Abstract

Over the past decade, DNA barcoding has provided new insight into fish ecology and biosystematics and led to new species' discovery. DNA barcoding is a method for the recognition and identification of species using short, standardised DNA fragments. The correct taxonomic identification of species is critical for the assessment and monitoring of biodiversity. This study applied DNA barcoding techniques to identify selected fish species from a mangrove-based estuary in Way Kambas National Park, Lampung Province, Indonesia. The gene encoding cytochrome c oxidase subunit I (COI) was amplified and bi-directionally sequenced from 22 specimens. The resulting 680 base pairs (bp) sequence was used to identify species, obtain phylogenetic information, and analyse genetic distances. A neighbour-joining tree was constructed based on the mitochondrial COI gene using the Kimura two-parameter model. This study also exhibits conservation status for those identified species. Our findings will facilitate future studies of fish species diversity in mangrove estuary-based ecosystems and provide preliminary data in policymaking in conservation areas such as National Park.

Abstract

Infertility is currently an important problem in the world, including in Indonesia. Infertility can occur in men and women. Inmen, factors causing infertility can come from external sources. One environmental factor that can cause infertility is exposureto heavy metals from the environment. One way to prevent and treat infertility includes consuming natural antioxidants derivedfrom plants. One plant that has antioxidant compounds and has antioxidant activity is the kabau plant. This research wasconducted to test the effect of kabau seed pod extract (Archidendron jiringoides Komariah) on male mice exposed to Pb metalto determine its effect on the concentration of spermatozoa in male mice. The methods used in this research include:preparation of test animals, making simplicia and kabau seed pod extract, exposing the extract to test animals, isolatingorgans, and making spermatozoa suspensions. Giving kabau seed pod extract to mice that had previously been exposed tolead (Pb) increased the concentration of mice spermatozoa with the most effective dose of kabau bark extract, namely 190mg/kg BW. However, statistical analysis shows that the increase is not significant.

Abstract

The Calotes genus presents a challenge due to the complexity of its species. However, research on the cryptic species complex within the Indonesian Calotes genus is still lacking. This study aims to determine the extent of genetic relationships and assess the potential existence of a species complex within the Indonesian genus Calotes (Daudin, 1802) (Squamata, Agamidae) using the partial Cytochrome c Oxidase Subunit 1 (COI) gene sequence as a molecular marker. Samples of the Indonesian Calotes genus in this study were collected from South Lampung (Lampung), Bogor (West Java), and Langkat (North Sumatra). By aligning 582 bp sequence similarities with reference sequences in GenBank, we confirmed that seven out of eight samples analyzed belonged to Calotes vultuosus, while one sample was identified as Calotes versicolor. The identity values ranged from 96 to 100%. The C. vultuosus samples in this study displayed lower genetic distances, ranging from 0 to 3%, with the reference C. vultuosus sequence from Indonesia compared to the reference sequence from India, which ranged from 6 to 9%. Phylogenetic tree reconstruction, utilizing both maximum likelihood with IQ-Tree and Bayesian Inference with BEAST methods, further supports these findings. It reveals distinct groupings between C. vultuosus samples from Indonesia and India. These results suggest the potential occurrence of a species complex within the Indonesian genus Calotes. Furthermore, the inclusion of eight COI gene sequences from two Calotes species in the GenBank database has the potential to confirm the existence of previously undocumented species in Indonesia.

Abstract

Abstract

Wick system is an easy-to-do and energy-efficient hydroponic system. Used plastic bottles can be used as planting media, thereby reducing the amount of plastic waste. Pahawang Village is facing the problem of plastic bottle waste and there has been minimal effort to reduce and manage the plastic waste. Training in leafy greens cultivation using the wick system is very much needed by residents because the production can be consumed to meet the nutritional needs of the family. This activity aims to improve community knowledge and skills through training in vegetable cultivation with wick system using plastic bottle waste. The training succeeded in increasing the understanding and skills of participants regarding cultivation with the hydroponic wick system. There are several factors that drive this outcome, including the easy-to-understand content of material presentation, the easy wick system, and the motivation of the participants. Continuous community assistance needs to be carried out as a follow-up so that cultivation efforts can be continued. In that case, the community can generate economic benefits after fulfilling the nutritional needs of the family.
