Abstract

ABSTRACTMongan et al., 2011. Production and fractionation of liquid smoke from corn cob to inhibit lipid peroxidation oflaying fish (Decapterus russelli).Corncob is a cellulose material and can be made to liquid smoke by pyrolysis. Liquid smoke from pyrolysis canbe purified using adsorbent. Chitin is an adsorbent produced by crustacea shell. Liquid smoke purified by chitincan act as antimicroba and antioxidant. Layang fish is very abundantly in Gorontalo Province, so preservativetechniques were needed to prevent lipids peroxidation in fish. The aims of this research was to determineantioxidative activity of liquid smoke from corncop to prevent lipids peroxidation on layang fish.This research using laboratory experiment methods to analyze total phenolic content, free radical scavengingactivity, total antioxidant and lipids peroxidation on layang fish by spectrophotometer UV-vis. This researchusing complete random design (rancangan acak lengkap, RAL), thus obtained data were analyzed usingstatistic. Significant difference between groups were analyzed using ANOVA with p<0,05. If there is significantdifference between groups, data were analyzed using Duncan’s multiple range test (DMRT).The results showed that liquid smoke can be produced using corncop by pyrolysis is 31,7 %. Fractioned liquidsmoke using chitin obtained 4 fractions were fraction I, II, III and IV. Liquid smoke and fractioned liquid smokecontain total phenolic content are 280, 273, 280, 286 and 301 mg/kg gallic acid, respectively. Free radicalscavenging activity from liquid smoke and fractioned liquid smoke are 87,39, 86,08, 89,29, 89,90 and 93,73%,respectively. Although, total antioxidant from liquid smoke and fractioned liquid smoke are 59, 35, 51,32, 51,57,52,74 and 53,58 ppm, respectively. Fraction IV of liquid smoke fractioned with chitin was used in antioxidativeactivity of laying fish test. Fraction IV with concentration 25% can inhibit lipid peroxidation on fresh layang fishby 19 mikromolar for two days. Although layang fish without liquid smoke shows high lipid peroxidation by 24,96mikromolar. Fraction IV with concentration 25% can inhibit lipid peroxidation on layang fish by 18,28 mikromolarfor two days. Antioxidative activity from liquid smoke caused by it phenolics compound that act as antioxidant.As conclusions of this research, liquid smoke can be produced from corncob and fractioned with chitin. Liquidsmoke contain phenolics and act as antioxidant. Liquid smoke can inhibit lipids peroxidation in layang fish. Thisresearch suggested that liquid smoke can be used to inhibit lipid peroxidation on layang fish..Keywords : liquid smoke, corn cob, peroxidation, layang fish

