Abstract

Application measures of inquiry conducted by teachers who do not meet the basic standards, so that students have trouble doing scientific work skills well. Teachers had not found the right method for learning the material Natural Phenomena. In addition, teachers do not monitor students affective value in a sustainable manner, while the affective value determines the success of student learning. This study aimed to analyze the application of scientific inquiry teaching model can improve student affective value and learning outcomes class VII in SMP N 2 Semarang. This research was classroom action research which consisted of 3 cycles of learning. The sample in this research were class VII D and VII E. Results of the analysis of students affective learning value showed the average scientific interests and attitudes of students  in learning increases in each cycle. Average learning outcomes student of class VII D and VII E on the first cycle of 46,32; second cycle of 49,63; and the third cycle 82,81. Conclusions from this research was the application of scientific inquiry teaching model can increase the value of affective and cognitive learning outcomes matter Natural Phenomena class VII D and E in the Junior High School 2 Semarang Penerapan langkah-langkah inkuiri yang dilakukan oleh guru tidak memenuhi standar yang baku, sehingga siswa kesulitan melakukan ketrampilan bekerja ilmiah dengan baik. Guru belum menemukan metode yang tepat untuk pembelajaran materi Natural Phenomena. Selain itu, guru belum memantau nilai afektif siswa secara berkelanjutan, sedangkan nilai afektif menentukan keberhasilan belajar siswa. Tujuan penelitian ini adalah analisis penerapan model pembelajaran scientific inquiry untuk meningkatkan kemampuan afektif dan hasil belajar siswa kelas VII di SMP N 2 Semarang. Penelitian ini merupakan penelitian tindakan kelas dengan kolaborasi antara guru dengan mahasiswa yang terdiri dari 3 siklus pembelajaran. Sampel dalam penelitian ini yaitu kelas VII D dan VII E. Hasil analisis nilai afektif siswa terhadap pembelajaran menunjukkan rata-rata minat dan sikap ilmiah siswa dalam pembelajaran meningkat di setiap siklusnya. Rata-rata hasil belajar siswa kelas VII D dan VII E pada siklus I sebesar 46,32; siklus II 49,63; dan siklus III 82,81.  Simpulan dari penelitian ini adalah penerapan model pembelajaran scientific inquiry dapat meningkatkan nilai afektif dan hasil belajar kognitif materi Natural Phenomena siswa kelas VII D dan E di SMP Negeri 2 Semarang.

Abstract

Identifikasi mRNA untuk mengukur ekspresi suatu gen dapat dijadikan sebagai alat diagnostik suatu penyakit. mRNA RECK menghasilkan protein RECK yang terbenam dalam membran glikoprotein sel sehingga memperkuat integritas sel. Ekspresi mRNA RECK dikaitkan dengan proses fisiopatologis dalam kanker ovarium.Metode one step qRT –PCR sangat sensitif dan akurat, namun membutuhkan kompleksitas yang tinggi. Salah satu faktor yang dapat mempengaruhi hasil kuantifikasi mRNA adalah suhu annealing primer mRNA RECK.Penelitian ini melakukan optimasi suhu annealing secara in-silico melalui http://tmcalculator.neb.com/ dan http://www.biophp.org/minitools/melting_temperature diperoleh suhu 49°C (57°C primer forward; 54°C primer reverse) dan 51,8°C primer forward; 51,1°C primer reverse. Proses selanjutnya yaitu melakukan running qRT-PCR menggunakan suhu bertingkat. Suhu 55,6°C pada awal optimasi menunjukkan hasil yang baik terlihat dari kurva meltpeak yang spesifik pada suhu 75°C. Namun setelah proses running menggunakan sampel yang berbeda, hasil meltpeak tidak spesifik. Proses optimasi kedua diperoleh suhu 52,5°C dengan meltpeak spesifik suhu 76,5°C. Optimasi annealing dilanjutkan dengan melakukan persamaan konsentrasi RNA template menggunakan pengenceran perbandingan 1:3 (RNA: Nuclease Free Water (NFW)) Kesimpulan dari penelitian ini diperoleh suhu annealing mRNA RECK sebesar 52,5°C dan pengenceran 1:3 RNA dengan NFW, serta penggunaan reagen KAPA one step PCR tanpa penambahan dUTP dan ROX high serta ROX low.

