Abstract

ABSTRACTSuji (Dracaena angustifolia Roxb.) has been used empirically by the people of NorthSulawesi to cure the disease. Flavonoids and steroids in suji leaves suspected to haveantiinflammatroy effects. This study aims to examine the antiinflammatory effects of ethanolextract from suji leaves (Dracaena angustifolia Roxb.) on edema of white males wistar footinduced with 5% white egg solution, and to determine an effective dose of extract to decreasethe edema volume ofrat foot. The test was done using rat hind paw edema or esthablished anartificial inflammation in left foot of white male rats.The treatments were carried out on fivegroups, the positive control group was administered with diclofenac sodium, the negativecontrol group was administered with aquadest, and the extract groups were administered with100 mg/KgBB, 300 mg/KgBB and 1000 mg/KgBB of suji leaf extract. Edema volume weremeasured every hour for 7 hours using a glass tube. The average of edema volume (mL) andtime (hour) was plotted in a curve to form Area Under Curve (AUC). The AUC values wereused to calculate the percentage of antiinflammatory on each group compared to the negativecontrol group. Obtained data were analyzed by one-way ANOVA followed by LSD test withconfident level was 0,05. The result of this study indicate that suji leaf can reduce edemavolume of rat foot that induced with 5% white egg solution and antiinflamatory of ethanolextract from suji leaf 100 mg/KgBB by 33,19 %, 300 mg/KgBB by 14,04 % and 1000mg/KgBB by 5,80 %.Keywords: Suji(Dracaena angustifolia Roxb.), antiinflammatory, edema

Abstract

ABSTRACTTunicate is an invertebrate that lives in a coral reef ecosystem and produces many compounds such as, antibacterial, antitumor and anticancer. This study aimed to determine the antimicrobial activity of extracts and fraction of tunicate (Polycarpa aurata) collected in the Lembeh Strait, Bitung against Escherichia coli, Staphylococcus aureus and Candida albicans. Tunicate (Polycarpa aurata) was extracted by maceration method using 96% ethanol solvent, fractination using liquid-liquid partition method with n-hexane, chloroform and methanol solvent, and antimicrobial testing using Kirby Bauer?s disk diffusion method. The results showed that ethanol extract of tunicate (Polycarpa aurata) had antimicrobial activity againts Escherichia coli with an inhibition of 15.12 mm, and againts Candida albicans with an inhibition of 15 mm. While the methanol fraction showed antimicrobial with a strong category and inhibition of 16.17 mm againts Staphylococcus aureus. Keyword: Tunicate (Polycarpa aurata), Extraction, Fractination, Antimicrobials  ABSTRAKTunikata merupakan invertebrata di ekosistem terumbu karang yang banyak menghasilkan senyawa seperti, antibakteri, antitumor dan antikanker.  Penelitian ini bertujuan untuk  mengetahui aktivitas antimikroba ekstrak dan fraksi tunikata (Polycarpa aurata) yang dikoleksi di Selat lembeh, Bitung terhadap Escherichia coli, Staphylococcus aureus dan Candida albicans.  Tunikata (Polycarpa aurata) diekstraksi menggunakan metode maserasi dengan pelarut etanol 96%, fraksinasi menggunakan metode partisi dengan pelarut n-heksan, kloroform dan metanol, dan pengujian antimikroba menggunakan metode difusi agar Kirby Bauer. Hasil penelitian menunjukkan bahwa ekstrak etanol tunikata (Polycarpa aurata) memiliki aktivitas antimikroba kategori kuat pada fraksi metanol dengan daya hambat sebesar 16, 17 mm terhadap Escherichia coli, pada ekstrak etanol dengan daya hambat sebesar 15, 12 mm terhadap Staphylococcus aureus sedangkan pada Candida albicans aktivitas yang sangat baik terjadi pada ektraksi etanol sebesar 15 mm. Kata Kunci: Tunikata (Polycarpa aurata), Ekstraksi dan Fraksinasi, Antimikroba

