Abstract

The tomato sauce is a complementary food that is often served with other foods, such as chicken noodles. Various factors can be the cause of contamination of tomato sauce by microbes like bacteria. These factors include environmental conditions that are not clean or container that was left open. Bacterial contamination in tomato sauce can result not suitable for consumption. Given by the high public interest in the consumption of tomato sauce with chicken noodle, it is necessary to observe the bacterial contamination of chicken noodle tomato sauce. To determine the bacterial contamination in tomato sauce of chicken noodle in traditional market of Condong Catur, Sleman, Yogyakarta. The samples used were tomato sauces that are used by chicken noodle stalls in the Condong Catur traditional market. Sampling was also carried out by recording the environmental conditions stalls and tomato sauce serving. To determine the bacterial contamination, samples examined in the laboratory based on the total bacterial count method using plate count agar (PCA) media. Seven of the ten samples showed the total number of bacteria <104 CFU/ml, whereas the other three samples are not suitable for consumption by the total number of bacteria >104 CFU / ml. The main factors of high contamination is poor hygiene of chicken noodle stall environmental, tomato sauce containers that are less clean and are often left open.Keywords: tomato sauce, total bacteria

Abstract

Potato (Solanum tuberosum L.) is one of the five basic sources of carbohydrates. It consumed by many people world spreads. One of the constraints in potato production is the presence of fungal diseases. Fungi that cause diseases in potato crops include Phytophthora infestans which causes late blight, Fusarium oxysporum which cause fusarium wilt, Alternaria solani Sor. which cause brown spot disease, and Aspergillus niger which infect bulbs and produce aflatoxin. Identification of potatoes-contaminating fungi can lead the farmers to get a better potatoes production. This research goal is to identify potatoes-contaminating fungi in traditional market of Condong Catur, District of Sleman, Yogyakarta. This research was using descriptive method with laboratory examination. We took 30 defected potatoes suspected of being infected by fungi. Samples were taken from eachpotato aseptically and then cultured in Saboraud’s Dextrose Agar (SDA) media. Observation was held for the fungi growth after 24 hours incubation. From the laboratory examination, there were fungal infections in all of potatoes and identified as Phytopthora infestans (26.67%), Fusarium oxysporum (86.67%), Alternaria solani Sor. (6.67%), and Aspergillus niger (13.33%). Phytophthora infestans, Fusarium oxysporum, Alternaria solani Sor., and Aspergillus niger identified as potatoes-contaminating fungi in Traditional Market of Condong Catur, District of Sleman, Yogyakarta.Keywords: contaminant, fungi, potatoes

Abstract

Chicken meat is a good source of protein for daily consumption. It is very easy decayed biologically by enzymes or microbial spoilage. White galangal (Alpinia galanga Linn Swartz) is a kind of spice crop that can live in the highlands and lowlands. Generally, people utilize white galangal as a blend of seasoning. Galangal’s role as a food preservative is inseparable from its anti-microbial activity and secondary metabolite contents, i.e. essential oils. The anti-microbial is a biological or chemical compounds that could interfere the growth and activity of microbes, particularly microbes as a food spoilage. This research goal is to determine the number of bacteria in chicken meat with the provision granting the white grated galangal rhizome (Alpinia galanga Linn Swartz).This was a descriptive study with laboratory testing. We use pour plate method for the bacteria number determination. Independent variables is the indwelling time with grated white galangal for 1-5 hours and the dependent variable is the number of bacteria in chicken meat.The result showed that total number of bacteria after smeared with white grated galangal rhizome for 1 hour 463.500 CFU/gr, 2 hour 130.250 CFU/gr, 3 hour 58.250 CFU/gr, 4 hour 142.500 CFU/gr and 5 hour 302.500 CFU/gr. This study showed that grated white galangal has proven to reduce the number of bacteria in chicken meat.

