secara in vivo pada mencit jantan galur Swiss Webster. Penelitian dilakukan dengan metode eksperimen, terdiri dari empat perlakuan serbuk biji pepaya yang dilarutkan dengan CMC 1% (dosis 0, 400, 600, dan 800 mg/kg BB). Semua dosis diberikan pada mencit secara gavage dengan volume 0,1 mL/10 g BB. Perlakuan diberikan setiap hari selama 28 hari. Pada hari ke-29, darah mencit diambil melalui retroorbital untuk diuji SGPT dan SGOT. Perlakuan dosis serbuk biji pepaya tidak berpengaruh terhadap berat badan, kondisi fisik, dan organ visceral mencit, namun meningkatkan kadar SGPT dan SGOT. Peningkatan tersebut masih di bawah ambang batas normal SGOT (70–400 U/L) dan SGPT (25–200 U/L).Abstract Papaya seeds have a bitter, spicy, and pungent aroma so they are less desirable for processing. The material has not been used optimally, even though its potential is to be used as a food preservative. Therefore, research is needed to examine its safety. This study aimed to compare the effect of subacute toxicity of 'Bangkok' and 'California' papaya seed powder in vivo on male mice of Swiss Webster strain. The research was conducted using an experimental method, consisting of four treatments of papaya seed powder dissolved in 1% CMC (dose 0, 400, 600, and 800 mg/kg BW). All doses were given to mice by gavage with a volume of 0.1 mL/10 g BW. The treatment was given every day for 28 days. On day 29, the blood of mice was taken retroorbitally to be tested for SGPT and SGOT. The dose of papaya seed powder treatment showed no effect on body weight, physical condition, and visceral organs of mice, but increased levels of SGPT and SGOT. The increase was still below the normal threshold for SGOT (70–400 U/L) and SGPT (25–200 U/L).

Abstract

The testing has been conducted research on the antibacterial activity of aqueous extracts of flowers kecombrang against E. coli and S. Aureus. This study was aims to provided scientific evidence of excellence kecombrang plants as functional food ingredients. Antibacterial activity assays performed using dispersive method. From the results of antibacterial testing kecombrang flower water extract against E. coli concentration of 20% = 0 mm, 40% = 0 mm, 60% = 4.8 mm, 80% = 5.2 mm, 100% = 7.3 mm and the test bacteria S. aureus concentration of 20% = 8.67 mm, 40% = 9.11 mm, 60% = 12:33 mm, 80% = 12:44 mm, 100% = 13.89 mm.

Abstract

The catfish is a fish that is widely cultivated and consumed in Indonesia. The Catfish growth depand on by the availability of food and water quality. EM4 and EM10 are an example of liquid biofertilizer. The addition of EM4 in the fish pond can help the growth of the fish and maintain of the water quality. While the test of EM10 has not been done on fishery fields. The purpose of this study was to determine the effect of EM10 fertilizers on of sangkuriang catfish (Clarias gariepinus Var. sangkuriang) growth and water quality. This research was conducted in February-March 2014. Research using completely randomized design with 7 treatments and 3 replications. The treatments tested were control (A), EM4 10 ml (B), EM4 20 ml (C), EM4 30 ml (D), EM10 10 ml (E), EM10 20 ml (F), and EM10 30 ml (G ). Analysis of data were using ANOVA followed by Duncan test if there is a real effect (ᾱ = 0.05). EM10 fertilizers at concentration of 20 ml could affect to the specific growth rate on catfish in 7 days maintenance, the concentration of 10 ml at 14 days of villa maintenance and the concentration of 30 ml at 28 days of maintenance. EM10 fertilizers can affect the weight growth of catfish. Fertilizer EM10 can maintain the temperature of the water.

