Abstract

This study aims to explain how bacterial contamination and the presence of Escherichia coli and Salmonella sp. on fried bananas sold in the UNIMA campus environment. This exploration used clear qualitative subjective observation with the TPC strategy for all total bacterial contamination and involved specific media for Escherichia coli and Salmonella sp. From the results of the review, it was found that 1 out of 3 experimental examples of complete bacterial contamination exceeded the threshold set by the Food and Drug Supervisory Agency of the Republic of Indonesia (BPOM RI) in 2016, with the most extreme obstacle being the number of microorganisms in food being 10-4 colonies/ g. Of the 3 samples tried, all samples were identified with Escherichia coli and Salmonella sp. Based on the research conducted, it can be concluded that, of all the observed fried samples, all detected bacterial colonies

Abstract

Jajanan (street food) merupakan makanan dan minuman yang dibuat oleh pedagang kaki lima dan dijual di sekolah, pasar dan tempat-tempat umum lainnya. Makanan dan minuman menjadi aspek penting dalam kehidupan manusia. Bakteri Escherichia coli merupakan salah satu bakteri yang ada dalam makanan dan minuman. Adapun tujuan dari penelitian ini yaitu untuk mengisolasi dan mengidentifikasi bakteri Escherichia coli jajanan Pasar Girian Kota Bitung. Metode yang digunakan dalam penelitian ini yaitu metode deskriptif kualitatif. Sampel yang digunakan sebanyak 3 sampel jajanan (minuman boba, kue pukis dan molen), masing-masing sampel diambil dari tempat dan penjual yang berbeda di Pasar Girian. Pengujian dilakukan menggunakan metode Most Probable Number (MPN) seri 5-5-5 dengan 3 pengenceran (). Sampel di ujikan ke dalam media Lauryl Tryptose Broth (LTB) kemudian dilanjutkan ke media Ec Broth, dan kemudian dilakukan penanaman bakteri pada media Eosin Metyle Blue Agar (EMBA), dilakukan pewarnaan gram pada setiap sampel positif dan uji biokimia. Hasil dalam penelitian ini menunjukan bahwa ke tiga sampel yang diperdagangkan di Pasar Girian Kota Bitung positif terkontaminasi oleh bakteri Escherichia coli. Jumlah cemaran terbesar terdapat dalam sampel minuman boba yaitu sebesar >160 MPN/gr.

Abstract

The low learning outcomes of students in Biology learning at SMA Negeri 1 Tutuyan is the background for the need for learning innovations that are able to increase active participation and conceptual understanding. This study aims to analyze the effectiveness of the Group Investigation learning model in improving student learning outcomes in Cell materials. The research used a pseudo-experiment method with a one group pretest-posttest design. The research sample amounted to 35 students from class XI Science who were selected purposefully. The instruments used were multiple-choice tests, pre-tests and post-tests of 25 questions for each test. The results showed an increase in the average score from 23.14 in the pre-test to 83.14 in the post-test. Inferential data analysis using the t-test showed a t-calculated value of 89.55, far above the t-table at a significance level of 5%. The category of student learning outcomes has also undergone significant changes, from a very low majority to a high and very high majority. The application of the Group Investigation model encourages students to be active in investigation, group discussions, and independent learning, which strengthens their collective and individual mastery of the material. These findings prove that the Group Investigation model is effective in improving learning outcomes, and can be implemented as a learning strategy in the Independent Learning Curriculum. This study contributes to the development of cooperative learning models in the context of Biology learning at the high school level, and can be replicated in other materials and levels to strengthen its effectiveness.

Abstract

Pendidikan biologi menuntut pendekatan pembelajaran yang mampu mengembangkan keterampilan berpikir kritis dan pemahaman konseptual secara kontekstual. Penelitian ini bertujuan untuk menguji pengaruh model pembelajaran Project Based Learning (PjBL) terhadap hasil belajar peserta didik pada materi ekosistem hutan hujan tropis. Penelitian ini merupakan eksperimen kuantitatif dengan desain posttest only control group. Subjek penelitian terdiri dari dua kelas X IPA di SMA Negeri 1 Tombatu yang dipilih secara acak, masing-masing terdiri dari 30 siswa. Kelompok eksperimen menerapkan pembelajaran PjBL melalui proyek diorama, sedangkan kelompok kontrol mengikuti pembelajaran konvensional. Hasil analisis data menunjukkan bahwa rata-rata nilai post-test kelompok eksperimen (86,1) lebih tinggi secara signifikan dibandingkan kelompok kontrol (78,8). Uji-t menghasilkan nilai t_hitung sebesar 5,276, yang melebihi t_tabel pada taraf signifikansi 0,05, mengindikasikan adanya perbedaan yang bermakna antara kedua kelompok. Temuan ini menunjukkan bahwa PjBL efektif dalam meningkatkan keterlibatan belajar dan hasil akademik siswa. Kegiatan proyek yang bersifat kolaboratif dan aplikatif mampu menciptakan pembelajaran bermakna dan membangun koneksi antara konsep biologi dan pengalaman nyata siswa. Implikasi dari penelitian ini menegaskan pentingnya penerapan model pembelajaran berbasis proyek dalam pengajaran biologi untuk mengoptimalkan hasil belajar siswa, serta membuka peluang untuk eksplorasi lebih lanjut pada aspek afektif dan keterampilan abad ke-21.

