Abstract

Tanaman yang diketahui memiliki potensi sebagai obat antihipertensi adalah daun sirih Keraton (C. discolor). Daun sirih Keraton telah digunakan dalam pengobatan tradisional untuk berbagai kondisi, termasuk hipertensi, berkat kandungan senyawa bioaktif seperti flavonoid, alkaloid, dan saponin. Penelitian ini bertujuan untuk memberikan gambaran tentang tingkat keamanan ekstrak dan juga membantu dalam merancang protokol dosis yang lebih aman untuk uji klinis atau penggunaan dalam produk kesehatan. Penelitian ini menggunakan metode animal tasing dengan mengukur nilai LD50 menggunakan 30 tikus wistar jantan sebagai subjek penelitian yang terbagi dalam 5 kelompok perlakuan dengan dosis 500 mg/kgBB, 2000 mg/kgBB dan 5000 mg/kgBB. Hasil dari penelitian ini menunjukan bahwa ekstrak daun sirih keraton hingga dosis tertinggi tidak menunjukan gejala toksisitas. Meskipun tidak terdapat tanda-tanda toksisitas pada hewan uji hingga dosis tertinggi hasil penelitian ini dapat dikatakan sebagai toksisitas semu, tetapi hasil ini tetap dapat dijadikan rujuan awal tentang keamanan ekstrak daun sirih keraton ini. Sehingga masih diperlukan penelitian lanjutan untuk menentukan nilai LD50 yang sebenarnya.

Abstract

Introduction: Lung cancer is characterized by high mortality rates and poor prognosis, further requiring alternative therapies owing to the adverse effects of conventional chemotherapy. Rambutan (Nephelium lappaceum L.) has antimicrobial, antiproliferative, and antioxidant properties and can cure cancer. Method: Compounds from rambutan peel were identified through PubChem and analyzed for physicochemical properties using SwissADME. Target proteins were identified using SuperPred and compared with lung cancer–related proteins from GeneCards via Venny. Protein interactions were examined and visualized using STRING and Cytoscape, respectively. Protein activity was analyzed using WebGestalt, and docking simulations were performed using Autodock 4.2.6. Result: Physicochemical analysis of 10 active rambutan compounds via SwissADME showed favorable properties based on Lipinski’s rule, suggesting good oral bioavailability. In total, 276 specific target proteins were related to lung cancer. Cytoscape visualized the 53 core proteins implicated in lung cancer. Gene ontology and Kyoto Encyclopedia of Genes and Genomes analyses indicated that rambutan compounds are active in lung cancer pathways, particularly nonsmall cell lung cancer. The analysis identified the top 10 interacting proteins STAT3 and CASP3 had the highest interaction value of 28, followed by PIK3CA and NFKB1 at 22, MAPK3, PIK3R1, PTGS2, GRB2, MAPK1, and CASP8. Per docking simulations, 1,2-diphenylethane-1-ol was the highest affinity to STAT3 (−4.78) and CASP3 (−6.75). Conclusion: Rambutan peel compounds have remarkable anticancer potential, particularly with high affinity for STAT3 and CASP3 proteins, based on in silico molecular docking studies.

Abstract

The purple yam plant (Dioscorea alata L.) is an economically important staple food for millions of people in tropical and subtropical regions. Dioscorea spp. anthocyanin chemicals have been demonstrated to have antioxidant and neuroprotective properties. The purpose of this study was to explore the potency of anthocyanin compounds in purple yam as antiparkinsonian agents via the monoamine oxidase B (MAO-B) receptor (pdb: 2V5Z) and the catechol-O-methyltransferase (COMT) receptor (pdb: 6I3C) using a molecular docking technique. The study was divided into four stages: (1) pharmacokinetic and Lipinski Rule evaluation, (2) protein (receptor) and ligand preparation, (3) docking method validation, and (4) molecular docking for MAO-B and COMT proteins. Pharmacokinetic prediction and Lipinski rule evaluation revealed that cyanidin, delphinidin, and delphinidin-3-glucoside had an ADMED profile and met Lipinski’s rule. The docking results showed that the binding energy (G) of the compounds cyanidin, delphinidin, and delphinidin-3-glucoside to the MAO-B receptor was lower (-9.50 kcal/mol) than that of the natural ligands ((-4.79 kcal/mol). The Cys172, Leu 171, Ile198, Phe168, Pro104, Trp119 and the 'gatekeeper' residue Ile199 are the amino acids that are majoring involved in MAO-B inhibitors. At the COMT receptor, all the tested compounds had a higher binding energy than native ligands (> -4.79 kcal/mol) except for Cyanidin 3,5-diglucoside (-4.64 kcal/mol). The amino acids Trp143 and Pro174, ensure correct substrate orientation, Mg2+ ions, and cofactor SAM, as well as residues Lys144 and Glu199. In conclusion, this study showed that based on the molecular docking approach, the active compounds of purple yam namely cyanidin, delphinidin, and delphinidin-3-glucoside have the potential to be developed as anti-parkinsonian agents through MAO-B and COMT.