Abstract

Pendahuluan: Jamur Candida sp dapat berperan sebagai mikroorganisme yang bersifat patogen yaitu menimbulkan infeksi primer dan sekunder. Penderita diabetes mellitus (DM) dapat mengalami infeksi jamur tersebut karena mekanisme pertahanan alami tubuh yang rendah. Komplikasi yang terkait DM dapat meningkatkan risiko infeksi, peningkatan kadar gula dalam darah, jaringan, dan urine. Penelitian ini bertujuan untuk mengetahui keberadaan jamur Candida sp dalam urine penderita diabetes mellitus tipe 2. Metode Jenis penelitian ini adalah deskriptif dengan desain penelitian cross sectional. Jumlah sampel sebanyak 30 spesimen urine penderita diabetes mellitus tipe 2 dengan kriteria inklusi berjenis kelamin laki-laki dan perempuan, pasien adalah peserta program pengelolaan penyakit kronis (prolanis) di Puskesmas Ngemplak 2 Kabupaten Sleman Yogyakarta. Tahap pengumpulan data dilakukan pada bulan Juli 2019 di Puskesmas Ngemplak 2 Kabupaten Sleman Yogyakarta. Pemeriksaan jamur Candida sp dilakukan secara makroskopis dan mikroskopis. Hasil: Hasil penelitian menunjukkan sebanyak 8 sampel (26,7%) ditemukan jamur Candida sp dan 22 sampel (73,3%) tidak ditemukan jamur Candida sp. Sampel dengan jamur Candida sp, sebagian besar berasal dari penderita diabetes mellitus berjenis kelamin perempuan yaitu sebanyak 5 sampel (16,3%). Kesimpulan: Jamur Candida sp dapat ditemukan dalam urine penderita diabetes mellitus terutama penderita dengan jenis kelamin perempuan. Hal ini dapat disebabkan oleh kondisi sistem kekebalan tubuh menurun, atau sering menggunakan pakaian ketat dari bahan sintetis.

Abstract

Demam tifoid yang disebabkan oleh bakteri Salmonella typhi merupakan salah satu infeksi global diseluruh dunia dan merupakan masalah kesehatan yang serius di negara-negara berkembang seperti di Asia dan Afrika, termasuk di Indonesia. Tingkat resistensi bakteri Salmonella typhi merupakan salah satu permasalahan serius dibidang kesehatan. Bawang putih (Allium sativum Linn.) yang mengandung senyawa allisin dapat berfungsi sebagai antibakteri dengan spektrum yang luas. Maka dari itu, bawang putih diharapkan dapat menjadi solusi dalam mengatasi permasalahan resistensi bakteri. Tujuan penelitian ini adalah untuk mengetahui pengaruh perasan umbi bawang putih (Allium sativum Linn.) dalam menghambat pertumbuhan bakteri Salmonella typhi. Penelitian ini menggunakan bakteri Salmonella typhi yang diberi perlakuan perasan umbi bawang putih dengan konsentrasi 20%, 40%, 60%, 80% dan 100%. Metode yang digunakan adalah difusi dengan sumuran pada media agar Mueller-Hinton. Masing-masing konsentrasi perasan bawang putih dimasukkan dalam sumuran agar dan dihitung diameter zona hambat pertumbuhan bakteri. Data yang didapat dianalisis secara deskriptif menggunakan Analisis Varians (ANOVA). Rata-rata diameter zona hambat yang paling tinggi yaitu 45,3 mm pada konsentrasi perasan 100% sedangkan pada konsentrasi 20% diperoleh rata-rata diameterzona hambat paling rendah 27,0 mm. Dengan nilai signifikansi 0,000 yang kurang dari á=0,05 pada hasil uji Anova menunjukkan adanya pengaruh antibakteri dari perasan bawang putih dalam menghambat pertumbuhan Salmonella typhi. Dari penelitian ini, dapat disimpulkan bahwa perasan bawang putih memiliki potensi sebagai alternatif pengobatan terhadap infeksi bakteri Salmonella typhi. Melihat adanya potensi tersebut, maka perlu dilakukan penelitian lebih lanjut untuk mengetahui daya hambat perasan bawang putih terhadap pertumbuhan bakteri Salmonella typhi secara in vivo.

Abstract

Infectious diseases are diseases caused by pathogenic bacteria that enter the body, multiply and cause disease. One cause of the disease is a bacterial infection of Staphylococcus aureus. Treatment of infectious diseases is usually done by giving antibiotic but some types of bacteria become resistant. Utilization of plants as traditional medicine is growing rapidly and many alternative medicine used by some communities, one of which is cabbage (Brasicca oleracea var. capitata f. alba) containing antibacterial compounds such as flavonoids, saponins, polyphenols, tannins and sulfur. Such compounds capable of inhibiting the growth of bacteria by inhibiting bacterial cell wall synthesis, protein synthesis, and forming complex compounds against extracellular proteins that interfere with bacterial cell membrane integrity. This research is an experimental research with the aim to determine the inhibitory ethanol extract of cabbage (Brasicca oleracea var. capitata f. alba) against Staphylococcus aureus in in vitro. Antibacterial test method used is that pitting diffusion method. Ethanol extract of cabbage (Brasicca oleracea var. capitata f. alba) can inhibit the growth of Staphylococcus aureus on the mark with the formation of a clear zone around the wells which starts from a concentration of 20%.

