Abstract

Lektin adalah protein yang mengikat monosakarida dan oligosakarida secara spesifik tetapi tanpa aktivitas katalisis (yaitu bukan enzim) dan, berbeda dengan antitibody yaitu bukanlah produk dari suatu respon sistim pertahanan tubuh.    Lektin dapat mengagglutinasi berbagai macam sel seperti limfosit, sperma, bakteri dan jamur.  Dengan sifat-sifatnya ini, lektin dapat berperan dalam pencegahan infeksi HIV dan AIDS dan pertahanan terhadap virus dan infeksi dari bakteri dan juga pencegahan terjadinya pembuahan.  Walaupun peningkatan telah dibuat melalui karakterisasi biokimia dari lektin alga laut ini, informasi tambahan diperlukan untuk pengertian yang lebih komprehensif tentang sifat-sifat, struktur bahkan fungsi biologis mereka. Penelitian ini menggunakan alga merah laut Eucheuma cotonii sebagai sumber lektin.  Pemurnian lektin menggunakan serangkaian metode yang dimulai dari ekstraksi, kolom kromatografi dan diakhiri dengan SDS-PAGE.  Penentuan karakteristiknya dilakukan melalui pengujian terhadap eritrosit dan beberapa jenis gula untuk menentukan aktivitas hemaglutinasi dan jenis gula spesifik.  Dari hasil Ekstraksi ekstrak kasar lektin diperoleh sebanyak 1.86% dari berat keringnya.  Setelah melewati kolom kromatografi dari 25 tabung yang dihasilkan  hanya diperoleh 5 tabung yang memiliki kandungan lektin.  Melalui SDS-PAGE diperoleh lektin murni dengan berat molekul 11,7 kD.  Konsentrasi terkecil lektin yang masih memiliki aktivitas hemagglutinasi adalah 2,75 µg.mL-1.  Aktivitas hemaglutinasi lektin ini dapat dihambat oleh gula L-Rhamnose dan Maltose.

Abstract

An experiment was designed to study the effects of curcumin and thyroxine hormone supplementation in the diet to improve the reproductive performance of catfish broodstock by improving egg quality that eventually increases the production of seedlings. Catfish used in this experiment were supplemented with curcumin and thyroxine hormone through their feeds for 12 weeks. The results showed that there was no significant difference (p > 0.05) in the hepatosomatic index (HSI), gonadosomatic index (GSI), percentage of gonad maturity, total cholesterol concentration in the spawned eggs, high density lipoprotein (HDL) concentration in the spawned eggs, fertilization rate of spawned eggs, and hatching rate of fertilized eggs.  However, there were significant differences (p < 0.05) in the concentration of vitellogenin in the spawned eggs, egg diameter of the spawned eggs, and the triglycerides contents of spawned eggs. It was concluded that curcumin and thyroxine supplementations of African catfish increased vitellogenin concentrations and diameters of spawned eggs that have great potential to improve the reproductive performance.

Abstract

Permasalahan utama dalam kegiatan usaha budidaya perikanan antara lain adalah ketersediaan benih ikan yang tepat waktu, tepat jumlah dan berkualitas. Ketersediaan benih ikan sangat tergantung pada jumlah telur yang dapat dibuahi dan ditetaskan. Semakin tinggi presentase pembuahan dan penetasan maka akan semakin tinggi pula jumlah larva yang dapat dihasilkan. Penelitian ini menggunakan ikan lele sebagai hewan coba dengan tujuan untuk mengetahui pengaruh suplementasi kurkumin dan tiroksin pada induk ikan lele terhadap kualitas dan performa telur yang dihasilkan. Suplematasi kurkumin dan hormon tiroksin melalui pakan selama 12 minggu pemeliharaan. Parameter yang dianalisis berupa konsentrasi vitelogenin telur, diameter telur, koefisien keragaman diameter telur, konsentrasi lipid dalam telur (triglserida, kolesterol dan HDL), derajat pembuahan dan derajat penetasan. Hasil penelitian menunjukkan bahwa kelompok perlakuan yang diberikan kombinasi penambahan kurkumin dan hormon tiroksin memiliki nilai konsentrasi vitelogenin (8.17+2.74 µg/mL) (p<0,05), diameter telur (1.43+0.00 mm) (p<0,05), dan konsentrasi trigliserida (4.89+0.53 mg/g) (p<0,05) tertinggi diantara semua perlakuan, sementara itu kelompok perlakuan yang baik hanya diberikan penambahan kurkumin, maupun kombinasi penambahan kurkumin dan tiroksin cenderung memiliki nilai derajat pembuahan dan penetasan yang lebih tinggi dibandingkan dengan kelompok perlakuan yang tidak diberikan penambahan kurkumin.  The main problem in aquaculture is the availability of fish seedlings on time, in the right quantity and quality. The availability of fish seeds is determined by the number of eggs that can be fertilized and hatched. The higher the percentage of fertilization and hatching, the higher the number of larvae that can be produced. This study uses catfish as experimental animals because catfish are one of the most consumed cultivation commodities. The aim of this study is to determine the effect of curcumin and thyroxine supplementation on catfish brooders on the quality and performance of eggs produced. Catfish used in this experiment were given the addition of curcumin and thyroxine hormone through the feed for 12 weeks of rearing. Parameters analyzed were egg vitellogenin concentration, egg diameter, coefficient of diversity in egg diameter, lipid concentration in eggs (triglycerides, cholesterol, and HDL), degree of fertilization and degree of hatching. The results showed that the group given the combination of the supplementation curcumin and the thyroxine hormone had the values of vitellogenin concentration (8.17+2.74 µg/mL) (p <0.05), egg diameter (1.43+0.00 mm) (p <0.05), and triglyceride concentration (4.89+0.53 mg/g) (p <0.05) the highest among all treatments, meanwhile the group is only given the addition of curcumin, and the combination of supplementation of curcumin and thyroxine tends to have higher values of fertilization and hatching compared to the group which is not given the addition of curcumin.