Abstract

ABSTRACT Sponges are multi-cell marine invertebrates whose tissue and organ functions are very simple which live in coral reef ecosystems. Sponges are known to produce bioactive compounds because of their symbiotic relationships with microorganisms so that they have the potential to be developed in the field of medicine including as an antimicrobial. This study aims to determine whether the fungus associated with Phyllospongia lamellose sponge taken from the waters of, Southeast Minahasa Regency has antimicrobial activity. This research includes sampling, isolation and fungal inoculation, fermentation, extraction with acetone and then fractionated with ethyl acetate solvent,dried until it gets crude extracts and is carried out as well as antimicrobial testing against Staphylococcus aureus, Escherichia coli, and Candida albicans. Antimicrobial activity was obtained from inhibitory zones formed around the paper disk against the test microbes. From the results of the study concluded that the fungus associated with the Phyllospongia lamellose sponge has antimicrobial activity against the bacteria Staphylococcus aureus, Escherichia coli, and Candida albicans. Keywords: Phyllospongia lamellose sponge, Antimicrobial, Stapylococcus aureu, Escherichia coli, Candida albicans. ABSTRAK Spons merupakan invertebrata laut multi sel yang fungsi jaringan dan organnya sangat sederhana yang hidup pada ekosistem terumbu karang. Spons diketahui menghasilkan senyawa bioaktif karena adanya hubungan simbiotik dengan mikro organisme sehingga berpotensi untuk dikembangkan dalam bidang pengobatan diantaranya sebagi antimikroba. Penelitian ini bertujuan untuk mengetahui apakah jamur yang berasosiasi dengan spons Phyllospongia lamellose yang diambil dari perairan Desa Tumbak, Kecamatan Posumaen, Kabupaten Minahasa Tenggara memiliki aktivitas antimikroba. Penelitian ini meliputi kegiatan pengambilan sampel spons Phyllospongia lamellose, isolasi dan inokulasi jamur yang berasosiasi dengan spons, fermentasi, ektraksi dengan aseton kemudian difraksinasi dengan pelarut etil asetat, dikeringkan hingga mendapat ekstrak kasar dan dilakukan serta pengujian antimikroba terhadap bakteri Staphylococcus aureus, Escherichia coli, dan jamur Candida albicans. Aktivitas antimikroba didapatkan dari zona hambat yang terbentuk disekitaran cakram kertas terhadap mikroba uji. Dari hasil penelitian disimpulkan bahwa jamur yang berasosiasi dengan spons Phyllospongia lamellose memiliki aktivitas antimikroba terhadap bakteri Staphylococcus aureus, Escherichia coli, dan jamur Candida albicans. Kata Kunci : Spons Phyllospongia lamellose, Antimikroba, Stapylococcus aureu, Escherichia coli, Candida albicans.

TERHADAP SEL KANKER PAYUDARA T47D Adithya Yudistira1)1)Program Studi Farmasi FMIPA UNSRAT Manado, 95115  ABSTRACT                This study aims to find out how the value of IC50 from plant extract to breast cancer cells T47D. This research is experimental laboratory using maseration extraction technique and activity test to breast cancer cell from sesewanua leaf extract. The results obtained are: sesewanua leaf extract has a result as a moderate anticancer because it only has a result of 359.44 μg / mL, which for strong anticancer should have an IC50 value below 200 μg / mL.  Keywords : sesewanua leaf, anti-cancer plant, breast cancer cell T47D  ABSTRAKPenelitian ini bertujuan mengetahui berapa nilai IC50 dari ekstrak tumbuhan tersebut terhadap sel kanker payudara T47D. Penelitian ini bersifat eksperimental laboratorium dengan menggunakan teknik ekstraksi maserasi dan uji aktivitas terhadap sel kanker payudara dari ekstrak daun sesewanua. Hasil yang diperoleh yaitu : ekstrak daun sesewanua memiliki hasil sebagai antikanker sedang karena hanya mempunyai hasil 359,44 µg/mL, dimana untuk antikanker kuat harusnya mempunyai nilai IC50 dibawah 200 µg/mL. Kata kunci : Daun sesewanua, tumbuhan anti kanker, sel kanker payudara T47D