Abstract

Problems related to child marriage occur due to several educational, economic, cultural, and environmental factors. This has more negative impacts than positive impacts, not only a serious problem in Indonesia but also in neighboring countries such as Malaysia. Therefore, this study aims to analyze the comparison of legal policies in Indonesia and Malaysia against child marriage. This research method is normative juridical by a examining legislative documents, books, journals, and other literature studies related to this research. The results show that the age limit for marriage in Indonesia is 19 years for men and women, while in Malaysia the age limit for marriage for men is 18 years and 16 years for women. The policy applied in Indonesia to prevent child marriage is to raise the age limit of marriage and fulfill PERMA Number 5 of 2019, while in Malaysia, if a child marriage is to take place, it must fulfill the Standard Operating Procedure of the Sharie Judge and the policy applied to prevent child marriage through 'urf which is formed through the level of education, economy, and psychology.  Keywords: child marriage, marriage age limit, policy

Abstract

Teki grass (Cyperus rotundus L.) tubers are medicinal plants that have been widely utilized for their anti-inflammatory, antioxidant, antibacterial, and anticancer properties. These pharmacological activities are attributed to the presence of polyphenolic compounds, particularly flavanones. This study aims to determine the flavanone content in teki grass tubers using UV-Vis spectrophotometry. The research was conducted as a laboratory-based experimental study, involving several steps: sample preparation, simplisia processing, extraction, and quantitative analysis of flavanone content by comparison with a flavanone standard. The UV-Vis spectrophotometric analysis revealed the presence of flavanones in the extract of teki grass tubers. Quantitative results showed that the flavanone concentration in the extract was 128.29 µg/mL. These findings confirm that teki grass tubers are a potential natural source of flavanones, supporting their traditional herbal medicine use.

Abstract

Cervical cancer is one of the second-leading causes of death in women. The discovery of cancer drug candidates continues to be carried out due to the resistance that occurs in cervical cancer therapy. Plant metabolite compounds are one of the sources used to explore new drug candidates. Red ginger rhizome is a candidate plant that has anti-cervical cancer activity. This study aims to determine the cytotoxicity of an ethanol extract of red ginger rhizomes on the growth of HeLa cancer cells and predict anti-proliferative activity via the jak2/stat3 and hedgehog pathways. The sample (red ginger rhizome simplicia) was extracted by remaceration using 75% ethanol. The MTT assay method is used to test the cytotoxicity and anti-proliferation of metabolite compounds using Autodock 4.2 software. The receptors used in the jak2, stat3, and smo pathways were obtained from the Protein Data Bank with the codes 6VGL, 6NUQ, and 5L7I, respectively. The ethanol extract produced is a thick yellowish brown extract with an aromatic smell and spicy taste, with an extract yield of 18.63% w/w. 75% ethanol extract of red ginger rhizomes has cytotoxic activity in HeLa cancer with an IC50 of 104.22 ± 6.18 µg/mL and an IC50 of cisplatin of 38.61 ± 3.66. Prediction of antiproliferative activity via the jak2 pathway shows a binding energy and Ki value of -7.47 kcal/mol, -7.48 kcal/mol, and 3.33 uM, 3.27 uM, as shown by alpha-cedrol and beta-eudesmol compounds. The highest inhibition on the stat3 and smo pathways was shown by the beta compound eudesmol, with binding energy and Ki values of -6.05 kcal/mol, -7.57 kcal/mol, and 36.48 uM, respectively; 2.81 uM.

