Abstract

The publicly available enhanced data of ligands and decoys for estrogen receptor alpha (ERα) which were recently published has made the retrospective validation of a structure-based virtual screening (SBVS) protocol to identify ligands for ERα possible. In this article, we present the retrospective validation of an SBVS protocol using PLANTS molecular docking software version 1.2 (PLANTS1.2) as the backbone software. The protocol shows better enrichment factor at 1% false positives (EF1%) value and the Area Under Curve (AUC) value of the Receiver Operator Characteristic (ROC) compared to the original published protocol. Moreover, in all 1000 iterative attempts the protocol could reproduce the co-crystal pose of 4-hydroxitamoxifen in ERα binding pocket. It shows that the protocol is not only able to identify potent ligands for ERα but also able to be employed in examining binding pose of known ligand. Thence, the protocol was successfully employed to examine the binding poses of α-mangostin, an ERα ligand found in the Garcinia mangostana, L. pericarp.

Abstract

Lipid nanoparticles, one of nanoparticle technology results has been developed both as a drug delivery system and as a research object. Complex lipid nanoparticle, in the drug delivery system development, has an advantage due to its less toxicity. It is important to develop a validated analytical method to determine the drug content in the system of nanoparticle lipid. This research aimed to develop a valid RP-HPLC method to determine free drug content of 4-n-butylresorcinol in the liposome matrix followed by the determination of the encapsulation efficiency (EE%). A system of the RP-HPLC method has been developed using a column of C18 and methanol-redistilled water-glacial acetic acid 79:20:1 (v/v) as the mobile phase with pH maintained at 2.1–2.6. This method was validated in the parameters of selectivity, sensitivity, linearity, accuracy, and precision. During 30 days of storage, the complex lipid nanoparticle system showed the increase in the EE% value during storage time.

Abstract

Soy milk, one of the soybean products, become more popular in recent years due to its benefit for human health. Biological activities of soybean products have been widely studied according to the presence of isoflavone aglycones content, including genistein, daidzein, and glycitein. Hence, it is important to develop an effective and efficient analytical method to provide guidance regarding appropriate isoflavone intake levels for soy milk. A reversed-phase high performance liquid chromatography (HPLC) method was developed and optimized in this study employed by R statistical software with the package of rsm. A C18 column was used for HPLC separation with the detection at 260 nm. Optimized condition for HPLC separation has been achieved with the mobile phase of methanol: water (63.26:36.74), a flow rate of 0.81 mL/min, and a column temperature of 45.31 °C. These conditions were applied in the HPLC system and successfully tested for the system suitability. Quantitative estimation was performed and resulted that the genistein, daidzein, and glycitein content in soy milk samples were 6.372, 6.273, and 2.853 µg/mL, respectively.

Abstract

Resorcinol and 4-n-butyl resorcinol have been used to improve skin health. However, these two compounds were unstable due to the oxidation process. Lipid nanoparticle formulation strategies were reported as the solution to overcome the stability problem for both resorcinol and 4-n-butyl resorcinol. Nevertheless, it is important to determine the content of resorcinol and 4-n-butyl resorcinol in lipid nanoparticle formulation. Aiming to develop the analytical method for resorcinol and 4-n-butyl resorcinol determination, a response surface methodology (RSM) was applied in the HPLC optimization stage. An optimized HPLC condition was obtained by generating a Box-Behnken design followed by multiple response analysis. It was obtained that optimized HPLC conditions due to the predictive multiple response optimization were methanol percentage of 50.0%, acetonitrile percentage of 18.1%, and flow rate of 0.6 mL min–1. This optimized condition was successfully applied and met the requirements of the system suitability test. Quantitative estimation was performed and resulted that the resorcinol and 4-n-butyl resorcinol content in lipid nanoparticles were 70.37 ± 0.47 and 95.07 ± 0.80 µg mL–1, respectively.

