Abstract

Association studies of SNPs have become very important in determining how genetic variants are linked to complex diseases, quantitative traits, and physiological responses. Genetic polymorphisms in the GLP-1R gene have potentially decreased protein stability and are associated with many diseases, especially diabetes and obesity. This study aimed to screen the expression and investigate the genetic polymorphism of the GLP-1R by using several beneficial bioinformatics tools. We observed database Ensembl, GTEx portal, and KEGG to identify the structure, expression, and molecular pathway of GLP-1R. In silico computational methods (SIFT, PolyPhen v2, PROVEAN, and PhD-SNP) were used to identify nsSNPs of the GLP-1R that potentially influence protein structure and function. I-Mutant was used to investigate possibly damaging nsSNPs, while GeneMANIA was used to investigate GLP-1R gene-gene interactions. GLP-1R is localized on chromosome 6p21, contains 13 exons, and has the regulation variant (CTCF, promotor, enhancer, and promotor flank region). GLP-1R is highly expressed in the pancreas to stimulate glucose-dependent insulin secretion and suppress glucagon secretion. Seven nsSNPs of the GLP-1R gene were found to be potentially deleterious: rs10305421, rs201672448, rs10305492, rs2295006, rs6923761, rs1042044, rs140642887, and rs10305510. I-Mutant server showed that nsSNPs rs140642887 was unstable, decreased GLP-1R protein stability, and impaired other genes' interaction and function (SP1, SP3, GNAS, and GCG). This study is the first in silico analysis of the polymorphic GLP-1R gene, and will serve as a great resource for developing precision medications to treat diseases associated with these polymorphisms.

Abstract

Kirsten rat sarcoma viral oncogene (KRAS) gene mutations lead to abnormal activation of the RAS signalling pathway and have been associated with poor prognosis and resistance to some therapeutics. This study aimed to identify mutation characteristics of the KRAS genes codon 12 and 13 in colorectal cancer patients in West Sumatra. KRAS mutations were analyzed in 20 DNA of colorectal cancer patients' tissue samples by using polymerase chain reaction (PCR) with specific primer and direct sequencing analysis. Our findings showed five samples (25%) with mutated KRAS at codons 12 and 13 (including three samples with GGT→GAT, one sample with GGT→GTT in codon 12, and one sample with GGC→GAC in codon 13). In conclusion, we found two variations of amino acid changes at codon 12 (G12D and G12V) and one at codon 13 (G13D). More research with many samples is required to obtain conclusive data on the relationship between these gene mutations and colorectal cancer response to therapy and prognosis.

Abstract

Diabetic foot ulcer (DFU) has been associated with genetic and environmental factors, which could potentially have a role in DFU development. Single nucleotide polymorphisms (SNPs) in genes linked to DFU, including inflammation. Bibliometric studies on the SNP on genes affecting DFU still have not been evaluated. This study aims to depict bibliographically and understand the topic trend of genetic polymorphism and the risk of DFU publications. A bibliometric methodology was applied in this study. The data were extracted through the Scopus database from 2011 to 2021. VOS viewer was used to classify and summarize Scopus articles. The 35 articles were evaluated. India topped the list of countries with the most publications, and Tehran University of Medical Sciences was the primary institution. Singh K and his team were the first contributing authors with 44 citations. Keywords analysis indicated that the research hotspots were DFU, type 2 diabetes mellitus (T2DM), diabetic foot ulcers, polymorphisms, hypoxia, vascular endothelial growth factor (VEGF), diabetic foot, diabetes mellitus, and oxidative stress. This study summarizes the current state, trends in genetic polymorphism and risk with DFU research. It may provide researchers with insight into the genetic polymorphism and risk associated with DFU research, as well as useful information for identifying possible collaborators and partner institutions.

