Abstract

Sargassum is a macroalgae in tropical and subtropical waters. Sargassum sp. contains Mg, Na, Fe, tannin, iodine, and phenol. This study aims to determine the antibacterial potential of Sargassum sp. extract in inhibiting the growth of Staphylococcus aureus. Samples were dried in the sun and then pureed using a blender and extracted by maceration method, for 7 days with methanol solvent at a ratio of 1: 3, evaporated using a rotary vacuum evaporator and water bath to get extract results. Antibacterial activity was tested using the disc diffusion method against various concentrations of extracts, namely 10,000, 5000, 1000, 100, 10, and the control. The results showed that this study implies that Sargassum sp. extract has potential as a natural antibacterial against Staphylococcus aureus. Still, its effectiveness is determined by the extraction method and concentration used.

Abstract

Pesawaran merupakan salah satu kabupaten dengan sebaran potensi objek wisata yang beragam di sejumlah daerah, salah satunya yaitu Pulau Pahawang. Penurunan luasan padang lamun yang terjadi setiap tahunnya disebabkan oleh berbagai aktivitas manusia. Aktivitas tersebut diantaranya yaitu pembangunan konstruksi pesisir, pembuangan limbah domsetik, alur perjalanan kapal, dan juga penangkapan ikan. Pelaksanaan kegiatan pengabdian kepada masyarakat dilaksanakan di Desa Wisata Pahawang, Kabupaten Pesawaran. Kegiatan ini melibatkan masyarakat sekitar Desa Wisata Pahawang dan Himpunan Mahasiswa Perikanan dan Kelautan Fakultas Pertanian Universitas Lampung. Kontribusi masyarakat sebagai peserta dalam kegiatan transplantasi lamun serta melakukan kegiatan monitoring persentase pertumbuhan lamun. Metode transplantasi yang dapat dilakukan yaitu metode terf dan sprig anchor. Jumlah bibit yang ditanam menggunakan metode terf sebanyak 350 bibit dengan 14 plot frame dimana masing-masing plot berisi 25 bibit. Pada metode sprig anchor, jumlah bibit yang ditanam sebanyak 475 bibit dengan 19 plot dimana masing-masing plot berisi 25 bibit. Jenis lamun yang ditanam yaitu Enhalus acoroides dengan ciri morfologi tumbuhan yaitu daun panjang menyerupai pita dengan ujung daun membulat. Pemulihan kondisi ekosisitem lamun dilakukan dengan kegiatan transplantasi lamun menggunakan dua metode yaitu terf dan sprig anchor. Kegiatan pengabdian masyarakat pelestarian ekosistem lamun diterima baik oleh masyarakat Desa pahawang dan dapat dilanjutkan oleh masyarakat untuk menjaga kelestarian lingkungan serta keberlangsungan makhluk hidup yang tinggal di wilayah pesisir.

Abstract

Caulerpa sp. (anggur laut) adalah jenis rumput laut hijau yang tumbuh di perairan tropis dan subtropis. Pengobatan jerawat umumnya menggunakan antibiotik, tetapi dapat menyebabkan efek samping dan resistensi bakteri. Penelitian ini bertujuan untuk mengeksplorasi potensi Caulerpa sp. sebagai alternatif pengobatan jerawat alami. Dilaksanakan di Laboratorium Oseanografi Universitas Lampung dari Oktober hingga November 2024, penelitian ini menggunakan metode maserasi dengan metanol 95% untuk ekstraksi. Aktivitas antibakteri diuji terhadap Propionibacterium acnes dengan metode difusi cakram. Hasil menunjukkan bahwa ekstrak metanol Caulerpa sp. memiliki aktivitas antibakteri yang lemah, dengan zona hambat tertinggi 4,03 mm pada konsentrasi 10.000 ppm. Beberapa faktor yang mempengaruhi aktivitas ini termasuk konsentrasi senyawa bioaktif dan karakteristik resistensi bakteri. Penelitian lebih lanjut diperlukan untuk meningkatkan efektivitas ekstrak ini sebagai agen antibakteri alami.

Optimum temperature, pH, and salinity can suppress the activity of the fucoidanase enzyme through the inhibition test. 4. Cytobacillus kochii GSD can produce fucoidanase enzyme with low molecular weight.     Abstract Extensive research has shown that low molecular weight fucoidan exhibits significantly greater biological activity than its high molecular weight. C. kochi GSD, a Sargassum symbiont bacterium, is proven to have the activity of hydrolyze fucoidan. This study proposes the growth optimization and fucoidanase enzymatic activity of C. kochii GSD bacteria under varying environmental conditions (temperature, pH, and salinity) cultured in basic liquid medium (BLM) for 48 hours. Based on Response Surface Methodology (RSM), the range of temperature, pH, and salinity for the growth optimization test of C. kochi GSD bacteria were 26.591, 30, 35, 40, and 43.49oC, the pH used starts from 3.636, 5, 7, 9, and 10.363, while the salinity to be used starts from 3.522, 5, 17.5, 30, and 38.522 ppt, respectively. The best conditions for growth of each environment were then continued with the test of fucoidanase enzyme activity in vitro. The results showed that C. kochii GSD bacteria grew optimally at temperature, pH, and salinity of 35oC, 7, and 30 ppt, respectively. The optimum enzyme activity of C. kochii GSD is at 72 hours with the forming of clear zones on media containing fucoidan and given Cetylpyridinium chloride (CPC) solution with clear zone diameters of 31.17 mm (temperature), 25.9 mm (pH), and 20.97 mm (salinity), respectively. The conclusion of this study is a high novelty finding to produce low molecular weight fucoidan enzymatically with C. kochii GSD bacteria to be used as an immunostimulant.

Abstract

This community service promotes a sustainable and environmentally friendly approach to modern fisheries, benefiting the surrounding community's economy. Probiotic technology applied for cultivating duck groupers in floating net cages at Sari Ringgung Beach, Hanura Village, Teluk Pandan District, Pesawaran. Utilizing probiotics in floating net cages enables fish growth to 3x3x3 m in size, with each cage holding a maximum of 300 fish. The average weight of the harvested fish is 400-500 grams, resulting in a total production of 150 kilograms of fish. The activity collaborated with lecturers from the Program Study of Marine Science, Aquaculture, and Agricultural Product Technology, involving students of the Program Study of Marine Science, and partners of the Bina Usaha II fish farmer group in Ringgung Village.  The partners' contribution was to prepare a place for training and manufacturing practices. Activities that the team has carried out include giving pretest and posttest questionnaires, coordinating with partners for tools and materials, and applying probiotics to fish. Thus, duck grouper farmers who use probiotics in floating net cages can minimize the operational costs of cage maintenance and increase survival rates. The application of the technology is supported by the optimization of the aquaculture process so that the production and marketing activities of the products can be increased.