Abstract

Resveratrol (RSV), a natural lipophilic phytoalexin, was reported as an antioxidant and anti-inflammatory agent, which has the potential to cure diabetic wounds. However, several studies suggested the limitation of RSV, such as poor aqueous solubility, poor stability, and poor oral bioavailability. To overcome the issues, RSV was formulated as a topical nanoemulsion. It is important to ensure the quality of the dosage form by evaluating RSV load in the nanoformulation and optimizing the formula. A reversed-phase HPLC method was developed and validated prior to the load determination of RSV in the nanoemulsion formulation. The composition of triacetin-eugenol, Kolliphor® RH 40, and Transcutol® was further optimized by employing a Box-Behnken Design (BBD) to achieve the optimum composition with expected viscosity and RSV load. The HPLC method for determining RSV load was successfully validated for parameters of selectivity with the resolution of 8.487, linearity and range (r = 0.9979), precision (0.12% of RSD), accuracy (109–110% of recovery), the limit of detection (0.574 µg/mL), and limit of quantitation (1.740 µg/mL). The result of formula optimization was promising, showing the optimum composition of triacetin-eugenol, Kolliphor® RH 40, and Transcutol® at 4.44 g, 30.97 g, and 11.39 g, respectively.

Abstract

Kanker payudara termasuk dalam kasus yang paling banyak terjadi di Indonesia. Berbagai upaya penanganan kanker telah dilakukan, namun dinilai belum memuaskan mengingat efek samping yang cukup serius. Keragaman hayati mendorong pengujian bahan-bahan alam yang berpotensi sebagai agen antikanker payudara. Bandotan (Ageratum conyzoides L.) secara empiris diketahui memiliki berbagai efek farmakologis termasuk sebagai agen antikanker. Penulisan review artikel ini menggunakan metode studi pustaka. Beberapa hasil penelitian menunjukkan adanya kandungan senyawa fitokimia pada herba bandotan seperti flavonoid, chromene, terpenoid, alkaloid, minyak atsiri, kumarin, sterol, glikosida, tanin dan saponin yang terbukti memiliki aktivitas antikanker. Oleh karena itu, herba bandotan (Ageratum conyzoides L.) diyakini memiliki potensi besar sebagai agen antikanker payudara. Namun masih perlu dilakukan pengujian lebih lanjut secara in vivo maupun in vitro mengenai efektivitas dan keamanan herba bandotan sebagai agen antikanker payudara.

Abstract

Garlic (Allium sativum L.) has various potentials in overcoming diseases, one of which is as an antibacterial. The purpose of this study was to explore, analyze and review the activity of garlic (Allium sativum L.) as an antibacterial agent. This study uses a literature search method using search sites such as Google Scholar, Pubmed, ScienceDirect, AYU, NCBI, Elsevier, PlosOne, and Google Scholar. by entering keywords such as antibacterial, Garlic, and Allium sativum L. Garlic contains secondary metabolites that function as antibacterial agents such as alkaloids, tannins, saponins, flavonoids, allicin. Garlic has been shown to inhibit the growth of gram-positive and negative bacteria species Escherichia, Salmonella, Staphylococcus, Streptococcus, Klebsiella, Proteus, Bacillus, Clostridium, P. aeruginosa, K. pneumoniae and, Bifidobacterium lactis predominantly using the paper disk and well diffusion method.

Abstract

Pada akhir tahun 2019, dunia dilanda wabah COVID-19. Penyakit yang disebabkan oleh virus Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) yang dapat menular secara cepat melalui droplet pada saat seseorang batuk, bersin, bahkan saat berbicara. Penyebaran virus ini dapat dicegah dengan meningkatkan sistem imun tubuh. Jahe merah merupakan salah satu tanaman yang mengandung banyak senyawa kimia yang dapat menjadi imunomodulator bagi tubuh. Jumlah senyawa minyak atsiri, gingerol, dan diarilheptanoid yang terkandung dalam jahe merah secara berturut-turut sebanyak 194, 85, dan 28 jenis. Studi in silico dapat digunakan untuk memprediksi potensi dari jahe merah sebagai imunomodulator. Metode ini terbukti dapat menganalisis senyawa yang memiliki potensi klinis dalam periode yang singkat dan biaya yang murah. Dari hasil penelusuran beberapa penelitian dengan pendekatan in silico, diperoleh hasil bahwa senyawa kimia dalam jahe merah memiliki aktivitas inhibitor SARS-CoV-2 diantaranya adalah cyanin, gingeronone A, dan quercetin. Kata Kunci: Jahe Merah, SARS-CoV-2, Immunomodulator, In Silico, Molecular Docking   At the end of 2019, the world was hit by the COVID-19 outbreak. A disease caused by the Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) virus which can be transmitted quickly through droplets when a person coughs, sneezes, or even talks. The spread of this virus can be prevented by increasing the body's immune system. Red ginger is a plant that contains many chemical compounds that can be an immunomodulator for the body. The number of volatile oil compounds, gingerols, and diarylheptanoids contained in red ginger were 194, 85, and 28 types, respectively. In silico studies can be applied to predict the potential of red ginger as immunomodulator. This method is proven to be able to analyze compounds that have clinical potential in a short period and low cost. It was found that cyanin, gingeronone A, and quercetin were potential compounds in the red ginger with SARS-CoV-2 inhibitory according to several reports using in silico approach.

