Abstract

Breast cancer shows the proliferation of malignant epithelial cells that limit the ducts and lobes of the breast. If this process is not controlled, it will cause lumps that can then spread to other parts of the body and cause death. High-mobility group box protein 1 (HMGB1) has been reported to play roles in promoting cell survival of breast cancer cells. The inhibition of HMGB1 could be a reasonable target for the treatment of breast cancer. Amaranthus tricolor has been found could reduce the viability of breast cancer cells. In this study, we aim to predict the ability of the active compounds in Amaranthus tricolor leaves to inhibit the HMGB1 through molecular docking study. The molecular docking was conducted by using the PyRx software. This study shows that the four active compounds in Amaranthus tricolor leaves, namely isorhamnetin, routine, myricetin, and quercetin, have the smallest bond energy, indicating that the four compounds are the most stable and have the highest potency as HMGB1 inhibitor.

Abstract

Advanced glycation end products (AGEs) have been thought to be a major cause of diabetic nephropathy (DN). The mechanisms underlying the involvement of AGEs antibody in diabetic nephropathy are not fully understood. The present study was designed to investigate the protective effect of AGEs antibody on AGEs-induced glomerular damage. Isolated glomeruli were pre-incubated either with 10 µg/mL polyclonal anti-AGEs antibody (AGE-pAb) or monoclonal anti-Nɜ -carboxymethyl-lysine antibody (CML-mAb) as a model of AGEs antibody to block interaction of AGEs with receptor for AGEs (RAGE) and incubated afterwards either with 100 µg/mL bovine serum albumin (BSA) or AGE-modified bovine serum albumin (AGE-BSA) for 48 h. Annexin V/nephrin doublestaining was performed to determine apoptosis. Using immunofluorescence, we found that administration of AGE-BSA not only significantly increased glomerular cells apoptosis and nuclear factor kappa B (NF-ĸB) p65 expression, but also reduced expression of nephrin, an important structural and signal molecule of podocytes slit diaphragm. Blocking the interaction of AGE-RAGE with AGEs antibody significantly protected glomerular cells from AGEs-induced apoptosis and NF-ĸB p65 overexpression. We found that AGE-pAb conferred superior protective effect compared with CmL-mAb for the same reduction in apoptosis and NF-ĸB p65 expression. In sharp contrast, CmL-mAb led to preserve expression of podocytes nephrin better than AGE-pAb. These results demonstrate that the antibody against AGEs may be beneficial for preventing the glomerular damage in DN.

Abstract

Head trauma or traumatic brain injury (TBI) gives most serious impact on the central nervous system. Several experimental models have been established to mimic different pathogenesis characteristics of TBI. The purpose of this study was to determine whether there is evidence of hystopathological lesions in the brain tissue after Marmorou TBI models. This study uses Rattus norvegicus Sprague Dawley strain. Macroscopic and microscopic observations on the brain tissue were done. Macroscopic lesions were observed in the brain. Microscopic observation was performed with Haematoxylin-Eosin (HE) staining and immunohistochemistry on the distribution of microglia cells and pyramidal cells in the cortex. Meanwhile, the distribution of NRG-1/ErbB, proliferation, and apoptosis were observed in the hippocampus. The results of macroscopic observation showed that there were wounds caused by falling loads and vasodilatation. On microscopic observation, the TBI group showed an increase in neutrophils distribution and distribution of activated microglia to produce TNF-α, and decrease in the number of cortical pyramidal cells significantly. The distribution of NRG-1 tended to decrease after exposure of TBI and had no effect on its receptor, erbB. Exposure of TBI appears to lower the activity of neuronal cells proliferation in dentate gyrus (DG) area and significantly increase the number of apoptotic cells. Marmarou model is a physiological model of TBI that spontaneously occurs following a trauma to the head, for example trauma due to an accident. This data can be used as a preliminary data of inflammation and tissue regeneration of disrupted adult brain. Therefore, this research could be used as the basis in the studies of therapeutic agents in the process of neurogenesis of brain cells.Keywords: traumatic brain injury, ERG-1/ErbB, dentate gyrus, Ki67, TNF-a, microglia

