Abstract

The ancient records about herbal medicine show traditional healing practices are deeply rooted in Indonesian culture. The medical use of plants decreased because of inheritance customs combined with the lack of written documentation on medicinal plants but dependent on oral information transmission. Traditional research and preservation activities of local healing plants which belong to specific cultural communities become possible through this phenomenon. Researchers conducted ethnopharmacological research about antidiarrheal medicinal plant uses that included traditional knowledge about flavonoid-containing plants within the Osing ethnic group of West Java Indonesia. The goal of this research was to verify flavonoid compounds in Osing's medicinal plants through Near Infra-Red (NIR) spectroscopy combined with chemometric methods. The evaluation of antidiarrheal potential for these compounds relied on flavonoid standard reference substances because these compounds demonstrate strong antioxidant properties which stabilize intestinal functioning. Chemometric techniques together with Near Infra-Red spectroscopy (NIR) were applied to analyze krangean leaves (Litsea cubeba) in order to establish their flavonoid compound levels. The quality assessment of multivariate calibration models shows that PLS regression produces RMSEC of 2.870 and RMSEP of 0.0124 with R2cal = 0.9931 and R2val = 0.9919. The analysis using linear discriminant analysis (LDA) succeeded in its completion with recognition rates above 90% for each discriminating class.

Abstract

This study aims to develop a method to authenticate akway bark (Drimys piperita Hook f.) infusion using UV spectroscopy and chemometrics techniques. The background initiative of this study is the consideration of the high economic value of akway bark which may potentially lead to the adulteration of the akway raw materials for traditional medicine. The sample used in this study was akway bark obtained from Manokwari, Papua, Indonesia. Samples of akway bark, cinnamon, and a mixture of both were prepared in the form of powder and then infused. The infusion was examined using UV spectroscopy to obtain the absorption value of each wavelength. Chemometrics techniques including principal component analysis (PCA) and multivariate calibration using principal component regression (PCR) and partial least squares (PLS) regression were carried out during the study. Additionally, computational discrimination using sparse partial least squares discriminant analysis (sPLS-DA) was performed afterwards. A total of 36 distinctive wavelengths were obtained. The absorption values were then used to form a PCA model. The best multivariate calibration model was derived from PCR data processing on the original spectra for both akway and cinnamon bark infusion samples. The AUC-ROC values obtained from the application of the sPLS-DA technique for each sample were 1.000, 0.956, and 0.633 for akway bark, cinnamon, and the mixture of both, respectively. Authentication of akway bark infusion has been successfully conducted on the presence of cinnamon as the adulterant.

Abstract

Alzheimer’s Disease (AD) is a leading cause of dementia, characterized by progressive cognitive decline driven in part by amyloid-β (Aβ) accumulation. The β-site amyloid precursor protein cleaving enzyme 1 (BACE-1) is responsible for initiating Aβ generation, making it a central therapeutic target. Yet, developing effective BACE-1 inhibitors has proven difficult due to structural complexity and pharmacological limitation. This study aimed to construct and validate a structure-based virtual screening (SBVS) workflow combining Vina and PyPLIF HIPPOS to facilitate the identification of promising BACE-1 ligands. The protocol was validated through 100 independent redocking experiments of the native ligand (PDB ID: 3L5F), all reproducing the crystallographic pose with RMSD < 2.0 Å. Large-scale screening of the DUDE dataset (283 active ligands; 18,100 decoys) generated interaction fingerprint, which were subsequently analyzed using Recursive Partitioning and Regression Trees (RPART) under varying prior probabilities. At the optimal prior ratio of 0.82:0.18, the model achieved an enrichment factor (EF) of 10.03, surpassing the DUDE benchmark (EF = 8.1). analysis consistently highlighted ionic interactions with Asp289 and hydrophobic contacts with Trp137 as key determinants of ligand activity. From 283 active ligands, 32 were classified as true positives, narrowing the pool of candidates and interpretable SBVS protocol and proposing a dual anchoring strategy involving Asp289 and Trp137 as a rational design principle for novel BACE-1 inhibitors in AD therapy.