Abstract

Tingginya curah hujan dan rendahnya sanitasi dapat mengakibatkan munculnya berbagai infeksi oleh bakteri. Untuk mengatasi hal tersebut, agen antibakteri yang sesuai dapat digunakan. Resistensi antibiotik merupakan masalah global yang mendesak menyebabkan berkurangnya efektivitas antibiotik dalam mengatasi infeksi bakteri. Permasalahan ini perlu diatasi dengan pencarian alternatif senyawa dengan aktivitas antimikroba, termasuk yang berasal dari bahan alam. Rambutan (Nephelium lappaceum L.) merupakan tanaman yang berasal dari Indonesia dan Malaysia, serta tersebar di Asia Tenggara. Tanaman rambutan telah terbukti memiliki khasiat dalam pengobatan, seperti antelmintik dan antidiare. Beberapa penelitian ilmiah telah menunjukkan bahwa daun, biji, dan kulit buah rambutan memiliki aktivitas antibakteri. Aktivitas tersebut ditunjukkan oleh ekstrak atau fraksi yang diperoleh melalui berbagai metode dan pelarut yang berbeda. Aktivitas antibakteri ini dapat timbul karena adanya kandungan berbagai senyawa, termasuk senyawa-senyawa fenolik, seperti geraniin, ellagic acid, quercetin, dan corilagin. Beberapa penelitian telah menunjukkan adanya aktivitas antibakteri dari tanaman rambutan terhadap bakteri yang resisten. Artikel review ini disusun menggunakan metode studi pustaka melalui penelusuran artikel penelitian, artikel ilmiah, dan review jurnal pada database elektronik seperti internet, Google Scholar, Pubmed, ScienceDirect, dan Elsevier. Artikel review ini bertujuan untuk merangkum informasi hasil penelitian terbaru terkait aktivitas antibakteri tanaman rambutan yang diharapkan dapat menjadi sumber informasi para peneliti di masa depan.

Abstract

Seledri merupakan salah satu tanaman yang digunakan sebagai penyedap olahan masakan. Diketahui juga seledri memiliki aktivitas sebagai antioksidan, antiinflamasi dan antibakteri. Oleh sebab itu, telaah pustaka ini bertujuan untuk mengkaji potensi antibakteri seledri (Apium graveolens L). Penelitian terkait aktivitas antibakteri pada Seledri sebagian besar dilakukan menggunakan sampel segar atau simplisia dari daun seledri yang diekstraksi, tetapi ada beberapa penelitian menggunakan biji sebagai bahan uji. Metode uji aktivitas antibakteri pada seledri dilakukan dengan metode difusi cakram disk, metode dilusi cair Kirby dan Bauer yang dimodifikasi, metode difusi cakram, metode dilusi, metode difusi sumuran, metode difusi wells. Hasil uji aktivitas antibakteri menunjukkan bahwa seledri mengandung senyawa flavonoid, fenolik, tanin dan saponin sebagai penghambat aktivitas antibakteri terhadap bakteri Staphylococcus aureus, Staphylococcus epidermidis, Escherichia coli, Streptococcus mutans, Actinomyces sp, dan Lactobacillus acidophilus.
