Abstract

Type 2 diabetes mellitus is a global health issue associated with insulin resistance and impaired glucose metabolism. Murraya paniculata L. Jack (kemuning) has shown antidiabetic potential, yet its role as a DPP-4 inhibitor remains underexplored. This study aimed to investigate the potential of kemuning leaf bioactive compounds as natural DPP-4 inhibitors through an in-silico approach. Compound structures were obtained from the PubChem database and analyzed using AutoDock Vina for molecular docking against the DPP-4 protein. Pharmacokinetic properties, toxicity, and drug-likeness were evaluated using pkCSM and SwissADME, while complex stability was assessed through molecular dynamics simulation (RMSF). The results showed that most compounds demonstrated favorable binding affinities (−6.8 to −7.9 kcal/mol), along with acceptable pharmacokinetic and toxicity profiles. Among the tested compounds, 1-aminoindan-1,5-dicarboxylic acid emerged as the most promising candidate for natural DPP-4 inhibition.

Abstract

Sugarcane (Saccharum officinarum L.) leaves contain various bioactive compounds, which exhibit antioxidant and pharmacological potential. However, their application in pharmaceutical preparations faces challenges due to low solubility, stability, and bioavailability. Therefore, the development of an emulsion formulation is needed to improve its stability and absorption. This study aims to determine the phytochemical content of sugarcane leaf extract and to evaluate the physical stability of its emulsion formulation. The extraction was obtained through maceration using 96% ethanol, followed by phytochemical screening. Emulsions were prepared using sugarcane leaf extract as the active ingredient, Virgin Coconut Oil (VCO) as the oil phase, Tween 80 as a surfactant, and PEG 400 as a co-surfactant. The formulations were tested for organoleptic properties, emulsion type, percent transmittance, pH, particle size, polydispersity index, zeta potential, centrifugation, and cycling stability. The results showed that sugarcane leaf extract contained flavonoids, alkaloids, tannins, phenols, triterpenoids, and saponins. Among the four tested formulas, F1 and F2 showed the best stability, with pH 6–7, high percent transmittance, uniform particle size with polydispersity index <0.3, and absence of phase separation. These findings suggest that F1 and F2 represent optimal emulsion formulations for further pharmaceutical development.

Abstract

The white race, especially the adult population, has a prevalence of type 2 diabetes ranging from 3% to 6%. The diabetes frequency in Singapore has greatly escalated in the past ten years. Despite being called a tropical country, Indonesia is the world's second-largest contributor to biodiversity (following Brazil). Approximately 2,500 species of plants are classified as herbs in Indonesia, which has about 30,000 species. An example of these herbs is snake fruit that gives numerous pharmacological effects, such as immunostimulator, antioxidant, antidiabetic, and it improves lipid profile. A lot of types of snake fruit can be found in Indonesia, and their various content are likely to appear due to the different types of snake fruit's physical conditions, as indicated in the previous research. Phenolic compounds and flavonoid groups can inhibit the α-glucosidase enzymes due to being capable of neutralize free radicals, and eventually, pancreatic beta cell apoptosis can be prevented. Methods use Systematic Review by conducting literature searches using PubMed and Ebsco. The literature discusses snake fruit, antidiabetic and cell apoptosis. There are six studies, concluding that snake fruit can be antidiabetic and it prevents cell apoptosis. Snake fruit can be an antidiabetic alternative in the treatment of diabetes, particularly in preventing cell apoptosis.

Abstract

Background: Vascular calcification is a significant contributor to cardiovascular complications in diabetes mellitus (DM), particularly affecting the prognosis of patients.   Objectives: To conduct a comprehensive analysis of the molecular mechanisms underlying vascular calcification in DM, with a focus on insights from human aortic smooth muscle cells (HASMCs). Methods: The search was conducted following the PRISMA guidelines, utilizing databases such as MEDLINE/PubMed, Science Direct, and Google Scholar. The search focused on articles published within the last 5 years that discussed the molecular mechanisms of vascular calcification in DM, specifically in HASMCs. Results: Five selected reviews were found in a total of 637 articles. DM significantly accelerates vascular calcification in HASMCs through the upregulation of osteogenic markers and activation of the Wnt/β-catenin signaling pathway. Other identified mechanisms include inflammation, ferroptosis, and endothelial dysfunction, contributing to the complex interplay of factors that drive vascular calcification in diabetic patients. Conclusion: It is concluded that DM significantly accelerates vascular calcification in enhanced expression of osteogenic markers and activation of the Wnt/β-catenin signaling pathway in human aortic smooth muscle cells. Patients with diabetes are more likely to have cardiovascular issues as a result of this pathological process.
