Abstract

Helminth infection remains one of health problems of Indonesian people for years. The tropical condition of Indonesian climates with high level humidity has believed to be the one of factors that increase the number of this infection cases. Based of many studies, the number of helminth infections is related to the poor of hygiene and sanitation. This study is conducted to know the percentages of helminth infections among 0-14 years old children in Dusun Ngemplak, Desa Ngemplak Seneng, Klaten and also their personal hygiene with questionaire instruments. To know helminth infection in subjects, we examine faeces sample with simple method and 1% eosin dye. As the results of examination, we found Trichuris trichiura egg in three samples. This might related to the poor of their hygienic behaviors. Some of the behavior such as wash hand before eating and after defecation, use sandal or shoes when playing, consume helminthic drug for last 6 months, and defecate in toilet has shown percentage under 70%. Other behaviors such as cutting and cleaning nails and use a spoon when eating has shown percentage above 70%.

Abstract

Klebsiella pneumoniae is one of the most bacteria found in urine of Urinary Tract Infection (UTI) Patients. Study in many countries has found that this species become resistant to more than one antibiotics. This research goal is to know the effectiveness of talok (Muntingia calabura) infused water inhibiting the growth of K. pneumoniae. This was an experimental study using K. pneumoniae bacteria isolated from urine samples using diffuse agar method. The concentration of talok infused water was varied from 25-100% and the antibiotics used was cefotaxime, meropenem and chloramphenicol. The inhibition activity was measured by the diameter of inhibition zone. The results show that K. pneumoniae is still give sensitive response to antibiotic treatments. While the inhibition zone from talok infused water treatments can be categorized in middle and strong activity. From this study we can conclude that talok infused water potential in inhibiting the growth of K. pneumoniae. Future study still needed to develop this potential, especially for the bacteria that has been resistant to some antibiotics.

Abstract

Lactobacillus has long been known to reconstruct the microbiome of the urogenital organs of healthy people. However, changes in lactobacillus abundance are known occurring in urogenital tract infections (UTI) patients. It is also known that there are Lactobacillus species that have specific genomic characteristics and nutritional requirements that cause them to turn into pathogens. Understanding population characteristics and taxa composition that reconstruct urine microbiome is very important for the diagnosis and treatment of urogenital infections. This review provides an update information of metagenomic sequencing to identify the urinary microbiome profile and the role of lactobacilli in healthy and UTI patients.

Abstract

Standard microbiological culture techniques can only identify a fraction of the urogenital microbiome. Meanwhile, identifying and characterizing infectious microorganisms are very important for the success of diagnosis and treatments, especially for Urinary Tract Infection (UTI) patients. This study aimed to characterize the urogenital microbiome of UTI patients using 16S rRNA gene sequencing. We sequenced two pooled DNA samples from voided urine of UTI patients (21 females and 13 males). To determine the structure and composition of taxa in the samples, 16S rRNA gene sequencing was performed using the Illumina Mi‐Seq paired‐end platform. The most abundant genera were Burkholderia‐Caballeronia‐Paraburkholderia (71%) followed by Prevotella (33%), Escherichia‐Shigella (24%), Klebsiella (23%) and Sneathia (10%). The female microbiome was dominated by Prevotella bivia (28%), Escherichia coli (24%), Sneathia sanguinegens (7%) and Klebsiella pneumoniae (4%). On the other hand, the male microbiome was dominated by K. pneumoniae (23%) and E. coli (2%). K. pneumoniae and E. coli were the most abundant species found in both microbiomes. The 16S rRNA gene sequencing used in this study successfully uncovered the composition of the urogenital microbiome, which might not have been possible with conventional culture methods.