Abstract

Improving the quality of biology learning in secondary schools requires pedagogical innovations that connect academic content with real-life contexts and leverage digital technology. This study aims to analyze the effect of a contextual teaching approach assisted by interactive PowerPoint on students’ learning outcomes in the human respiratory system topic. A true experimental design with a pretest–posttest control group was employed at SMA Negeri 1 Langowan, involving two eleventh-grade classes selected through simple random sampling. The experimental class received contextual instruction integrated with interactive PowerPoint, while the control class was taught using conventional methods. Data were collected through validated achievement tests and analyzed using normality and homogeneity tests, followed by independent and paired t-tests. The results indicate a significant improvement in learning outcomes for students in the experimental class compared to the control group. The experimental group’s posttest scores were substantially higher, and the difference was statistically significant. Integrating a contextual approach with interactive visualization effectively enhanced students’ understanding of abstract biology concepts, promoted active participation, and strengthened learning motivation. These findings support constructivist theory and highlight the relevance of contextual teaching in science education aligned with the Merdeka Curriculum. The study implies the importance of implementing context-based strategies combined with simple digital media to improve biology learning effectiveness. Future research may explore its impact on non-cognitive skills such as critical thinking and scientific attitudes. Furthermore, the results of this study provide opportunities for teachers to be more creative in designing teaching materials that are not only informative but also interactive and relevant to students' lives. By utilizing media such as interactive PowerPoint, teachers can present complex biological content in a more engaging and understandable way.

Abstract

The utilization of pineapple peel in the fermentation process produces bacteria and probiotics that are beneficial for human health. The fermentation process is carried out to enhance the nutrition, shelf life, and organoleptic properties of food products. The presence of microbes, especially bacteria, is believed to play an important role in the fermentation process of pineapple peel. However, the specific bacteria involved in the fermentation of pineapple peel are not yet fully known. The aim of this research is to study the bacterial succession during the fermentation of pineapple peel (Ananas comosus (L.) Merr) and to identify the types of bacteria obtained during the fermentation process. This research uses a qualitative descriptive laboratory-based method by conducting morphological identification of bacteria and biochemical testing to determine the types of bacteria present in the fermentation of pineapple peel. Based on the identification data, 11 bacterial isolates were found during the fermentation of pineapple peel. The isolates were identified macroscopically and then subjected to a series of biochemical tests. The results of the research indicate that the microorganisms involved in the fermentation of pineapple peel belong to the genera Staphylococcus sp., Bacillus sp., and Lactobacillus sp.. The microbial succession during pineapple peel fermentation shows that the bacterial population of the genus Staphylococcus sp. dominates on day 0 of fermentation, while the genus Bacillus sp. undergoes a succession pattern, experiencing growth on day 1 and continuing to increase until day 3. Meanwhile, bacteria from the genus Lactobacillus sp. only appeared on day 1 of fermentation.

Abstract

Tujuan penelitian ini adalah untuk mengetahui ciri fisik dan ukuran Apis dorsata Binghami asal Kabupaten Minahasa Tenggara. Selain itu, kami akan membuat pohon filogenetik untuk Apis dorsata menggunakan data komputasi yang diperoleh dari bank gen NCBI. Penelitian ini menggunakan metodologi deskriptif dengan pendekatan kualitatif, bertujuan untuk menggambarkan secara sistematis dan akurat fakta, kualitas, dan korelasi fenomena yang diteliti dengan menggunakan deskripsi, foto, atau penggambaran secara rinci. Penelitian dilakukan di Desa Maulit, Kecamatan Pasan, Kabupaten Minahasa Tenggara. Setelah menganalisis hasilnya, ditentukan bahwa analisis morfologi, morfometrik, dan molekuler menghasilkan identifikasi yang identik. Penelitian ini menggunakan gen mitokondria 16S rRNA dan CO1 untuk tujuan analisis hubungan. Pemanfaatan gen mitokondria 16S rRNA dan CO1 dalam metode analisis dibenarkan karena banyak keunggulannya dibandingkan gen lain. Hal ini menunjukkan bahwa ciri-ciri morfologi spesies A. dorsata Binghami sebanding dengan spesimen dari tempat lain, meskipun morfometrinya menunjukkan variasi ukuran dibandingkan dengan spesies yang sama yang tercatat di wilayah lain. Selain itu, ciri-ciri molekuler terlihat jelas dalam jarak filogenetik dan genetik yang ditetapkan.

Abstract

This study aims to evaluate the effectiveness of applying the Problem-Based Learning (PBL) model in improving student learning outcomes on the ecosystem material at SMA Negeri 2 Tondano. This study uses a Classroom Action Research (CAR) design with two cycles, involving 30 students in the experimental class using the PBL model and 30 students in the control class applying a conventional learning model based on discussions and question-and-answer sessions. Data were collected through affective observations and cognitive evaluations using posttests at the end of each cycle. The results of the study show that the PBL model can improve student learning outcomes in both cognitive and affective domains. In the cognitive domain, the classical completeness percentage increased from 80% in Cycle I to 100% in Cycle II, with the average class score increasing from 80.15 to 90.35. In the affective domain, all students showed significant improvement, with observation scores in the high category in both cycles. The application of the PBL model motivated students to be more actively engaged in learning and improved their positive attitudes, such as discipline, responsibility, and self-confidence. This study shows that the PBL model is effective in improving both cognitive understanding and affective development in students. These findings suggest that applying PBL in Biology learning can improve the quality of teaching and student learning outcomes, with important implications for the development of problem-based teaching methods in schools