Abstract

Sea grass is a group of flowering plant, leaved, has its own roots and live in a shallow water.It usually used to capture sediment, compost, and as antioxidant. The use of sea grass onhuman needs to be conducted. Because of its potency and its component such as flavonoids, aresearch to identify kind of flavonoids in sea grass must be conducted. The aims of thisresearch are to isolate and identify flavonoids in sea grass (Syringodium isoetifolium) usingthin layer chromatography and spectrophotometer UV-Vis. Extraction was done withmaceration using ethanol 96% as solvent. Isolation was done by Thin Layer ChromatographyGF 6054, and n-butanol : acetic acid : water (BAA) (4:1:5). There are two spot after isolatewith TLC, the first is yellow spot with Rf 0,4 and second is red with Rf 0,8. Identification offlavonoids was done using spectrophotometer UV-Vis. Abstract

ABSTRACTTunicate Didemnum molle is a marine biota that has bioactive components that can be used as raw material for medicines. This study aims to determine the antimicrobial activity of extracts and fractions of Didemnum molle on microbial growth of Escherichia coli, Staphylococcus aureus and Candida albicans. The sample was extracted by maceration using ethanol and fractionated using, n-hexane, chloroform and methanol. Antimicrobial activity was tested using the disc diffusion test method (Kirby and Bauer). Ethanol extract of Didemnum molle inhibited the growth of microbes of Staphylococcus aureus (8.12 mm) and Candida albicans (9.00 mm). Chloroform fraction inhibited the growth of Staphylococcus aureus (9.10 mm), Escherichia coli (10.00 mm) and Candida albicans (9.65 mm). While the methanol fraction is only able to inhibit microbial growth of Candida albicans (10.00 mm). Penelitian ini bertujuan untuk mengetahui aktivitas antimikroba dari ekstrak dan fraksi tunikata Didemnum molle terhadap pertumbuhan mikroba Escherichia coli, Staphylococcus aureus dan Candida  albicans. Sampel diekstraksi dengan cara maserasi menggunakan etanol dan difraksinasi menggunakan, n-heksan, kloroform dan metanol. Aktivitas antimikroba diuji menggunakan metode disc diffusion test (Kirby dan Bauer). Ekstrak etanol Didemnum molle menghambat pertumbuhan mikroba Staphylococcus aureus (8,12 mm) dan Candida albicans (9,00 mm). Fraksi kloroform menghambat pertumbuhan mikroba Staphylococcus aureus (9,10 mm), Escherichia coli (10,00 mm) dan Candida albicans (9,65 mm). Sementara fraksi metanol hanya mampu menghambat pertumbuhan mikroba Candida albicans (10.00 mm). Fraksi heksan tidak menunjukkan aktivitas terhadap semua mikroba uji. Kata Kunci : Tunikata Didemnum molle, antimikroba, Escherichia coli, Staphylococcus aureus, Candida albicans.

Abstract

ABSTRACTUlva lactuca Algae habitat was found in shallow marine waters and its morphology is thin and flat thallus. Ulva lactuca Algae, known as sea lettuce, contains a lot of chlorophyll in its cells. Chlorophyll has the ability as a free radical scavenger and also has antioxidant activity. Antioxidants are compounds that could inhibit oxidation reactions by binding to free radicals and highly reactive molecules. This study was conducted to determine the antioxidant activity of ethanol extract of Algae Ulva lactuca obtained from the Lembeh Strait, Bitung City. Ulva lactuca Algae was extracted by maceration method using ethanol solvent. Testing of antioxidant activity was carried out using the DPPH method, which was measured using a UV-VIS spectrophotometer. The results showed that the ethanol extracts of Ulva lactuca Algae had antioxidant activity with a percentage value of 51.63% at a concentration of 100 mg/L. Keywords : Ulva lactuca Algae, Antioxidants, Ethanol, DPPHABSTRAKHabitat tumbuhan Alga Ulva lactuca terdapat di air laut dan morfologinya berupa thallus tipis dan gepeng. Alga Ulva lactuca yang dikenal dengan selada laut, banyak mengandung klorofil dalam sel-selnya. Klorofil memiliki kemampuan sebagai penangkap radikal bebas dan juga memiliki aktivitas antioksidan. Antioksidan merupakan senyawa yang dapat menghambat reaksi oksidasi dengan cara mengikat radikal bebas dan molekul yang sangat reaktif. Penelitian ini dilakukan untuk mengetahui aktivitas antioksidan dari ekstrak etanol Alga Ulva lactuca yang diperoleh dari Peraian Selat Lembeh, Kota Bitung. Alga Ulva lactuca diekstrak dengan metode maserasi menggunakan pelarut etanol. Pengujian aktivitas antioksidan dilakukan dengan metode DPPH yang di ukur menggunakan Spektrofotometer UV-VIS. Hasil penelitian menunjukkan bahwa ekstrak etanol Alga Ulva lactuca memiliki aktivitas antioksidan dengan nilai presentase sebesar 51,63% pada konsentrasi 100 mg/L. Kata kunci : Alga Ulva lactuca, Antioksidan, Etanol, DPPH

