Abstract

A diet high in advanced glycation end products (AGEs) has been linked to an increased risk of hypertension and other vascular degenerative diseases. Some intervention such as moderate intensity exercise, yogurt probiotic intake and anthocyanin supplementation had been conducted separately, and showed promising effects. The aim of this study is to analyse the effect of combining rose petal yoghurt and moderate intensity exercise in lowering blood pressure and sRAGE level in high AGEs diet induced model. This research was experimental study, using post test-only control group design. The animals that were used were white male rats (Rattus norvegicus) Wistar strain, divided into 5 groups. Group KN was given standar diet, KP was treated with high AGEs diet, KA was treated with high AGEs diet and combination of rose yoghurt and moderate exercise, KB was given high AGEs diet dan rose yoghurt, and KC group was treated with high AGEs diet and moderate exercise. After 12 weeks systolic and diastolic blood pressure was examined using Kent Scientific CODA blood pressure analyzer and sRAGE examined with ELISA technique. The result shows most decline in systolic and diastolic BP was at KC group which got a high AGEs diet and moderate intensity exercise. KC’s systolic BP was measured 120 ± 2.82 mmHg and diastolic at 77.25 ± 9.5 mmHg. The results of ELISA examination show KN has higher sRAGE level at 1865 ± 503.4 ng/L on the contrary KP has lower sRAGE level at 530 ± 78.6 ng/L.

Abstract

Understanding the molecular mechanisms underlying skeletal muscle adaptation to different training regimens is essential for advancing muscle health and performance interventions. The aim of this study was to investigate molecular and genetic adaptations in the resting skeletal muscle of sedentary individuals compared to strength- and endurance-trained athletes using bioinformatics and in vitro validation. Differentially expressed genes (DEG) analysis of the GSE9405 dataset was conducted. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses were performed, followed by protein-protein interaction (PPI) network analysis and receiver operating characteristic (ROC) analysis. To validate the bioinformatics findings, the expression of two identified genes was assessed using real-time polymerase chain reaction (PCR) in professional athletes and age-matched non-athletes. Analysis of RNA expression profiles from the GSE9405 dataset identified 426 DEGs, with 165 upregulated and 261 downregulated in trained individuals. Enrichment analysis highlighted pathways related to metabolic efficiency, mitochondrial function, and muscle remodeling, all crucial for athletic performance. PRKACA and CALM3 were identified as key upregulated genes in trained individuals with central roles in these pathways. The area under the curve (AUC) values for CALM3 and PRKACA were 0.8558 and 0.8846, respectively, for differentiating the two groups. Validation in human samples confirmed that CALM3 expression was significantly higher in athletes (p=0.001), suggesting its critical role in muscle adaptation. However, PRKACA expression differences between the groups were not statistically significant (p=0.321). These findings provide insights into gene-level responses to long-term training, offering a basis for targeted interventions to enhance muscle health and athletic performance.

Abstract

Diabetes mellitus is a chronic metabolic disorder, and maternal hyperglycaemia during pregnancy increases the risk of fetal developmental complications. This study aimed to evaluate the protective role of L-carnitine against glucose-induced embryotoxicity in zebrafish (Danio rerio). Fertilized embryos were exposed to 5% glucose with or without L-carnitine supplementation (50 or 100 uM). Developmental outcomes including survival, hatching, malformations, body length, and heart rate were assessed using microscopy and analyzed by ANOVA and Kaplan–Meier survival curves. High glucose significantly reduced survival (hazard ratio = 6.86; p < 0.0001), delayed hatching (median hatching time 84 vs. 60 hpf; HR = 0.17, 95% CI = 0.08–0.39; p < 0.0001), and induced growth retardation and bradycardia (p < 0.0001). L-carnitine did not rescue survival or hatching but partially ameliorated growth impairment (p < 0.05) and restored heart rate in a dose-dependent manner, with 100 uM supplementation approaching control values (p < 0.001). Morphological abnormalities such as yolk sac edema and spinal curvature remained present but less severe with co-treatment. In conclusion, L-carnitine confers partial protective effects on growth and cardiac function under hyperglycaemic stress, although it does not prevent early lethality.