Abstract

ABSTRAK Penelitian ini bertujuan untuk mengisolasi dan mengkarakterisasi fraksi flavonoid dari ekstrak etil asetat tongkol jagung serta menentukan aktivitas singlet oxygen quenching. Tongkol jagung diekstraksi dengan cara refluk menggunakan etanol 80% selama 2 jam. Setelah itu disaring dan filtratnya diuapkan dengan rotary evaporator. Ekstrak etanol disuspensikan dalam air dan diekstraksi berturut turut dengan petroleum eter, etil asetat, butanol dan air. Fraksi pelarut terbaik difraksinasi dengan kromatografi kolom menggunakan silika gel 60 dan eluen n-heksana: etil asetat (4:6). Fraksi dievaluasi dalam singlet oxygen quenching dengan sistem reaksi fotooksidasi asam linoleat (0,03 M) dalam sistem emulsi yang mengandung 5,68 x 10-3 mM eritrosin sebagai sensitiser dan campuran reaksi disinari cahaya fluoresen (4000 lux) selama 5 jam. Hasil karakteristik senyawa hasil isolasi (F2) berdasarkan hasil analisa UV-Vis terdapat 2 pita serapan pada panjang gelombang 289 nm (pita I) dan panjang gelombang 313 nm (II) yang mengindikasikan bahwa senyawa hasil isolasi termasuk golongan flavonoid jenis flavanon atau dihidroflavon. Spektra UV senyawa terisolasi dengan pereaksi geser kemungkinan memiliki gugus hidroksi yang terletak pada atom C-5 atau C-7. Analisis spektrofotometer infra merah (IR) menunjukkan adannya bilangan gelombang 3375 cm-1 (OH, fenol), 2920 cm-1 (CH), 1462 cm-1 (C=C aromatik), 1604 cm-1 (C=0) dan 1031 cm-1 dan 1167 cm-1 (C-O-C). Isolat (F2) memiliki aktivitas singlet oxygen quenching (antiphotooxidation) terhadap fotooksidasi asam linoleat yang mengandung eritrosin sebagai sensitizer.ABSTRACT The objective of this research was to isolate and characterized flavonoid fractions from corn cob ethyl acetate extract and to determine the singlet oxygen quenching activity. Corn cob was extracted with a reflux method using 80% ethanol solvent for two hours. After the extraction, filttering was occurred and the filtrate was evaporated with rotary evaporator. The ethanol extract was suspended in water and continually extracted with petroleum ether, ethyl acetate, buthanol and water. The best solvent fraction was fractionated with column chromatography using silica gel 60 and n-hexane and ethyl acetate (4:6) as eluent. The fraction was evaluated in singlet oxygen quenching with the photooxidation of linoleic acid (0.03 M) reaction system inside an emultion system that contains 5,68 x 10-3 mM of eritrosin as sensitizer and the mixed reaction was lit by a fluorescent light (4000 lux) for 5 hours. The characteristic result of the isolated compound (F2) based on the UV-Vis analysis result there are two absorptopn band at 289 nm (band I) and 313 nm wavelength (band II) that indicate that the isolated compound belongs to the flavonoid group as a flavanon or a dihydroflavon type. The UV spectra of isolated compound with shifting reagent showed having a hydroxyl group located at the C-5 or C-7 atom. The infrared (IR) spectrophotometer analysis showed the wave number 3375 cm-1 (OH, phenol), 2920 cm-1 (CH), 1462 cm-1 (C=C aromatic), 1604 cm-1 (C=O) and 1031 cm-1 and 1167 cm-1 (C-O-C). The isolate (F2) has the singlet oxygen quenching activity (antiphotooxidation) on the photooxidation of linoleic acid that contain erythrosine as sensitizer.

Abstract

ABSTRAK Penelitian ini bertujuan untuk mempelajari komposisi proksimat dan potensi antioksidan dari biji jagung Manado kuning. Tahap pendahuluan penelitian ini yaitu analisis proksimat. Kemudian biji jagung Manado kuning ini diektraksi menggunakan cara refluks selama dua jam dengan masing-masing pelarut etanol, etil asetat dan n-heksan. Pada tahap kedua yaitu ditentukan kandungan fenolik dan total karotenoid. Pada tahap ketiga penelitian ini yaitu pengujian aktivitas antioksidan menggunakan radikal bebas DPPH dengan spektrofotometer UV/Vis. Hasil penelitian menunjukkan pada pengujian proksimat bahwa serbuk biji jagung Manado kuning  memenuhi syarat Standar Nasional Indonesia, tetapi berbeda dengan kadar abu. Kandungan total fenolik tertinggi pada ekstrak etanol diikuti ektrak etil asetat dan ekstrak n-heksan dengan hasil berturut-turut adalah 54,69 mg/kg; 51,83 mg/kg; dan 37,75 mg/kg. Pada pengujian aktivitas antioksidan sebagai penangkal radikal bebas DPPH diperoleh hasil ekstrak etanol memiliki aktivitas antioksidan yang baik dibandingkan dengan ekstrak etil asetat dan ekstrak n-heksan.ABSTRACT This research aim was to study the proximate phenolic and antioxidant activity of Manado yellow corn kernels. The preliminary stage of this research was the proximate analysis. Then the Manado yellow corn kernels were extracted by using the reflux way for two hours determined the content of total phenolic, and carotenoids. In the third satge of this research was examined the antioxidant activity by using DPPH free radical by UV/Vis. This results from this research showed that the seed powder of Manado yellow corn kernels at the proximate testing was qualified in Indonesia National Standard, but in contrast to the ash content.The highest total phenolic content of the extract ethanol followed by ethyl acetate extract and n-hexane extract with successive result 54.69 mg/kg; 51.83 mg/kg; and 37.75 mg/kg. In examining the antioxidant activity as an antidote to free radicals DPPH results obtain ethanol extract has antioxidant activity compared with extract ethyl acetate and n-hexane.