Abstract

Kegiatan mencuci tangan dengan air mengalir dan sabun atau menggunakan hand sanitizer berbasis alkohol merupakan bagian dari protokol kesehatan sebagai upaya yang dilakukan untuk mencegah penyebaran virus penyebab COVID-19. Untuk menanggapi kebutuhan masyarakat mengenai ketersediaan sabun dan hand sanitizer, maka dilakukan kegiatan pengabdian kepada masyarakat yang bertujuan untuk meningkatkan kemampuan masyarakat dalam memproduksi sabun dan hand sanitizer secara mandiri. Kegiatan ini dilakukan selama bulan Juni hingga Oktober 2021 dengan target sasaran yaitu kelompok Karang Taruna Ketup4t dan PKK di Dusun Gondang, Desa Randuagung, Kecamatan Singosari, Kabupaten Malang, Jawa Timur. Metode pelaksanaan kegiatan adalah participation action research dengan cara pemberian edukasi pembuatan produk dan pendampingan anggota kelompok kader dalam proses pembuatan produk. Hasil utama kegiatan ini adalah kader mampu memproduksi sabun dan hand sanitizer homemade. Kesimpulan dari kegiatan ini adalah Kelompok Kader di Dusun Gondang mampu memproduksi sabun dan hand sanitizer homemade.

Abstract

A population growth rate of 1.22% has become a national problem and makes Indonesia one of the most densely populated countries in the world. Meanwhile, the population growth rate in Malang city, which has been successfully decreasing, needs to be sustained. This community service aimed to help maintain this achievement by providing training on contraception in the pharmaceutical care aspect, especially in clinical pharmacy. It is because if the contraceptive preparation chosen is birth control pills, the choice of hormonal contraception needs to be adjusted to the family planning acceptor’s clinical condition and emphasizing the importance of medication adherence is required. The problem-solving method used in this community service activity was Participatory Action Research (PAR). Techniques for delivering material were done through presentation, training, and practices using visual aids. The activity was located at the meeting hall of RW 08, Bunulrejo Village, Blimbing District, Malang City and was carried out for two months with the highlight of the event in March 2022. Three types of questionnaire instruments used were personal data and data related to the history of contraceptive methods used, knowledge instrument, and activity evaluation instrument. The knowledge instrument has been declared valid and reliable through the results of the Pearson and Cronbach’s Alpha correlation tests. The results of this community service activity showed that 43.48% of the participants experienced class 1 obesity, and 9% suffered from comorbid hypertension. The pre-test and post-test results related to knowledge showed an increase of 32.46 points. Besides, the evaluation questionnaire of the three aspects of the assessment, including process, benefits, and training presenters, showed an average score of ≥ 3.50. In short, class 1 obesity and hypertension comorbidity are the conditions to consider when choosing hormonal contraceptive preparations for the community, although the prevalence was relatively small. Participant knowledge related to contraception increased after the training. From the evaluation questionnaire results, the participants considered that the series of training activities went very well.

