Abstract

Tujuan penelitian ini adalah untuk mengetahui pengaruh perlakuan superovulasi selama 48 jam sebelum pemotongan ternak terhadap jumlah folikel,  kualitas oosit, dan residu hormon gonadotropin eksogen. Penelitian ini menggunakan domba betina dewasa sebanyak delapan ekor yang dibagi menjadi dua kelompok masing-masing terdiri dari empat ekor domba.  Siklus estrus diamati dengan cara melakukan pengamatan/deteksi estrus baik secara visual maupun dengan menggunakan pejantan pengusik (teaser). Perlakuan superovulasi dilakukan dengan memberikan injeksi intramuskular 500 i.u. PMSG (Folligon, Intervet) pada hari ke-10 dari siklus estrus.  Pemotongan dilakukan 48 jam kemudian, kemudian diambil ovariumnya dan segera dievaluasi. Pengujian residu hormon dilakukan menggunakan metode HPLC (High Performance Liquid Chromatography), sampel diambil dari serum darah. Parameter yang diamati meliputi (1) Jumlah folikel berdasarkan ukuran dan, (2)  Jumlah oosit berdasarkan kualitas, (3) Residu hormon gonadotropin eksogen (PMSG) pada ternak yang diberi perlakuan.. Metode penelitian yang digunakan adalah metode eksperimental.  Data yang diperoleh dianalisa menggunakan uji t dengan ulangan sama.  Hasil penelitian menunjukan bahwa tidak terdapat perbedaan yang nyata antara domba kontrol dengan domba yang mendapat perlakuan injeksi PMSG dalam hal jumlah folikel, jumlah oosit maupun kualitas oosit, namun jumlah folikel ukuran sedang (2-6 mm) dan oosit grade A pada domba yang diberi PMSG lebih banyak dibandingkan pada domba kontrol.  Hasil pengujian dengan HPLC ditemukan kadar hormon PMSG sebanyak 240,62 i.u/ml.Kata Kunci :  superovulasi, pmsg, domba

Abstract

The aims of the research was to determine the effect of antioxidants addition on liquid semen quality of young ram and knowing which antioxidants have the most excellent effect. The research was conducted experimentally with completely randomized design (CRD). There were three treatments, that were P0 = semen added diluent without the addition of antioxidants as a control, P1 = semen added diluent have been added 9,3 mM glutathione, P2 = semen added diluent have been added 9,3 mM alpha tocopherol. Each treatment was repeated six times. The parameters consist of sperm motility and intact plasma membrane (IPM). The result showed that the addition of antioxidants can maintain liquid semen motility of young ram on day 6, 9, and 11, while addition of antioxidant has no significant effect on IPM. Result showed that glutathion is the best antioxidants that can maintain motility on day 6, 9 and 11 is glutathione with the average 72,28%, 62,47%, and 54,43%, nevertheless for alpha tocopherol with average 58,11%, 48,13%, and 41,95% gave result is not significant to P0 motility with average 62,29%, 53,08%, and 42,36%. It is concluded that addition of antioxidant can maintain liquid semen motility on day 6, 9, and 11 but can not give effect to the IPM, and the addition of antioxidant glutathione give the most excellent effect to liquid semen motility.

Abstract

The aims of this study was to determine the right time for breeding after hormonal induction with the application of Artificial Insemination to increase pregnancy. The research material were 20 local goat. Estrous synchronization technique is performed using an intravaginal implant of EAZI-BREED ™ CIDR®, then injecting Pregnecol TM serum Gonadotrophin 400 IU intramuscularly 48 hours before the removal of EAZI-BREED ™ CIDR®. The length of intravaginal implant of EAZI-BREED ™ CIDR® is divided into two groups, 8 (A) and 16 (B) days. To analyze the hormone progesterone (P) blood samples are taken once every twelve hours to 72 hours after the removal of EAZI-BREED ™ CIDR®. Artificial insemination is done by cervical technique using frozen semen. The results of the research showed that estrous synchronization with the duration of intravaginal implant EAZI-BREED ™ CIDR® in the two groups was not significantly difference in terms of response, onset or duration of estrus. However, the percentage of pregnancy rate after insemination was different, where the pregnancy rate in group A was lower than group B (37.50 vs 50.00%).