Abstract

ABSTRAK  Tujuan penelitian ini yaitu menganalisis aktivitas antioksidan beras analog sagu baruk dan ubi jalar ungu. Uji warna menggunakan chromameter, senyawa antioksidan yang diuji yaitu total fenolik dan antosianin, sedangkan aktivitas antioksidan yaitu total antioksidan yang diuji secara spektrofotometri. Hasil penelitian menunjukkan bahwa warna beras analog memiliki nilai L= 62,01, a*= 12,49 dan b*= 1,54. Beras analog mengandung total fenolik sebesar 73,98 mg/L dan antosianin sebesar 7,01 mg/L dan memiliki total antioksidan sebesar 62,01 mg/mL. Sagu baruk dan ubi jalar ungu dapat dibuat beras analog yang memiliki potensi antioksidan. Kata kunci: Antosianin, total antioksidan, beras analog, sagu baruk, ubi jalar ungu  ABSTRACT  The aim of this research was to analyze antioxidant activity of rice analog from baruk sago and purple sweet potatoes. Color was evaluated using chromameter, antioxidant compound which evaluated was total phenolic and anthocyanin, and antioxidant activity was total antioxidant which evaluated using spectrophotometry. The result showed that rice analogue possess L= 62,01, a*= 12,49 dan b*= 1,54. Rice analogue possess total phenolic 73,98 mg/L and anthocyanin 7,01 mg/L, and exhibit total antioxidant 62,01 mg/mL. Abstract

The objective was to determined antioxidant activity of water and ethanol extracts from sago baruk trunk pith. After that, the extracts of trunk pith were analyzed total phenolic and condensed tannin content. Antioxidant activity of each extracts were evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical at various concentrations (1-5 mg/ml). The extracts were identified its compounds group by spectrophotometry UV at 200-400 nm. This result showed that water extract (EA) has highest total phenolic content than ethanol extract 80%. The phenolic total content of EA and EE80 were 124.62; and 86.52 μg/mL, respectively. Whereas, ethanol extract 80% has condensed tannin no different with water extract. The condensed tannin total content of EE80 and EA were 39.911 and 39.74 μg/mL, respectively. Result was evaluation of DPPH free radical scavenging, ethanol extract 80% showed highest free radical scavenging activity than water extract. Result was IC50 calculation of EE80 and EA have IC50 value were 56.23 and 1949.84 μg/mL, respectively. UV spectra of phenolic compounds in the water and ethanol extracts exhibited a maximum absorbance at 275 nm and indication present in tannin condensed component. It was concluded that ethanol extract 80% having tannin condensed component and potential as antioxidantPenelitian ini bertujuan untuk menentukan aktivitas antioksidan ekstrak air dan ekstrak etanol dari empelur batang sagu baruk. Empelur batang sagu baruk diekstraksi dengan menggunakan pelarut air dan etanol. Setelah itu, setiap ekstrak dianalisis kandungan total fenolik dan tanin terkondensasi. Aktivitas antioksidan dari ekstrak empelur batang dievaluasi dengan radikal bebas 1,1-difenil-2-pikrilhidrazil (DPPH) pada berbagai konsentrasi (1-5 mg/mL). Ekstrak diidentifikasi golongan senyawanya dengan spektrofotometer UV-vis pada panjang gelombang 200-400 nm. Hasilnya menunjukkan bahwa ekstrak air (EA) memiliki kandungan total fenolik tertinggi daripada ekstrak etanol 80% (EE). Kandungan total fenolik EA dan EE berturut-turut adalah 150,31 dan 88,92 μg/mL sedangkan kandungan total tanin terkondensasi tidak menunjukkan perbedaan antara ekstrak air dan ekstrak etanol 80%. Kandungan total tanin terkondensasi EA dan EE berturut turut adalah 39,91 dan 39,74 μg/mL. Hasil pengujian aktivitas penangkal radikal bebas DPPH, ekstrak etanol menunjukkan penangkal radikal bebas DPPH yang lebih tinggi daripada ekstrak air. Hasil penghitungan IC50 untuk ekstrak etanol dan ekstrak air berturut-turut adalah 56,23 dan 1949,84 μg/mL. Spektra UV dari senyawa fenolik yang terdapat dalam ekstrak air dan atanol menunjukkan absorbansi maksimum pada 275 dan petunjuk adanya komponen tannin terkondensasi. Hasil penelitian ini menyimpulkan bahwa ekstrak etanol 80% memiliki tanin terkondensasi dan berpotensi sebagai antioksidan.

