Abstract

This study aims to analyze the effectiveness of the Direct Administration Service Program (Pelangi Adminduk) in Tegal Harum Village, West Denpasar District. This study uses a quantitative method, with a population of all households in Tegal Harum Village totaling 3,325 households. A simple random sampling technique was used, resulting in 357 households as the sample. Data were analyzed using research instrumentation tests and descriptive/efficiency. The research results indicate that: (1) The Pelangi Adminduk program in Tegal Harum Village is already effective, as demonstrated by organizational factors and high service recipient satisfaction; (2) The constraints affecting the effectiveness of the Pelangi Adminduk program include existing regulations and employee awareness; (3) Solutions to address these constraints involve (a) improving existing regulations to make them easier to understand and apply, and (b) enhancing employee awareness through intensive socialization.

Abstract

Background: The availability of safe and adequate sanitation can reduce high infant and under-five mortality rates. Access to sanitation in Bali Province reached 97.12% in 2021; however, the achievement of Open Defecation Free (ODF) villages remained very low at only 45.5%, below the national average of 48.7%. Even by the second quarter of 2022, out of 716 villages/ward, only 330 (46.09%) had achieved ODF status. Accordingly, this study was conducted to determine strategies for achieving ODF villages in Bali Province. Method: The study used a cross-sectional design. It was exploratory in nature, examining internal and external conditions in the form of strengths and weaknesses (internal factors) as well as opportunities and threats (external factors). These strategic factors were analyzed using a SWOT model. Results: The presence of rewards/incentives for ODF villages and an STBM monitoring system were showed significant strengths (score 0.63). The main weakness factor was the lack of cross-sectoral coordination (score 0.52). The highest-scoring opportunity factor was the triggering of behavior change and health promotion by community health centers (Puskesmas) (score 0.67), while the most significant threat factor was the limited availability of clean water facilities (score 0.73). Conclusion: Strategies for achieving ODF villages include: strengthening programs through regulations, budget allocation, monitoring-evaluation systems, cross-sectoral collaboration, sanitation technology development, capacity building of potential human resources, integration of STBM with other priority programs, one of which is the Settlement Sanitation Development Program. Abstract

Penelitian bertujuan untuk mengetahui efektifitas dosis dan usia biakan dari isolat Trichoderma spp. dalam pengendalain penyakit akar gada pada tanaman sawi. RAK Faktorial digunakan pada penelitian dengan 2 faktor dan  3 ulangan. Faktor pertama adalah dosisdari isolat Trichoderma spp. yang terdiri dari 3 level dosis, yaitu 500 ml; 250 ml dan 0 ml. Faktor kedua adalah usia biakan dari isolate Trichoderma spp. yang terdiri dari 3 lever usia biakan isolate Trichoderma spp yaitu usia biakan 1 minggu; usia biakan 2 minggu dan usia biakan 3 minggu. Pada tanaman sawi yang terinfeksi penyakit akar gada, perlakuan beberapa dosis isolat Trichoderma spp. dan perlakuan beberapa usia biakan dari Trichoderma spp. memberikan pengaruh yang berbeda nyata terhadap variabel tinggi tanaman sawi, daun tanaman sawi dan persentase penyakit. Perlakuan dari isolat Trichoderma spp. dosis 500 ml menghasilkan tinggi tanaman 21,68% dan  daun tanaman 26,63 %  lebih tinggi secara nyata dibandingkan  dengan tinggi tanaman dan  daun pada perlakuan  tanpa pemberian isolat Trichoderma spp. Perlakuan  dari isolat Trichoderma spp. dengan usia biakan 2 minggu,  menghasilkan tinggi tanaman 12,80 % dan daun 16,04% nyata lebih tinggi, dibandingkan dengan  tinggi dan daun tanaman pada perlakuan isolat Trichoderma spp. usia biakan 1 minggu . Persentase penyakit akar gada pada tanaman sawi pada perlakuan kombinasai isolat Trichoderma spp. dosis biakan 500 ml dan usia biakan isolat Trichoderma spp 2 minggu,  mencapai 0 %,  yang berarti tanpa adanya serangan penyakit akar gada. DOI: 10.37637/ab.v3i1.422

High impact of Clerodendrum paniculatum leaf extract to suppress zucchini yellow mosaic virus infection in zucchini plants. Biodiversitas 23: 2914-2919. Zucchini yellow mosaic virus (ZYMV) is an important virus in zucchini plants (Cucurbita pepo L.). ZYMV infection is prevalent in C. pepo in Bali and is extremely difficult to eradicate. This study aimed to determine the efficacy of pagoda leaf extract (Clerodendrum paniculatum L.) in reducing yield loss in C. pepo. A factorial randomized block design with two variables was employed in this study. The application time of the C. paniculatum extract was the first variable, and concentration was the second. Plant extract was applied during seed immersion, one day before and after inoculation, and on the same day of virus inoculation. The concentrations of C. paniculatum extract employed were 10%, 30%, and 50%. The crude extract was sprayed onto the C. pepo leaves. Plants treated with extract one day after virus inoculation at 10%, 30%, and 50% concentrations exhibited yellowing, yellowing with cupping, mosaic, mosaic with cupping, and stunting. While application of extract one day before virus inoculation at 30% and 50% concentrations resulted in yellowing and yellowing with cupping signs. Plants infected with ZYMV first showed flowering at 37 DAP, whereas plants treated with C. paniculatum leaf extract one day before virus inoculation started flowering at 32 DAP. Application of C. paniculatum leaf extract one day before ZYMV inoculation on C. pepo effectively suppressed 40% yield loss. The findings of present investigation revealed information about the potential of C. paniculatum leaf extract as a plant virus control agent through the mechanism of induction in C. pepo.

