Abstract

Attention on chitin degrading enzymes has been growing since their ability to reduce the waste of shrimp/other crustaceans processing industries and converting them into value added products such as biologically active chitin and chitosan oligomer. Previous experiment found that KLU 11.16 isolate was one of the potential bacteria isolated from shrimp waste producing chitinolytic enzymes including chitosanases. A study on the identification of KLU 11.16 extracellularcrude enzyme was carried out by cultivating the bacteria on chitin medium. Due to the wide application of chitosanase, the characterization of the crude chitosanase was carried out after an identification of the enzymes secreted. Based on assessment using zymogram technique, this bacteria secreted a mixed extracellular chitinolytic enzyme and other hydrolytic enzyme. The crudechitinolytic enzyme degraded 85% deacetylated (DA) better than 100% DA chitosan, and slightly degraded glycol chitin, indicating that KLU 11.16 secreted chitosanases and chitinases enzyme. In addition to the chitinolytic enzyme, the bacteria also secreted protein and carbohydrate degrading enzymes when running at SDS-PAGE enriched with casein, soluble starch and CMC substrates.Crude chitosanases enzyme was performed well at pH 6 and temperature of 300C, and the activity can be increased by addition of 1 mM Fe 2+ in form of chloride salt. Addition of detergent, i.e1mM of Triton X-100 and SDS slightly decreased the activity. Future application of the crude chitosanase from KLU 11.16 was on producing chitosan derivative such as chitosan oligomer using substrateof 85% DA chitosan, which is more digestable by other enzymes secreted by KLU 11.16

Abstract

Application of biocontrol agent is an environmental-ly-friendly method to overcome the Fusarium wilt in chilli due to Fusarium oxysporum. This research aimed to isolate actinomycetes antagonist to F. oxysporum FU3 as biocontrol agent from the soil of chilli plantation in Kulon Progo, Yogyakarta. The actinomycetes were isolated using SCA medium and followed by screening for anti-F. oxysporum FU3 on PDA. The filtrate from isolates which showing the highest inhibition zone was produced and tested against strain FU3. The best isolate was then characterized. There are four iso-lates of actinomycetes showing antagonistic activity against F. ox-ysporum (AK4, AK5, AK7, and AK9). The highest antagonistic ac-tivity was shown by AK5 filtrate with inhibitions on PDA and PDB are 41.71 ± 3.5 % and 81.77 ± 5.1 %, respectively. Further obser-vation showed that filtrate of AK5 caused abnormality and lysis on FU3 hyphae. From the results, we concluded that AK5 is a poten-tial biocontrol agent against F. oxysporum. Further studies to opti-mize the production and formulize the bio-fungicide are suggested

Abstract

Oleaginous fungi are one of the microorganisms that can accumulate a high number of biomasses quickly (within 96-130 hours) and are often used to produce lipids. The growth of fungi depends on the chemical composition of the environment in which it grows. The growth media of fungi must contain high carbohydrates as a source of nutrients and high nitrogen content. One of the carbon sources that fungi can use in the growth process is glucose. BR 2.2 isolate is an oleaginous fungus capable of accumulating 28.44% lipids from the total dry biomass with glucose as a carbon source in 50 mL of growth media. Therefore, this study was conducted to determine the effect of variations in the volume of media and incubation time on the production of biomass and lipid isolate BR 2.2. Biomass and lipid production were analyzed at media with additional water volumes of 10, 20, 30, 40, and 50 mL with 48, 96, and 144 hours of incubation times. The results showed that lipid accumulation and biomass production increased with the reduction of water content in the growth media and reached the highest number in the media volume of 20 mL with an incubation time of 144 hours, i.e., 0.87±0.04 g/L and 12.53±0.29 g/L. It can be concluded that biomass and fungal lipid increased along with incubation time and nutrient concentration.

Abstract

The school is an educational institution that runs with tuition fees used by its students. During the Covid 19 pandemic which limits human mobility, this does not affect the obligation to pay tuition fees by students, in a payment information system that is able to overcome this problem, it is urgently needed. An Android-based Payment Information System with a midtrans payment gateway is the solution to this problem. This information system provides payment applications for education without direct meetings between the parents of students and the school. This application has two levels of users, namely admin user and student user. The admin user has the responsibility to manage the application as a whole, including more new students, more billing for each student, payment reports made by students on the system. Users are given access rights to view billing that must be started, view transactions that have been made and make changes to personal data if an error occurs in data input. This application is expected to be a solution for educational institutions in overcoming problems that are now commonly found and able to open people's minds in the process of digitizing life. Abstract

Oleaginous yeasts, such as Lipomyces starkeyi, can convert carbon and nitrogen sources into lipids up to 50% of the cell's dry weight. A high lipid percentage was achieved (53.5%) using Nitrogen Limited Medium (NLM), which indicates that the components of NLM play a role in the production of biomass and lipids. Nevertheless, the statistical analysis of the role of NLM components on lipid production has not yet been conducted. Thus, this research was designed and carried out to determine the role of NLM components in lipid production using Plackett-Burman Design. The results show that the variables had an insignificant impact on lipid production based on the ANOVA test (P >0.05). Additionally, main effect plots were generated to identify the negative and positive effects of the components. The graph indicates that peptone and yeast extract (YE) are essential components in high concentrations to increase lipid production. This result was due to the insufficient concentration used in this experiment (YE= 0.5 g/L; peptone= 0.3 g/L) compared to the optimal conditions (YE= 8 g/L; peptone= 3 g/L). Therefore, further research should be conducted with the addition of external factors (pH, temperature, shaker speed) to acquire more significant results on biomass lipid production.

Abstract

Broiler chickens are the most popular livestock commodity due to their high protein content. Improving chicken rearing quality is needed to produce high quality chickens. Feed is one of the determining factors for broiler growth. This research was conducted to study the potential of adding broiler feed with fermented coconut pulp using Mucor irregularis on morphometric development and lymphoid organs (bursa fabricius and lien), pectoralis major muscle growth, and small intestine length. Total 180 male broiler chickens (Gallus gallus gallus) post-hatch (DOC) were separated into 5 groups with 3 replicates and each replicate amounted to 12 chickens. The control group (K) was given basal feed, treatment 1 (P1) and P2 were given non-fermented coconut pulp (1% and 2%), while P3 and P4 were given fermented coconut pulp (1% and 2%), chickens rearing were done until 16 days old (3 days of acclimation). Body morphometry measurements were taken and 3 chickens from each group were dissected to measure muscle organ index, bursa fabricius and lien, and small intestine length. Data were analyzed using One-way ANOVA with Duncan's test (α = 0.05). The highest increase in chicken body morphometry, organ index of bursa fabricius, lien, pectoralis major muscle, and length of small intestine is owned by P4 group chickens. Based on the results of this study, it can be seen that the addition of fermented coconut pulp in feed has a good impact on the growth of chicken body morphometry, increased weight and index of lymphoid organs (bursa fabricius and lien) and pectoralis major muscle, and lengthening of the small intestine.