Abstract

Bioflok adalah teknik budidaya ikan untuk mengelola lingkungan budidaya dengan memanfaatkan mikroorganisme untuk meningkatkan penggunaan pakan dan sisa hasil metabolisme dengan penambahan sumber karbon organik. Penelitian ini bertujuan untuk mengetahui ketahanan hidup bibit ikan mujair (Oreochromis mossambicus) dan nilai parameter kimiawi lingkungan pada media pemeliharaan bioflok dengan debris daluga (Crytosperma merkusii) sebagai sumber karbon. Penelitian ini menggunakan Rancangan Acak Lengkap (RAL) dengan 4 perlakuan dan 3 ulangan. Perlakuan yang dilakukan adalah tanpa bioflok, C:N=15, C:N=20, dan C:N=25. Hewan uji ikan mujair dengan panjang rata-rata 5,5 cm di tebar 10 ekor setiap ember. Parameter yang diamati adalah kualitas air yang meliputi suhu, pH, konduktivitas, dan redoks dan Survival Rate (SR) ikan mujair. Dari hasil penelitian diketahui bahwa perlakuan terbaik pada C:N=25 dengan persentase SR tertinggi yaitu, 86,67%. Kualitas air selama penelitian ada pada kondisi optimal di setiap perlakuan.

Abstract

The availability of quality seeds, on time with large amounts is a major problem in freshwater aquaculture. Supplementation of curcumin in female carp aims to increase the reproductive productivity of fish through increased activity of the liver in the synthesis of vitellogenin, a precursor of egg yolk, that play a role in the growth and development of eggs until larvae. This study used 25 female carp, which supplemented with curcuminat several doses: 0; 2.5; 5; 10; and 20 g of curcumin/kg of feed. The collected data was analyzed using a completely randomized design (CRD). The results showed that supplementation of curcumin female carp significantly (p<0.05) affected the malondialdehyde(MDA) concentration of liver, estradiol hormone in the blood, gonadosomatic index, relative fecundity, and larval survival, but did not significantly (p>0.05) affect the superoxide dismutase (SOD), vitellogenin concentration in blood and egg diameter. The study showed that the supplementation of curcumin prevents liver damage showed by a decreased level of liver MDA. The supplementation of curcumin with a dose of 5 g/kg of feed optimizes the productivity and reproductive performance of carp by increasing the production of vitellogenin, the diameter of egg, and the number of survival larva through optimizing the performance of the fish liver.

Abstract

Rattus norvegicus is one of the experimental animals that is widely used in research. Knowing the hematological status of R. norvegicus rats during reproduction is the aim of this study. In this study was used 12 female rats were sexually mature. Data were collected at the rats that were not yet pregnant (T0); 10 days pregnant mouse (T10); 20 days pregnant mouse (T20); 7-day lactated rat (L7); and 20-day lactating mice (L20). This study shows that during pregnancy and breastfeeding there is an increase in the level of erythrocytes in the blood. During pregnancy, there is a decrease in hemoglobin levels and higher during lactation. The hematocrit value during lactation shows a higher value than in the non-pregnant and pregnant states.

Abstract

RNA / DNA ratio is a parameter that shows the activity of protein synthesis that is taking place in a cell. In mammals, during the reproductive period, the ovaries and uterus are the organs that play an active role. The purpose of this study was to know the activity of protein synthesis that occurs in the ovaries and uterus in female white rats (Rattus norvegicus) in various physiological conditions during reproduction. In this study, female rats that were sexually mature were used. Data were collected on rats with physiological status not yet pregnant (T0); pregnancy 10 and 20 days (T10, T10), and lactation 7, 10, and 20 days (L7, L10, L20). The results showed that there was a decrease in the value of the ovary RNA / DNA ratio during pregnancy and lactation. In contrast, the uterine RNA / DNA ratio increases during pregnancy and lactation. Based on the results of the study, it can be concluded that the increase in the uterine RNA: DNA ratio that occurs during pregnancy is because it is used for the synthesis of various structural and functional components for fetal growth and development