Abstract

Background: Health science development is currently rapidly changing due to the Covid-19. Changes have taken place not only in terms of scientific development but also in terms of learning methods. Almost all learning methods have shifted into online learning, requiring students to be more active.Objectives: This study aims to determine the level of SDLR among pharmacy students in Indonesia, especially during the pandemic.Methods: The sampling was conducted using an proportional sampling technique based on the distribution of Pharmacy Schools (PTF) in all the provinces in Indonesia. The level of readiness of these pharmacy students was measured using a questionnaire that consisted of four domains. The questions were scored according to the characteristics of the questions (negative or positive).Results: The results of this study showed that 1884 respondents participated in this study. The basic characteristics showed that 89.1% participants were private pharmacy schools and 84.4% of them were female respondents. The mean of each domains of SDLR for self-management, the desire to learn, the emotional competency, and the responsible decision-making was 3.24 ± 2.65, 3.37 ± 2.09, 3.04 ± 1 87, 3.16 ± 2.16, respectively. Based on gender, the difference of mean in sub domain was p=0.000 (need for learning), p=0.002 (hope for outcome), and p=0.000 (identifying problem).Conclusion: In conclusion, the Self-Directed Learning Readiness (SDLR) among pharmacy students in Indonesia is quite high during the pandemic with a score above 3 (scale 1-4).

Abstract

Bajakah tampala (Spatholobus littoralis), a medicinal plant traditionally used in Indonesia, particularly on Kalimantan Island, has garnered interest for its potential health benefits. However, scientific evidence remains scarce. This study investigated the antioxidant activity of S. littoralis extract and its total phenolic and flavonoid content. Ethanol extraction and evaporation were used to prepare the extract. The DPPH method assessed antioxidant activity, while Folin–Ciocalteu and AlCl3 complexation methods quantified total phenolics and flavonoids, respectively. The S. littoralis extract exhibited strong antioxidant activity with an IC50 value of 54.19 ± 8.15 µg/mL. Additionally, the extract contained substantial levels of phenolics (0.649 ± 0.026% GAE) and flavonoids (1.084 ± 0.043% QE). These findings suggest a link between the high phenolic and flavonoid content of S. littoralis extract and its observed strong antioxidant activity.

Abstract

Penyakit parkinson adalah gangguan pada neurodegeneratif yang ditandai dengan hilangnya progresif neuron dopaminergik otak tengah di Substansia Nigra Pars compacta (SNpc). Pencegahan penyakit Parkinson dapat dilakukan dengan enzim inhibitor MAO-B. Senyawa flavonoid dan polifenolik telah terbukti memiliki aktivitas sebagai antiparkinson melalui penghambatan MAO-B. Salah satu tanaman endemik Indonesia, Bajakah Tampala (Spatholobus littoralis Hassk.) diketahui memiliki komponen senyawa flavonoid dan polifenolik. Tujuan: Untuk mengeksplorasi potensi ekstrak etanol Bajakah Tampala yang kaya flavonoid dan polifenolik sebagai inhibitor MAO melalui studi in silico. Metode: Uji in silico terdiri dari evaluasi Lipinski dan studi farmakokinetik (ADMET) menggunakan program pkCSM, dan uji molekular docking terhadap 15 senyawa flavonoid dan polifenolik dari data HRMS ekstrak etanol kayu Bajakah Tampala pada enzim inhibitor MAO-B (kode PDB: 2V5Z) menggunakan Software Autodock 4.2. Hasil: Berdasarkan prediksi studi profil farmakokinetik maupun hasil evaluasi Lipinski Rule of Five senyawa yang terkandung didalam tanaman Bajakah Tampala (Spatholobus littoralis Hassk.) memiliki profil farmakokinetik yang cukup baik dan memenuhi kriteria dari evaluasi Lipinski Rule of Five, dan memiliki afinitas yang baik untuk validasi docking molekuler. Lima belas senyawa aktif flavonoid ekstrak etanol 70% memiliki potensi menghambat enzim MAO-B dengan binding energy berkisar antara -8,34 hingga -10,28 kcal/mol. Sedangkan native ligan safinamide memiliki binding energi -9,78 Kcal/mol. Kesimpulan: Senyawa tanaman Bajakah Tampala (Spatholobus littoralis Hassk.) memiliki kandungan senyawa aktif yang berpotensi dapat dikembangkan menjadi senyawa obat, dan dapat dilakukan penelitian lebih lanjut secara in vitro dan in vivo sebagai obat antiparkinson.
