Abstract

Abstract

Widal examination is a serological method to diagnose typhoid fever used by Godean II Public Health Center, Sleman Regency, Yogyakarta Special Region (DIY). However, this method is difficult to hold onto because there is no agreement on agglutination standards. A positive Widal test result is not necessarily typhoid fever. Therefore, the results really need to be confirmed by other supporting examinations. IgM anti Salmonella IgG rapid test is one of the serological tests to support the diagnosis of typhoid fever which has better sensitivity and specificity to handle specific antibodies against Salmonella typhi in serum. This study aims to examine the results of the serological test of Immunoglobulin Miu (IgM) Anti Salmonella gamma immunoglobulin (IgG) on positive widal samples at Godean II Public Health Center and to see the lowest positive widal titers IgM antisalmonella in the sample. This research is a descriptive study with accidental sampling technique. The method of examination used a rapid test immunochromatography to examine IgM IgG anti Salmonella typhi on positive widal serum samples at Puskesmas Godean II, Sleman, DIY. The results showed that 21.7% of the samples were positive IgG negative IgM; 8.7% of IgM samples were positive IgG positive, and 69.6% IgM negative IgG negative. Anti Salmonella IgM in widal positive samples appeared at titer 320. Anti Salmonella typhi IgM and IgG were detected in positive widal samples at Godean II Public Health Center. Immunoglobulin Miu (IgM) appears at titer 320.

Abstract

The microbiome that inhabits the human body plays a role not only in maintaining health but also in causing diseases, such as urinary tract infections (UTIs). This occurs due to changes in the host's body and causes changes in the composition of the microbiota that make up the microbiome. This case report contains the results of 16S rRNA gene sequencing in the urine specimens of a 41-year-old female patient diagnosed with UTI. Illumina MiSeq was carried out for 16S rRNA gene sequencing in the V3-V4 hypervariable region. The 16S rRNA sequence yielded a total of 98,754 high-quality reads with 283 species and 299 operational taxonomic units (OTUs) identified. Based on the sequencing results, we found an overabundance (93%) of Lactobacillus iners as the most abundant and dominant taxa. Lactobacillus iners is one of the bacteria that is difficult to culture. However, with the 16S rNA gene sequencing technology, these bacteria can be detected along with their abundance in the structure of the urogenital microbiome. Bacterial overabundance may have occurred as a result of changes in the host ecology, resulting in UTI clinical symptoms. This result indicates that, in addition to maintaining the health of the urogenital organs, L. iners can also be the causative agent of UTI. An ecological perspective appears to be necessary to determine the composition and function of the microbiota, especially in patients with UTI. The data set has been sent via NCBI as a Sequence Read Archive (SRA).

Abstract

Background: Pneumonia is an acute infection of the lower respiratory tract characterized by coughing and shortness of breath caused by Klebsiella pneumoniae bacteria. Sansevieria contains flavonoids, saponins, alkaloids, and steroids known to have antibacterial properties that can destroy the bacterial cell walls.  Objectives: This study aimed to determine the antibacterial activity of Sansevieria leaf extract against the growth of K. pneumoniae.  Materials and Methods: Detection of antibacterial activity of Sansevieria was conducted with disc-diffusion test. The presence of antibacterial activity is indicated by the appearance of inhibition (clear) zone after applying the extract at a concentration of 60-100% to the culture medium. As a positive control, the antibiotic chloramphenicol was used.  Results: The higher the extract concentration, the higher the inhibition zone formed. The inhibition zone formed from extract concentrations of 60%, 70%, 80% is classified as medium (d = 8; 8.5; 9 mm). Meanwhile, the inhibition zones at concentrations of 90% and 100% are 10.5 and 13.5 mm and are classified as strong. The clear zone of the positive control was 29 mm (very strong). Conclusions: Sansevieria leaf extract has the potential to inhibit the growth of K. pneumoniae bacteria.

Abstract

Staining is an important stage in preparing microscopic preparations so that the histological structure can be observed clearly. Hematoxylin-Eosin is the most widely used dye. Unfortunately, eosin is a synthetic chemical compound that is carcinogenic. To reduce the health and environmental impacts of eosin compounds, it is important to look for alternative dyes from natural ingredients. Hibiscus (Hibiscus rosa-sinensis L.) is a plant known to contain anthocyanins which can be used as eosin substitutes. This study aims to determine the potential of hibiscus extract as a substitute for eosin in histological staining. In this study, unstained liver tissue preparations from mice (Mus musculus) were used. Hibiscus extract dye was compared with eosin as a control. The color quality of the tissue preparations was used as an experimental parameter. The results of the microscopic assessment showed that liver tissue stained with Hibiscus extract at concentrations of 60% and 80% was less good than eosin. However, because tissue can stain reddish, hibiscus extract has the potential to be used as an eosin substitute. The right extraction method may be able to produce intact anthocyanin so that it can color the tissue optimally.
