Abstract

This study examines the effectiveness of biology instruction integrating climate change issues in enhancing preservice biology teachers' climate literacy and systems thinking. The study employed a quantitative approach using a quasi-experimental pretest-posttest control group design. The sample comprised 60 preservice biology teachers assigned to an experimental class and a control class, with 30 participants in each group. The experimental class received biology instruction integrated with climate change issues, whereas the control class received conventional instruction. Data were collected through a climate literacy test and a systems thinking test and analyzed using descriptive statistics, multivariate analysis of covariance (MANCOVA), and follow up univariate analysis of covariance (ANCOVA). The results reveal a significant difference between the two groups in the combined posttest scores of climate literacy and systems thinking after controlling for pretest scores, indicated by Wilks' Lambda = 0.302, F(2, 55) = 63.48 p < 0.001 Follow up analyses showed significant effects on climate literacy, F(1, 57) = 84.3 p < 0.001 , partial ?² = 0.597 and systems thinking, F(1, 57) = 56.23 p < 0.001 partial ?² = 0.497 The findings confirm that integrating climate change issues into biology instruction effectively strengthens preservice biology teachers' climate literacy and systems thinking.

Abstract

This study aimed to analyze the antibacterial activity of ethanolic extracts of bidens pilosa L. leaves and ginger rhizomes, both individually and in combination, and to evaluate their interaction against the growth of Escherichia coli in vitro. This study was a laboratory experiment employing a posttest-only control group design. The extracts were obtained through ethanol maceration. Antibacterial activity was tested using the agar well diffusion method to measure inhibition zone diameter, the dilution method to determine the Minimum Inhibitory Concentration (MIC) and Minimum Bactericidal Concentration (MBC), and the checkerboard assay to assess extract interaction through the Fractional Inhibitory Concentration Index (FICI). The ethanolic extracts of bidens pilosa L. leaves and ginger rhizomes exhibited antibacterial activity against E. coli, with stronger inhibition observed at higher concentrations. The extraction yields of ketul leaves and ginger rhizomes were 9.4% and 12.8%, respectively. The MIC values of ketul leaf and ginger rhizome extracts were 25 mg/mL and 12.5 mg/mL, respectively, while their combination decreased to 6.25 mg/mL. The combined extracts also produced larger inhibition zones than the single extracts. A FICI value of 0.75 indicated an additive interaction. The combination of ethanolic extracts of bidens pilosa L. leaves and ginger rhizomes has potential as a natural antibacterial agent against Escherichia coli. Aktivitas Antibakteri dan Interaksi Ekstrak Etanol Daun Bidens pilosa L. dan Rimpang Jahe terhadap Pertumbuhan Escherichia coli secara In Vitro ABSTRAK: Penelitian ini bertujuan menganalisis aktivitas antibakteri ekstrak etanol daun bidens pilosa L. dan rimpang jahe, baik secara tunggal maupun kombinasi, serta menilai interaksinya terhadap pertumbuhan Escherichia coli secara in vitro. Penelitian ini merupakan eksperimen laboratorium dengan rancangan posttest-only control group design. Ekstrak diperoleh melalui maserasi menggunakan etanol. Uji antibakteri dilakukan dengan metode difusi sumuran untuk mengukur diameter zona hambat, metode dilusi untuk menentukan Minimum Inhibitory Concentration (MIC) dan Minimum Bactericidal Concentration (MBC), serta checkerboard assay untuk menilai interaksi kombinasi ekstrak melalui Fractional Inhibitory Concentration Index (FICI). Ekstrak etanol daun bidens pilosa L. dan rimpang jahe menunjukkan aktivitas antibakteri terhadap E. coli, dengan daya hambat yang meningkat seiring peningkatan konsentrasi. Rendemen ekstrak daun ketul dan jahe masing-masing sebesar 9,4% dan 12,8%. Nilai MIC ekstrak daun ketul dan jahe berturut-turut sebesar 25 mg/mL dan 12,5 mg/mL, sedangkan kombinasi keduanya menurun menjadi 6,25 mg/ml. Kombinasi ekstrak juga menghasilkan zona hambat lebih besar dibandingkan ekstrak tunggal. Nilai FICI sebesar 0,75 menunjukkan interaksi aditif. Kesimpulan singkat: Kombinasi ekstrak etanol daun bidens pilosa L. dan rimpang jahe berpotensi dikembangkan sebagai agen antibakterii terhadap Escherichia coli.

Abstract

Kota Dua Beach is a tourist destination located in East Sarotari Village, Larantuka District, East Flores Regency. Until now, the number and types of gastropods in the intertidal zone of Kota Dua Beach are not yet known. The aim of this research is to determine the diversity and abundance of gastropod species on Kota Dua Beach and develop biology learning media in the form of videos. This research is development research using the ADDIE model. 17 species of gastropods were found, namely Haliotis squamata, Conus miles, Conus rosiae, Bostycapulus aculeatus, Cypraea tigris, Cypraea annulus, Tenguella granulate. Platydoris cruenta, Onchidium melakense, Patella caerulea, Trochus maculates, Turbo cornutus, Astralium calcar, Cypraea talpa, Lunella smaraga, Muricanthus nigritus and Cellana nigrolineata. The diversity index of 2.714527275 is in the medium category. The highest species abundance index was 0.85 ind/m², while the lowest was 0.1 ind/m². The results of video validation by media experts reached a percentage of 71.76%, and by material experts, 93.33% with an average of 82.54% categorized as very valid as learning media. The conclusion of this research is as follows: 17 species of gastropods were identified at Kota Dua Beach with a diversity index in the medium category, the highest species abundance was 0.80 ind/m², and the video developed about the diversity and abundance of gastropods at Kota Dua Larantuka Beach is classified as valid for use as a learning medium.
