Abstract

Penelitian bertujuan untuk menguji aktivitas antibakteri ekstrak Eucheuma cottonii terhadap Streptococcus mutans menggunakan metode difusi cakram. Streptococcus mutans merupakan penyebab utama karies gigi, dan pemanfaatan bahan alam seperti alga merah dapat menjadi alternatif pengganti antibiotik sintetis. Sampel Eucheuma cottonii yang diperoleh dari Pesisir Desa Ruguk, Lampung Selatan, dimaserasi dengan pelarut metanol, kemudian diekstraksi menggunakan metode rotary evaporator dan water bath. Ekstrak diuji pada berbagai konsentrasi (10.000, 5.000, 1.000, 100, 10 ppm) untuk mengukur zona hambat terhadap bakteri uji. Hasil menunjukkan bahwa metode water bath menghasilkan zona hambat maksimal sebesar 2 mm pada konsentrasi 10.000 ppm dalam 24 jam pertama, tetapi efek antibakteri melemah seiring waktu. Metode rotary evaporator menunjukkan hasil yang berfluktuasi, dengan zona hambat terbesar pada konsentrasi 5.000 ppm sebesar 0,90 mm dan anomali pada konsentrasi rendah setelah inkubasi lebih lama. Secara keseluruhan, aktivitas antibakteri tergolong lemah (<5 mm), menunjukkan sifat bakteriostatik, yakni menghambat pertumbuhan tanpa membunuh bakteri secara permanen. Metode water bath lebih unggul dalam menghasilkan ekstrak yang stabil dengan aktivitas antibakteri konsisten dibandingkan rotary evaporator. Penelitian ini menyimpulkan bahwa ekstrak Eucheuma cottonii memiliki potensi antibakteri, tetapi optimasi lebih lanjut diperlukan untuk meningkatkan efektivitasnya terhadap Streptococcus mutans.

Abstract

Sea grape (Caulerpa sp.) is a green seaweed species with bioactive content consisting of flavonoids, terpenoids, alkaloids, and phenols. This study evaluates the antimicrobial potential of Sea*Grape macroalgae extract (Caulerpa sp.) obtained from Pasaran Island, Lampung. The diffusion assay method assessed the antimicrobial activity of Caulerpa sp. extract against Staphylococcus aureus. Caulerpa sp. samples were extracted using the maceration extraction method and evaporated using two methods: water bath and rotary evaporator. The results showed that Caulerpa sp. extract had antimicrobial activity at a concentration of 10,000 ppm, which was resistant to Staphylococcus aureus. These findings suggest that Caulerpa sp. from the waters of Pasaran Island has the potential to be a source of natural antimicrobial and anti-inflammatory components if the concentration of the extract is more significant, which can be further developed for applications in the health and pharmaceutical fields.

Abstract

Eucheuma cottonii is one type of macroalgae that has been widely cultivated. Potential resources that can be developed is the use of macroalgae as antibacterial. Bioactive compounds in Eucheuma cottonii namely alkaloids, flavonoids, terpenoids, tannins, and saponins can be utilized as antibacterials. This study aims to see the antibacterial activity of Eucheuma cottoni seaweed extract against Pseudomonas aeruginosa bacteria using the disc diffusion method. The test was conducted from October 7 to December 14, 2024, at the Aquatic Productivity Laboratory, and Oceanography Laboratory, Marine Science study program, Department of Fisheries and Marine, Faculty of Agriculture, University of Lampung. Samples were taken from Lontar Village, Tirtayasa District, Serang Regency, Banten Province. Dilution of Eucheuma cottonii extract with a concentration of 10,000 ppm 5,000 ppm, 1000 ppm, 100 ppm and 10 ppm. The variable observed was the magnitude of the inhibition zone of Eucheuma cottoni against Pseudomonas aeruginosa bacteria. The results showed that Eucheuma cottonii extract had weak antibacterial activity, with the highest inhibition zone of 13.93 mm at a concentration of 10,000 ppm. This study shows that Eucheuma cottonii extract is only effective in inhibiting the growth of Pseudomonas aeruginosa bacteria in the first 48 hours. Further research is needed to improve the effectiveness of this extract as a natural antibacterial agent.

Cytobacillus kochii GSD can produce fucoidanase enzyme with low molecular weight.     Abstract Extensive research has shown that low molecular weight fucoidan exhibits significantly greater biological activity than its high molecular weight. C. kochi GSD, a Sargassum symbiont bacterium, is proven to have the activity of hydrolyze fucoidan. This study proposes the growth optimization and fucoidanase enzymatic activity of C. kochii GSD bacteria under varying environmental conditions (temperature, pH, and salinity) cultured in basic liquid medium (BLM) for 48 hours. Based on Response Surface Methodology (RSM), the range of temperature, pH, and salinity for the growth optimization test of C. kochi GSD bacteria were 26.591, 30, 35, 40, and 43.49oC, the pH used starts from 3.636, 5, 7, 9, and 10.363, while the salinity to be used starts from 3.522, 5, 17.5, 30, and 38.522 ppt, respectively. The best conditions for growth of each environment were then continued with the test of fucoidanase enzyme activity in vitro. The results showed that C. kochii GSD bacteria grew optimally at temperature, pH, and salinity of 35oC, 7, and 30 ppt, respectively. The optimum enzyme activity of C. kochii GSD is at 72 hours with the forming of clear zones on media containing fucoidan and given Cetylpyridinium chloride (CPC) solution with clear zone diameters of 31.17 mm (temperature), 25.9 mm (pH), and 20.97 mm (salinity), respectively. The conclusion of this study is a high novelty finding to produce low molecular weight fucoidan enzymatically with C. kochii GSD bacteria to be used as an immunostimulant.