Abstract

ABSTRACTSesewanua leaves contain flavonoids which can be potentially as antidiabetic. Flavonoids reduce blood sugar levels by capturing free radicals that cause damage to pancreatic beta cells and inhibit pancreatic beta cell damage. The purpose of this study was to determine the effect of antidiabetic activity on the administration of Sesewanua (Clerodendron squamatum Vahl.) Leaf extracts in wistar white rats (Rattus norvegicus) induced by Aloxan. This type of research is a laboratory experiment using a completely randomized design. Test animals are grouped into 5 groups each consisting of 3 test animals. The negative control group (K-) was not treated, the positive control group (K +) was given metformin, groups 2, 3, and 4 were treated with different extract concentration of 200, 400, and 800 mg. Before the treatment was done the rats were examined the initial blood sugar level, then induced alloxan monohydrate 120 mg / kg BW intraperitonially (ip), and checked the blood sugar level on the 3rd day and continued with the administration of the leaf extracts of the animal every two days until the 14th day. The results showed that sesewanua leaf extracts with a concentration of 200, 400, and 800mg had a decreased activity in blood sugar levels from day 4 to day 14 with a decrease of 13.66 mg / dL in the concentration group 1, 43 mg / dL in the dose concentration 2, and 103 mg / dL in the concentration group 3.Keywords: Blood Glucose, Clerodendron squamatum Vahl., Flavonoids,  Rattus norvegicus ABSTRAKDaun sesewanua mengandung flavonoid yang dapat berpotensi sebagai antidiabetes. Flavonoid menurunkan kadar gula darah dengan cara menangkap radikal bebas penyebab kerusakan sel beta pankreas dan menghambat kerusakan sel beta pankreas. Tujuan penelitian ini adalah mengetahui efek aktivitas antidiabetes terhadap pemberian ekstrak Daun Sesewanua (Clerodendron squamatum Vahl.) pada Tikus Putih Galur Wistar (Rattus norvegicus) yang telah diinduksi Aloksan. Jenis penelitian ini adalah eksperimen laboratorium menggunakan rancangan acak lengkap. Hewan uji dikelompokan dalam 5 kelompok masing-masing terdiri dari 3 ekor hewan uji. Kelompok kontrol negatif (K-) tidak diberi perlakuan, kelompok kontrol positif (K+) diberikan metformin, Kelompok 2, 3, dan 4 diberi perlakuan dengan konsentras ekstrak yang berbeda-beda yaitu 200, 400, dan 800 mg. Sebelum perlakuan dilakukan tikus diperiksa kadar gula darah awal, kemudian diinduksi aloksan monohidrat 120 mg/kgBB secara intraperitonial (ip), dan diperiksa kadar gula darah pada hari ke-3 dan dilanjutkan dengan pemberian ekstrak daun sesewanua setiap dua hari sekali sampai hari ke-14. Hasil penelitian menunjukkan bahwa ekstrak daun sesewanua dengan konsentrasi 200, 400 , dan 800mg memiliki aktivitas penurunan kadar gula darah dari hari ke-4 sampai hari ke-14 dengan penurunan sebesar 13.66 mg/dL pada kelompok konsentrasi 1, 43 mg/dL pada kelompok konsentrasi 2, dan 103 mg/dL pada kelompok konsentrasi 3.Kata Kunci: Clerodendron squamatum Vahl., Flavonoid, Glukosa Darah, Rattus norvegicus

