Abstract

Local krai cucumber (Cucumis sativus L.) accessions represent valuable genetic resources for the development of cultivars with desirable agronomic and fruit characteristics. This study aimed to determine the agronomic characteristics of three local krai cucumber accessions as a basis for selecting potential accessions. The study was conducted from December 2025 to March 2026 at the Karangsari experimental field, Sleman, Yogyakarta, at an altitude of approximately 200 m above sea level. The study used a one-factor randomized complete block design (RCBD), consisting of three local krai cucumber accessions, namely CBT-246 Lamongan, CBT-248 Sidoarjo, and CBT-267 Rembang, with six replications. The observed parameters included fruit weight per plant, number of fruits per plant, fruit weight per plot, number of fruits per plot, fruit length, fruit diameter, leaf blade length, leaf blade width, flowering time, and first harvest time. Data were analyzed using ANOVA at the 5% significance level, followed by Tukey’s HSD test at α = 0.05. The results showed that the three accessions differed significantly in fruit weight per plant, fruit length, fruit diameter, leaf blade width, and flowering time. Accession CBT-267 Rembang showed superior performance in the highest fruit weight per plant, reaching 841.72 g, the greatest fruit length, reaching 39.42 cm, and the earliest flowering time, at 22.00 days after planting (DAP). Meanwhile, CBT-246 Lamongan had the largest average fruit diameter, at 5.78 cm, and the widest leaf blade, at 17.90 cm. Based on these results, CBT-267 Rembang has potential as a promising accession for the selection of long-fruited types with high fruit weight per plant. However, further multilocation and multiseason trials are required to confirm the stability of its agronomic characteristics.

Abstract

White jack bean (Canavalia ensiformis L.) is a promising underutilized legume with potential applications as an alternative food crop and genetic resource for crop improvement. However, information on the morpho-agronomic performance of local accessions remains limited. This study evaluated morpho-agronomic variation among four local white jack bean accessions and identified accessions with desirable traits for preliminary selection. The experiment was conducted from November 2025 to March 2026 at the Teaching Factory Garden of Politeknik Pembangunan Pertanian Yogyakarta–Magelang, Indonesia. A randomized complete block design was used, comprising four accessions, namely FBKR-1060, FBKR-1062, FBKR-1064, and FBKR-631, with six replications. Quantitative traits included leaf length, leaf width, plant height, days to flowering, days to harvest, pod length, pod width, and seed diameter. Qualitative traits comprised growth habit, leaf apex shape, leaf base shape, pod shape, and seed color. Quantitative data were subjected to analysis of variance followed by Tukey’s honestly significant difference test at the 5% significance level. Significant accession effects were observed for days to flowering, days to harvest, pod length, pod width, and seed diameter. FBKR-1060 and FBKR-631 flowered and reached harvest earlier than FBKR-1062 and FBKR-1064. FBKR-1064 and FBKR-631 produced the longest pods and largest seed diameters, while FBKR-631 had the widest pods. All accessions exhibited a determinate growth habit, pointed leaf apex, obtuse leaf base, slightly curved pods, and white seeds. These findings indicate that quantitative traits are more informative than the observed qualitative descriptors for differentiating the evaluated accessions. FBKR-631 is a promising accession for further multi-environment and yield evaluations because it combines earliness with relatively large pod and seed dimensions. Keywords: accession; Canavalia ensiformis; earliness; morpho-agronomic traits; preliminary selection

Abstract

Long beans (Vigna unguiculata var. sesquipedalis) require superior strain development to support increased productivity. This research aims to identify the morphological diversity of the F4 generation long bean strain and determine the potential strain for further selection. The study used the individual observation method without repetition to evaluate the phenotypic segregation in three strains F4 (G1 = Strain 1, G2 = Strain 2, and G3 = Strain 3) with the Parama and Pertiwi varieties as comparisons. A total of 19 morphological characters were observed in 231 individuals of long bean plants of the F4 generation. Each plant is treated as an individual selection, analyzed using Hierarchical Cluster Analysis (HCA) with the UPGMA method. The main distinguishing characters between clusters include the coloring of anthocyanin pods, the color of the old pods, the color and pattern of the seeds, the color of the seeds in the hilum circle, and the color of the flower petals. G2, G3 strains, and most of the G1 strains are recommended as further selection materials based on the suitability of the observed phenotype character. Meanwhile, G1.8, G1.9, G1.12, G1.52, G1.74 and G1.79. have the potential to be used as cross elders because they show differences in morphological characteristics that can support the formation of diversity in the segregan generation. Keywords: Morphological Diversity, Cluster Analysis, Dendrogram, F4 Generation, Advanced Selection

