Abstract

Screening of glucoamylase enzyme producing fungi obtained from sago dregs soil at Tapakis district of Padang Pariaman have been done. The screening was conducted by incubating of sample on Potato Dextrose Agar media previously added with starch 1% as an inducer. Incubation was performed during 120 hours at temperature of 20-250C. The existence of glucoamylase enzyme producing fungi were indicated with the disappearance of lugol indicator solution colour around the colonies. It was found that there were 2 isolates of fungi which produced glucoamylase. Following identification, it could be concluded that isolates were Aspergillus sp. and Penicillium sp.

Abstract

A Study on the isolation and qualitative test  of cellulose producer fungus from waste product soil of Saccharum officinarum in Tabek Patah West Sumatera has been done by using  Czapek Dox Agar (CDA) selective media. The selulolitic ability test of this isolated fungus by giving the congo red solution on the surface of medium that was enriched with Carboxy Methyl Cellulose, the formed of transparent area around the colonies showed that the fungus has the capability to decomposed the cellulose. The hydrolitic enzyme in glucose form was analyed using Fehling solution and Thin Layer Chromatograph. The result from this research showed that the sample consist of two species of decomposed cellulose fungus Aspergillus terreus Thom and Rhizopus sp.

Abstract

The skin of Mangosteen fruit (Garcinia mangostana L.) is widely used as traditional medicine, for it contains a lot of xanthone compounds. Today, there are many products from mangosteen pericarp extract being sold on the market, one of which is the preparation of mangosteen pericarp capsules. Determining the total xanthone content in the preparation of mangosteen pericarp capsules (Garcinia mangostana L.) circulating on the market needs to be done to calculate the levels of xanthone that are not included in the preparation label. The results of the analysis with UV-Visible Spectrophotometry obtains the maximum absorption wavelength at 243 nm and the linear regression equation of the calibration curve that is y=0,0568+0,0727x with a value of r=0,9999. Validation of the analysis method showed the results of intraday precision obtained % RSD 0,34%, 0,17%, and 0,16%, interday precision of 0,23%, 0,35%, and 0,16%. The accuracy obtained was mean % recovery 95,55%, limit of detection 0,35 μg/ml and limit of quantitation 1,15 μg/ml. The results of the determination of the total xanthone content in the mangosteen pericarp capsules were 100,8 μg/mg in sample A; 197 μg/mg in sample B; and 50,2 μg/mg in sample C.

Abstract

The research aims to determine the effect of repeat frying and duration of storage to rancidity coconut oil used thiobarbituric acid (TBA) method. The sample used were coconut oil with three different brands namely coconut oil A, B, dan C. The highest TBA number of samples coconut oil A, B, dan C was after three frying and 2 days of storage, TBA number was 0.879, 0.694, and 1.336 mg malonaldehyde/Kg sample. Based on data, can conclude samples coconut oil A, B dan C has under gone rancidity, but still below the maximum limit set by Badan Standarisasi Nasional (1991) was 3 mg malonaldehyde/Kg sample. The quality coconut oil A, B, dan C still below the maximum limit set by Badan Standarisasi Nasional (2013) such as acid number was 0.6 mg KOH/g, peroxide number was 2 meq/kg, saponification number was 196-206, iod number was 45-46, and free fatty acid was 0.3 %.

Abstract

It has been the research about determination level of vitamin B1 in soy bean and tempe. The determination level of vitamin B1 was measured by spectrophotometry visible method. The result is level of vitamin B1 in soy bean higher than level of vitamin B1 in tempeh. Which level of vitamin B1 in soy bean and tempeh is about 1.0213 % ±0.0188% and 0.2960 % ±0.0095%. It show that the proces manufacturing, like washing, bathing, boiling can affect the level of vitamin B1. Statistic analyze anova one way by using SPSS 17 program shows there are significant different (p<0.05) in the level from each sample.

Abstract

A study on protein content of fresh mussel shells, curry soup mussel shells and fried mussel shells by using Kjeldahl method had been researched. The procedur involve destruction, destilation and titration. Each sample were destructed with consentrated sulfuric acid and catalyst mixture of selenium. The solution of sodium hydroxyde was then added to the resulting solution and the amonia liberated was destilled into solution of hydrochloric acid prepared in excess. Its then the hydrochloric solution was titrated with solution of sodium hydroxide 0.1 N using methyl red as and indicator. Protein content can be calculated by multiplying the total nitrogen content of the sample with protein conversion factor (total nitrogen X 6.25). The highest protein content is fried mussel shells 7.1491 % ± 0.0249, than curry soup mussel shells 6.5771 % ± 0.1095 and the last fresh mussel shells 6.3927 % ± 0.0206.

Abstract

A study on the preparation of sodium carboxymethil cellulose (Na-CMC) from celulose powder that was isolated from the stems elephant grass (Pennisetum purpureum Schumach) has been performed. Cellulose was purified using 17.5% of sodium hydroxide solution, then bleach, dried and crushed to obtain alpha cellulose. Alfa cellulose was eterified with sodium chloroacetat resulting sodium carboxymethyl cellulose, furthermore characterized: organoleptic, losses on drying, chemical identification, pH test and test with fourier transform infrared (FT-IR). The results showed that of 300 g stems elephant grass yield alpha cellulose as much as 92,.54 g and 25 g of alpha cellulose produced  Na-CMC as 16.92 g. Based on the characterization made Na-CMC and Na-CMC Merck®  meet Indonesian Pharmacopoeia 3th edition requirement.

Research has been done on the determination of caffeine that leaves Kawa Coffee  ( Coffea robusta Linden ex DE Wildem ) by using  Soxlhetation method. Abstract

The determination of phenolic compounds and it’s antioxidant activity of a few tea samples of sacked tea and unsacked tea which is found at the market has been done. The Acquisition of phenolic compound content done by using Folin-Ciocalteau method and it’s antioxidant activity measurement using DPPH as an oxidant (free radical), gallat acids used as the comparison. The results showed that four samples tested unsacked tea KA has the strongest antioxidant activity among the other three samples, because It has the lowest IC50 value. The  IC50 value influenced by it’s phenolic concentration. The phenolic content from unsacked tea KA obtained 39.5487 mg/g, while on samples of tea sacked KA, sacked SW, and unsacked 2K respectively:  19.754 mg/g; 33.9279 mg/g ; and 27.9814 mg/g.

Abstract

A study concerning the manufacture of calcium lactate from the limestone hills Tui Leopold. Limestone sample of  30 g was dissolved in 6 M HCl, filtered and the filtrate was added 6 M NH4OH to pH 10. Then added lactic acid 1 M. The precipitate is filtered, washed, dried in an oven at a temperature of 1050 C. The results obtained calcium lactate as 6. 2084 g. To determine the calcium content in limestone identification and assay of calcium, limestone is used as much as 1 g of calcium levels and obtained percentage is 36.08 %. Examination of limestone calcium lactate include organoleptic examination, solubility, qualitative identification, examination and the melting point of the assay by the method complexometry. The third difference in the weight of calcium lactate calcium percentage of the results obtained, for a weight of 1 g as much as 77.13 % w/w , weight of 2 g as much as 90.28 % w/w, while the weight of  3 g as much as 81.42 % w/w. From the data of calcium lactate assay with a sample of 7.93 g drying results have calcium levels that the percentage purity 82.94 % w/w, and the results of testing the melting point is 2280 C calcium lactate. So natural limestone hill Tui Leopold has a high purity calcium levels that can be made of calcium lactate.