Abstract

Sago Baruk (Arenga microcarpha) is one of endemic crop type of Archipelago of Sangihe Talaud, North Sulawesi and potential as source of alternative food. The objectives of this research were to determine the phenolics composition and antioxidant activity of fresh and dry sago baruk trunks pith sago which was extracted by sequentially with aquades and filtrate. Flour sago baruk extract were analysed phenolic total, flavonoid and tannin condensed content by spectrophotometer. Antioxidant activity of each sago flour are evaluated in 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging, total antioxidant and reducing power. Result of analysis in phenolic dan flavonoid total content indicate that dry sago baruk higher compared fresh sago flour of baruk wet while tannin condensed content do not show difference. Result of examination of antioxidant activity indicated that fresh sago baruk having higher DPPH free radical scavenging activity compared to dry sago baruk (the average values of 83,08% and 81,49%, respectively). In contrast, total antioxidant and reducing power of dry sago baruk presented higher antioxidant activity compared to fresh sago baruk with average value (the average values of 109,48 and 92,52 μmole/100 g, respectively). Futhermore, correlations analysis revealed that total phenolic, flavonoid, tannin content with DPPH and FRAP scavenging activities were well positively correlated with each other (R= 0.7526; 0,7467 and 0,8146, respectively). Result of this research concluded that dry and fresh sago baruk trunks pith having phenolic compound and antioxidant activitiesSagu baruk (Arenga microcarpha) merupakan salah satu jenis tanaman endemik Kepulauan Sangihe Talaud, Sulawesi Utara yang potensial sebagai sumber pangan alternatif. Tujuan penelitian ini adalah untuk menentukan komposisi senyawa fenolik dan aktivitas antioksidan dari sagu baruk segar dan kering yang diekstraksi secara sekuensial dengan akuades dan filtrat. Empelur batang sagu segar dan kering diekstraksi secara sekuensial dengan akuades dan filtrat. Ekstrak tepung sagu baruk dianalisis kandung total fenolik, flavonoid dan tannin terkondensasi dengan spektrofotometer. Aktivitas antioksidan dievaluasi dengan penangkalan radikal bebas difenil pikrilhidrazil (DPPH), total antioksidan dan kemampuan mereduksi. Hasil analisis terhadap kandungan total fenolik dan flavonoid menunjukkan bahwa sagu baruk kering lebih tinggi dibandingkan dengan tepung sagu baruk basah sedangkan kandungan tannin terkondensasi tidak menunjukkan perbedaan. Hasil pengujian aktivitas antioksidan menunjukkan bahwa sagu baruk segar memiliki aktivitas penangkal radikal bebas DPPH lebih tinggi dibandingkan dengan sagu baruk kering (dengan persentase rata-rata berturut-turut adalah 83,08 dan 81,49%). Sebaliknya, total antioksidan dan kemampuan mereduksi sagu baruk kering memberikan aktivitas antioksidan tertinggi dibandingkan dengan sagu baruk segar (dengan rata-rata 109,48 dan 92,52 μmol/100 g). Selanjutnya, analisa korelasi mengungkapkan bahwa kandungan total fenolik, flavonoid, tannin dengan aktivitas penangkalan DPPH dan FRAP secara positif berhubungan dengan satu sama lainnya (R= 0.7526; 0,7467 dan 0,8146). Hasil penelitian ini menyimpulkan bahwa empelur sagu baruk kering dan segar memiliki senyawa fenolik dan aktivitas antioksidan.