Abstract

Daging kerbau memiliki nilai gizi protein dan susunan asam amino lengkap. Kandungan air dan protein yang tinggi menyebabkan daging mudah busuk, sehingga perlu dilakukan pengawetan penggaraman serta pengasapan. Protein daging kerbau dapat dipengaruhi oleh pengolahan bahan pangan, seperti kadar pemberian garam dan pengasapan. Tujuan penelitian untuk menganalisis profil protein daging kerbau dengan variasi konsentrasi garam  10%b/b, 20% b/b, 30% b/b, 40% b/b penggaraman 3 jam serta pengasapan 2 jam. Profil protein daging kerbau dapat dianalisis menggunakan metode SDS – PAGE 12%. Hasil penelitian dari profil protein daging (kontrol), penggaraman 3 jam konsentrasi garam 10%, 20%, 30% dan 40% berturut – turut 28 sub unit protein, 26 sub unit protein,  25 sub unit protein, 23 sub unit protein, dan 21 sub unit protein, sedangkan daging yang diasapkan tanpa garam, daging penggaraman 3 jam serta pengasapan 2 jam konsentrasi garam 10%, 20%, 30% dan 40% berturut -turut 22 sub unit protein, 24 sub unit protein, 20 sub unit protein, 13 sub unit protein dan 12 sub unit protein. Berdasarkan hasil penelitian ini dapat disimpulkan bahwa semakin tinggi konsentrasi garam maka kandungan protein yang terdapat dalam daging kerbau akan rusak dan semakin sedikit hal ini menyebabkan denaturasi protein yang ditandai dengan berkurangnya sub unit protein. Namun protein daging kerbau lebih banyak rusak apabila diasapkan.

Abstract

Thermal stability and lifetime stability for immobilization lipase on zeolit by absorption technique has been carried out. Immobilization of lipase on zeolit was performed via physical adsorption. This reseacrh aimed to determine the effectiveness of pancreatic lipase immobilization in term of thermal and lifetime stability of the hydrolysis activity. The result of this research proved that pancreatic lipase was able to immobilized to zeolit by using the physical adsorption. Immobilized lipase shows better lifetime stability than the free lipase. Immobilized enzyme catalyzed reaction has a lower decreases free fatty acid than that of the free enzyme. Lower decline of value suggests the enzyme is more stable for repeated use. The immobilized lipase was able to maintain its activity after multiple-uses up to four reaction cycles. Similar trend was also observed for thermal stability study. Immobilized lipase shows a higher thermal stability that that shown by free lipase. Immobilized lipase on zeolit was also able to maintain its activity at 40oC.

Abstract

Caulerpa racemosa is a green algae consumed by people in northern coastal areas. C. racemosa has a habitat attached to the shallow seabed. C. racemosa usualy consumed fresh without any cooking process so that the contamination of microorganisms can be eaten. Molecular identification using 16S rRNA is needed to determine the type of bacterial contaminants in C. racemosa. The isolates of C. racemosa were cultured in HIA, BAP, and BHI media. Bacteria from BHI media were isolated by DNA, PCR for 16S rRNA gene, and sequencing. Bacteria isolate C. racemosa was found to have the α-hemolytic ability in BAP media. The sequencing analysis showed that the three bacterial colonies of C. racemosa isolate had high similarity with V. parahemolyticus, Caldalkalibacillus mannanilyticus, and Exiguobacterium profundum.

Abstract

Kanker ovarium mengalami peningkatan insidensi dan kematian yang disebabkan oleh kesulitan dalam mendeteksi kanker pada stadium awal dan belum ada uji skrining yang efektif untuk kanker ovarium. Dibutuhkan metode deteksi yang memiliki sensitivitas dan spesifisitas yang tinggi berupa biomarker yang bersirkulasi di dalam plasma darah berupa mikroRNA. Deteksi melalui mikroRNA diharapkan dapat meminimalkan rasa sakit atau minimal invasif. Penelitian analisis profil mikroRNA yang berperan dalam kanker ovarium perlu dilakukan untuk mendapatkan biomarker deteksi awal minimal invasif kanker ovarium yang tepat. Analisis profil ekspresi mikroRNA pada kanker ovarium dilakukan dengan isolasi plasma darah, sintesis cDNA, dan qRT-PCR sampel yang disatukan, dan qPCR miR-320c. Profiling mikroRNA pada kanker ovarium dilakukan dengan metode pooling sampel, kemudian dilanjutkan dengan kuantifikasi mikroRNA yang mengalami peningkatan dan penurunan paling signifikan secara individu sampel. Hasil analisis profil ekspresi mikroRNA pada kanker ovarium menunjukkan peningkatan ekspresi miR-320c baik pada serous maupun pada mucinous. Hasil kuantifikasi ekspresi individu menunjukkan penurunan ekspresi miR-320c pada stadium awal dan peningkatan yang signifikan miR-320c pada stadium lanjut kanker ovarium. Penelitian ini menunjukkan bahwa ekspresi mikroRNA dapat berbeda-beda pada masing-masing orang. Kata kunci : kanker ovarium, miR-32Oc, profil ekspresi, minimal invasif

Abstract

HCG hormone is a hormone produced by the placenta in early pregnancy with the aim of maintaining pregnancy. The Elisa method is a method for examining HCG levels which has the principle of specific antigen and antibody reactions. The work stage in the laboratory that has the biggest error is in the pre-analytic stage reaching 60%, one of which is the reagent temperature. The ELISA method recommends the reagent temperature to be stored at 2-8°C and when used at 25°C. Examination reagents Elisa method uses reagents that use enzymes in the examination. Enzymes are very sensitive to temperature. This study aims to determine the ratio of HCG levels at reagent temperatures of 25°C & 8° using the ELISA method. This type of research is analytic supported by experiments and literature studies. Serum samples of 6 pregnant women were taken from the Kedungmundu Public Health Center using a limited population count formula. The examination was carried out by comparing the reagents at a temperature of 25°C and 8°C using 6 serums of pregnant women and in duplicate. Data analysis only used primary data and statistical tests were carried out using normality, homogeneity and T-Test tests. The average result at a temperature of 25°C was 380.851 and the average temperature of 8°C was 321.99 and the T-test statistic test showed p value = 0.469 and 0.470. Based on the results of the study, it can be concluded that there is an insignificant difference in the concentration of HCG by comparing the reagents at 25°C and 8°C.