Abstract

Perkembangan awal larva sangat dipengaruhi oleh ketersediaan makanan yang berasal dari kantung telurnya. Tidak terpenuhi nutrisi pada masa awal perkembangan dapat mempengaruhi perkembangan larva. Tujuan penelitian ini adalah untuk mengukur diameter telur hasil pemijahan  dan panjang badan pada awal perkembangan larva ikan mas dari induk yang diberi pakan dengan penambahan kurkumin. Penelitian ini menggunakan induk betina ikan mas yang diberikan suplementasi kurkumin pada beberapa dosis yaitu 0%; 0,25%; 0,5%; 1%; dan 2%.  Ikan yang telah matang gonad kemudian dipijahkan. Telur yang telah terbuahi kemudian diambil untuk ditetaskan dan diamati perkembangan larvanya selama 2 hari setelah penetasan (hsp). Hasil pengamatan menunjukkan bahwa perlakuan suplementasi kurkumin dengan dosis 0,5% yang diberikan dalam pakan induk betina ikan mas memberikan nilai yang optimal terhadap diameter telur hasil pemijahan (p<0,05) dan panjang badan larva sampai pada umur 2 hsp (p<0,05), namun panjang mutlak larva sampai pada 2 hsp tidak menunjukkan adanya perbedaan antar perlakuan (p>0,5). Berdasarkan hasil penelitian ini dapat disimpulkan bahwa suplementasi kurkumin dengan konsentrasi 0,5% pada induk betina ikan mas dapat mengoptimalkan pertumbuhan larva pada awal kehidupannya.Kata kunci : Cyprinus carpio; kurkumin; larva Early Development of Common Carp Larva from Broodstock Fed Diet Supplemented with CurcuminABSTRACTThe early growth of the larva is greatly influenced by the availability of food from the egg yolk. Insufficiency of nutrition in the early stages of development can affect the development of larva. The purpose of this study was to know the early growth of larva from broodstock supplemented with curcumin. This study used the female broodstock of common carp that supplemented with curcumin at several doses; 0%; 0,25%; 0,5%; 1%; and 2%. The fishes that had been mature were spawned, and the fertilized eggs were taken to be hatched. The larva was observed for 2 days after hatching (dah). The results showed that the treatment of curcumin supplementation with a dose of 0,5% given in the common carp feed gave an optimal value for egg diameter (p <0,05) and body length of the larva until the age of 2 dah (p <0,05), but did not differ for absolute length growth (p>0,05).Keywords: curcumin; Cyprinus carpio; larva

Abstract

Feed is an important factor that influences the chicken's development and growth. Meanwhile, dalugha debris is a by-product of dalugha processing which is expected to be a source of feed for the growth and development of broiler chickens. The good feed can optimize metabolic processes in the animal so that it can support the process of growth and development of the animal. This study aims to determine the levels of hemoglobin, cholesterol, and blood glucose in broiler chicken that is given dalugha debris supplementation. This study used a completely randomized design (CRD), where as many as 60 docs of unsex chicken used were divided into four treatments and three replications. Each replication consisted of five chickens. The treatment given was the concentration of dalugha debris at several doses: 0%, 0.065%, 0.13%, and 0.26%. The parameters observed were hemoglobin, cholesterol, and blood glucose levels. The results showed that the provision of dalugha debris in the feed significantly (p<0.05) affected the blood glucose level, but did not affect the cholesterol and hemoglobin levels (p>0.05) of the broiler chicken. Abstract

Ants are a group of social insects that belong to the order Hymenoptera and the Family Formicidae. This study aims to determine the type of ants (Formicidae) in the Mangrove Area of Tatengesan Village, Southeast Minahasa. This research occurred in Tatengesan Village, Posumaen District, Southeast Minahasa Regency, North Sulawesi. The research method uses a purposive sampling method with the placement of the paths determined based on considerations taking into account the physical and biological conditions that affect the presence of ants in the field. Observation lines were made in as many as three transects, each with four observation plots. Data was collected using adhesive paper traps (Fly Sheet) and direct capture method (hand collecting) and analyzed using the Shannon-Weiner diversity index, species richness index, and species evenness index. Placement of self-adhesive paper on each transect, including lower tree stratum, highest tide limit, middle tree stratum, and upper tree stratum. Based on the results of the research on the observation transect, there were five genera, namely Anoplolepis with 25 individual ants, Camponotus with 31 individual ants, Crematogaster with 98 individual ants, Oecophylla 2168 ants, and Tetraponera 6 individual ants from 6 species of ants consisting of Anoplolepis gracilipes, Camponotus atriceps, Camponotus sp., Crematogaster scutellaris, Oecophylla smaragdina, and Tetraponera nigra. The highest number of genera obtained was Oecophylla, the highest number of species was Oecophylla smaragdina, and the number of individuals caught was 2168.