Abstract

Pasundan cattle spread across the West Java region, but their population decreased from 2014 to 2019. Accelerated population increase has been carried out through the artificial insemination (AI) and it is hoped can help meet the need for meat. AI program can be accompanied by the application of sperm sexing so it can meet the sex according to purpose. The method of albumin column is sexing method based on morphometric data. The aim of this research was to identify the X and Y crhomosome bearing sperm morphometrically as a basis for sperm sexing in Pasundan cattle semen. This research used frozen semen from seven Pasundan cattle. Sperm morphometric observations were carried out using an Olympus microscope equipped with the DP2-BSW-E application. The parameters observed included the length, width and area of the sperm head and the proportion of X and Y chromosome bearing sperm. Two straws were used from each bull, each straw was counted as 200 sperm, so the number of sperm measured was 400 sperm per bull. The data was analyzed quantitatively descriptively. The results of measurements on seven males showed that the sperm head of Pasundan cattle had a minimum-maximum length of 7.68 - 11.36 µm; width 3.97 - 6.30 µm; sperm head area 31.09 - 51.12 µm2. The percentage 41.00-55.50% for X- and 44.50-59.00% for Y- chromosome bearing sperm, with average 50.25% for X- and 49,75% for Y-chromosome bearing sperm. It was concluded that the X and Y chromosome bearing sperm have balanced natural proportion, so the sperm sexing needs to be done to increase the chances of birth of the sex according to purpose.

Abstract

Placental retention or retensio secundinae is one of the reproductive disorders that often occurs in dairy cows. This will interfere reproductive efficiency, including Days Open and Calving Interval. Retained placenta is defined as the failure to completely pass all of the placenta from the uterus (cotyledon villi) for more than 8 hours after parturition. This study aimed to determine the relationship between parity and placental retention and its impact on lambing days and lambing distance in dairy cows. The research was conducted at North Bandung Cattle Farming Cooperative (KPSBU) Lembang. The method used was data analysis using descriptive analysis and path analysis. The results showed that parity has a significant relationship with placenta retention, and placenta retention with increasing parity can affect days open and calving interval in dairy cows at KPSBU Lembang. Path analysis found that the t-statistical results had exceeded 1.96 with a significant p-value that did not exceed 0.05, so there was a significant relationship between the variables in the study. Dairy cows that have experienced retained placenta at North Bandung Cattle Farming Cooperative (KPSBU) Lembang have low reproductive efficiency, characterized by long open days and calving intervals exceeding the expected time.

Abstract

Kambing Peranakan Etawah adalah ternak yang populer dikembangbiakan di Indonesia dikarenakan dapat memenuhi kebutuhan protein hewani masyarakat.  Dalam menunjang program perkembangbiakan kambing tersebut, usaha yang dapat dilakukan adalah memprogram keturunan sesuai dengan tujuan pemeliharaan. Sexing adalah cara terbaik yang dapat membantu dalam program perkembangbiakan. Penelitian dilakukan untuk mengetahui pengaruh dari pemberian kadar Resiquimod (R848) pada keutuhan membran plasma hasil sexing dengan metode swim up. Metode ini dapat menyeleksi spermatozoa berkromosom X dan Y, dengan prinsip akan menguji motilitas spermatozoa terbaik dengan cara bergerak naik ke lapisan atas. Pengujian kualitas spermatozoa tersebut meliputi parameter keutuhan membran pada setiap lapisan. Penelitian ini merupakan percobaan dengan analisis statistik metode Rancangan Acak Lengkap (RAL). Penelitian ini diberikan empat perlakuan, yaitu P0 (tanpa R848), P1 (R848 0,3 μM), P2 (R848 0.6 μM), dan P3 (R848 0,9 μM). Setiap perlakuan dilakukan pengulangan sebanyak tujuh kali. Hasil penelitian menunjukkan bahwa keutuhan membran plasma lapisan atas dan bawah memiliki nilai persentase dengan rentang 80,10% - 83,53%. Dapat disimpulkan bahwa penambahan resiquimod tidak memberi pengaruh nyata terhadap keutuhan membran plasma spermatozoa kambing Peranakan Etawah. Etawah crossbreed goats are popular livestock bred in Indonesia because they can meet people's animal protein needs. In supporting the goat breeding program, efforts that can be made are to program the offspring according to rearing objectives. Sexing is the best way that can help in the breeding program. The research was conducted to determine the effect of administering levels of Resiquimod (R848) on the integrity of the plasma membrane resulting from sexing using the swim up method. This method can select spermatozoa with X and Y chromosomes, with the principle that it will test the motility of the best spermatozoa by moving up to the top layer. Testing the quality of spermatozoa includes membrane integrity parameters in each layer. This research is an experiment using statistical analysis using the Completely Randomized Design (CRD) method. This research was given four treatments, namely P0 (without R848), P1 (R848 0.3 μM), P2 (R848 0.6 μM), and P3 (R848 0.9 μM). Each treatment was repeated seven times. The results showed that the integrity of the upper and lower layers of the plasma membrane had a percentage value in the range of 80.10% - 83.53%. It can be concluded that the addition of resiquimod does not have a significant effect on the integrity of the plasma membrane of spermatozoa in Etawah crossbreed goats.