Abstract

Abstrak: Salah satu masalah yang dihadapi oleh Pimpinan Cabang Istimewa Muhammadiyah (PCIM) Thailand adalah semangat berdakwah menegakkan dan menjunjung tinggi ajaran Islam sehingga terwujud masyarakat Islam yang sebenar-benarnya menurun, hal ini disebabkan karena: (a) masa peralihan kepemimpinan Cabang Istimewa yang baru, dan (b) kondisi sosial-politik dan budaya yang berbeda dengan kondisi di Indonesia. Pengabdian ini bertujuan untuk memperkuat semangat berdakwah di kalangan PCIM Thailand Muhammadiyah melalui peningkatan pemahaman ideologi Muhammadiyah sebagai gerakan pemurnian dan pembaruan. Metode yang digunakan meliputi sosialisasi, penyuluhan, dan praktek yang melibatkan sepuluh orang PCIM Thailand. Kegiatan dilakukan dalam bentuk ceramah, diskusi interaktif, dan simulasi ibadah sesuai Himpunan Putusan Tarjih Muhammadiyah. Evaluasi dilakukan melalui pengamatan, dan self assessment. Dari hasil angket responsi dengan skala lima dapat disimpulkan bahwa kegiatan penguatan ideologi Muhammadiyah bagi PCIM Thailand ini rata-rata berada pada kategori sangat baik (90%) yaitu: semua materi pelatihan penting, menambah pengetahuan tentang ideologi Muhammadiyah, mencerahkan dan relevan untuk dikaji lebih lanjut, memuat pengetahuan baru yang bermanfaat bagi PCIM Thailand dalam dakwah di tengah-tengah masyarakat yang pluralistik, dan mereka berada pada kondisi minoritas.Abstract: One of the challenges faced by the leadership of Pimpinan Cabang Istimewa Muhammadiyah (PCIM) Thailand is the declining spirit of da'wah to uphold and promote Islamic teachings for establishing a truly Islamic society. This decline is attributed to: (a) the transitional period of new leadership within the PCIM, and (b) the socio-political and cultural conditions in Thailand, which differ significantly from those in Indonesia. This community service initiative aims to revitalize the spirit of da'wah among PCIM Thailand members by enhancing their understanding of Muhammadiyah ideology as a movement of purification and renewal. The methods employed include socialization, counseling, and practical activities involving ten PCIM Thailand members. The activities were conducted through lectures, interactive discussions, and worship simulations following the Himpunan Putusan Tarjih Muhammadiyah. Evaluation was carried out through observation and self-assessment. Based on a five-point scale questionnaire, the findings revealed that the Muhammadiyah ideology reinforcement program for PCIM Thailand was rated as "excellent" (90%). Participants highlighted that the training materials were essential, enriched their knowledge of Muhammadiyah ideology, were enlightening and relevant for further study, and provided new, valuable insights. These materials are particularly significant for PCIM Thailand's da'wah efforts in a pluralistic society where they are a minority.

Abstract

Penyakit tidak menular seperti diabetes melitus, hiperkolesterolemia, dan hiperurisemia merupakan tantangan utama kesehatan masyarakat di Indonesia, terutama karena rendahnya deteksi dini di tingkat komunitas. Pengabdian masyarakat ini bertujuan mendeskripsikan hasil deteksi dini risiko penyakit metabolik melalui skrining gula darah, kolesterol, dan asam urat pada Ibu PKK di Kampung Wisata Bukit Nobita, serta memberikan rekomendasi upaya promotif dan preventif. Kegiatan dilakukan melalui observasi, koordinasi, penyuluhan, dan pemeriksaan kesehatan pada 15 anggota PKK, dilanjutkan evaluasi pengetahuan dan kepuasan peserta. Hasil menunjukkan mayoritas peserta memiliki lingkar pinggang di atas normal, kadar gula darah, kolesterol, dan asam urat melebihi batas normal, serta beberapa kasus tekanan darah tinggi, yang menandakan tingginya risiko sindrom metabolik di komunitas ini. Kegiatan ini meningkatkan pengetahuan peserta tentang status kesehatan dan pentingnya perubahan gaya hidup sehat. Temuan ini menegaskan perlunya skrining rutin dan edukasi berkelanjutan di komunitas, serta kolaborasi dengan fasilitas kesehatan untuk menurunkan risiko penyakit tidak menular. Hasil pengabdian ini penting sebagai dasar intervensi kesehatan masyarakat berbasis komunitas di masa mendatang.

Abstract

The Thai government has realized the serious problem of environmental pollution due to plastic waste. In everyday life, plastic has become an important and integral part of societies around the world due to its various uses, such as packaging, agriculture, auto parts, electronic applications and medical devices. This results in approximately 2 million tons of plastic waste being generated per year. Managing organic waste in the form of stacked buckets does not require large areas of land and equipment is easy to obtain. This activity includes: a. training and practice in making stacked buckets, b. training in making crafts from inorganic waste, and c. digital marketing training for product sales. The expected output of this activity is that Muhammadiyah residents in Thailand have an understanding regarding making stacking buckets independently so that organic waste management is more optimal. The next output is increasing the community's skills in making crafts by utilizing inorganic waste and turning it into a product to be marketed and can increase family income and well-being.
