Abstract

Increasing cassava production is required to maintain supply sustainability and meet seed demand. The use of splice grafting and the application of auxin IBA are expected to accelerate rooting and enhance the productivity of cassava. The objective of this research is to study the effect of IBA application on the growth of cuttings and grafted seedlings of cassava. The research was conducted from October 2024 to February 2025 at the Integrated Field Laboratory of the Faculty of Agriculture, University of Lampung, Natar District, South Lampung. The experiment was designed according to randomized block design (RBD) with factorial treatments arrangement (3×2). The first factor was IBA concentration consisted of three levels, namely without IBA (A0), 1000 ppm (A1), and 2000 ppm IBA (A2). The second factor consisted of 2 types of plant materials, namely cuttings seedlings (B1) and slanted graft seedlings (B2). The results show that the application of IBA significantly increases the growth and production of cassava compared to without IBA application. IBA at 2000 ppm is the best concentration for the observed variables. The use of planting materials, whether cuttings or slanted grafts, both enhance the yield of cassava.

Abstract

Ubi kayu merupakan tanaman pangan penting dengan beragam fungsi. Perbaikan genetik ubi kayu dibutuhkan untuk menghasilkan tanaman dengan kuantitas dan kualitas yang lebih baik. Salah satu cara yang dapat dilakukan adalah dengan melakukan perbaikan genetik secara bioteknologi melalui transformasi genetik. Proses transformasi genetik memerlukan sel target yang dapat diperoleh melalui embriogenesis somatik. Penelitian ini bertujuan untuk mengetahui pengaruh jenis dan konsentrasi auksin terhadap induksi kalus primer dan embriogenesis somatik ubi kayu klon SN. Penelitian ini menggunakan rancangan acak lengkap faktor tunggal berupa berbagai jenis dan konsentrasi ZPT, yaitu: kontrol (MS + NAA 6 mg/l), MS + Picloram (8 mg/l, 12 mg/l, dan 15 mg/l) + NAA 6 mg/l, dan MS + 2,4-D (8 mg/l, 12 mg/l, dan 15 mg/l) + NAA 6 mg/l. Hasil penelitian berdasarkan perhitungan rata-rata menunjukkan bahwa waktu muncul kalus tercepat adalah pada media dengan penambahan 2,4-D 8 mg/l + 6 mg/l NAA (9,79 hsi) dengan bobot kalus sebesar 0,091 g. Perlakuan picloram 12 mg/l, 2,4-D 8 mg/l, dan 2,4-D 12 mg/l menghasilkan persentase eksplan berkalus yang sama yaitu sebesar 92±1,1%. Persentase kalus berembrio perlakuan 2,4-D 8 mg/l adalah sebesar 44±1,8% dengan jumlah embrio sebanyak 35 embrio. Perlakuan picloram 12 mg/l memiliki persentase kalus berembrio sebesar 17±1,8% dan jumlah embrio sebanyak 8 embrio.