Abstract

Eichhornia crassipes is a weeds to people. The objectives of this research were to analyze phytochemicalcontent and photoreduction activity. Phytochemical content that analyzed were total phenolic, total flavonoidand condenced tannin. Photoreduction activity were done using reduction of Fe3+ to Fe2+. The highest totalphenolic, flavonoid and condenced tannin were extracted with water in leaves, 26.327, 3.191, 25.300 mg/kgrespectively. Photoreduction activity of enceng gondok decreases Fe3+ concentration from 9,111 to 1,500 ppm.Enceng gondok posses photoreduction activity because it has phenolic, flavonoid and condenced tannincontent.

Abstract

The objectives of the present research were evaluated effect of solvent extraction on antiphotooxidative from cloveleave (Eugenia aromatic (L). A certain quantity of powdered sample was extracted with methanol 80%, ethanol 80%and aseton 80%. After evaporation the extracts were dissolved in methanol and assayed for phenolic, flavonoid andcondensed tannin content. Singlet oxygen quenching and free radical activity of each extracts of spices at 500 ppmlevel were evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical. Anti-photooxidative activity test wasconducted using linoleic acid as substrates each containing 5 ppm erythrosine as a photosensitizer. The resultshowed that solvent was significantly having effect on total phenolic, flavonoid and condensed tannin content.Methanol extract possessed highest total flavonoid and condensed tannin content than aseton and etanol. Methanolextract of clove leave was found to have the highest anti-photooxidative effect, followed by aseton and ethanol. Inaddition, ekstrak methanol showed any difference in it free radical scavenger effect than that of ethanol and asetonextracts. This results show that clove methanol extract has the best activity on photooxidation of linoleic acid andscavenger of free radical DPPH. These results suggest that the methanolic extract of clove leave contains somecompounds having singlet oxygen quenching and free radical scavenging activities on linoleic acid and free radicalDPPH.Penelitian ini bertujuan untuk mempelajari pengaruh pelarut ekstraksi terhadap aktivitas antifotooksidan dankomposisi fenolik yang terdapat pada ekstrak daun cengkeh. Serbuk daun cengkeh diekstraksi dengan metanol 80%,etanol 80% dan aseton 80%. Setelah evaporasi, ekstrak daun cengkeh dilakukan pengujian untuk kandungan fenolik,flavonoid dan tannin terkondensasi. Aktivitas penstabilan oksigen singlet dan radikal bebas dari ekstrak dievaluasidengan menggunakan asam linoleat sebagai substrat yang mengandung 5 ppm eritrosin sebagai fotosensitiser danradikal bebas 1,1-diphenyl-2-picrylhydrazyl (DPPH). Hasil penelitian ini menunjukkan bahwa pelarut ekstraksimemiliki efek sangat signifikan terhadap kandungan fenolik, flavonoid dan tannin terkondensasi. Pelarut ekstrakmetanol memiliki kandung flavonoid dan tannin terkondensasi paling tinggi daripada aseton dan etanol. Ekstrakmetanol daun cengkeh ditemukan mempunyai efek antifotooksidatif diikuti ekstrak etanol etanol dan aseton. Selainitu, pelarut methanol juga menunjukkan suatu perbedaan dalam penangkalan radikal bebas DPPH. Berdasarkananalisis komposisi fenolik dalam daun cengkeh dengan HPLC teridentifikasi senyawa eugenol, asam galat, kuersetin,rutin dan kaempferol. Hasil ini menyimpulkan bahwa ekstrak metanol daun cengkeh mengandung beberapasenyawa fenolik yang mempunyai aktivitas sebagai penstabilan oksigen singlet dan penangkalan radikal bebas.