The antiviral potential of macroalgae in suppressing Sweet potato leaf curl virus (SPLCV) infection in sweet potatoes. Biodiversitas 24: 4079-4086. Sweet potato leaf curl virus (SPLCV) was first found in sweet potatoes in Indonesia in 2022. Prevention of spread of virus is essential, especially by using macroalgae extract which is environmentally friendly and has antiviral activity. The aim of present research was to test the potential of sea macroalgae to suppress SPLCV infection and to analyze phytochemicals of potential macroalgae containing an antiviral substance. Macroalgae extract was sprayed on the test plants that were infected by SPLCV. The observed parameters were changes in symptoms, disease incidence and severity, virus confirmation by PCR, and phytochemical analysis. The test results up to day 21 showed that Eucheuma spinosum was found to be effective in suppressing SPLCV infection in sweet potatoes, up to symptomless infection in young leaves. E. spinosum and E. cottonii suppressed disease incidence by 80% and 40% and lower disease severity as much as 71% and 48%, while E. serra showed less ability to suppress SPLCV infection. The two macroalgae had flavonoid, saponin, and steroid content which may be the reason to suppress the viral infection. The results of PCR analysis showed that microalgal extract had the highest nucleotide and amino acid homology with Gianyar (LC586170) isolate with values of 99.7 and 100%. The macroalgae with the highest ability to suppress the virus were E. spinosum and E. cottonii. This showed that the application of macroalgae extract did not change the amino acid sequence of SPLCV isolate.

Beneficial interaction between rice stunt virus and its insect vector Nilaparvata lugens Stal based on life table. Biodiversitas 24: 4690-4698. The brown planthopper (Nilaparvata lugens) causes direct and indirect damage as a stunting virus vector in rice. Insect vectors and viruses are closely related with respect to disease transmission to host plants. Thus, this research aimed to determine the specific relationship between a virus and its insect vector on the biology and demography statistics of viruliferous brown planthopper (BPH). Research method included the propagation of N. lugens and stunt virus inoculum, the observation of cohort, stadia duration and egg survival rates, imago life span, and N. lugens life table. The BPH life table was arranged with the jackknife method for two treatments: plants infected by stunt virus and healthy plants. The research result showed that plants infected by stunt virus can shorten the BPH nymphal development stage from instar 2 to instar 5, life cycle, preoviposition period, and double time. Plants infected by stunt virus did not influence the BPH hatching pattern but influenced the total number of hatching eggs. The next BPH generation of stunt virus inflected plants increased 29.51 times more from the previous generation, while on healthy plants, the next BPH generation only increased 27.51 times. Virus-infected plants generally appeared to be superior quality hosts for vectors compared to uninfected plants, thus enhancing vector life table and virus spread.

Begomovirus diversity and distribution on melon plants in Bali, Indonesia. Biodiversitas 25: 572-582. Melon crops in Bali, Indonesia often exhibit curling and yellowing symptoms in young plants, the cause of which remains unidentified. This research aimed to determine the presence of Begomovirus isolates from Bali and their distribution among melon plants. The research methods involved a comprehensive survey, meticulous sampling, precise virus detection with PCR, thorough sequencing analysis, and accurate calculation of viral disease incidence based on DNA sequencing results. Sampling and symptom observation were conducted in seven Bali regencies: Denpasar City, Gianyar, Badung, Tabanan, Buleleng, Jembrana, and Bangli. The PCR method with Begomovirus universal primers SPG1/SPG2 was used for virus identification, followed by nucleotide sequencing. The research identified three viruses infecting melon plants in Bali: Squash leaf curl virus (SLCuV), Squash leaf curl China virus (SLCCNV), and Squash leaf curl Philippines virus (SLCuPV). The highest disease incidence for SLCuV, SLCCNV, and SLCuPV was found in Denpasar, Buleleng, and Badung, at 40%, 30%, and 30%, respectively. SLCCNV has spread to seven regencies in Bali, namely Denpasar City, Gianyar, Tabanan, Buleleng, Bangli, and Jembrana. SLCuV has spread to six regencies in Bali, all except Badung Regency. SLCuPV was only found in Denpasar City, Badung, and Buleleng. The molecular characteristic of SLCuV Bali isolate has the closest nucleotide and amino acid homology with East Timor isolate (KY652743) which is 98.4% (99.0%), SLCCNV Bali isolate is closest with Malaysia isolate (EF197940) which is 98.8% (99.8%), and SLCuPV is closest with Taiwan isolate (JF746195) which was 98.8% (99.5%). The novelty of this research lies in the first report of SLCuPV infection in Curcubitaceae plants in Indonesia, a finding that could significantly impact our understanding of viral infections in crops.

Tuktuk) from different source materials applied with liquid biofertilizers. Nusantara Bioscience 12: 127-133. This research was to examine growth and yield of shallots using different sources of propagation material, namely true shallot seed (TSS) and bulbs. Soil biological fertility, which was generally low, was improved by the addition of liquid organic fertilizer. The purpose of this study a) to determine the differences in the propagation of plants from seeds and bulbs of shallot Tuktuk varieties, and b) to determine the effect of liquid biofertilizer maxigrow and rhizobacteria. The study used a one-factor randomized design. The results showed that the growth and yield of shallots propagated with bulbs were better than the origin of the seeds. The treatment of the two types of liquid biofertilizer produces tangible growth and yield, but there was no significant difference between the two kinds of liquid organic fertilizer.