Abstract

The study aims to determine the effect of resiquimod (R848) levels on the motility of spermatozoa of Etawah Crossbreed goats resulting from separation, and  to determine the opimum  level of use of resiquimod (R848) on the motility of spermatozoa of  Etawah Crossbreed goats resulting from separation. The object used in this research was fresh semen from 4 years old Etawah Crossbreed goats. This research used a Completely Randomized Design method with 4 treatments (P0 = 0 µM R848, P1 = 0,3µM R848, P2 = 0,6 µM R848, dan P3 = 0,9 µM R848). Based on the research results, it shows that R848 has a significant effet on sperm motility in the lower layer but has no effect on sperm motility in the upper layer resulting from the separation. The research data was followed by the Duncan test. The result showed that motility of P3 (80,70%) was significantly lower than P0 (89,77%), P1 (89,52%), dan  P2 (89,38%). It was cocluded that resiquimod (R848) had an effect on lower layer spermatozoa motility, with an optimum level of 0,3 µM in Etawah Crossbreed goat spermatozoa.

Abstract

Penelitian ini bertujuan untuk mengetahui pengaruh pemberian glukosa dan inhibitor glikolisis terhadap viabilitas sperma domba lokal. Penelitian ini dilaksanakan pada bulan Januari - Mei 2024 dengan objek penelitian berupa semen dari tiga pejantan domba lokal yang berumur 2-3 tahun. Metode Rancangan Acak Lengkap (RAL) digunakan pada penelitian ini dengan 4 perlakuan yaitu tanpa pemberian glukosa (P0), pemberian glukosa 27 mM (P1), 54 mM (P2), dan 108 mM (P3) dengan inkubasi pada 37°C selama 0-60 menit. Setiap perlakuan diulangi sebanyak 5 kali. Kemudian setelah perlakuan optimal pada eksperimen pertama diketahui, dilanjutkan dengan eksperimen kedua dengan pemberian 3-MCPD 0, 0,5, 1, dan 2 mM pada media bufer yang mengandung glukosa 27 mM. Setiap perlakuan diulang 5 kali. Data hasil penelitian dianalisis menggunakan sidik ragam dan dilanjutkan dengan uji Tukey HSD. Hasil penelitian menunjukkan bahwa pemberian glukosa pada dosis tertentu secara signifikan mempertahankan viabilitas sperma. Pemberian glukosa 27 mM (P1) nyata mampu mempertahankan viabilitas lebih tinggi yaitu 75 ± 2,7%, dibanding P0 (67 ± 2,4%), P2 (74 ± 0,71%), dan P3 (72 ± 0,69%) yang berarti bahwa sperma menggunakan glukosa untuk mempertahankan integritas membrane. Namun, pengaruh glukosa dalam mempertahankan viabilitas sperma terhambat dengan kehadiran 3-MCPD (0,5 mM, 65,8 ± 1%; 1 mM, 61,6 ± 1,2%; dan 2 mM, 55,4 ± 2,2%). Kesimpulan dari penelitian ini yaitu pemberian glukosa dapat mempertahankan viabilitas sperma kambing melalui proses glikolisis.The purpose of this research was to determine the effect of glucose supplementation and 3-MCPD (a glycolysis inhibitor) on sperm viability of Local Ram. This research was conducted from January - May 2024 with the object of research was sperm from 2 to 3-year-old male Local Ram. The research applied Completely Randomized Design (CRD) with four treatments i.e.: sperm without glucose supplementation (P0), 27 mM glucose (P1), 54 mM glucose (P2), and 108 mM glucose (P3) with incubation at 37°C for 0-60 min. Each treatment was replicated five times. Then after the optimal treatment in the first experiment was known, continued with the second experiment by adding 0, 0.5, 1, or 2 mM 3-MCPD in a buffer medium containing 27 mM glucose. Each treatment was replicated five times. The obtained data were analyzed using the ANOVA and Tukey HSD test. The results showed that addition of glucose at certain doses significantly maintained the viability of sperm. The addition of 27 mM glucose (P1) has the highest sperm viability by 75 ± 2.7%, compared to P0 (67 ± 2.4%), P2 (74 ± 0.71%), and P3 (72 ± 0.69%), suggesting sperm utilise glucose to support their membrane integrity. However, the supporting effect of glucose on sperm viability was abolished by the presence of 3-MCPD (0.5 mM, 65.8 ± 1%; 1 mM, 61.6 ± 1.2%; and 2 mM, 55.4 ± 2.2%). In conclusion, ram sperm utilise glucose to maintain their viability via glycolysis.