Abstract

This study explores the use of embryo culture as a strategic approach to rescue maize (Zea mays L.) embryos derived from interspecific crosses that fail to develop normally. Not all crosses result in viable seeds; in many cases, embryos remain immature or undergo developmental arrest, which increases the risk of embryo abortion before they can develop into complete plants. Embryo development failure in maize crosses, especially in combinations involving genetically distant genotypes, represents a major constraint in plant breeding programs. Without appropriate intervention, the resulting hybrid genotypes may be lost and consequently slow down the development of new superior varieties. To address this limitation, immature embryo culture represents an effective in vitro technique. Embryo culture involves the excision of young or incompletely developed plant embryos from seeds or ovaries and their subsequent cultivation on nutrient-rich artificial media to support further growth and plant regeneration. The objective of this study was to identify the optimal Murashige and Skoog (MS)-based culture medium composition for immature maize embryo growth using a factorial completely randomized design with three replications. Treatments consisted of combinations of agar concentration (4, 6, 8, and 10 g L⁻¹) and medium strength (0.5 MS, 0.75 MS, and 1 MS). The results indicated no significant interaction between MS medium strength and agar concentration. 1 MS medium produced the best results for shoot height, root length, and root number, whereas 0.75 MS medium resulted in the highest germination percentage. No significant differences were observed among treatments for days to germination and leaf number. An agar concentration of 6 g L⁻¹ yielded the highest germination percentage and shoot height, while other variables were not significantly affected by agar concentration. In conclusion, 1 MS medium or an agar concentration of 6 g L⁻¹ can be recommended for the culture of immature maize embryos

Abstract

This study aimed to analyze the effects of pruning at different crop ages on the physical, physiological, and biochemical quality of rice seeds, as well as to identify seed responses to variations in pruning timing. The experiment was conducted using a non-factorial Completely Randomized Design on the rice variety Inpari 32. Four pruning treatments were applied, namely no pruning (P0), pruning at 25 days after transplanting (DAT) (P1), 35 DAT (P2), and 45 DAT (P3), with three replications for each treatment. Observations focused on physical seed quality parameters, including moisture content, seed purity, and 1000-seed weight, as well as physiological seed quality parameters, namely germination percentage, vigor index, growth rate, rapid emergence (RE), percentage of normal seedlings, percentage of non-emerged fresh weight, and seed biochemical composition (total sugars, starch, and carbohydrates). Data were analyzed using analysis of variance (ANOVA) at a 5% significance level, followed by Duncan’s Multiple Range Test. The results revealed that pruning treatments did not significantly affect moisture content, seed purity, growth rate, germination percentage, vigor index, rapid emergence, percentage of abnormal seedlings, non-emerged fresh weight, starch content, or carbohydrate content. However, the highest 1000-seed weight was observed in the non-pruned treatment. In contrast, pruning at 35 DAT resulted in the highest total sugar content compared to other treatments, indicating its potential contribution to improved seed physiological quality. These findings suggest that pruning at an appropriate growth stage may influence specific seed quality attributes, particularly biochemical characteristics.

Abstract

This study aimed to characterize the morphological and agronomic traits of five local tomato accessions from Kutoarjo (A, B, C, D, and E) as a foundation for plant breeding programs. The experiment was conducted using a randomized complete block design with six replications. Observations included qualitative and quantitative characters based on the IPGRI and UPOV descriptor guidelines. The analysis results proved that vegetative traits, such as plant height, stem diameter, and leaf size, did not show significant differences among the accessions. In contrast, generative traits (days to flowering, fruit size, productivity, and seed weight) showed highly significant differences. The Kutoarjo E accession exhibited the best agronomic performance with a productivity of 2.20 kg/plant, 67.83 fruits/plant, and a 1000-seed weight of 3.47 g. In conclusion, these local tomato accessions possess phenotypic and agronomic variations with great potential for sustainable utilization as basic breeding materials.
