Abstract

Carrots (Daucus carota L.) are known to contain bioactive compounds, particularly phenolic compounds, which act as antioxidants. Objective: This study aimed to determine the total polyphenol content in carrot ethanol extract and evaluate its potential as a natural antioxidant source. This study was a quantitative experimental laboratory study. Carrot simplicia was obtained through a process of sorting, washing, chopping, drying, and grinding, then extracted using 70% ethanol solvent. The total polyphenol content was determined using UV-Vis spectrophotometry with Folin-Ciocalteu reagent and gallic acid as a standard, and the results were expressed in mg Gallic acid equivalent per gram of extract (mg GAE/g). Results: Gallic acid solution was used to obtain a standard curve. The analysis results showed a linear regression equation of y = 0.0161x + 0.042, R2 = 0.9947, with a correlation coefficient (r) value of 0.997. The total phenol content of each sample was determined with three replications. The results showed that carrot ethanol extract had a total polyphenol content of 42,205 mg GAE/g extract. This value indicates that carrot extract contains polyphenol compounds that have the potential to be a source of natural antioxidants.

Abstract

Staphylococcus epidermidis is a bacteria that causes mild skin infections accompanied by abscesses, with a high  prevalence in Indonesia, requiring natural alternatives due to antibiotic resistance. This study aimed to evaluated antibacterial activity of 96% ethanol extract and n-hexane, ethyl acetate, water fractions of pineapple peel (Ananas comosus L. Merr.) against S. epidermidis ATCC 12228 and to determined MIC/MBC of the most active fraction. This quantitative and experimental study used the disk diffusion and liquid/solid dilution. methods. Population: pineapple peels from Blitar, East Java; sample: 700 g Simplicia powder. The nstruments included rotary evaporator, incubator, autoklaf, laminar air flow; data were analyzed using the Kruskal-Wallis (SPSS 25). Results showed that extract produced inhibition zones ranging from 5.63–11.43 mm, ethyl acetate fraction 3.80–7.55 mm, water 2.35–6.88 mm, n-hexane was inactive. The MIC and MBC values of the extract were 25%; ethyl acetate fraction MIC and MBC were 50%. Extract, ethyl acetate, and water fractions exhibited antibacterial activity aginst S. epidermidis, with extract being the most effective.

Abstract

Staphylococcus aureus merupakan bakteri patogen gram positif yang seringmenyebabkan berbagai infeksi pada manusia dan menunjukkan peningkatan resistensi terhadapantibiotik, sehingga diperlukan alternatif antibakteri yang lebih aman dan berbasis bahan alam.Salah satu tanaman yang berpotensi sebagai antibakteri alami adalah bunga kamboja merah(Plumeria rubra L.) yang diketahui mengandung metabolit sekunder seperti flavonoid, alkaloid,saponin, tanin, steroid, dan terpenoid yang secara teoritis memiliki mekanisme kerja antibakterimelalui kerusakan dinding sel, gangguan permeabilitas membran, serta penghambatan sintesisasam nukleat bakteri. Tujuan: Penelitian ini bertujuan untuk mengetahui kandungan metabolitsekunder dari ekstrak bunga kamboja merah (Pulmeria rubral L.) terhadap bakteriStaphylococcus aureus, dan untuk mengetahui aktivitas antibakteri ekstrak bunga kambojamerah (Pulmeria rubral L.) terhadap bakteri Staphylococcus aureus, serta manakah konsentrasiekstrak bunga kamboja merah (Pulmeria rubral L.) yang mampu menghambat bakteriStaphylococcus aureus. Metode: Penelitian menggunakan metode eksperimental laboratoriumdengan desain in vitro. Sampel berupa bunga kamboja merah dideterminasi, dikeringkan, dibuatsimplisia, distandarisasi, kemudian diekstraksi menggunakan metode maserasi dengan pelarutetanol 96%. Kesimpulan: Ekstrak yang diperoleh dilakukan skrining fitokimia dan diujiaktivitas antibakterinya menggunakan metode difusi cakram pada media Mueller Hinton Agardengan variasi konsentrasi 30%, 40%, dan 50%. Parameter yang diamati adalah diameter zonahambat yang terbentuk. Hasil: Data dianalisis secara statistik menggunakan SPSS meliputianalisis deskriptif, uji normalitas, uji homogenitas, dan uji ANOVA.

Abstract

The use of synthetic colorats in cosmetics, particularly eyeshadow, may pose risks of irritation and allergic reactions when used excessively. Therefore, safer alternatives are needed, including natural colorants such as anthocyanins from hibiscus flowers (Hibiscus rosa sinensis L.), which provide natural color and antioxidant activity. This study aimed to formulate hibiscus flower extract as a natural colorant in an eyeshadow cream preparation, determine ist anthocyanin content, and evaluate the physical quality and stability of the preparation at various extract concentrations. The study employed a laboratory experimental method involving the preparation of crude drugs, extraction using 70% ethanol by maceration, standardization of the crude drug and extract, phytochemical screening, formulation of eyeshadow cream with F0 (0%), F1 (10%), F2 (15%), and F3 (20%) extract concentrations, physical quality and stability testing, and determination of anthocyanin content using UV-Vis spectrophotometry. The result showed an extract yield of 30% and a moisture content of 1,24%. The extract met the requirements for ethanol and heavy metal-free testing and contained alkaloids, flavonoids, saponin, triterpenoid, tannins, and anthocyanins. All formulations met the physical quality and stability requirements. Anthocyanin contents were 16,53, 17,19, 23,21, and 25,88 mg/L for F0,F1,F2, and F3, respectively. Increasing the extract concentration resulted in a more intense eyeshadow cream color. These findings indicate that hibiscus flower extract has potential as a natural colorant in eyeshadow cream preparations.