Abstract

Abstrak: Suplemen kesehatan adalah produk yang dapat melengkapi, meningkatkan dan memelihara kebutuhan zat gizi, serta dapat memperbaiki fungsi kesehatan karena memiliki nilai gizi dan atau efek fisiologis. Banyaknya beredar suplemen yang diklaim mampu mencegah dan mengobati penyakit di kalangan masyaratakat membuat penggunaan suplemen yang tidak rasional dan memiliki potensi resiko yang tinggi. Sasaran kegiatan ini adalah tim penggerak PKK kelurahan di Kota Malang. Pada kegiatan ini setiap kelurahan akan diundang 3 (tiga) orang perwakilan TP PKK dari 5 kecamatan. Tujuan kegiatan pengabdian masyarakat berupa edukasi penggunaan suplemen kesehatan pada tim penggerak PKK Kelurahan Kota Malang yaitu untuk meningkatkan pengetahuan masyarakat tentang penggunaan suplemen kesehatan yang rasional. Rangkaian kegiatan diadakan pada bulan Agustus - September 2022. Kegiatan meliputi : Penyuluhan mengenai imunitas dan sistem daya tahan tubuh; penyuluhan mengenai penggunaan suplemen kesehatan yang Rasional; penyuluhan mengenai bentuk sediaan khusus dari suplemen kesehatan yang beredar di masyarakat; permainan bagi peserta, yaitu Hoax Buster Games; Pretest dan posttest, kegiatan Konsultasi setelah edukasi serta evaluasi kepuasan peserta. Hasil kegiatan adalah : dari 180 peserta yang ditargetkan, 146 peserta atau 81,11% peserta hadir di acara penyuluhan; Rata-rata nilai pretest adalah 60 dan nilai rata-rata posttest sebesar 75, dapat terlihat adanya peningkatan nilai rata-rata peserta berturut-turut sebesar 25%. Peserta dengan tingkat pengetahuan “Baik” naik secara signifikan dari 10,95% menjadi 50%. Selain itu, peserta dengan tingkat pengetahuan “Cukup” juga turun secara signifikan dari 89,04% menjadi 50%. Hasil rata-rata kepuasan masyarakat adalah 4,68 atau 93,52 atau dapat disimpulkan peserta merasa keseluruhan kegiatan pengabdian masyarakat telah berjalan dengan sangat baik. Dari kegiatan ini diketahui telah terjadi peningkatan pengetahuan tim penggerak PKK Kelurahan Kota Malang mengenai penggunaan suplemen kesehatan. Diharapkan pada kader akan dapat mengedukasi masyarakat.Abstract: Health supplement is a product to supplement nutritional needs, increase, maintain, have nutritional value and/or physiological effects to improve health function. The large number of supplements circulating which are claimed to be able to prevent and treat disease among the public makes the use of supplements irrational, which had high potential risk. The target of this activity is Team for Empowerment and Family Welfare (TP-PKK) in Malang City. In this activity, 3 (three) TP PKK representatives from 5 sub-districts will be invited to each sub-district. The purpose of community service activities in the form of education on the use of health supplements in the PKK Team of Malang City Village is to increase public knowledge about the rational use of health supplements. The series of activities will be held in August - September 2022. Activities include: Counseling on immunity and the immune system; counseling regarding the rational use of health supplements; counseling regarding special dosage forms of health supplements circulating in the community; games for participants, namely Hoax Buster Games; Pretest and posttest, Consultation activities after education and evaluation of participant satisfaction. The results of the activity were: of the 180 targeted participants, 146 participants or 81.11% of the participants attended the counseling event; The average pretest score was 60 and the posttest average score was 75. It can be seen that there was an increase in the average participant score by 25% respectively. Participants with a "Good" level of knowledge rose significantly from 10.95% to 50%. In addition, participants with an "Enough" level of knowledge also fell significantly from 89.04% to 50%. The average result of community satisfaction was 4.68 or 93.52 or it could be concluded that the participants felt that the overall community service activities had gone very well. From this activity it is known that there has been an increase in the knowledge of the Malang City Urban Village PKK driving team regarding the use of health supplements. It is hoped that the cadres will educate the community.

Abstract

Vaccination is an effective method to suppress COVID-19 transmission, but injection-based vaccination is less effective due to its inability to induce mucosal immunity. This study aimed to determine the effects of vaccine formulations on bacteria viability and antigen expression to find the optimal formulation. Three intranasal preparation formulations (F1, F2, and F3) were created with different ingredient compositions, along with a control. Physicochemical tests were conducted on day 0 and day 14 to assess bacterial viability, and antigen expression was evaluated using the western blot method. Formula 2, containing sodium alginate (0.615%), trehalose (4.125%), polyvinyl alcohol (0.1%), and calcium chloride (5%), exhibited the best viability test results, although no significant differences were observed among the groups. The study concluded that variations in composition concentrations could affect bacterial stability, with Formula 2 showing the best results in terms of bacteria viability and antigen expression up to 14 days after formulation.