Abstract

Diabetes mellitus is a big public health problem in Indonesia. The length of time of suffering from DM is associated with a decrease in the function of pancreatic beta cells resulting in insulin resistance, which occurs in type 2 diabetes mellitus. Insulin resistance can be determined by measuring copeptin levels. The aim of this study was to determine differences in levels of human CPP protein (copeptin) in diabetes mellitus patients based on length of suffering. This type of research is observational analytic with cross sectional research design. Samples were taken from diabetes mellitus outpatients at Roemani Muhammadiyah Hospital as many as 20 samples, then the samples were examined for levels of human CPP (copeptin) protein using the ELISA (Enzyme Linked Immunosorbent Assay) method. The results showed that the average copeptin level in patients with diabetes mellitus ≤ 3 years was 1054 pg/mL and the average copeptin level in patients with diabetes mellitus > 3 years was 899 pg/mL. Statistical tests using the Independent Sample T Test obtained a significant sig value of 0.027 with a significance level of 0.05, namely 0.027 3 years, namely 899 pg/mL

Abstract

Suhu serum pada uji ELISA merupakan faktor pra analitik yang dapat mempengaruhi kadar parameter yang diukur. Serum yang tidak diperiksa secara langsung akan disimpan pada suhu 4°C, sedangkan pemeriksaan ELISA menggunakan spesimen serum suhu ruangan (25°C). Masa tunggu suhu serum menjadi 4°C hingga 25°C seringkali tidak tepat sehingga pemeriksaan tetap dilakukan meskipun serum masih berada pada suhu 4°C. Untuk mengetahui perbedaan suhu serum pada suhu 4°C dan 25°C pada uji ELISA maka dilakukan penelitian dengan menggunakan parameter Human Chorionic Gonadrotopin (hCG). hCG adalah salah satu parameter untuk mendiagnosis kehamilan. Tujuan dari penelitian ini adalah untuk mengidentifikasi apakah terdapat perbedaan kadar hCG serum pada suhu 4°C dan 25°C menggunakan metode ELISA. Sampel yang digunakan adalah ibu hamil trimester II sebanyak 10 orang yang akan diperiksa secara duplo. Masing masing 10 spesimen akan diperiksa kadar hCG menggunakan metode ELISA dalam kondisi suhu serum 4°C dan 25°C. Hasil penelitian menunjukkan bahwa rata-rata hCG serum yang diperiksa pada suhu 25°C (385.677 IU/L) lebih tinggi dibandingkan dengan serum pada suhu 4°C (381.912 IU/L), namun hasil statistik menunjukkan tidak terdapat perbedaan yang signifikan antara kadar HCG diperiksa pada suhu 4°C dan 25°C (p >0,05).

Abstract

Atherosclerosis is triggered by cholesterol accumulation in endothelial layers that induces the production of inflammatory cytokines, including TNF-α, IL-6, and IL-10. This condition disrupts microRNA homeostasis like miR-29b-3p, generally maintaining artery health. Steamed tomatoes contain higher antioxidant properties than raw, which might be better against atherosclerosis. However, its influence on inflammation and miR-29b-3p balance remains unclear. This study aims to elucidate the effect of steamed tomatoes on the TNF-α, IL-6, and IL-10 levels and miR-29b-3p expression under atherosclerosis conditions. Sprague Dawley male rats were equally divided into K1 group of healthy rats given a placebo; K2 group was atherosclerosis rats induced with 2 m of cholesterin/ 200 g/KgBW per day, then K3 and K4 groups were atherosclerosis rats supplemented with 20 mg/ 200 g/KgBW per day of atorvastatin and 16 mg/ 200 g/KgBW per day of steamed-tomato extract for 60 days. Steamed tomato decreases cytokine level in the K4 group and significantly differs from all groups (p < 0.050). This study showed that increased proinflammatory cytokine, including TNF-α and IL-6, is hypothetically involved in plaque formation and lamina layer destruction in endothelial. Steamed tomato supplementation also significantly increases miR-29b-3p expression to 0.98 ± 0.33 log10 fold change higher than K2 and K3 (p = 0.000). This study demonstrated the potential of steamed tomatoes to improve dyslipidemia, atherosclerosis-related inflammation, and miRNA homeostasis. This research provides additional knowledge regarding determining the expression of miR-29b-3p, which has the potential to be developed as a diagnostic or therapeutic target.