Abstract

Penyakit menular yang disebabkan oleh vektor seperti demam berdarah   dengue,   malaria,   filariasis,   encephalitis,   masih   menjadi   masalah kesehatan masyarakat di dunia termasuk Indonesia. Demam berdarah dengue adalah penyakit yang disebabkan oleh virus Dengue yang ditularkan ke manusia melalui gigitan nyamuk Aedes sp yang terinfeksi virus Dengue. Nyamuk Aedes sp berkembangbiak di tempat-tempat penampungan   air   yang   mengandung   air   jernih   atau   air   yang   sedikit terkontaminasi.  Penelitian  ini  bertujuan  untuk  mengidentifikasi  jenis  larva nyamuk yang ditemukan di tempat penampungan air dan menghitung kepadatan larva nyamuk di RT 04 RW 15 Padukuhan  Dero, Desa Condongcatur. Jenis penelitian ini adalah deskriptif dan desain penelitian adalah Cross sectional.  Pengambilan sampel dilakukan di 50 rumah yang berada di RT 04 RW 15 Padukuhan Dero Desa Condongcatur. Hasil penelitian menunjukkan bahwa jenis larva nyamuk yang ditemukan di   tempat   penampungan   air   di   RT   04   RW   15   Padukuhan   Dero,   Desa Condongcatur adalah larva Aedes aegypti. Wilayah ini  berisiko tinggi untuk penularan DBD dengan kepadatan larva nyamuk masing-masing untuk House Index (HI) adalah 44%, Container Index (CI) adalah 17,8% dan Angka Bebas Jentik (ABJ) yang di bawah 95%, yaitu sebesar 56%. Kata Kunci: larva, nyamuk, penampungan air

Abstract

Madura has at least 22 genotypes of local tobaccos (Nicotiana tabacum L.). This diversity could potentially produce new genotype of tobaccos with superior characters. However, information of the genetic diversity of Madura tobaccos is still limited. The aim of this study was to determine the genetic variation and relationship of 24 genotypes of Madura tobaccos with Random Amplified Polymorphic DNA (RAPD) analysis. In this research we were used 6 single primers for amplification: (OPA-18, OPB-12, OPB-14, OPC-1, OPC-8 and OPC-19) and 2 mixture primers ((OPB-12+OPC-8) and (OPC-1+OPC-19)). Genetic similarity and clustering was analyzed with Unweighted Pair Group Method Arithmetic (UPGMA) method with Numerical Taxonomy and Multivariate Analysis System (NTSYS) version 2.10 software. From this research we found that OPA18425, OPB12450, OPC8500, (OPC19+OPC1)550 and OPC8800 can be used as specific markers. Polymorphic bands percentage with mixture primers was relatively equal with single primers (<60%). The dendogram showed that Madura tobacco genotypes consist of 2 main clusters: cluster A (22 genotypes) and cluster B (2 genotypes: Bukabu Sa’ang and Prancak-95). Madura tobaccos had high genetic similarity between genotypes ranging from 0.80-1.00.

Abstract

Indonesian tropical climate is a country and rich in marine products. Shrimp petis is a fermented products of shrimp. Shrimp petis is stored for too long risk for overgrown microorganisms. Therefore, during storage by a factor of humidity, shrimp petis can be polluted and overgrown by the fungus Aspergillus species. Fungus Aspergillus flavus commonly known as the cause of contamination of various types of food stored. Aflatoxin are toxins produced by fungi Aspergillus flavus. Aflatoxins can invade the central nervous system, some of which are carcinogenic cause cancer of the liver, kidneys, and stomach. The study objective was to determine the growth of the fungus Aspergillus flavus on various brands of shrimp paste are sold in supermarkets and traditional markets. Methods This study is a descriptive method of research objects shrimp paste with different brands sold in supermarkets and traditional markets with a single variable, namely the growth of Aspergillus flavus. The data obtained were analyzed descriptively using SPSS 17.0. Research shows the growth of the fungus Aspergillus flavus on various brands of shrimp paste are sold in supermarkets and traditional markets. The percentage growth of the fungus Aspergillus flavus on various brands of shrimp paste is 20% of the 10 samples identified shrimp paste.Keywords: Petis shrimp, Aspergillus flavus, aflatoxin.