Abstract

Excessive exposure to UV radiation results in skin photoaging, which may be prevented or treated using natural plant compounds. Herbal cosmetics and medicines have grown in popularity due to the abundance of relatively safe compounds. This research aims to explore the network pharmacology of Glycyrrhiza glabra (GG) and Curcuma domestica (CD) against skin photoaging. Active compounds from GG‐CD were sourced from databases including TCSMP, KnapSack, TCMID, and published literature, while disease targets were collected from GeneCards and OMIM databases. The STRING database was utilized to construct the protein‐protein interaction (PPI) network. Enrichment analyses for Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway were performed using Metascape. The herb‐compounds‐target‐pathway‐disease (H‐C‐T‐P‐D) network was visualized using Cytoscape software. A total of 529 compounds, 2,335 active compound targets, and 120 skin aging targets were obtained. GO enrichment revealed 1,635 biological processes, 67 cellular components, and 121 molecular functions. The study suggests that GG and CD have the potential to treat skin photoaging by targeting multiple targets, such as TP53, TNF, AKT1, IL6, and IL‐1B, as well as multiple pathways, such as those in cancer, apoptosis, TNF, IL‐17, and the AGE‐RAGE signaling pathway. Experiment validation is necessary to confirm the preliminary network pharmacology results.

Abstract

Kanker payudara adalah kanker dengan angka kejadian tertinggi pada wanita. Protein yang terlibat dalam proses metastasis pada kanker adalah high mobility group box 1 (HMGB1). Licorice (Glycyrrhiza glabra) dilaporkan memiliki efek farmakologi antikanker dan ekstrak etanol roasted licorice dapat mengurangi viabilitas metastasis sel kanker. Sistem penghantaran mikrosfer merupakan serbuk padat terdiri dari polimer, crosslinked, dan bahan aktif yang dapat memberikan efek terapeutik diperpanjang. Penelitian ini bertujuan untuk memprediksi kemampuan licorice menghambat protein HMGB1 melalui molecular docking study dan mengetahui konsentrasi crosslinked natrium tripolifosfat yang paling optimal dalam formula kapsul mikrosfer ekstrak licorice. Metode penelitian dilakukan dengan uji in silico  berupa molecular docking dan uji ADMET, formulasi kapsul mikrosfer ekstrak licorice dengan variasi konsentrasi natrium tripolifosfat 3%, 6%, dan 9%, dan uji evaluasi mutu sediaan meliputi morfologi dan ukuran, sifat alir, penetapan kandungan, organoleptik, kadar air, waktu hancur, dan keseragaman bobot. Data hasil uji evaluasi dianalisis secara statistik menggunakan uji Kruskal Wallis melalui IBM SPSS 28.0. Hasil yang diperoleh yakni senyawa glycyrrhetic acid, liquiritin apioside, dan liquiritin berpotensi menghambat protein HMGB1, glycyrrhetic acid dan licochalcone A merupakan substrat CYP450 3A4, dan delapan senyawa merupakan substrat P-gp dengan kelas toksisitas rendah. Kemudian tidak ada perbedaan yang signifikan pada hasil uji ketiga formula namun konsentrasi crosslinked natrium tripolifosfat yang paling optimum dapat terlihat melalui uji evaluasi mutu sediaan. Kesimpulan penelitian ini adalah senyawa glycyrrhetic acid, liquiritin apioside, dan liquiritin dalam licorice berpotensi menghambat HMGB1 dan formula mikrosfer yang memenuhi uji evaluasi paling baik adalah formula dengan konsentrasi natrium tripolifosfat 3%.
