Abstract

Brown rice (BrR) is a nutrient-rich staple food and a potential source of γ-oryzanol, which has the potential to promote health effects in hypercholesterolemia. However, the availability and biological mechanism of γ-oryzanol in Indonesian BrR remain unexplored. This study aimed to characterize γ-oryzanol content in three Indonesian BrR varieties, Black Madras, Lawang, and UB BrR, and evaluate its biological functions for hypercholesterolemia. γ-Oryzanol was identified using HPLC, antioxidant activity by the ferric reducing antioxidant power (FRAP) method, and anticholesterol activity by the Lieberman-Burchard method. In silico analysis was performed to assess γ-oryzanol derivatives' interaction with microsomal triglyceride transfer protein (MTP). Black Madras BrR showed the highest γ-oryzanol content (0.3007 ± 0.0011 g). γ-Oryzanol was comprised of four main derivatives: cycloartenyl ferulate, 24-methylenecycloartanyl ferulate, campesteryl ferulate, and β-sitosteryl ferulate. Black Madras BrR exhibited strong antioxidant (IC50 18.89 ± 0.37 μg/mL) and anticholesterol (IC50 14.02 ± 0.23 µg/mL) activities. Docking simulations revealed that γ-oryzanol derivatives interact with MTP lipid-binding residues (Leu643, Ile666, Phe813, Val817) with the same binding energy as lomitapide. Molecular dynamics indicated γ-oryzanol stable interaction, closer to lomitapide. These findings proposed the potential bioactivity of γ-oryzanol from Black Madras BrR as antioxidant, anticholesterol, and a natural MTP inhibitor for hypercholesterolemia treatment.

Abstract

Marga Citrus (jeruk) merupakan salah satu marga yang anggotanya beragam yang terdiri dari 255 jenis dengan 44% diantaranya termasuk kelompok Mandarin (Mandarins group). Hubungan kekerabatan secara fenetik antar jenis di dalam marga Citrus sp. dapat diamati menggunakan karakter morfologi maupun anatomi. Tujuan dari penelitian ini adalah mengetahui hubungan fenetik antara jeruk purut (C. hystrix), jeruk kasturi (C. microcarpa), dan Citrus sp. menggunakan karakter morfologi dan anatomi. Pada penelitian ini digunakan sampel jeruk purut, jeruk kasturi, dan Citrus sp. koleksi Kebun Raya Bogor (KRB). Karakter organ vegetatif (batang dan daun) serta organ generatif (bunga, buah dan biji) dilakukan pada ketiga spesimen. Data yang diperoleh selanjutnya dianalisis secara fenetik menggunakan program komputer PAST v4.03. Hasil penelitian menunjukkan bahwa Citrus  sp. dan C. hystrix memiliki tingkat kemiripan yang lebih tinggi antar keduanya jika dibandingkan dengan C. microcarpa. Beberapa karakter yang menjadi pembeda utama antara C. microcarpa, C. hystrix, dan Citrus sp. koleksi KRB adalah ujung dan pangkal daun, warna kulit buah, ukuran buah, bentuk biji, dan kerapatan stomata.

Abstract

Konyal (Passiflora suberosa L.) is a wild and invasive plant belonging to the Passifloraceae. The existence of P. suberosa as an invasive species can threaten the existence of local plants. As a scientific conservation area, the Kebun Raya Bogor (KRB) is a habitat for Konyal. One effort that can be made to minimize the threat of invasive species is through mapping. The aim of this research was to map the distribution of konyal (P. suberosa) in the scientific conservation area of Kebun Raya Bogor and measure environmental factors in the habitat of konyal (P. suberosa). The research was carried out using an exploratory method throughout the Kebun Raya Bogor area in January 2024. Documentation of komyal (P. suberosa) and measurements of environmental factors in its growing habitat were carried out during exploring activities. In addition, distribution coordinate data was recorded and used for mapping using QGIS v3.36.2 program. The research results showed that the konyal  (P. suberosa) in the KRB was spread over six areas, including II A, II C, II D, II F, III I, and  XIII A. Environmental factors at the location where P. suberosa was found in the Kebur Raya Bogor area, such as air humidity (70 – 77%), temperature (26 – 33 ˚C), and light intensity (2250 – 16200 lux). Distribution of konyal (P. suberosa) in KRB area showed high density in vak II (II A, II C, II D, and II F). The location vak II has vegetation with sparse canopy and low shade, thereby increasing the sunlight intensity compared to other areas.

Abstract

Syzygium zeylanicum (Myrtaceae) is a species that grows in the Indo-Malayan region. Different growing environments for S. zeylanicum can influence morphological characteristics and reflect genetic diversity. The objective of this study was to describe the morphological characteristics of S. zeylanicum and optimize Inter Simple Sequence Repeats (ISSR) primers as an initial step toward studying the genetic diversity of S. zeylanicum. This study was conducted by collecting S. zeylanicum samples from the Bogor Botanical Garden at six locations: V.B.84, V.B.84a, II.Q.87, II.Q.87a, V.C.54a, and V.C.97a. Leaf and branch samples were collected for morphological characterization. DNA isolation was performed using 2% CTAB. Optimization was performed for 12 ISSR primers (ISSR1, ISSR2, ISSR3, ISSR4, ISSR5, ISSR6, ISSR7, ISSR8, ISSR9, ISSR10, ISSR11, and ISSR12) with annealing temperature ranges of 52 – 54 °C, 52.1 – 5 3.1 °C, 55 – 58 °C, and 45 – 50 °C. Characterization showed that S. zeylanicum leaves are simple, lanceolate, caudate tips, aequilateral bases, entire margins, brochidodromous venation, opposite leaf arrangement, petiolate attachment, abaxially glabrous, and adaxially lustrous. The leaf length, leaf width, and branch diameter of S. zeylanicum ranged from 6 – 8.7 cm, 2.3 – 3.03 cm, and 1.45 – 1.96 cm, respectively. In addition, 11 of the 12 ISSR primers successfully obtained optimal annealing temperatures, including ISSR1 (57.7 °C), ISSR2 (52.8 °C), ISSR3 (56.6 °C), ISSR5 (52.7 °C), ISSR6 (52.9 °C), ISSR7 (47.8 °C), ISSR8 (47.8 °C), ISSR9 (52.6 °C), ISSR10 (53.0 °C), ISSR11 (49.7 °C), and ISSR12 (49.0 °C). Only ISSR4 primer did not produce DNA bands after optimization at all temperature ranges used.