Abstract

Gene of C/EBPα is a gene associated with lipid composition and distribution, the gene therefore contributes in enhancing of meat quality. There is no information of sequence variations of C/EBPα gene investigation in local and exotic beef cattle in Indonesia. This research was aimed to detect the presence of sequence variations of C/EBPα gene in local beef and exotic cattle. Twelve samples were used in this study such as Bali cattle (2), Ongole grade (2), Pasundan (2), Friesian Holstein (1), Angus (1), Simmental (2), Limousin (1) and Banteng (1). Several steps of the research were DNA extraction and quantification, amplification (PCR), electrophoresis, sequencing and sequence analysis of C/EBPα gene. DNA was collected either from blood or hair bulbs.  Determination of homolog bases of C/EBPα gene was based on BLAST result while base sequence variation was analyzed by using MEGA 6. The result showed that all cattle samples were confirmed bearing the C/EBPα gene that amplified along of 1,339 bp. and annealing temperature of 54°C. It was obtained three nucleotide variations at the position of 870 (G→A), 931 (A→G) and 1196 (G→A) only found in Bali cattle and Banteng, but there were no nucleotide variations in the local beef cattle, exotic cattle and the sequence reference of Bos taurus (Japanese Black/Acc No. DQ068270.1; Hanwoo/Acc No. D82984.1; Qinchuan/Acc No. NM_176784.2). Those nucleotide variations caused changing of amino acid of Alanine to Threonine at 931 and Asparagine to Serine at 1196. Mutation at position 271 changed nucleotide Cytosine to Adenine (C271A) that found in PO and Simmental cattle. Further research is needed to confirm the changing of amino acid of C/EBPα gene in Bali cattle might affect to meat quality.

Abstract

Water distillation and purification of drought clove (Syzygium arimaticum) interest yields clove interest essential oils 6,5 % (v/w), rust colored ofchocolate, refractive index (25°C) 1,52874, specific mass (25°C) 1,0636 g/ml and condensation in ethanol 70% (1:2) limpid. Oil of clove haves the character of antibacterial to five test bacteriums ( B. subtilis, B. cereus, S. aureus, P. aeroginosa and E. coli). Result of analysis GCMS shows existence of eugenol compound (72,98%), kariaphilena (10,40%), a-humulena (1,04%) and acetate eugenol ( 15,58%). SpectraphotometerJKTI, VOL. 12, No.1, Juni 2010 analysis UV- Vis existence of absorption at Amaks 207,63 nm (ethanol) and 283,76 nm (eugenol). Analysis FTIR existence of bunch - OH (3543,39 em'), aliphatic C-H (1605,79 - 1766,90 em'), aromatic C=C ( 1433,60 - 1514,74 em'), C-Caril (1433,60-1514,74em-I) and cO (1268,44 em'). Keyword: Antibacterial, clove (Syzygium arimaticum) and essential oils.

Abstract

Kelelawar merupakan salah satu spesies mamalia yang memiliki habitat cukup berdekatan dengan aktivitas manusia dan mampu terbang dalam jarak jauh. Habitat kelelawar dengan kondisi lembab, suhu rendah dan intensitas cahaya rendah berpotensi sebagai distribusi mikroba dan menyebabkan penyakit yang disebabkan oleh cendawan dan khamir yang ditemukan di kelelawar. Penelitian ini bertujuan untuk mengetahui jenis cendawan pada habitat kelelawar. Metode tangkapan menggunakan kelelawar untuk ditangkap dan dilepaskan dengan mist net dan sampel diambil secarapurposive sampling. Sampel yang dikumpulkan adalah rambut hidung dan air liur di mulut kelelawar. Hasil penelitian menunjukkan bahwa 4 jenis cendawan dan 2 spesies khamir diidentifikasi seperti Aspergillus niger, A. fumigatus, Fusarium sp., Mucor sp., Candida sp. dan Rhodotorula sp. Cendawan dan khamir yang ditemukan di kelelawar belum diketahu efek negatif terhadap kesehatan, namun beberapa genera cendawan memiliki patogen potensial pada organisme hidup lainnya.