Abstract

The objective of the research was to determine the estrous profile of Etawah Crossbred goats after estrous synchronization with different methods. Eighteen does aged 12-24 month old were divided in three groups to receive estrous synchronization treatments (T1 = 14 days intravaginal implant of 60 mg of progesterone (MPA), T2 = two times injection of 5 mg PGF2α (lutalyse) in 11 days interval, and T3 = 10 days of intravaginal implant of 60 mg of progesterone (MPA) + injection of 5 mg PGF2α 48 hours before removal) with six replications. The parameters consisted of estrous behaviour, changes in size and colour of vulva, and duration of estrus when the number of superficial and keratin cells were dominating in the vaginal mucus cell. Data from estrous behaviour and score of vulvar colour was analyzed using Kurkal Wallis test, while onset of estrus, size of vulva slit and estrous duration was analyzed using ANOVA and Duncan test. The result showed that estrous behaviour and changes in color and size of vulva were not significantly different, but estrous duration was significantly different. Estrous duration in T1 (31.30 hour) and T2 (31.10 hour) was significantly longer than that of T3 (11.36 hour). It is concluded that different methods of estrus synchronization affected estrous quality equally but it affected the estrous duration differently based on vaginal mucus cells. Treatment implant vaginal sponge content progesterone for 14 days and double injection of PGF2α with 11-day interval given longest estrous duration.

Abstract

The research aims to evaluate proportion of X-Y chromosom bearing sperm after sexing of local ram semen with different combination of BSA concentration. The research object was ten ejaculated semen of local ram, three years old. The research design used CRD (completely randomize design) with four treatments of BSA concentration combination on upper and bottom layer (T1: 3% & 6%, T2: 4% & 6%, T3: 5% & 10%, and T4: 6% & 12%), and 10 repetitions. Data analysis using analysis variance and Duncan’s Multiple Range Test. The result showed that combination of BSA concentration was significantly effect on proportion of X-Y chromosome bearing sperm of local ram sperm. The higher average proportion of X- and Y- chromosome bearing sperm was obtained at combination 5% (75.55±1.09% for X) at upper layer and 10% BSA (76.45±1.12% for Y) of bottom layer. The conclusion is that combination of BSA concentration significantly effect on proportion of X-Y chromosome bearing sperm, and concentration of 5% and 10% BSA at upper layer and bottom layer gave the higher proportion of X-Y chromosome bearing sperm of local ram sperm.