Abstract

Rhodamin B is a synthetic dye that was banned for use in foods and is certified as a hazardousmaterial according to Minister of Health of Indonesian Republic No. 722/Menkes/Per/IX/1988because it can cause liver, kidney, and spleen damage, and followed by anatomical changes in theform of organ enlargement. Rhodamin B dye is often used for a food and beverage products. Thestudy was conducted to identify Rhodamin B on cakalang fufu in the market of Manado city.Cakalang fufu samples were taken from four markets in the city of Manado, namely : the Paal 2market, Bahu market, Karombasan market and Bersehati 45 Market. Samples were extracted by twomethods, the method of extraction followed by purification and extraction method using wool yarnand absorption followed by identification using thin layer chromatography (TLC) and detected withUV light 254 nm and 366 nm. Rhodamin B content is read using a UV-Vis spectrophotometry. Theresult showed that out of eight samples examined with two tests, there are four samples containingRhodamin B with higher levels of the average value of Rhodamin B in samples, that is from themerchant two of Bahu Market is 0,054 ug/ml, at merchant two Karombasan market is 0.184 ug/ml,34merchant one at Bersehati 45 Market is 0.103 ug/ml and merchant two at Bersehati 45 is 0.514 ug/ml.Based on these results, some of cakalang fufu circulating in Manado City Markets are not safe forconsumption.Keywords: Rhodamin B, Cakalang fufu, TLC and UV-Vis spectrophotometry

Abstract

ABSTRACTAntioxidant are compounds that are able to inactivate the development of the oxidation reactions of the molecules or neutralize free radicals. Mahogany bark (Swietenia mahagoni Jacq.) has properties as a medicinal plant. Through the phytochemical screening, the mahogany bark (Swietenia mahagoni Jacq.) contains antioxidant compounds such as alkaloids, saponins and phenolics. The aim of this study was to determine the presence of antioxsidant activity in the extract of mahogany bark (Swietenia mahagoni Jacq.). using the DPPH method with a concentration of 250 mg / L, 200 mg / L, 150 mg / L and 100 mg / L and Vitamin C p.a as a positive control. Each sample made three repetitions of the test. The test uses a UV-Vis Spectrophotometer. The result of the study showed that mahogany bark (Swietenia mahagoni Jacq.) showed antioxidant activity with the percentage of 21,50% at the concentration at 250 mg / LKeywords : Swietenia mahagoni Jacq., Antioxidant, Ethanol, DPPHABSTRAKAntioksidan ialah senyawa yang mampu menginaktivasi berkembangnya reaksi oksidasi molekul lain atau menetralisir radikal bebas. Kulit Batang Mahoni (Swietenia mahagoni Jacq.) memiliki khasiat sebagai tanaman obat. Melalui skrining fitokimia, Swietenia mahagoni Jacq. Mengandung senyawa antioksidan seperti alkaloid, saponin dan fenolik. Penelitian ini bertujuan untuk mengetahui adanya aktivitas antioksidan didalam ekstrak tanaman Kulit Batang Switenia mahagoni Jacq. menggunakan metode DPPH dengan konsentrasi 250 mg/L, 200 mg/L, 150 mg/L dan 100 mg/L dan Vitamin C p.a sebagai kontrol positif. Masing-masing sampel dibuat tiga kali pengulangan uji. Pengujian menggunakan alat Spektrofotometer UV-Vis. Hasil penelitian menunjukkan bahwa ekstrak Kulit Batang Swietenia mahagoni Jacq. memiliki aktivitas antioksidan dengan presentase sebesar 21,50% pada konsentrasi 250 mg/L Kata Kunci : Swietenia mahagoni Jacq., Antioksidan, Etanol, DPPH
